1.Expression of Dkk-3 and Cyclin D1 protein in hepatocellular carcinoma and their clinical significance

Purpose To investigate the expression of Dkk3 and Cyclin D1 protein in human hepatocellular carcinoma (HCC) and clinical significance.Methods Immunohistochemistry was used to detect Dkk-3 and Cyclin D1 protein expression level in 80 cases of hepatocellular carcinoma and corresponding para-cancer tissue.Results The expression of Dkk-3 in hepatocellular carcinoma was significantly lower than those in corresponding para-cancer tissue (P < 0.05) and the expression of Cyclin D1 in hepatocellular carcinoma was significantly higher than those in CoTesponding para-cancer tissue (P < 0.05).The up-regulation of Cyclin D1 and the down-regulation of Dkk-3 proteins were correlated with pathologic differentiation degree (P <0.05).There was a significant inverse correlation between Dkk3 and Cyclin D1 expression (P =0.044,rs =-0.226).Conclusion The abnormal expression of Dkk-3 and Cyclin D1 gene in human hepatocellular carcinoma suggest that Dkk-3 and Cyclin D1 gene may play an important role in the development and progression of the cancer.The combination deteetion of the two biomarkers may provide valuable data for diagnosis and prognosis estimation of HCC.
2.Launching ‘ interest-oriented research team’ to promote professional competence among public health students

Practicing and learning platform was preliminary established by teachers to explore the feasibility and availability of ‘ interest-oriented research team’ in promoting professional competence among public health students.Public health students interested in interest-oriented research were recruited.Through this platform,students can learn professional knowledge and conduct practice earlier and complete their study facing the society,community and public.After four years' practice and exploration,students indeed improved the abilities of researching and practicing,therefore,the training model of ‘interest-oriented research team’ needs promoting.
3.Study of Mechanism of Proinsulin Gene Mutations in Diabetes Mellitus

Objective To construct several human proinsulin mutants plasmid related to diabetes and to express in INS-1 (Insulin secreting beta cell derived line) cell. Methods Human mild proinsulin gene was used as template , and site-directed mutagenesis PCR was employed to generate four human proinsulin plasmid mutants. Each mutant plasmid was sequenced then transfected with empty plasmid and mild plasmid into INS-1 cell by liposome 2000. Insulin value in each cell solution was determined by radioimmunoassay. Results Proinsulin mutants plasmid were confirmed by sequencing. In-sulin values in culture solution of H-C(B19)G、H-L(B11)P、H-R(S6)C mutants are less than those in wild type and H-F (B25)L(P<0.05). Comparison of insulin values between H-C(B19)G、H-L(B11)P、H-R(S6)C groups were not statistically significant(P>0.05), and all these three groups showed no significant differences with empty plasmid group statistically (P>0.05).Insulin value of H-F(B25)L was of no significant differences statistically with empty plasmid(P>0.05). Conclu-sion Four human proinsulin mutants plasmid were constructed and expressed successfully in INS-1 cell, and different mu-tants plasmid result in diabetes through different mechanism.
4.Effect of Atorvastatin on the levels of adiponectin and leptin and adiponectin-leptin ratio in the elderly and middle-aged patients with type 2 diabetes and combined hyperlipidemia

Objective To evaluate effects of Atorvastatin on the levels of adiponectin and leptin and adiponectin-leptin ratio in the elderly and middle-aged patients with type 2 diabetes and combined hyperlipidemia Methods Totally 80 the elderly and middle-aged patients with type 2 diabetes and combined hyperlipidemia were treated with Atorvastatin (10mg/d) for 12 weeks.Fasting plasma levels of glucose(FPG),glycosylated hemoglobin(HbA1 c),total cholesterol (TC),triglyceride (TG),high-density lipoprotein cholesterol (HDL-C),low-density lipoprotein cholesterol(LDL-C),adiponectin(APN) and leptin(LEP) were measured at baseline and 12 weeks after treatment.And the adiponectin/leptin(A/L)ratio was calculated.Results Before versus 12 weeks after medication,TG[(4.92±1.23)mmol/L vs.(4.26±1.07)mmol/L,t=11.124,P< 0.001],TC[(2.69 ± 1.17) mmol/L vs.(1.66 ± 1.01) mmol/L,t=24.816,P<0.001] and LDL-C[(2.79 ± 1.02) mmol/L vs.(1.91 ± 0.92) mmol/L,t=21.508,P<0.001]were decreased obviously;HDL-C increased obviously [(1.03 ± 0.30) mmol/L vs.(1.33 ± 0.26) mmol/L,t =-12.011,P < 0.001];APN increased slightly without statistical significance (P =0.064);LEP level decreased slightly without statistical significance(P=0.068),and A/L increased obviously(P< 0.05).Conclusions Atorvastatin treatment can significantly decrease TG,TC and LDL-C level in the elderly and middle-aged patients with type 2 diabetes and combined hyperlipidemia,and significantly increase HDL-C as well as A/L level,while it has no obvious influence on APN and LEP.
5.The clinical application of vestibular diagnosis and treatment system in benign paroxysmal positional vertigo.

OBJECTIVE:
To evaluate the effect of the vestibular diagnosis and treatment system (SRM-IV ) in diagnosis and treatment of patients with benign paroxysmal positional vertigo (BPPV).
METHOD:
Patients who were diagnosed as BPPV by SRM-TV in the clinic of our hospital from November 2013 to October 2014 were retrospectively analyzed in this study.
RESULT:
Among 425 suspected cases, 230 BPPV-positive patients were diagnosed including 131 cases of posterior SC (57.0%), 95 cases of horizontal SC (41.3%) and 4 cases of more than two SC (1.7%). The cure rate by SRM-V was 94.6% and the effective rate was 100.0%. The relapsed occurred in 10 patients (4.8%), which contained 4 men and 6 women.
CONCLUSION
SRM-V can realize 360° reasonable repositioning procedure while Canalish reposition procedure cannot. SRM-V can improve both the corrective rate of diagnosis and the cure rate, especially for the patients who suffered from complex BPPV.

6.Practice of case-based PBL teaching model in clinical immunology and laboratory teaching

Objective To train medical students' competence of independent thinking and problem solving and to improve the quality of teaching.MethodsThe case-based PBL teaching model was introduced to teaching practice for 25 undergraduates majoring in medical laboratory.The teaching process included pre-class mobilization,question,self-study,discussion and summary.Effectiveness of teaching was evaluated with questionnaire.Results For these medical students,the enthusiasm of learning ( 83.3% ),ability of self-learning and information seeking ( 79.2% ),ability of discussing and problem solving ( 79.2% ),ability of scientific thinking and clinical reasoning ( 79.2% ) were significantly improved.Teaching satisfaction rate of case-based PBL teaching model was up to 95.8%.ConclusionBetter results of teaching were achieved and the case-based PBL teaching model is worth spreading.
7.The value of MR in the diagnosis of primary splenic lymphoma

Objective To evaluate the value of MR in the diagnosis of primary splenic lymphoma (PSL).Methods The MR imaging features of 3 PSL cases proved by pathology were retrospectively reviewed. Results Three cases were all pathologically diagnosed as B-cell non-Hodgkin's lymphoma(NHL). Two cases were multiple node shape and one was massive shape. Unenhanced MR imaging revealed heterogeneous splenic enlargement, with large nodules showing iso-hyperinternse on T1WI and hypointense on T2WI. Linear hyperintense on T1WI and T2WI was seen on the spleen peripherous. The spleen vascular was infiltrated. CE-MRI showed heterogeneous enhancement of spleen with iso-hypointense. The focus of postperitoneal showed medial enhancement. Immunohistochemistry-showed 2 cases of diffuse B cell. The CD20 and CD19 α of tumor cell showed diffuse( + ) ,CD3 ,CD5 individual( + ) ,CD43 ( + ) ,CD45RO( + ), CD10 ( +/- ), Mum ( + ), MAC387 individual ( + ), 1 case of B lymphocell type, CD79α ( + ) ,CD23( + ) ,CD38( + ) ,λ( + ). Conclusion The MR imaging features of PSL were characteristic and helpful in the diagnosis of PSL, but the correct diagnosis was still dependent on the pathology and immunohistochemical staining.
8.The effect evaluation of a nutrition and food safety science experience among primary and secondary school students

Objective To evaluate the effectiveness of an activity of nutrition science experience on nutrition and food safety cog-nition among primary and secondary school students .Methods With stratified cluster sampling ,students of grade 5 and 6 in one primary school and students of grade 1 and 2 in one middle school in Chongqing were selected ,and randomly divided into interven-tion group(n= 501) and control group(n= 522) .Only conducted the activity in the intervention group .Baseline data of all the students were investigated before the intervention .Effect evaluation was performed instantly in the intervention group and control group ,and a follow-up survey carried out in the intervention group after 9 months(n= 472) .Results The nutrition knowledge scores of instant intervention group were 9 .03 ± 2 .75 and 14 .70 ± 3 .28 before and after intervention respectively (U=29 .78 ,P<0 .01);the knowledge scores of the nine months later intervention group were 12 .35 ± 2 .89 ,which were lower than instant interven-tion group(U=12 .40 ,P<0 .01) ,but higher than before intervention(U=18 .04 ,P<0 .01) .The food safety scores of instant inter-vention group ,which were higher than control group ,nine months later intervention group and before intervention(P<0 .01) .Con-clusion It is feasible and effective to conduct a nutrition science experience among primary and secondary school students .
9.Strontium ranelate promotes osteogenic differentiation of rat bone mesen-chymal stem cells through Hedgehog/Gli1 signaling pathway

AIM: To explore whether strontium ranelate ( Sr ) promotes osteogenic differentiation of rat bone mesenchymal stem cells (BMSCs) through the Hedgehog/Gli1 signaling pathway.METHODS:BMSCs were isolated from 4-week-old rats by adherent culture and induced to differentiate into osteoblasts .According to the experimental purposes , the cells were exposed to different concentrations of Sr , cyclopamine ( Cy, an inhibitor of Hedgehog receptor ) or Gli1-siR-NA.The expression of Gli1 and Runx2 in the cells was detected by Western blotting .The activity of alkaline phosphatase ( ALP) was measured by the method of colorimetry , and the mineralized nodules were observed under microscope with aliz-arin red staining .RESULTS:Exposure to Sr at concentrations of 0.1 to 5 mmol/L for 7 d markedly increased the expres-sion of Gli1 in the BMSCs , and the increase in Gli1 expression was the most obvious following Sr exposure at concentration of 3 mmol/L.Cy at concentration of 10 μmol/L inhibited Sr-induced up-regulation of Gli1 expression.Transfection of the BMSCs with Gli1-siRNA not only obviously inhibited Sr-induced up-regulation of Gli1 and Runx2 ( a downstream protein of Gli1) expression, but also antagonized Sr-induced enhancement of ALP activity and the formation of mineralized nodules . CONCLUSION:The Hedgehog/Gli1 pathway is involved in Sr-induced osteogenic differentiation of rat BMSCs .
10.Role of JAK/STAT signaling pathway in high glucose-induced damage in human umbilical vein endothelial cells

AIM: To investigate whether Janus kinase/signal transducer and activator of transcription ( JAK/STAT) signaling pathway mediates high glucose-induced damage in human umbilical vein endothelial cells ( HUVECs ) . METHODS:The cell viability was examined by CCK-8 assay.The expression levels of JAK2, STAT3, caspase-9 and en-dothelial nitric oxide synthase ( eNOS) were detected by Western blot .The intracellular levels of reactive oxygen species ( ROS) were tested by DCFH-DA staining followed by photofluorography .Mitochondrial membrane potential ( MMP) was measured by rhodamine 123 staining followed by photofluorography .RESULTS:Treatment of HUVECs with high glucose (40 mmol/L glucose) for 6~12 h enhanced the protein level of phosphorylated JAK 2, peaking at 9 h.Treatment of the cells with high glucose for 6~12 h also increased the protein level of p-STAT3 with the peak value at 12 h.Pretreatment with the inhibitor of JAK/STAT pathway AG490 for 1 h before exposure of the HUVECs to high glucose significantly inhibi-ted the high glucose -induced injury , as evidenced by an increase in the cell viability , decreases in the expression of caspase-9 and the intracellular ROS production , and increases in MMP and the expression of eNOS .CONCLUSION:JAK/STAT signaling pathway is involved in the high glucose-induced damage in HUVECs .
