1.Diabetes pill in combination with metformin on elderly patients with type 2 diabetes mellitus with cerebral microvascular lesion blood viscosity, the effects of cognitive dysfunction

Objective To summarize analysis in elderly patients with type 2 diabetes mellitus with cerebral microvascular lesion with diabetes pill in combination with metformin on patients after treatment the blood viscosity, the effects of cognitive dysfunction.Methods Our hospital during the period of July 2014 to September 2016 received 100 cases of elderly patients with type 2 diabetes mellitus with cerebral microvascular lesions, according to the random allocation method divided into observation group(50 cases) and control group(50 cases) and control group using metformin treatment, the observation group USES the diabetes pill metformin treatment, treatment effect of two groups of patients and related indicators for comparative analysis.Results After treatment the cognitive function score and blood viscosity related indicators, statistically significant differences between observation group and control group.Conclusion Elderly patients with type 2 diabetes mellitus with cerebral microvascular lesions in clinical application in diabetes, pill metformin, help to improve the patient's cognitive dysfunction, improve the patient's blood viscosity, promote the prognosis of patients, is worthy of popularization and application.

3.Experimental Study of Percutaneous Absorption of Neostigmine Cream in Vitro

OBJECTIVE:To study the in vitro percutaneous absorption of neostigmine cream.METHODS:Using isolated mouse skin as permeation barrier,the influence of different concentrations of azone on percutaneous permeation of neostigmine was studied.RESULTS:Azone could obviously enhance the percutaneous permeation of neostigmine,and the accumulative permeation quantity of neostigmine cream containing2%and5%azone increased7.17%and11.87%respectively compared with that of cream without azone.CONCLUSION:The percutaneous permeability of neostigmine cream is satisfactory and the best concentration of azone in enhancing permeation seems to be5%.

OBJECTIVE:To establish a HPLC method for determination of the concentration of Buspirone in serum.MET_ HODS:The chromatographic system consisted of ZORBA SB-C 18 column and mobile phase of methanol-acetonitrile-water-0.1mol/L NaAC buffer solution(pH4.5)(10∶38∶51∶1),with a detection wavelength of237nm,and the content was calcu?lated by peak area internal standard method.RESULTS:The linear range of Buspirone was1~80ng/ml,r=0.9979.CONCL_ USION:This method is simple and accurate for determination of Buspirone in serum.
5.Effects of chronic cerebral ischemia on hippocampal microvascular architecture and ChAT expression in rats

Objective To investigate the damage of hippocampal neurons induced by chronic cerebral ischemia in the rats,and to clarify its mechanism.Methods 90 healthy adult male SD rats were randomly divided into sham operation group (n=30 ) and experimental group (n=60 ). The chronic cerebral ischemia rat models were established by permanently ligating the common carotid arteries on both sides of the rats in experimental group.The rats in sham operation group were established by incising the cervical median,only the common carotid arteries on both sides were separated without ligating. The rats in sham operation group and experimental group were respectively sacrificed at the 7th,14th and 21st day after operation.At each time point 10 rats in sham operation group and 20 rats in experimental group were selected and sacrificed.Immunohistochemistry staining was used to observe the dynamic changes of the expression levels of choline acetyltransferase(ChAT)in hippocampus neurons, and TA-FE method was used to observe the dynamic changes of hippocampal microvascualr architecture. MiVnt image analytical system was used to quantitatively analyze the immunohistochemistry result,the microvessel density (MVD)and micorvessel area density (MVA)of horizontal part of hippocampus in the rats. Results Compared with sham operation group,the ChAT expression levels in hippocampus neurons of the rats in experimental group at different time points were significantly decreased(P<0.05);and with the prolongation of time,the ChAT expression levels were gradually decreased;the ChAT expression level in 14-day experimental group was significantly lower than that in 7-day experimental group (P<0.05 );the ChAT expression level in 21-day experimental group was significantly lower than those in 7-day and 14-day experimental groups(P<0.05).The MVD and MVA of hippocampus of the rats in experimental group at different time points were obviously decreased compared with sham operation group(P<0.05);the MVA was gradually decreased with the prolongation of time, and the MVA of hippocampus of the rats in 21-day and 14-day experimental groups were obviously decreased compared with 7-day experimental group(P<0.05);the MVD was gradually decreased with the prolongation of time,the MVD of hippocampus of the rats in 21-day and 14-day experimental groups was obviously decreased compared with 7-day experimental group(P<0.05);the MVD of hippocampus of the rats in 21-day experimental group was obviously decreased compared with 14-day experimental group(P<0.05).Conclusion Chronic cerebral ischemia can lead to the progressive decrease of the ChAT expression level,MVD and MVA of hippocampus of the rats to aggravate gradually the learning and memory dysfunction, which may be one of the reasons of vascular dementia.
6.Comparison of Dispersibility Between Plasdone K30 and Plasdone S630 in Nimesulide Dispersible Tablets

OBJECTIVE: To compare dispersibility between two kinds of plasdone used as the hydrophilic adhesive in nimesulide dispersible tablets.METHODS: Nimesulide dispersible tablets were prepared with Plasdone S630 and Plasdone K30 as the hydrophilic adhesive respectively which were dissolved in 5% alcohol solution or 5% water solution.By friability,hardness and dispersibility tests,the influence of S630 and K30 in quality of nimesulide dispersible tablets were observed.RESULTS: Plasdone S630 alcohol solution or aqueous solution showed same or better results in friability,hardness and dispersibility compared with Plasdone K30 alcohol solution or aqueous solution.CONCLUSION: Plasdone S630 could be used as a alternate of K30 for hydrophilic adhesive in dispersible tablets.
7.Effect of Chitosan Wound Repair Membrane Gel on Serum Levels of TNF-α,IL-10 and ICAM-1 of Patients with Second Degree Burns

Objective:To explore the effect of chitosan hydrogel on the serum levels of tumor necrosis factor (TNF-α),interleukin (IL-10) and intercellular adhesion molecule (ICAM-1) of patients with burns.Methods:96 cases with second degree burns who were treated in our hospital from June 2015 to November 2016 were selected and randomly divided into two groups,with 48 cases in each group.The patients in the observation group were treated with chitosan hydrogel,while the patients in the control group were treated with conventional vaseline gauze wound.Then the wound healing,the serum levels of tumor necrosis factor (TNF-α),interleukin (IL-10) and intercellular adhesion molecule (ICAM-1) and the incidence of adverse reactions were observed and compared between the two groups before and after the treatment.Results:The healing rate of superficial burns at the 7th day in the observation group was significantly higher than that of the control group (P<0.01).The healing rate of deep burns at the 15th day and 25th day were significantly higher than those of the control group (P<0.05).The healing time of the superficial burns and deep bums in the observation group were significantly shorter than those of the control group,and the SI score was significantly lower (P<0.01).After treatment,the serum levels of TNF-α,IL-10 and ICAM-1 of patients with superficial burns significantly decreased in the two groups,and the observation group was lower than that of the control group (P<0.05).The serum levels of TNF-α,IL-10 and ICAM-1 of patients with deep burns were significantly lower than before,and the observation group was lower than that of the control group (P<0.01).The incidence of wound infection in the observation group was lower than that of the control group (P<0.05).Conclusion:Chitosan wound repair membrane gel could accelerate the wound healing,prevent the wound infection,and reduce the inflammation with high safety.
8.Diagnosis and treatment of lumbar disc herniation in teenagers: a clinical analysis of 40 cases

Objective:To investigate the pathogenesis, clinical feature and alternative approach of lumbar disc herniation in teenagers. Methods: The clinical data of 40 inpatients (age ≤21 years, admitted in Changhai Hospital from 1993 to 2001) with lumbar disc herniation were retrospectively reviewed. Of them, 7 patients received conservative treatment and the remaining underwent operation, including interlaminar partial laminotomy on one side (16 patients), hemilaminar laminotomy (14 patients) and interlaminar partial laminotomy on both sides (3 patients). All patients were followed up for 3 to 10 (4.2?0.6) years. Follow-up evalutions were performed via clinical visits. The Japanese Orthopaedic Association's evaluation system for lower back pain syndrome (JOA score) was used to evaluate the outcomes. Results: The lumbar and leg pain was released in all 40 patients after therapy. There was no deterioration of symptoms in the operative groups. During the follow-up, 8 patients had occasional low back pain but it did not affect their lives, 2 patients in the non-operative group had numbness, and no patients had lumbar degeneration in the operative groups. Objective: To study the antioxidative and hepatoprotective activities of low molecular fucoidan oligosaccharides(LMFO) from Laminaria japonica in mice.Methods: Mice were pretreated with LMFO(50?100?150 mg/kg ig respectively, 10 days),and then 0.2 % CCl 4 10 ml/kg ig and D-GalN(600 mg/kg)+LPS(lipopolysaccharide,1 ?g/kg) ig respectively in two model groups to induce liver injury. Liver injury was assessed by quantifying activities of plasma GPT, SOD, GSH-Px and MDA content.Results: The increase of plasma GPT activity was significantly inhibited by LMFO in two liver injury models, suggesting that LMFO had good protective effect on the hepatocytes. LMFO had good antioxidative effect in mice with liver injury induced by CCl 4 and D-GalN+LPS as indicated by decreased MDA content and increased activities of plasma SOD and GSH-Px. Conclusion: LMFO is protective against CCl 4-induced and D-GalN+ LPS induced liver injury in mice and its effect may be due to its antioxidative activities in vivo.
10.Optimization of the Prescription of Traditional Chinese Medicine for MS by Uniform Design

OBJECTIVE: To optimize the prescription of traditional Chinese medicine for motion sickness (MS).METHODS: Zingiber officinale,Herba pogostemonis,Aucklandia lappa were extracted respectively,and rotating-inducing MS mice were enrolled in uniform design.The prescription was optimized with MS index as the parameters.RESULTS: The optimal prescription was as follows:60 g Z.officinale,45 g Herba pogostemonis,5 g A.lappa.The extractive of prescription was significantly better than dimenhydrinate in the treatment of MS.CONCLUSION: MS index is a stable and sensitive parameters and it is suitable for screening and evaluation of anti-MS drugs.R.zingiberis,H.pogostemonis,A.lappq are potential drug for MS.
