1.EFFECTS OF IL-1 AND IL-4 ON THE EXPRESSION OF p27 AND CDK_2 IN RAT GLOMERULAR MESANGIAL CELLS IN VITRO

To evaluate the effects of interleukin-1 and interleukin-4 on the expression of p27 (one of the negative cell-cycle proteins) and CDK 2 (cell-cycle depending kinase, one of the positive cell-cycle proteins) on renal mesangial cells of rat in vitro, and to analyze their significance. The results showed that the proliferation of mesangial cells was induced markedly by interleukin-1, while the expression of p27 was down-regulated and that of CDK 2 was up-regulated. All of above changes were prevented by interleukin-4. The results suggested that the responses of rat renal mesangial cells to interleukin-1 and interleukin-4 were closely related to the changes in the expression of p27 and CDK 2.
2.Evaluation of inhalation of budesonide bitween suspension and aerosol in the treatment of infant asthma

Objective To evaluate the efficacy of inhalation of budesonide bitween suspension and aerosol in the treatment of infant asthma. Methods 80 patients less than 3 years old were divided in three groups randomly,in group A the patients received budesonide suspension inhalation therapy,in group B the patients received budesonide aeroso inhalation therapy, in group C the patients received route treament. The patients were followed 1,3,6,12 months after teeament,symptom assessment scores, acute episode of asthma and administration of medication were compared beteen two groups. Results Compared with group C, symptom assessment scores became lower (P <0.01 ) ,acute episode of asthma reduced and use of receptor β2 receptor agonist and steroid reduced in group A and B(P<0.01);after 1,3,6 montha, the indicators observed in group A was better than in group B(P< 0.05),but there was no difference between two group after 12 months( P > 0.05). Conclusion Budesonide inhalaton is effec-tire in the treatment of asthma, in patients with infant asthma, budesonide suspension aerosol inhalation should be the first choice of treatment.
3.The clinical observation of early micro-dose heparin in the children with systemic inflammatory reaction syndrome

Objective To observe the clinical effects of the children with svstemic inflammatorv re- active syndrome(SIRS)receiving micro-dose heparin at early stage of the diseases.Methods The 53 cas- es diagnosed as SIRS were included in the randomized control trial.They were divided into two groups,26 cases in control group and 27 cases in therapeutic group.The children in control group received therapy for their primary diseases and other routine managements for SIRS.The children in therapeutic group received both above therapy and micro-dose heparin(5-10 U/kg,1 fime/6hours)via subcutaneous injection at earlv stages of diseases for 3 days.Results There were improvements in both control and therapeutic group, platelets count increased,C-reactive protein decreased (P<0.01),there were significant diffemnce in platelets and C-reactive protein between two groups,the time of platelets recovery in therapeutic group [(28±9)h]Was less than that in control group[(55±14)h](P<0.01).In therapeutic group,the dumtion of SIRS was shortened (P<0.05),mortality and the incidence of multiple organ dysfunction svndrome (MODS)and disseminated intravascular eoagultion(DIC)were decreased significantly(P<0.05).Con- clusion Early micro-dose heparin in SIRS can shorten its duration and decrease the mortality and the inci- dence of MODS in the children with SIRS.
4.Effect of valsartan on P27 expression in mesangial cells of Thy1 glomerulonephritis rats

Objective To observe P27 expression and proliferation of mesangial cells of rats with Thy1 glomerulonephritis and to evaluate the interfering effect of valsartan, a specific angiotensin Ⅱ receptor antagonist, on Thy1 glomerulonephritis. Methods Rats were divided into normal control group, Thy1 glomerulonephritis group and valsartan treatment group. Following the onset of glomerulonephritis, renal morphological changes were observed on days 1, 3, 5 and 7, and proliferating cell nuclear antigen(PCNA) and P27 expression in glomerulus were measured by immunohistochemistry and Western blotting. Results High expression of P27 was found in quiescent mesangial cells of normal rats, but decreased expression in mesangial cells of rats with Thy1 glomerulonephritis, accompanying with the proliferation of mesangial cells. In valsartan treatment group, glomerular hypercellularity, mesangial matrix expansion and PCNA expression were significantly reduced as compared with those of the untreated rats with Thy1 glomerulonephritis from day 3 to day 7( P
5.The Clinical Application in Rapid Diagnosis Flu Virus A and Flu Virus B by Fluorescent Immunoassay Analysis

Objective To evaluate the clinical application value of Sofia Influenza A+B fIA in detecting flu virus A and flu virus B.Methods Flu virus A and flu virus B was detected in 168 samples of nasopharyneal secretions which collected from children with influenza infection by Sofia Influenza A+B FIA,GICA and RT-PCR methods.Results Sofia Influenza A+B FIA positive detection rate was 17.86%(30/168),higher than GICA method.Compared with RT-PCR,its sensitivity and specificity for flu virus A and flu virus B were 83.3%,100% and 71.4%,100% respectively.Conclusion Sofia Influenza A+B FIA is a rapid clinical application method with high sensitivity and specificity.
6.Value of color Doppler ultrasonography in the evaluation of intraocular tumor

Objective To evaluate color Doppler ultrasonography(CDU) in the diagnosis of intraocular tumor. Methods Two hundred seventy-three cases(301 eye) of intraocular space-occupying lesions were examinated by high-resolution CDU to observe the two-dimensional gray-scale image and vasculature of intraocular tumors. Results Intratumor calcifying spot could be detected in 138 cases(149 eye) of 153 cases(178 eye) with retinoblastoma,and intratumor blood flow with branchy pattern could be found in 152(177 eye) of 153 cases(178 eye) with retinoblastoma. In 4 Coats disease and 2 persistent hyperplastic primary vitreous and 3 retinopathy of prematurity, intratumor calcifying spot couldn′t be detected,and the band blood flow of the detached retinal vessels could be detected. Intratumor blood flow with branchy pattern could be detected in all 66 choroidal melanomas and 4 melanomas of ciliary body with infiltrated choroid and 15 of 19 choroidal metastases. Intratumor blood flow with “simulating vessel pool” or conglomerate sparkled pattern could be found in 12 choroidal hemangioma. No intrinsic flow was detected in 8 choroidal hemorrhages or subretinal hemorrhages and 2 teratoid medulloepitheliomas. Conclusions CDU is of significance in the diagnosis and differention of intraocular tumors.
7.Changes of serum fibroblast growth factor 23 in patients with end stage renal disease and its clinical significance

Objective:To observe the serum level of fibroblast growth factor-23(FGF-23) in patients with end stage renal disease (ESRD) and study its association with phosphorus and vitamin D metabolism.Methods: Serum FGF-23 level was determined by enzyme-linked immunosorbent assay (ELISA) in ESRD patients undergoing haemodialysis (HD, n=50) and peritoneal dialysis (PD, n=24) and in twenty healthy controls (control group,n=20).Serum level of 1,25-(OH) 2VitD was measured by enzyme immunoassay(EIA).Serum intact parathyroid hormone (PTH), creatinine, and calcium and phosphorus were also measured.Results: Serum FGF-23 was obviously higher in HD group([88.51?35.01] ng/L vs [11.76?3.63] ng/L)and PD group([87.85?33.65] ng/L vs [11.76?3.63] ng/L)than in control group. Moreover, the serum level of 1,25-(OH)2VitD was lower in HD and PD groups than in control group ([19.82?4.99] pmol/L vs [48.37?3.47] pmol/L; [24.31?7.11] pmol/L vs [48.37?3.47] pmol/L ), and the level of 1,25-(OH)2VitD was much lower in HD group than in PD group. 9.Expression and Significance of Neural Cadherin in Perihilar Cholangiocarcinoma

Objective To explore the expression and the clinical significance of neural cadherin (N-cadherin) in perihilar cholangiocarcinoma. Methods The expression of N-cadherin was detected by immunohistochemical method in 26 patients with perihilar cholangiocarcinoma and 5 samples of normal bile duct tissues. The expression of N-cadherin was also compared by Western blotting assay between perihilar cholangiocarcinoma and paraneoplastic tissues. Results The im-munohistochemical results showed that positive expression of N-cadherin was observed in 14 of 26 patients with perihilar cholangiocarcinoma and in 6 of 26 paraneoplastic samples (53.84%vs 23.08%, P<0.05). There was 0 N-cadherin expres-sion in normal bile duct tissues. The positive rate of N-cadherin expression was higher in low differentiated group than that in middle and well differentiated groups. The result of Western blotting assay showed that N-cadherin expression was signifi-cantly higher in perihilar cholangiocarcinoma than that in the paraneoplastic tissues (0.88 ± 0.12 vs 0.43 ± 0.09, P < 0.05). Conclusion N-cadherin may be involved in the occurrence and development of perihilar cholangiocarcinoma.
10.Roles of interleukin-17 and RORγt in a murine model with paraquat- induced acute lung injury

Objective To explore the roles of Th17 cells in paraquat-induced acute lung injury(ALI)in a murine models through investigating the expression of Th17 type cytokine IL-17 and the specific transcription factor of Th17 cells-RORγt mRNA. Methods Male Kunming mice were randomly divided into the normal control group and the poisoned group. The ALI model induced by PQ poisoning was produced by intraperitoneal injection PQ solu-tion (20 mg/kg),and the normal control group was given normal saline solution with the same volume. After treatment for 48 hours,the animals were sacrificed. H&E staining and TUNEL staining were done to measure histo-logical changes and apoptosis. Serum level of Th17 related-cytokines was assayed by ELISA. The mRNA ex-pression of RORγt and the protein expression of IL-17 in the lung tissue were detected by real time-PCR assay and western blot,respectively. Results Compared with the normal control group,the lung inflammation and apopto-sis were significantly higher in the poisoned group. The serum level of IL-17A was significantly increased (P <0.05) and the lung parenchyma RORγt mRNA and IL-17 protein levels were significantly increased. Conclu-sion IL-17 plays an important role in the pathogenesis of ALI induced by paraquat poisoning.
