3.Analysis of antioxidative activity of extract from Arca subcrenata Lischke

Objective To prepare active components from extract of Arca subcrenata Lischke and analyze its antioxidative activity.Methods The antioxidative component(P3)from Arca subcrenata Lischke was isolated by chromatography on Superdex-75 column followed by a Sephadex LH-20 column,and antioxidative activities were assayed using potassium ferricyanide and DPPH methods,respectively.The characters of this component were determined by ninhydrin reagent,anthrone reagent,Coomassie Brilliant Blue G250 reagent as well as thin-layer chromatography.Results and Conclusion A component with strong antioxidative activity was isolated and identified as glycosylated peptide.
4.Dynamic observation on bone mineral density of unsexed rabbits with QCT

Objective: The purpose of this tudy was to dynamicly observe the weight and the bone mineral density (BMD) of the unsexed rabbits with a few self-made standardized phantoms. Methods:The eighteen healthy adult female rabbits were measured for their weight and BMD in preunsexed and postunsexed 5 months, 10 months with quantitative CT(QCT). Results:There were 61.1% of rabbits whose weight and BMD decreased after 5 months of the postunsexed and 100% of rabbits whose weight and BMD decreased after 10 months of the operation. Conclusion:QCT can be used to dynamicly observe curative effect of drugs in various periods as well,and it is a good method to study osteoporosis.
5.Study on the Preparation and in Vitro Dissolubility of Ofloxacin Hollow Suppository

OBJECTIVE:To prepare ofloxacin hollow suppository(OHS) and to observe its in vitro dissolubility.METHODS:OHS was prepared with PEG 6000,PEG 400 and Carbopol-940 as wase material and wax as retardant.The content of ofloxacin in OHS was determined with the first order derivative UV-spectrophotometry.RESULTS:The wax,as a retardant,could retard the release of drug.The in vitro dissolution of OHS revealed the first order dissolution pattern:K0~1=27.81/h,indicating a speedy effect,K2~6=5.94/h,indicating sustained-release effect.CONCLUSION:This preparation is feasible in technology and controllabe in quality.The preparation of OHS extends the kinds of dose-form of ofloxacin

Objective To study the inhibitive effect and mechanism of PPAR?1 on the extracellular matrix (ECM) accumulation of mesangial cells induced by Ang Ⅱ .Methods The plasmid of PPAR?1/WT (wild type) was transfected into mesangial cells. After 48 hours of Ang Ⅱ stimulation, the gene expression of TGF-?1, PAI-1, c-fos and c-jun was examined by RT-PCR. Protein levels of p-ERK, I-?B and nucleus/cytosol ratio of NF-?B were estimated by Westen-blot. The concentrations of FN and TGF-?1 were estimated by ELISA. The activity of PPAR?1 was examined by specific PPRE binding activity. Plasmid expressing non-functional dominant negative type of mPPAR?1, pIRES2-EGFP-mPPAR?1/DN (DN), and blank plasmid, pIRES2-EGFP (Blank) were used as controls. 7.Protective effect and mechanisms of overexpressed PPAR? against fibrosis induced by high glucose in cultured mesangial cells

0 05).Among them,WT but neither DN, Blank, nor Pio significantly ameliorated the increased concentrations of TGF-a1 and fibro nectin in culture medium induced by HG. Compared with DN and Blank, WT transfect ion significantly attenuated high glucose-caused elevation in c-fos, c-jun and p-ERK expression, reduction in Ⅰ-?B level, and incretio n in nucleus/cytosol ratio of NF-?B, while Pio demonstrated similarly signific ant changes only in p-ERK and NF-?B. No significant difference of GLUT-1 mRN A level was detected between HG groups. Conclusions Overexpressed PPAR?1 can su ppress increased ECM production from glomerular mesangial cells cultured in HG c ondition. Its inhibitory effects on activation of ERK/AP-1 and NF-?B pathways are involved. The further increase of PPAR?activity may have direct anti-fibr otic effect in diabetic kidney.
8.Tilling in Rice Breeding

Targeting induced local lesions in genomes (TILLING) is a reverse genetics method for functional genomics research.It is possible to screen for point mutations in the populations of EMS mutagenesis with highthroughput and lowcost. EcoTILLING a method based on TILLING ,was developed for detecting multiple types of polymorphisms in germplasm collections,such as single nucleotide polymorphism,small deletion and insertion etc.Rice is a very important food crop and a model plant for genome research also. There are complete genome sequence and a lot of other bioinformatics resources about it.So the markerassisted breeding is becoming more and more important in rice breeding. Some issues based on TILLING about identifying germplasm based on gene sequence,EMS mutagenesis breeding,developing functional marker in rice breeding in future were discussed.
9.Treatment of 54 cases of primary malignant duodenal tumor.

OBJECTIVETo study the treatment of primary malignant duodenal tumor.
METHODThe data of 54 cases of primary malignant duodenal tumor during 1993 approximately 2003 were analyzed retrospectively.
RESULTSClinical manifestations were jaundice, abdominalgia, obstruction of digest tract and bleeding. Correct diagnosis rates of image examination were endoscopic retrograde cholangiopancreatography 92.8%, air barium double radiography 70.8%, gastroscopy 50.0%, CT 21.9%, MRI 21.4%. Tumor location was 1 in duodenal bulb, 45 in descending portion, 3 in horizontal part and none in ascending portion. 48 malignant tumors were operated, 31 pancreaticoduodenectomy, 1 pancreaticoduodenectomy and partial resection of superior mesenteric vein, 6 radical segmental duodenal resection, 1 palliative segmental duodenal resection, 3 duodenal wedge resection, 5 bypass operation (gastrojejunostomy and/or cholangiojejunostomy), 1 jejunostomy. Adjuvant chemotherapy was given in 13 cases. The survival rates were 5-year 45.4%, 3-year 45.4%, 1-year 63.2%. Median survival months were 24, 10, 38 and 16 respectively for radical operation group, palliative operation group, with postoperative adjuvant therapy group and without postoperative adjuvant therapy group. No significant survival time was found between radical operation group and palliative operation group, adjuvant therapy group and without postoperative adjuvant therapy group, pancreaticoduodenectomy group and radical segmental duodenal resection group in statistics. Among lymphyaden metastasis, tumor size, tumor depth, tumor thrombi, pathologic type and operative methods, only tumor thrombi had prognostic significance in multivariate analysis.
CONCLUSIONSPancreaticoduodenectomy and radical segmental duodenal resection should be selected for primary malignant duodenal tumor. Bypass operation can prolong survival and improve life-quality. Postoperative adjuvant treatment is advocated.

10.Effects of Nourishing Yin Removing Fire Chinese Herbs on Gene Expression of Hypothalamic Ghrelin and its Receptor in Female Precocious Rats.

OBJECTIVETo observe the effect of nourishing yin removing fire Chinese herbs (NYRF-CH) on the gene expression of hypothalamic growth hormone secretion peptide (Ghrelin) and its receptor growth hormone secretion peptide receptor 1alpha (GHSR1-alpha) at the puberty onset of danazol induced female precocious rats.
METHODSForty female SD rats were randomly divided into 4 groups, i.e., the normal group (N), the model group (M), the normal saline intervention group (NS), and the NYRFCH intervention group (NI), 10 in each group. 300 microg danazol was subcutaneously injected to all rats except those in the N group to prepare precocious rat model. NYRFCH and normal saline was respectively administered to rats in the NI and the NS group from the 15th day old for 7-10 days. No treatment was given to rats in the N group. Time of rats' vulva opening was recorded. Ovary index and uterus index were calculated. Peripheral blood levels of estradiol (E2), follicle stimulating hormone (FSH), and luteinizing hormone (LH), and hypothalamic contents of gonadotropin releasing hormone (GnRH) as well as the gene expression of hypothalamic Ghrelin and GHSR1-alpha were determined. Results Compared with the N group, the vulva opening time was advanced in the model group; peripheral blood levels of E2 and LH, uterus index, hypothalamic contents of GnRH increased; peripheral blood FSH levels and mRNA levels of hypothalamic Ghrelin and GHSR1-alpha decreased (P < 0.05, P < 0.01). Compared with the M group and the NS group, the vulva opening time was not advanced in the NI group; peripheral blood levels of E2 and LH, uterus index and hypothalamic contents of GnRH obviously decreased (P < 0.05, P < 0.01); mRNA levels of hypothalamic Ghrelin and GHSR1-alpha increased (all P < 0.01). But there was no statistical difference in the hypothalamic contents of Ghrelin, or the number and activity of GHSR1-alpha (P > 0.05).
CONCLUSIONNYRFCH had regulatory effect on regulating hypothalamic Ghrelin and GHSR1-alpha at gene transcription levels.
