1.Value of 18F-FDG PET/CT radiomics in predicting the prognosis of non-small cell lung cancer

Objective:To investigate the value of baseline 18F-fluorodeoxyglucose (FDG) PET/CT radiomics features in predicting the prognosis of non-small cell lung cancer (NSCLC) before treatment. Methods:From January 2016 to August 2018, a total of 300 patients with solitary NSCLC (189 males, 111 females, age (62.3±9.0) years) who underwent 18F-FDG PET/CT imaging before treatment in Fourth Hospital of Hebei Medical University were retrospectively analyzed. According to the ratio of 7∶3 (R language), 300 patients were randomly divided into training group ( n=210) and validation group ( n=90). LIFEx software package was used to extract the PET and CT radiomics features of primary focus in 300 NSCLC patients. The least absolute shrinkage and selection operator (LASSO) algorithm combined with Cox proportional hazard regression model were used to select radiomics features and clinical features for predicting overall survival (OS) and progression-free survival (PFS) in training group. Then radiomics model, clinical model and complex model which integrated the two were established and the radiomics score (Rad-score), clinical score and complex score of each patient were calculated. Data of validation group was used to validate each training model. Efficiencies of each model in predicting the prognosis of patients with NSCLC were further evaluated by the concordance index (C-index), and a nomogram was developed based on the best prediction model. Results:In training group, the C-indices of predicting OS and PFS in NSCLC patients of radiomics model were 0.762 and 0.724 respectively, which were 0.834 and 0.780 respectively in clinical model, and were 0.842 and 0.787 respectively in complex model. Cox multivariate analysis showed that both Rad-score and complex score were independent prognostic factors for OS (hazard ratio ( HR): 1.804, 9.996, 95% CI: 1.023-3.184, 4.582-21.808, both P<0.05) and PFS ( HR: 1.771, 5.627, 95% CI: 1.138-2.756, 3.429-9.234, both P<0.05). Conclusions:Pre-treatment 18F-FDG PET/CT radiomics can predict OS and PFS of NSCLC patients. The complex model based on the combination of radiomics and clinical model is effective in predicting the prognosis of NSCLC patients, and the nomogram of complex model is simple and convenient to assist clinical decision-making.
2.The application of three statistical methods for reference intervals establishment of coagulation assays during pregnancy

Objective:To compare the practical value of three statistical methods in establishing reference intervals by the indirect method.Methods:This is a methodological evaluation study. The data of coagulation parameters were obtained from laboratory information system, which were from pregnant women who had done prenatal examination in Peking University First Hospital from January 2017 to December 2019. The test results from 32 401 pregnant women were collected. Those healthy pregnant women were divided into three groups: early pregnancy group(n=11 151), middle pregnancy group(n=4 872) and late pregnancy group(n=16 378). Statistical analysis was performed for the result of PT, APTT, FIB, TT, D-D and FDP, and the necessity of stratification based on age in different periods of pregnancy was analyzed. Stratification based on age was necessary in three pregnancy groups for PT and APTT, in late pregnancy group for FIB and TT, and in early pregnancy group for D-D. The reference intervals of coagulation parameters were calculated by three statistical methods: non-parametric method, Hoffmann method and Q-Q plot method. Forty-two healthy pregnant women from October 2019 to January 2020 were enrolled as reference individuals for the validation of the reference intervals.The proportions of test results outside the reference intervals in the reference population calculated and compared.Results:The levels of the six coagulation assays vary significantly during the three periods of pregnancy, stratification based on age was necessary in three pregnancy groups for PT and APTT, in late pregnancy group for FIB and TT, and in early pregnancy group for D-D. If the number of test results was large, non-parametric and Hoffmann method provided more similar results, while the reference intervals calculated with Q-Q plot method was slightly wider than Hoffmann method. If the number of test results was small, reference intervals calculated with Hoffmann and Q-Q plot method were more reliable. For pregnant women during early pregnancy under the age of 35, the reference intervals of PT, APTT, FIB and TT calculated by this method were (10.44-13.11)s, (25.29-35.88)s, (2.61-4.64)g/L and (11.53-15.58)s.Conclusion:When establishing the reference interval, stratification according to pregnancy period and age was needed. Hoffmann method can be used as an alternative to the direct method.
3.Effects of Different Trunk-Restraint Squatting Postures on Human Lower Limb Kinematics and Dynamics

Objective To investigate the effect of trunk control on the biomechanical characteristics of lower limb movements during Asian squats(AS)and Western squats(WS)in young adults to provide empirical support for the application and promotion of deep squat training.Methods Twenty-four healthy young male collegiate students performed AS and WS with and without bar control,and their lower limb kinematic and kinetic characteristics were collected using an infrared light-point motion capture system and a three-dimensional(3D)dynamometer.The 3D angles of the lower limbs were obtained using Cortex-642.6.2 software,based on the calculation of Euler angles,and the 3D moments were obtained by applying the inverse dynamics method.The effects of trunk control and deep squatting posture on the lower limb kinematic characteristics were examined using a two-factor analysis of variance with a 2×2 repeated design.Results There was no significant interaction between trunk control and the deep squatting posture for either kinematic or kinetic parameters(P>0.05).The WS group had a large knee flexion angle,peak patellofemoral contact force,and ratio of peak hip and knee extension moments,and small ankle dorsiflexion and hip flexion angles(P<0.05).The deep squat with a bar had a large ankle dorsiflexion angle,peak patellofemoral contact force,and hip flexion angle as well as a small knee flexion angle and ratio of peak hip and knee extension moments(P<0.05).Conclusions WS is helpful for training hip extension muscle groups,whereas AS is helpful for training knee extension muscle strength.The peak patellofemoral joint contact force of the WS is significantly greater than that of the AS;therefore,it is recommended that patients with patellofemoral joint pain use the AS.A squat with a bar can compensate for the body's balance;thus,people with limited ankle dorsiflexion range of motion or anterior tibial muscle weakness may consider trunk control training,such as a deep squat with a bar.This may help improve lower limb stability during squats.
4.Correlation Between Cutaneous Adverse Reactions Caused by Aromatic Antiepileptic Drugs and Human Leukocyte Antigen Gene Polymorphism

Aromatic antiepileptic drugs such as carbamazepine are the first-line treatment for epilepsy. The adverse reactions have greatly limited their clinical application. The occurrence rate of severe skin adverse reactions such as Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) is low,but they are often fatal.Human leukocyte antigen (HLA) gene polymorphisms are reported to be related with skin adverse reactions caused by aromatic antiepileptic drugs,but the exact mechanism is unclear.This article will perform a review about the correlation between skin adverse reactions caused by aromatic antiepileptic drugs and HLA gene polymorphisms published in recent years,in order to provide theoretical basis for further study of HLA susceptibility genes in Chinese Han population,and provide a reference for achieving individualized treatment of epilepsy.
