1.An assessment of nutritional status of 25-hydroxy vitamin D and PTH in puerperal and pregnant women

Objective To investigate the serum calcium absorption level ,distribution level and nutritional status of 25‐hydroxy vitamin D and parathyroid hormone (PT H ) in pregnant and puerperal women .Methods A total of 740 healthy pregnant women and puerperal women in the perinatal clinic of our hospital from September 2014 to August 2015 were recruited as the observation group and divided into second trimester group ,third trimester group and puerperal group .Contemporaneous 100 healthy non‐preg‐nant women were chosen as the control group .The levels of serum 25‐hydroxy vitamin D and PTH in all research subjects were de‐tected by using the electrochemiluminescence method .Then the differences of detection results were compared among various groups .Results The serum 25‐hydroxy vitamin D level was (57 .9 ± 18 .2)nmol/L in the second trimester group ,(39 .4 ± 15 .0) nmol/L in the third trimester group ,(65 .1 ± 19 .6)nmol/L in the puerperal group and (78 .6 ± 16 .2)nmol/L in the control group , the differences among groups were statistically significant (P<0 .05) .The serum PTH level in the third trimester group was higher than that in the second trimester group ,the difference between the two groups was statistically significant (P<0 .05) .Serum calci‐um ,magnesium and alkaline phosphatase levels in the observation group were lower than those in the control group with statistical‐ly significant (P<0 .05) .Conclusion Pregnant and puerperal women usually have low levels of calcium and magnesium ,and 25‐hy‐droxy vitamin D deficiency .It is necessary to supply vitamin D and other mineral elements when taking calcium supplement .

Objective To establish the feasibility and clinical applications of intravenous myocardial perfusion imaging with Levovist in assessing the coronary microcirculaton reserve.Methods Intravenous myocardial contrast echocardiography (IMCE) was performed pre and post dipyridamole injection in 10 cases with normal coronary angiography (group I) and 8 cases with 50% to 70% stenosis in the lift anterior descending branch(LAD) (group Ⅱ) respectively.A set of special instrumental setting and an integrated imaging approach including power Doppler imaging,incremental triggering imaging and harmonic imaging were utilized to record the apical four chamber view.The power Doppler signals of the septum were analyzed and the time amplitude curves were plotted during Levovist infusion and incremental triggering.The maximal amplitude score (A),the mean ascending slope of the curve(?) and the product of A?? were measured.The reserve of A,? and A?? were also calculated.Results At baseline,both group I and group Ⅱ had normal myocardial perfusion.After dipyridamole infusion,contrast score A increased slightly in group I and decreased in group Ⅱ without statistical significance.Both ? and A?? were increased significantly after dipyradamole (P

Objective To study the clinical application of intravenous myocardial contrast echocardiography(IMCE) in assessing myocardial reperfusion by using intravenously infused Levovist before and after percutaneous coronary interventions(PCI). Methods IMCE was performed in 20 patients with coronary artery disease(CAD) before and after PCI. AIM To evaluate the anti-angiogenesis action of Bevacizumab on corneal neovascularization(CNV) in rats induced by alkali burns.·METHODS: 20 Health Wistar rats, aging from 6 to 8 weeks and weighting from 170g to 190g from 170g to 190g were prepared for CNV animal models. Both corneas of each animal in experimental were cauterized with alkali, then all rats were randomly divided into four groups (each group have 5 rats and 10 corneas), the both corneas of each rats were received subconjunctival Bevacizumab in different dosage (group 2, 0.5rag; group 3, 1.0mg; group 4, 2.0mg)and the group 1 received carrier solution. The occurrence and development ofCNVwereobservedbyslit-lamp microscope, and length and area of CNV were calculated. All rats were followeded up 16 days after alkali burns. The 40 corneas were taken for histopathological examination. Vascular endothelial growth factor (VEGF) were detected in all rat corneas by immunohistochemistry method. ·RESULTS: In the bevacizumabotreated eyes, the vascular area was lower than in the control eyes. The treated group was statistical differences compared with the control group; when vascular area were compared between the treated groups, no statistical differences were observed. The histopathological findings showed that the inflammation cells and the neovascularity in each treated group were significantly fewer than that in the control group. The expression of VEGF markedly increased in CNV control group compared with bevacizumab-injected group. ·CONCLUSION: Subconjunctival application of a certain concentrations Bevacizumab could inhibit angiogenesis in rats corneas induced by alkali burns.
6.Bioinformatical analysis of the specific allergen pTSX1 cDNA clone of Humulus pollen in allergic asthma

Objective To make a bioinformatical analysis of the specific allergen pTSX1 cDNA clone obtained from Humulus pollen in allergic asthma by immunological screening and gene cloning. Methods Bioinformatical approaches, including sequence analysis, sequence alignment, genetic makeup, protein structure and function prediction, were used. Results The analysis of the specific allergen pTSX1 cDNA showed that the gene had no reading frame and coded a protein with 204 amine acids. The analysis of amine acids sequence coded by the pTSX1 cDNA showed that the protein identity of this gene product might be one of ribosomal proteins. Conclusion The pTSX1 cDNA cloned from Humulus pollen in allergic asthma is a new gene, and the protein coded by pTSX1 gene may be one of ribosomal proteins.
8.AChR subunit mRNA expression in human muscles of patients with myasthenia gravis

Objective:To investigate extraocular muscles(EOM) susceptibility to myasthenia gravis(MG).Methods:Acetylcholine receptor(AChR) ?, ?, ?, ? and ? subunits from human EOM and other different muscle groups from 8 health controls and intercostal muscles from 10 MG patients were studied by RT-PCR System. Serum AChR antibodies from 83 patients with MG were assayed by ELISA.Results:①AChR?, ?, ?, ? and ? subunit mRNA could be amplified from normal human EOM and all muscle groups studied. ②AChR? P3A+/?-actin mRNA was expressed at higher level in general MG than in health controls(P=0.028). ③Ocular and general MG patients had higher AChR antibodies against AChR from adult gastrocnemius muscle than that from fetus(P=0.001,0.016).Conclusion:Susceptibility of EOM to MG is likely associated with muscle AChR subunit mRNA expression and intrinsic susceptivity of EOM.
9.Evaluation of vascular function in patients with Fabry disease

Objective:To evaluate the endothelial functions and autoregulation capacity of cerebral blood flow in patients with Fabry disease .Methods:Brachial artery vasodilation was assessed in 8 pa-tients with Fabry disease and 14 healthy controls by means of flow-mediated dilation ( FMD) and Nitro-glycerin-mediated dilation ( NMD) .Cerebrovascular reactivity was calculated in terms of breath-holding index ( BHI) and vascular motor reactivity ( VMR) by TCD-CO2 test in 4 patients and 14 healthy con-trols.Results:Compared with the controls , brachial artery vasodilation experiment showed no difference (the patients:FMD 15.94%±5.03% and NMD 23.92%±7.23%, the controls: FMD 14.57%± 5 .84% and NMD 22 .64%±6 .96%) , there was no relationship between FMD or NMD and the age , course of disease , MSSI or enzyme activity .In respect of cerebrovascular autoregulation capacity , there was no difference in anterior circulation , while cerebrovascular reactivity tended to be impaired in posteri-or circulation .Conclusion:Endothelial function showed no decline in patients with Fabry disease , but cerebrovascular autoregulation capacity tended to be impaired in posterior circulation .
10.Tandem Mass Spectrometry Acquisition Strategy Based on Exclusion of Precursor Ions

Due to the complexity of proteome samples, comprehensive analysis to characterize all proteins was still not possible with present methodologies. It has been shown that replicate runs could increase the number of identified) proteins. However, the redundancy of protein identifications was high. High-abundant peptides tended to be analyzed repeatedly in different runs. To reduce the redundancy and improve the efficiency of identification), we studied the MS/MS acquisition method of linear ion trap Fourier transform ion cyclotron resonance)-mass spectrometry(LTQ-FT) and an acquisition strategy based on exclusion of precursor ions was developed). It proved that the strategy could extremely reduce the redundancy of MS/MS acquisition and improve) the efficiency of protein identifications.
