Objective To investigate the level change and clinical significance of serum IL-6, IL-8 and IL-13 in the patients with lupus nephritis(LN). Methods The levels of serum IL-6,IL-8 and IL-13 were measured by enzyme linked immunosorbent assay (ELISA) in 56 patients with LN. Results The levels of serum IL-6,IL-8 and IL-13 in the patients with active LN were significantly higher than those in the patients with non-active LN and the controls, but had no significant difference between the patients with non-active LN and controls. The levels of serum IL-6,IL-8 and IL-13 in the patients with active LN were positively correlated with anti-dsDNA antibody titre and erythrocyte sedimentation rate, but negatively correlated with the serum levels of complements C 3 and C 4. Conclusion IL-6,IL-8 and IL-13 may be involved in the pathogenesis of lupus nephritis, can serve as a clinical reference index for activity of LN, and have important clinical significance in guiding LN therapy and improving LN prognosis.

Objective To study the effects and clinical significance of tea pigment on 24 hour urinary endothelin(UET) excretion in patients with early stage diabetic nephropathy.Methods 65 cases of early stage diabetic nephropathy were randomly divided into expermental group and control group.Control group was treated by routine treatment and the expermental group was treated by tea pigment,in addition to routine treatment,and was orally given tea pigment capsule 0 24g three time daily,for 8 weeks.The amounts of 24 hours UET and urinary albumin excretion rate(UAER)in the two groups patients before and after treatment were measured using radioimmunoassay method.Results The amounts of 24 hour UET in patients with diabetic nephropathy were elevated significantly as compared with those in normal control group(P0 05).After 8 week's treatment,the amounts of 24 hour UET and UAER in expermental group were significantly lower than those of control group(P

Objective To investigate the levels of serum tumor necrosis factor-alpha (TNF-?琢) and soluble tumor necrosis factor receptor (sTNF-R) and sTNF-R/TNF-?琢 ratios in patients with severe lupus nephritis (SLN) and the impact of double pulse therapy with methylprednisolone (MP) combined cytoxan (CTX). Methods Serum TNF-?琢, soluble tumor necrosis factor receptor type Ⅰ(sTNF-RⅠ) and soluble tumor necrosis factor receptor type Ⅱ (sTNF-RⅡ) levels were determined in 38 cases of SLN patients and 35 health controls by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) before and after the double pulse therapy with MP and CTX. Objective To investigate the mechanism of capsaicin in treating active psoriasis vulgaris. Methods A total of 42 patients with active psoriasis vulgaris diagnosed by histology and clinical features were given either placebo or 0.025% capsaicin ointment four times daily for 30 days randomly by double-blind method. Vasoactive intestinal polypeptide receptor 1(VIPR1) gene translation in active psoriatic lesions before and after treatment with capsaicin ointment was detected by in situ hybridization. Results There was positive staining of VIPR1 gene in all the layers of psoriatic epidermis (95.5%) before the treatment with capsaicin ointment, but nearly no dyeing in epidermis (18.2%) after the treatment for 30 days. There was nearly no brown staining before and after treatment in control group. Conclusion VIPR1 gene translation in psoriatic epidermis is down-regulated after capsaicin treatment for 30 days.

Objective To investigate the relationship of serum soluble CD40L with diabetic retinopathy(DR) in patients with type 2 diabetes mellitus(DM). Methods Serum soluble CD40L level was measured by ELISA in 30 diabetic patients without diabetic retinopathy (NDR), 29 patients with background diabetic retinopathy (NPDR), 31 patients with proliferative diabetic retinopathy (PDR), and 32 normal control(NC) subjects. Results The serum soluble CD40L levels were higher in NDR,NPDR and PDR groups [(4.55±3.66), (6.65±4.24), (8.31±5.23) μg/L,P<0.01] than in NC group[(2.01±1.35)μg/L]. Objective To investigate the effect and mechanism of lipoxin A4 (LXA4) on uric acid (UA) induced oxidative stress of human umbilical vein endothelial cells (HUVECs).Methods The HUVECs was treated with uric acid to constructing the model of oxidative stress,and intervene the model with LXA4 and xylene based iodine (DPI),rotenone.Reactive oxygen species (ROS) of HUVEC were detected by a fluorescence probe 2',7'-dichlorofluorescin diacetate (DCFH-DA).The activity of reduced nicotinamide adenine dinucleotide phosphate (NADPH) oxidase and p47phox protein was measured by Lucigenin enhanced chemiluminescence and Western blotting among control,uric acid (UA),LXA4 and UA +LXA4 groups,respectively.All the results were described by the relative expression of the control group,repeated measure variance analysis and least significant difference test (LSD) were used for statistical analysis.Results UA could stimulate HUVEC to generate ROS with different concentrations and times (F=7.286,F=4.532,P＜0.05).Compared with the control group(100±l 1),the ROS production of group with 80 mg/L UA (177±18),120 mg/L (226±29) and 160 mg/L (225±16) increased significantly (t=4.127,t=7.591,t=7.236,P＜0.05).Compare with baseline(100±8),the ROS production increased significantly (t=3.688,t=3.513,t=4.526,t=8.269,t=3.829,P＜ 0.05) at 3 h(143±16),6 h(140±17),12 h(183±20),24 h(240±29) and 48 h(160±22).LXA4 could inhibit ROS generation at different concentrations and times (F=4.008,F =4.497,P ＜0.05).Compared with LXA4 concentration of 0 nmol/L,the LXA4 concentrations of 10 nmol/L (162±16) and 100 nmol/L (132±15) could significantly inhibit ROS generation(t=3.712,t=4.083,P＜0.05).Compared with pretreatment (269±39),the ROS generation decreased significantly (t=6.373,t=6.426,t=7.125,t=6.981,P＜0.05).with LXA4 pretreated for 15 min (160±16),30 rain(158±21),1 h (136±13) and 2 h(140±13).Compared with the UA group(252±31),LXA4 and DPI could significantly inhibited ROS generation (145±29,154±27;t=6.356,t=5.853,P＜0.05),but Rot was not significantly intervented (241±32;t=1.027,P＞0.05).The NADPH oxidase activity in the UA group was significantly higher than that in the control group (144±16,100±13;t=3.659,P＜0.05),but the group of LXA4+ UA was significantly lower than that of the UA group (119±14;t=3.124,P＜0.05).The cytoplasmic expression of NADPH oxidase subunit p47phox of UA group was significantly lower than that in the control group (47±6,100±8;t=7.562,P＜0.05),but the LXA4+UA group was significantly increased compare with the UA group (83±6,t=5.386,P＜0.05).The cytomembrane expression of p47phox of UA group was significantly higher than that in the control group (328±36,100±4,t=12.817,P＜0.05),but the LXA4+UA group was significantly decreased compared with the UA group (183±30,t=5.129,P＜0.05).Conclusion LXA4 inhibits UA induced ROS production in HUVECs.This mechanism might be through inhibiting p47phox trafficing from cytoplasm to cytomembrane,results in inhibiting the activation of NADPH oxidase.

Objective To investigate the changes of blood and urine endothelin levels in the aged patients with primary nephrotic syndrome(PNS) and their clinical significance. Methods The levels of blood and urine endothelin were measured by radioimmunoassay at different stages(active and alleviated stages) in 40 elderly and 40 non elderly patients with PNS and 40 elderly healthy controls(control group). The levels of endothelin in blood and urine of the aged patients with PNS, and serum creatinine(Scr), endogenous creatinine clearance rate(Ccr), 24 hour urinary protein and urinary retinal binding protein(uRBP) were determined. Results The contents of endothelin in blood and urine of the aged PNS patients in the active stage were (229?81) ng/L and (143?58)ng/24 h,which were apparently higher than those in the alleviated stage 〔(60?11) ng/L, (53?13)ng/24 h〕 and the healthy control group〔(53?9) ng/L, (50?14 ng)/24 h, P 0 05). Conclusions Endothelin may be involved in the pathogenesis of the aged PNS. The increased urine endothelin level may be an important factor in glomerulus and tubular damage in the elderly PNS patients.
