Chinese Journal of Tissue Engineering Research 2005;9(24):242-243
BACKGROUND: The metabolism of acetylcholine in hippocampus reflects the function of cholinergic nervous system whose function is associated with learning and memory as well as intelligence.OBJECTIVE: To observe the changes of choline acetyltransferase activity in rat hippocampus after ischemia-reperfusion.DESIGN: Randomized controlled trial based on rats.SETTING: A Science and Research Department of Chengde Medical College.MATERIALS:The trial was conducted in the Central Laboratory of Chengde Medical College in 2002 and the subjects were 24 clean grade Wistar rats in equal number of the two sexes(weighting 260-280 g).METHODS:The 24 rats were randomly assigned into 3 groups: ① Model group:In this group the rats were made hyperlipemia and underwent bilateral carotid arteries blocking followed by reperfusion. ② Sham operation group:In this group the rats were made hyperlipemia and underwent only exposure of bilateral carotid arteries without ischemia-reperfusion. ③ Normal control group:In this group the rats received no intervention. The brains after the rats decapitated were harvested on the 1st 7th and 15th day respectively for colorimetric determination of the choline acetyltransferase activity in hippocampus.MAIN OUTCOME MEASURES:Determination of the choline acetyltransferase activity in the groups.RESULTS:None of the 24 rats was lost in the trial. ① The choline acetyltransferase activity in the model group on the 1st and 7th day was acetyltransferase activity in the model group on the 7th day was lower than that on the 1st and 7th day was lower than that in the normal controls[(0.037±0.006) μmol/g ·s, (0.017±0.006) μmol/g·s in model group vs (0.054±0.003) μ mol/g·s,(0.058±0.006) μmol/g·s in normal control group,P < 0.01].② The choline acetyltransferase activity in the model group on the 7th day was lower than that on the 1st day. With repairing of ischemia-reperfusion injury,it recovered partially[(0.039±0.007) μnmol/g.s].③ Choline acetyltrans-ferase activity in sham operation group was not different from that in normal control(P > 0.05).CONCLUSION:Simple exposure of carotid arteries does not change choline acetyltransferase activity. While ischemia-reperfusion can change the choline acetyltransferase activity and cause disorders of cholinergic nervous system function, which may be the reason for rat's intellectual disorders.

The potency of an improved recombinant multi-epitope vaccine against FMDV type Asia1 was evaluated in this study .A multi-epitope gene based on FMDV type Asia1 was designed and a recombinant expression plasmid (pRE-oIgG) was constructed .The proteins ,RE-oIgG and 3D were expressed in E .coli cells and purified with Ni-NTA agarose resin by affinity chromatography .The proteins ,RE-oIgG ,3D and RE-oIgG plus 3D ,were emulsified in an oil adjuvant ISA 206 .Twenty-five female guinea pigs were randomly divided into five groups and intramuscularly vaccinated for with RE-oIgG ,3D ,RE-oIgG plus 3D ,an inactivated FMDV vaccine (type Asia1) ,and PBS .All animals were vaccinated for two times .Anti-FMDV specific an-tibodies ,neutralization antibodies ,protection potency ,and lymphoproliferation assay were detected by ELISA ,virus neutrali-zation assay ,challenge test ,and flow cytometry ,respectively .Results showed that RE-oIgG plus 3D elicited significant high-level anti-FMDV specific antibodies compared to RE-oIgG alone (P<0 .05) .All the vaccinated animals induced higher level lymphoproliferation responses in vitro except PBS .Both 3D alone and PBS produced the negligible neutralizing antibodies and anti-FMDV specific antibodies .RE-oIgG plus FMDV 3D not only elicited high levels of anti-FMDV neutralizing antibodies ,but also induced significant lymphoproliferation responses .More importantly ,RE-oIgG plus 3D conferred complete protection to guinea pigs against challenge with 1 000 GPID50 .Interestingly ,two of five vaccinated animals with 3D alone were full protected against challenge ,and other three animals significantly showed a delay of 2-3 days in the onset of clinical signs .Therefore ,we considered that RE-oIgG plus 3D induces strong humoral and cellular immune responses ,which may be used for control and prevention of FMD in the future .

AIM: To detect the expression of microRNA (miRNA)-29a in pulmonary arteries of monocrotaline (MCT)-induced pulmonary hypertensive rats, and to investigate the effects of miRNA-29a on pulmonary hypertension.METHODS: MCT-induced pulmonary hypertensive model was established in Wistar rats. The expression of miRNA-29a in the lung tissue was determined by qPCR.miRNA-29a was overexpressed in the pulmonary hypertension rats by tail vein injection of miRNA-29a-mimic.Pulmonary artery pressure (PAP) and systemic arterial pressure (SAP) were measured. The morphological changes of the pulmonary arteries were observed by HE staining. The protein levels of p-Akt and p-eNOS were detected by Western blot.RESULTS: The mRNA expression of miRNA-29a was significantly decreased in the pulmonary arteries of MCT-induced pulmonary hypertensive rats.Furthermore, after overexpression of miRNA-29a, PAP was remarkably reduced, while SAP remained unchanged. In addition, the increased thickness of tunica media, the remodeling of pulmonary arteries and the decreased protein levels of p-Akt and p-eNOS in the pulmonary hypertensive rats were dramati-cally changed after miRNA-29a overexpression.CONCLUSION: Overexpression of miRNA-29a ameliorates pulmonary hypertension in rats. These effects may be associated with the activation of PI3K/Akt-eNOS signaling pathway.

Objective To detect the expression of KiSS-1, KiSS-1R and MMP-9 in hepatocellular carcinoma (HCC). To study the correlation of KiSS-1, KiSS-1R and MMP-9 expression with invasion and metastasis of HCC, and to explore the underlying mechanisms. Methods The expression of KiSS-1 , KiSS-1R mRNA in 33 HCC samples, 26 non-neoplastic adjacent liver tissue samples and 13 non-neoplastic distant liver tissue samples were detected by RT-PCR. Tissue chips were constructed by modified manual tools, which contained HCC, non-neoplastic adjacent liver tissues, non-neoplastic distant liver tissues, normal liver tissues and intrahepatic metastasis lesions. The expression of KiSS-1 and MMP-9 protein was determined by tissue chips, immunohistochemistry and semi-quantitative image analysis in 150 HCC, 137 non-neoplastic adjacent liver tissue, 98 non-neoplastic distant liver tissues, 16 normal liver tissues and 37 intrahepatic metastasis lesion samples. Results The results of RT-PCR showed that compared with the non-neoplastic adjacent liver tissues and the non-neoplastic distant liver tissues, the expression of KiSS-1 mRNA in HCC was significantly lower (P<0.01). The expression of KiSS-1R mRNA did not changed in HCC and non-neoplastic liver tissues (P>0.05). The expression of KiSS-1 protein was lower in HCC with metastasis and in clinical stage Ⅲ than that in those with non-metastasis, and in clinical stages Ⅰ and Ⅱ . It was also higher in the primary than in the metastasis lesions (P<0.01, respectively). The expression of MMP-9 was higher in tumors having peplos invasion and metastasis than in those with negative peplos invasion and non-metastasis. It was lower in the primary than the metastasis lesions (P<0. 01, respectively).Negative correlation between KiSS-1 and MMP-9 expression was found in HCC(r=- 0.340,P<0.01). Conclusions The imbalance between KiSS-1 and MMP-9 expression might play an important role in enhancing the invasive and metastatic capacity of HCC. Loss of KiSS-1 expression might predict an aggressive clinical behavior and was associated with metastatic potential in HCC.

Chinese Journal of Tissue Engineering Research 2024;28(2):295-301
BACKGROUND:Exercise as a viable non-pharmacological treatment has the potential to reverse skeletal muscle aging that deteriorates with age.The role of autophagy in the skeletal muscle aging process is indispensable.During skeletal muscle aging,Atg genes involved in regulating autophagy regulate the autophagic process in either a facilitative or inhibitory manner to improve the physiological morphology of skeletal muscle. However the specific molecular mechanisms of autophagy in the exercise regulation of skeletal muscle aging remain puzzling. OBJECTIVE:To search for general patterns of the effects of autophagic mechanisms on skeletal muscle aging during exercise through a review of articles in this field. METHODS:(1)CNKI and Web of Science were searched, reviewed, and screened for relevant literature using the keywords of"Atg genes(proteins),autophagy, exercise, and skeletal muscle aging"to lay the theoretical foundation for the full-text analysis.(2)The comparative analysis method was used to compare the similarities and differences among the included documents to provide reasonable theoretical support for the arguments.By the further comparative analysis of the literature, the relationship between relevant indicators was clarified,to provide the ideas for the full-text analysis. RESULTS AND CONCLUSION:Atg family-mediated autophagy is indispensable for delaying skeletal muscle aging.Atg genes involved in regulating autophagy regulate the autophagic process in either a facilitative or inhibitory manner to improve the physiological morphology and function of skeletal muscle.Different exercise patterns, such as age, time,or intensity at initiation, may have heterogeneous effects on the expression of autophagy-related proteins, but long-term aerobic exercise regulates Atg-related proteins, induces skeletal muscle autophagy, and delays the loss of muscle mass.

Chinese Journal of Tissue Engineering Research 2024;28(4):574-580
BACKGROUND:Aging is an irreversible process that is characterized by genes, diet and environment. As a central regulator of growth and development, mammalian target of rapamycin(mTor)can regulate the negative effects caused by aging, exercise and poor diet, which are correlated with the activity of mTor and its complexes. However, the relationship between these factors, such as mTor and the effect of exercise on aging,is still unclear. OBJECTIVE:To study the relationship between exercise, high fat/high salt diet and mTor in aging,so as to have a more comprehensive understanding of the prevention and treatment mechanism of aging. METHODS:(1)Literature retrieval was conducted in the core database of Web of Science and CNKI,using the keywords of"mTor gene, exercise, high fat/high salt diet, aging,"thereby providing theoretical support for this review.(2)Comparative analysis provided a theoretical basis for this thesis by carefully reading the obtained effective literature and comparing the differences among various literatures.(3)Through the comparative analysis of similarities and differences between the included articles,we could define each index and their relationship,so as to clarify the ideas of this review. RESULTS AND CONCLUSION:mTor is closely related to aging.Through the literature analysis,we believe that two complexes of mTor,mTorC1 and mTorC2,play important roles in aging, exercise and skeletal muscle growth and development. In addition,mTor-mediated S6K1,Akt,FOXO,and 4E-BP1 signaling pathways are strongly associated with exercise,high-fat diet,high-salt diet, and skeletal muscle/heart aging.

Background:
Feline panleukopenia virus (FPV) is a widespread and highly infectious pathogen in cats with a high mortality rate. Although Yanji has a developed cat breeding industry, the variation of FPV locally is still unclear.
Objectives:
This study aimed to isolate and investigate the epidemiology of FPV in Yanji between 2021 and 2022.
Methods:
A strain of FPV was isolated from F81 cells. Cats suspected of FPV infection (n = 80) between 2021 and 2022 from Yanji were enrolled in this study. The capsid protein 2 (VP2) of FPV was amplified. It was cloned into the pMD-19T vector and transformed into a competent Escherichia coli strain. The positive colonies were analyzed via VP2 Sanger sequencing. A phylogenetic analysis based on a VP2 coding sequence was performed to identify the genetic relationships between the strains.
Results:
An FPV strain named YBYJ-1 was successfully isolated. The virus diameter was approximately 20–24 nm, 50% tissue culture infectious dose = 1 × 10 −4.94 /mL, which caused cytopathic effect in F81 cells. The epidemiological survey from 2021 to 2022 showed that 27 of the 80 samples were FPV-positive. Additionally, three strains positive for CPV-2c were unexpectedly found. Phylogenetic analysis showed that most of the 27 FPV strains belonged to the same group, and no mutations were found in the critical amino acids.
Conclusions
A local FPV strain named YBYJ-1 was successfully isolated. There was no critical mutation in FPV in Yanji, but some cases with CPV-2c infected cats were identified.
