1.Preparation and characterization of heparin-modified gold nanoparticles

BACKGROUND: Macromolecules modified gold nanoparticles can be used in therapeutics, diagnostics, and other aspects, but most reports mainly focuses on the preparation of protein, DNA and small molecules of sugar modified gold nanoparticles, and the research on polysaccharide modified gold nanoparticles is superficial. OBJECTIVE: To prepare heparin-modified gold nanoparticles, studying their particle size and ultraviolet absorption spectra, and to determine the optimum reaction condition. DESIGN, TIME AND SETTING: The comparative observation of the experiment was performed at the Biotechnology Pharmaceutical Laboratory of School of Chemical Engineering in Wuhan University of Technology from April 2007 to September 2008. MATERIALS: Gold nanoparticles were prepared and preserved at 4 ℃. METHODS: Heparin with an aldehyde group (HEP-CHO) as reducing end was prepared by nitrous acid degradation method. HEP-CHO was further reacted with dimethyl sulfoxide solution containing 0.05 mL and 0.5 mL glacial acetic acid to synthesize heparin with thiol-group (HEP-SH) through reductive amination reaction. Heparin was then orientated immobilized in four bottles of dimethyl sulfoxide solution containing 0.5 mL glacial acetic acid by adding the thiol-group of HEP-SH and sodium cyanoborohydride for 2, 4, 6, and 24 hours. The obtained products were added in gold nanoparticle solution, and the heparin was fixed on the surface of gold nanoparticles by Au-S bonds. MAIN OUTCOME MEASURES: ① Maximum ultraviolet absorbance of gold nanoparticles before and after heparin modified; ② Average grain size of gold nanoparticles and heparin-modified gold nanoparticles. RESULTS: Gold nanoparticles with good particle size were prepared by using a rate of sodium citrate to chloroauric acid was 6:1 (mol/mol). The reductive amination reaction yield was higher when the more acetic acid was added to the solvent. The products were increased within 6 hours and 24 hours, but the difference was not significant. The average grain size and maximum ultraviolet absorbance of gold nanoparticles were 10 nm and 522 nm, and those of heparin-modified gold nanoparticles were 20 nm and 529 nm. CONCLUSION: Heparin-modified gold nanoparticles with good particle size are prepared, and the size can effect on the maximum ultraviolet absorbance. The optimum reaction condition for heparin immobilization is using a rate of dimethyl sulfoxide to acetic acid is 8:1 (mol/mol) as solvent of heparin, and the reaction time is 6 hours.
2.Construction of mIL-12 recombinant retrovirus vector and establishment of viral package cell line

Objective To construct mIL-12 recombinant retrovirus vector and evaluate the effect of mIL-12 expression on glioma in rats.Methods mIL-12 DNA was cloned into vector pLEGFP using standard procedures to develop recombinant plasmid pLEGFP-mIL 12,then it was transferred into PA317 cells.Results Products of enzyme digestion and PCR of recombinant plasmid pLEGFP-mIL12 were analysed using electrophoresis and a(2 290 bp) fragment of DNA as same as mIL-12 gene in size was found,transfected NIH3T3 expressed GFP-mIL12 fusion protein.Conclusion Transfected PA317 cells can produce retrovirus which can infect NIH3T3 cells,and the expression of mIL-12 gene can be detected in NIH3T3 cells.
3.Study of electroneurophysiology on Charcot-Marie-Tooth disease

Objective To explore the diagnostic value of electroneurophisology in patients with charcot-Ma-rie-Tooth disease(CMT).Methods Electromyogram(EMG),Motor nerve conduction velocity(MCV),Sensory nenre conduction velocity(SCV) and F wave were measured in 31 cases with Charcot-Marie-Tooth disease(CMT).Resuits The abnormality rate of EMG was 90%,MCV was 100%,SCV was 90%,and F wave was 34%.Conclu-sion The electroneurophysiology is very practical and valuable in diagnosing Charcot-Marie-Tooth disease.

observation group than in the control group at T1 and T2 (P < 0.01). These findings indicate that dexmedetomidine for sedation after hip replacement can lessen inflammatory response and contribute to the recovery of hip function.
6.The relationship between vitamin D receptor gene TaqⅠ polymorphism and diabetic retinopathy

Objective To investigate the polymorphism of the vitamin D receptor gene (VDR)TaqⅠin relation to diabetic retinopathy. Method Fragment length discrepant allele specific PCR(FLDAS-PCR) were used to determine VDR genetypes in 158 patients with diabetic retinopathy and in 198 normal subjects. Results The frequency distribution of VDR genotypes in diabetic retinopathy patients was 106 (67.1%) in TT, 33(20.9%) in Tt, 19(12.0%) in tt; and in normal persons was 165 (83.3%) in TT, 23(11.6%) in Tt, 10 (5.1%) in tt. 7.The variability of parameters of multi-focal electroretinogram

Objective To evaluate the variability of four parameters of multi-focal electroretinogram (mERG) a-wave amplitude, b-wave amplitude, a-wave latent period, b-wave latent period. Methods Sixty normal eyes of 46 volunteers were divided into 3 groups of different ages. RETIscan 3 12 system was used to carry out mERG examination. The stimulus matrix of 61 hexagonal elements spanning the central 24?of the visual field. These hexons were scaled with eccentricity and divided into 5 rings. First-order kernel was selected. Results The variability of four parameters of mERG was great. The variability of b-wave latent period was the smallest, its coefficient of variatian was 4.52%~15.62%;that of a-wave latent period held the second place:10.29%~48.67%;that of b-wave amplitude was greater:25.92%~76.11%;that of a-wave amplitude was the greatest:43.82%~88.23%. The results of three groups showed that b-wave amplitude of ring 1 had the smallest variability. Conclusions The variability of latent period is smaller than that of amplitude; the variability of b-wave was smaller than that of a-wave. The longer the centrifugal distance, the lower the amplitude density of a-wave and b-wave. Physiological and anatomical factors might be important for the variability of parameters of multi-focal electroretinogram.
8.Distribution and Antibiotic Resistance of Escherichia coli in Hospital

OBJECTIVE To understand the distribution and antibiotic resistance of E.coli isolated from our hospital and offer suggestions for clinic.METHODS The antibiotic susceptibility to 21 kinds of antibiotics were tested by K-B method and analyzed by Whonet software.RESULTS The antibiotic resistance of 301 E.coli strains to ampicillin,quinolones,piperacillin,gentamicin,cefazolin,cefuroxime,ceftriaxone,ceftazidime,aztreonam,cefepime, ampicillin/sulbactam were 83.39%,69.44%,62.79%,44.19%,42.86%,36.88%,35.22%,31.23%,29.90%,29.57% and 26.91%,respectively.CONCLUSIONS E.coli isolated from our hospital show high resistance to several kinds of antibiotics.The drugs should be choosed reasonably according to their antibiotic suscepitibity results.

Ezrin,a member of the ERM(Ezrin/Radixin/Moesin)protein family,fuctions as a molecular linker between the cytoskeleton and the plasma membrane.Recently,it has been found to be expressed abnormally in many malignant tumors,which indicats that it plays indicative of an important role in tumor infiltration and metastasis.Ezrin influences tumor metastasis in many ways,aspects including altering tumor cell movement,regulating cell-cell adhesion,participating in signal transduction in tumor cells,inhibiting cell apoptosis and regulating the function of phagocytes.
10.Making Monoclonal Antibodies by Phage Antibody Libraries

The emergence of repertoire cloning using phage antibody libraries has revolutionized the antibody engineering technology. We prepared phage antibody libraries from both human breast cancer cell immunized murine spleen cells and human peripheral lymphocytes from volunteer received HBsAg vaccination. Mouse anti-human breast cancer cell antibodies and human anti-HBsAg antibodies were successfully cloned by panning of the phage antibody libraries against breast cancer cells and immobilized HBsAg. The resulting Fab fragments were expressed in E.coli cells and the specificity, affinity and DNA sequences were analyzed. The results showed the potential of the phage display system in antibody preparation.
