1.Effect of beclin1 on vincristine-induced dopaminergic neurons injury in zebrafish.

To investigate vincristine-induced dopaminergic neurons toxicity and mechanism, and explore the molecular target to reduce the toxicity, zebrafish was chosen as a model animal, based on RT-PCR, Western blotting, whole mount in situ immunofluorescence and other technical means. The results showed that the transcription levels of tyrosine hydroxylase gene and dopamine transporter protein gene were inhibited. Furthermore, the number of dopaminergic neurons was decreased by vincristine. Autophagy was suppressed and beclin1 gene expression was inhibited in a dose-dependent manner by vincristine in larval zebrafish. Up-regulated beclin1 partly reduced vincristine-induced neurotoxicity, and down-regulated beclin1 increased toxicity. Beclin1 plays an important role in vincristine-induced dopaminergic neurons toxicity.

4.Comparison of human cell IFN-β production induced by bluetongue virus dsRNA and polyinosinic polycytidylic acid

Objective To investigate the capability of bluetongue virus(BTV)dsRNA inducing IFN-β from human cells.Methods Artificial complex interfemn inducer polyinosinic-polycytidylic acid (PolyI:C).BTV and BTV dsRNA were added to A549(human lung cancer cell)and HEL(human lung normal cells)culture system in difierent concentrations.IFN-β in culture median was detected by ELISA.Results Though all of the 3 reagents could induce IFN-β,BTV dsRNA significanay induced the highest level of IFN-β.The production of IFN-β was induced by BTV dsRNA in dose dependence.BTV dsRNA induced IFN-β level from HEL Was higher than that from A549(P<0.05).Conclusion BTV dsRNA Can induce IFN-β from human cells effectively,which shows its potential of an endogenous IFN-β inducer.
6.A qualitative research on posttraumatic growth in patients with cervical cancer who had a hysterectomy at the childbearing age

Objective We aimed to explore the experience of posttraumatic growth in patients with cervical cancer who had a hysterectomy at the childbearing age.Methods The phenomenological methodology was used in the study.An in-depth interview was conducted among 12 patients chosen by purposive sampling method.Results Through careful analysis and collation,three themes were identified:smashing the original assumption,searching the meaning of existence and rebuilding the new cognitive schemata.The experience of patients' was conformed to the model which was put forward by Tedeschi,and the theme changes in personal relationships,personal strength,altruism born from sufferings and new philosophy of life were conformed to theoretical connotation of PTG.Conclusions Clinical medical staff should update psychological nursing concept timely and taking advantages of PTG to help patients pass through their difficult period.
7.Study on Quality Standard of Tongluo Tangtai Granules

Objective To establish the quality standard for Tongluo Tangtai Granules. Methods TLC was applied to the qualitative identification of Rhizoma Coptidis, Semen Sinapis Albae and Radix Scrophulariae, and HPLC was applied to the quantitative determination of berberine in the prescription. Results Rhizoma Coptidis, Semen Sinapis Albae and Radix Scrophulariae could be detected by TLC, the spots were clear and there was no interference for the negative sample. A good linearity of berberine was showed in the range of 0.037 29-2.610 3 μg (r=1.000 0). The average recovery of berberine was 98.85%, and RSD was 0.70% (n=6). Conclusion The methods are simple, sensitive, accurate with good reproducibility. It can be used for the quality control of Tongluo Tangtai Granules.
8.Effects of Metformin on Mesenchymal Stem Cells cultured in Adipogenic Medium

Objective To investigate the effect of metformin on proliferation and differentiation of bone marrow mesenchymal stem cells (MSCs) cultured in adipogenie medium. Methods Bone marrow MSCs were isolated from SD rats and cultured in adipogenic medi- um with or without 100?mol/L metformin. MTT test was performed to evaluate the proliferation of cells, while oil red O staining and real time RT - PCR were performed to evaluate the differentiation of cells. Results The number of bone marrow MSCs in two groups both increased over time, and metformin induced more cells. However, the cells affected by metformin showed smaller and less cytoplasmic lipid droplets compared with the cells in control group. Moreover, real time RT - PCR has shown the inhibitory effect of metformin on adipocyte differentiation, with significantly decreased mRNA levels for adipogenic markers. Conclusion Metformin may promote proliferation, but inhibit differentiation of bone marrow MSCs cultured in adipogenic medium.
9.Effects of Levocarnitine on myocardial ischemia in rats

Objective:To study the effect of Levocarnitine preconditionon on the myocardial ultrastructure of ischemic rats and the long last calcium channel. Methods: We randomly divided 30 rats into a control group, a Levocarnitine group and a Dihydrochloride group. After the establishment of ischemic models, we observed the ultrastructure of the ischemic myocardium with optical and transmission electron microscopes, and detected the influx of calcium in the long last calcium channel with the patch clamp. Results: Levocarnitine protected the ultrastructure of ischemic myocardium and inhibited the influx of calcium in the long last calcium channel. Conclusion: Levocarnitine can reduce the severity and extent of ischemia-induced damage to the myocardium, protect mitochondria, stabilize its oxidation, inhibit the long last calcium channel of ischemic ventricular myocytes and lessen the injury induced by calcium overload.
10.Expression of survivin, CDK4, Ki-67 and clinical significance in pediatric acute leukemia.

The expression of Survivin, CDK4 and Ki-67 and the clinical significance in pediatric acute leukemia (AL) were investigated. The expression of Survivin, CDK4 and Ki-67 proteins was detected by using immunohistochemical assay in 37 children with AL and 10 children with normal bone marrow as controls. The positive expression rate of Survivin, CDK4 and Ki-67 was 45.9 %, 56.8 %, and 40.5 % respectively in 37 AL children, which was significantly higher than in control group accordingly (P<0.05). The expression of Survivin was positively correlated with CDK4 (P=0.007) and Ki-67 (P=0.008). In conclusion, all Survivin, CDK4 and Ki-67 proteins are over-expressed in pediatric AL and involved in the modulation of apoptosis and proliferation in pediatric AL.
