1.Effects of Lanthanum on Excitatory Amino Acids and Calcium Homeostasis of Cerebral Cortex in Rats

Objective To observe the effects of lanthanum on learning and memory,and explore the corresponding mechanism on the basis of the levels of excitatory amino acids and calcium homeostasis of cerebral cortex in rats.Methods Forty female Wistar rats were randomly divided into control,low-,moderate-and high-dose lanthanum groups(ten rats in each group).The offspring of lanthanum exposure groups were administrated with lanthanum from weaning for one month.The capability of learning and memory was measured by neuroethology testing.The levels of excitatory amino acids,intracellular calcium ion and Nissl body expression of cerebral cortex were also detected.Results The capabilities of learning and memory in lanthanum exposure groups were significantly lower compared with the control,with a dose-dependent manner.The levels of glutamate and intracellular calcium ion of cerebral cortex in low-and moderate-dose group were significantly higher than those in controls,and the levels of glutamate,aspartate and intracellular calcium ion of cerebral cortex in high-dose group were increased significantly as compared with the control group,low dose group and moderate dose lanthanum group.Besides,Nissl body expression of cerebral cortex in low-dose group was lower than that in control group,and the levels of Nissl body expression of cerebral cortex in middle dose lanthanum group and high dose groups decreased further.Conclusion The increased levels of excitatory amino acids,calcium homeostasis unbalance and cellular injuries in cerebral cortex may be involved in lower capacity of learning and memory in the rats exposed to lanthanum.

3.2-ME-induced apoptosis of K562 cells and its mechanism

Objective To study the effect of 2-methoxyestradiol (2-ME) on proliferation,apoptosis and its mechanism.Methods K562 cells were treated with 2-ME of different concentrations and time in vitro.Cell apoptosis rate was measured by flow cytometry(FCM).Activity of NF-Kappa B in nucleus was detected by electrophoretic molility shift assay(EMSA).Results After treatment with 2-ME,K562 cells apoptotic rate increased significantly.After treatment with 4μmol/L 2-ME for 24h、36h、48h,the activity of Caspase-3 and Caspase-9 were significantly higher and activity of NF-Kappa B in nucleus was significantly lower.Conclusion The present study showed that 2-ME induced apoptosis of K562 cells via active Caspase-3 and Caspase-9 and inhibit the activity of NF-kappa B in nucleus.This study provided useful experimental data for clinical application of 2-ME.
4.Preparation and Quality Control of Luhuifangba Burn Cream

OBJECTIVE:To prepare Luhuifangba burn cream and establish a method for its quality control.METHODS:The burn cream was prepared from freeze-dried powder of aloe barbadensis miller gel,borneol,hexadecanol,cosmolin,sodium dodecylsulfate,glycerol,ethylparaben and distilled water.Polysaccharide in the preparation was identified by paper chromatography,and borneol was identified by means of the color reaction between1%vanillin-sulfuric acid and bornyl alcohol and isoborneol.The content of polysaccharide in the cream was determined using anthrone-sulfuric acid method.RESULTS:The preparation was a cream in ivory white color,which spread easily and smelled slightly oily.Polysaccharide and borneol in aloe were able to be identified by the above-mentioned method.The linear range of polysaccharide was0.02~0.2mg/ml,and the average recovery was106.3%(RSD=2.5%).CONCLUSION:The preparation method is feasible,and the quality control method is simple,accurate,and reproducible.
5.Perioperative anesthetic management for patients who received percutaneous nephroscopic peripancreatic necrectomy for acute necrotizing pancreatitis

Objective To review our experiences on perioperative anesthetic management for patients who received percutaneous nephroscopic peripancreatic necrectomy for acute necrotizing pancreatitis.Method The clinical data on 18 patients with acute necrotizing pancreatitis who received percutaneous nephroscopic peripancreatic necrectomy in our hospital from August 2008 to January 2011 were retrospectively analyzed.Results There was a marked fluctuation in hemodynamic status of the patients which required the use of vasoactive drugs during perioperative period.PETCO2 significantly increased after pneumoperitoneum.Tracheal extubation was possible in 11 patients immediately after surgery in the operation room,while 7 patients required tracheal intuhation to be transported back to the surgical intensive care unit (SICU).Conclusion Most patients who underwent percutaneous nephroscopic peripancreatic necrectomy had a varying degree of shock and multi-organ injury before operation.Proper anesthetic induction and maintenance,correct use of vasoactive drugs and lung protective ventilation strategy,and active fluid resuscitation are the keys to good perioperative anesthetic management and to improve treatment results.
6.Expression and significance of Th17 cells in peripheral blood of patients with rheumatoid arthritis

ObjectiveTo investigate the expression and significance of Thl7 cells in peripheral blood of patients with Rheumatoid Arthritis (RA).MethodsThirty-three active RA and 27 stable RA patients and 30 healthy individuals were enrolled in the study.Intracellular flow cytometry detection of TH17 cells in peripheral blood was established.The correlations between Th17 cells and RA disease activity (DAS28 scale) as well as bone erosion were analyzed.ResultsThe expressions of Th17 cells in the patients with RA were significantly higher than those of healthy controls [ ( 1.60 ± 0.51 ) % vs (0.81 ± 0.21 ) %,t =10.30,P < 0.001 ].The Th17 levels in active RA patients were also higher than that of stable RA patients and healthy subjects [ ( 1.94 ±0.38 ) % vs ( 1.19 ± 0.27) %,t =8.85,14.60,P < 0.01 ].And the Th1 7 levels in stable RA patients were also higher than that of healthy subjects (t =5.88,P <0.01 ).Active RA patients with bone erosion ( 19 cases) also had higher levels of Th17 cells than those without bone erosion ( 14 cases) [ ( 2.09 ± 0.39 ) % vs.( 1.74 ±0.29 ) %,t =2.83,P < 0.01 ].The expression of Th17 cells was positively correlated with the DAS28 score ( r =0.86,P < 0.01 ).ConclusionThere is a high expression of Th17 cells in peripheral blood of RA patients,which is related to disease activity and bone erosion.Th17 cells might play an important role in the pathogenesis of RA.
7.Expression and significance of B lymphocyte stimulator and BAFF-receptor in peripheral blood mononuclear cells of patients with systemic lupus erythematosus.

Objective To investigate the expression and significance of B lymphocyte stimulator (Blys) and its receptor BAFF (BAFF-R) in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE). Methods The expression of Blys and BAFF-R was measured by flow cytometry in 90 pa-tients with SLE,which was compared with that of 45 healthy controls. The relationships between the expression of Blys, BAFF-R and other laboratory parameters as well as disease activity were analyzed. Results The expression of Blys and BAFF-R in PBMCs from patients with SLE was significantly elevated compared to healthy controls (P <0.001), so did the active group (P < 0.001) and inactive group (P < 0.001 and P < 0.01). The expression of Blys in PBMCs from active SLE patients was higher than that of inactive patients (P <0.05). However,there was no statisti-cal difference of BAFF-R between the two groups. The expression of Blys in PBMCs was positively related to SLEDAI (r =0.728,P <0.001) ,IgG and IgM(r=0.691,P<0.001 and r =0.453,P<0.01) ,but negatively related to C3 and CA (r = -0.510, P < 0.001 and r = -0.312, P < 0.05). The expression of Blys in dsDNA positive group was higher than those of dsDNA negative group (P < 0.01). The expression of Blys and BAFF-R in Cl qAb positive group was higher than those of ClqAb negative group as well (P <0.01). Conclusion The expression of Blys and its receptor BAFF-R in PBMCs from SLE is elevated ,which may reflect the disease activity and is related to the pro-duction of autoantibody. They might be involved in the pathogenesis of SLE.

Objective To investigate the risk factors of disease progression and adverse pregnancy outcome in patients with systemic lupus erythematosus (SLE) during pregnancy.Methods Clinical data of 118 pregnant women with SLE admitted from June 2004 to October 2015 were retrospectively analyzed.The patients were divided into selective pregnant group (group A,n =72 cases) and non-selective pregnant group (group B,n =46) according to the disease activity of SLE.The disease progression and pregnancy outcomes were compared between two groups.Results The various system damages occurred in group B,including hematological damage in 16 cases,kidney damage in 19 cases,erythra in 10 cases,arthritis in 10 and serositis in 12 cases;while the corresponding cases in group A were 10,14,6,4 and 4(x2 =7.133,P=0.008;x2 =6.658,P =0.010;x2 =4.304,P =0.038;x2 =7.030,P =0.008;x2 =10.095,P =0.001).SLE exacerbation occurred in 28 cases (74%) of group B and 12 cases (17%) of group A (x2 =34.944,P =0.000).The logistic regression analysis showed that hypocomplementemia,proteinuria,SLEDAI score before pregnancy and positive anti-dsDNA antibody were the risk factors of SLE disease exacerbation during pregnancy.The maternal complications occurred in group B,including pregnancyinduced hypertension in 7 cases,preeclampsia in 10 cases and infections in 11 cases/times;while the corresponding cases (case/time) in group A were 2,6 and 4 (x2 =4.526,P =0.033;x2 =4.304,P =0.038;x2 =8.525,P =0.004).There were 14 cases of therapeutic induced labor,7 case of stillbirth and 27 cases of total fetal loss in group B,while the corresponding cases in group A were 2,0 and 4 (x2=18.317,P =0.000;x2 =9.080,P =0.003;x2 =40.920,P =0.0300).The logistic regression analysis showed that positive anticardiolipin antibody,proteinuria,SLEDAI score before pregnancy and renal dysfunction during pregnancy were risk factors of fetal loss.There were 87 cases of successful delivery (73.7%),the successful delivery rates were 41.3% (19/46) in group B and 94.4% (68/72) from group A,respectively.The infant complications occurred in group B,including premature birth in 15 cases,low birth weight in 13 cases,neonatal jaundice in 5 cases and mild asphyxia in 5 cases;while the corresponding cases in group A were 24,18,4 and 2 (x2 =11.442,P =0.001;x2 =11.395,P =0.001;x2 =4.664,P =0.031;x2 =8.035,P =0.005).Conclusion SLE patients whose disease conditions are not well controlled would lead to higher percentage of disease deterioration during pregnancy and worse pregnancy outcomes.
10.Clinical outcomes of implant denture application following mandibular defect repair with fibula flaps in 58 cases

Objective:To explore the therapeutic effect of implant denture after repairing mandibular defect with fibula flaps.Methods:The patients who received implant dentures following mandibular defect repair with fibula flaps to treat mandibular ameloblastoma were included as study subjects. These patients were admitted to the First Affiliated Hospital of Zhengzhou University from March 2015 to January 2018. The free fibular flap was used to repair the mandibular defect after the diseased tissue was removed. The Straumann soft tissue level implants were placed 6-9 months after bone grafting, and the upper porcelain crown was repaired 3 to 4 months after implantation to restore the occlusal relationship and chewing function. The patients were followed up 6 and 12 months after surgery to observe the effect of surgical repair. European Organization for Research and Treatment of Cancer quality of life questionnaire head and neck module (EORTC QLQ-H&N35) and the 14-item Oral Health Impact Profile (OHIP-14) questionnaires were applied to investigate and evaluate the quality of life of the patients before surgery and 6 months and 12 months after surgery.Results:There were a total of 58 patients, 36 males and 22 females, from 18 to 62 years old, with an average of 31.6 years old. In all of the 58 cases, the free flaps survived with restored curvature of the patient’s mandibular defect, symmetrical maxillofacial region and occlusal relationship. Their functions of speech, chewing, and swallowing have all returned to normal. The EORTC QLQ-H&N35 questionnaire survey showed that the score in terms of diet, social contact and teeth was the highest at 6 months after surgery, with statistically significant difference from the preoperative level ( P<0.05); and the score decreased at 12 months after surgery, without statistically significant difference from the preoperative level ( P<0.05). Compared to the preoperative level, the scores in terms of mouth opening both gradually decreased at 6 months and 12 months after surgery, without statistically significant difference ( P<0.05). The score in terms of language at 6 months after surgery was higher than the preoperative level and then fell to the preoperative level at 12 months after surgery. There were no significant changes of the scores in terms of swallowing at each time point ( P>0.05). The OHIP-14 showed that the scores in terms of functional limitation, physical impairment and disability increased at 6 months after surgery with statistically significant difference compared with the preoperative level ( P<0.05), and decreased at 12 months after surgery without statistically significant difference compared with the preoperative level ( P>0.05). The scores in terms of psychological discomfort, psychological disorder and social disorder increased at 6 months after surgery with statistically significant difference compared with the preoperative level ( P<0.05). The scores decreased at 12 months after surgery and were significantly lower than the preoperative level ( P<0.05). The physiological pain gradually decreased at 6 and 12 months after surgery compared with the preoperative level with statistically significant difference ( P<0.05). However, there was no statistically significant difference between 6 and 12 months after surgery( P>0.05). Conclusions:The application of free fibular flaps to repair mandibular defects and the use of the implant dentures can not only restore the patients’ facial contour, but also restore the patients’ occlusal relationship and chewing function, which better improve the patients’ quality of life.
