Objective To study a method for the determination of heterophyllin B inTaizishen- Huangqi oral solution.Methods The HPLC system consisted of Phenomenex Luna-C18 (4.6 mm × 250 mm, 5μm) column, the mobile phase consisted of acetonitrile and 0.1% acetic acid, the gradient elution flow rate was 1.0 ml/min, the column temperature was 30℃, and the UV detector was set at 203 nm.Results The linear response range was 0.016-0.320 mg/ml (r=0.999 8). The average recovery of heterophyllin B was 98.92%, and RSD was 0.80%. The average determination of Heterophyllin B within 5 batches of preparation was 1.63 mg/ml. Conclusions The method showed its high sensitivity, accurate, repeatable and high secificity. It could be used as a method to control the standard ofTaizishen-Huangqi oral solution.

Objective To establish the RP-HPLC method for the determination of myricetrin and quercitroside in Euphorbia Hirta L.Methods The ZORBAX SB-C18 (250 mm × 4.6 mm,5 μn) column was used,the mobile phase consisted of acetonitrile:0.1％ H3PO4(21 ∶ 79),the flow rate was 0.8 ml/min,the column temperature was 30℃ the detecting wavelength was at 256 nm.Results The cablibration curve was linear within a range of 0.013～0.26 mg/ml and 0.008～0.16 mg/ml,the average recovery was 99.1％,98.9％and the RSD was 0.91％,1.55％,respectively.Conclusion The method is simple,repeatable and accurate,it can be applied in quantitative determination of myricetrin and quercitroside in Euphorbia Hirta L..

Objective To optimize the flash type extraction ofDanzhuye Granules.Methods With the total flavonoids and extract yield as indices, the orthogonal test was adopted to optimize the extraction times, the ethanol concentration, solid-liquid ratio, and the duration of extraction forDanzhuye Granules.Results The factors that infuenced the extract efficiency from high to low were the extraction times, volume fraction of ethanol, the ratio of liquid to solid, and the extraction time. The optimum extracting condition included that the origin was extrated by the ethanol volume fraction 80% with the material to liquid ratio 30:1, total of 3 times, with each time of 3 minutes. Validation experiments of the potimal condition showed that the extraction of total flavonoids was 29.1 mg/g, and the extract yield was 23.6%.Conclusions The flash type extraction method is suitable for extraction, which provides a new method for the development and utilization ofDanzhuye Granules.

Objective To investigate the feasibility of 3D-printing coplanar coordinate template (3D-PCT) for guiding 125I radioactive seed implantation in the treatment of pelvic wall recurrence of cervical cancer on ensuring the accuracy of dose.Methods From Oct 2016 to Dec 2017 in Peking University Third Hospital,totally 10 patients with pelvic wall recurrent cervical cancer after radiotherapy were treated with 125I radioactive seed implantation assisted by 3D-PCT.The median age was 53.5 years old (37-71 years old).KPS score of the cohort were more than 70.All patients had received pelvic radiation therapy previously.The median volume of the lesion was 31.9 cm3 (3.5-58.0 cm3).The prescription dose was 120-180 Gy.The activity of seeds was 0.55-0.67 mCi(1 Ci =3.7 × 1010Bq),while the number of seeds was 12-81 (median 50) on preoperative plan.Radioactive seeds implantation was performed under 3D-PCT guidance according to the preoperative plan.The actual number of implanted seeds was 53 (10-82).Dosimetry parameters of preoperative plan and postoperative plan including D90,D100,V100,V150,V200,external index (EI),conformal index (CI),heterogeneity index (HI),and organat-risk doses of D2 cm3,D1 cm3 and D0.1 cm3 were compared using the nonparametric test.Results The seed number of postoperative plan was more than that of preoperative plan (Z =-2.255,P ＜ 0.05),but all of the dosimetric parameters showed no significant difference (P ＞ 0.05).D2 cm3 and D1 cm3 of rectum for postoperative plan were lower than that of preoperative plan (Z =-2.100,-2.240,P ＜ 0.05),while other dose parameters of normal tisssues showed no statistically significant difference (P ＞ 0.05).Conclusions Assisted by 3D-PCT for 125I radioactive seed implantation in pelvic wall recurrence of cervical cancer,the actual postoperative dose could meet the requirement of the preoperatie plan through the intraoperative optimization of dose.3D-PCT could ensure the precise of delivered dose of 125I radioactive seed implantation.

Objective To investigate the expression of vascular cell adhesion molecule-1 (VC.AM-1) in oxidative stress induced hypertrophic chondrocytes,in Kaschin-Beck disease (KBD) patients and in rat fed with T-2 toxin under selenium deficient conditions in order to analyze the relationship between VCAM-1 biological function and the dysregulation of chondrocyte differentiation in KBD.Methods The ATDC5 was cultured in 1％ ITS solution (10 mg/L insulin,5.5 mg/L transferrin,and 6.7 μg/L sodium selenite) for 21 days,and stimulated with 3-morpholino-sydnonimine (SIN-1,a nitric oxide [NO] donor) to obtain the oxidative stress induced hypertrophic chondrocytes.Real-time PCR was used to detect VCAM-1 mRNA in hypertrophic chondrocytes induced by different concentrations of SIN-1.The expressions of VCAM-I in articular cartilage of child and adult KBD patients and KBD animal model were determined via the immunohistochemical method,and KBD cartilage samples were obtained in KBD areas from KBD child who had died or from adults who had had surgery.Results After treatment of hypertrophic chondrocytes (ATCD5 cells) with SIN-1 (0,1,3,5 mmol/L),VCAM-1 mRNA levels (1.00 + 0.00,1.22 ± 0.20,0.71 ± 0.22,0.37 ± 0.16) were decreased in a dose-dependent manner when compared with the control group (F =27.788,P ＜ 0.05).The densities of VCAM-1 positive cells in superficial and middle zones of the articular cartilage of children KBD patients [(16.08 ± 5.20)％,(19.20 ± 9.71)％] were higher than those of control group [(0.00 ± 0.00)％,(0.00 ± 0.00)％],while that in the deep zone [(7.00 ± 4.40)％] in children KBD patients was significantly lower than that of control [(51.60 ± 20.58)％,tS/M/D=-10.972,-6.249,6.564,P ＜ 0.05].The positive cell density of VCAM-1 in the adult patients was significantly increased in the superficial zone [(7.92 ± 4.29)％ vs (3.12 ± 1.12)％] but significantly decreased in the middle zone [(17.54 ± 8.27)％ vs (31.75 ± 13.30)％] of articular cartilage when compared with that of control group (tS/D =-3.824,3.037,P ＜ 0.05).In articular cartilage of the four groups of KBD rats,the density of VCAM-1 positive cells in the superficial zone was significantly higher in low selenium diet group,T-2 toxin diet group and selenium deficient plus T-2 toxin diet group [(4.11 ± 1.90)％,(5.00 ±2.02)％,(2.78 ± 1.48)％ vs (1.89 ± 1.76)％,P ＜ 0.05].But the density of VCAM-1 positive cells in the deep zone was significantly lower in rat feed with selenium diet and selenium deficient plus T-2 toxin diet [(13.67 ± 2.45)％,(20.56 ± 7.42)％] than that of control group [(33.00 ± 12.57)％,P ＜ 0.05] in the epiphyseal cartilage of KBD rats.Conclusions The level of VCAM-1 is decreased both in the SIN-1 induced hypertrophic chondrocytes and in the deep zone of articular cartilage in KBD patients and in rat fed with T-2 toxin and selenium-deficient diets.VCAM-1 may be associated with the death of deep zone chondrocytes and differentiation disorder in cartilage.
