1.Correlation of Pregnant Metaphase Serum CRP and Premature Rupture of Membranes and Preterm Birth.

Objective To explore the correlation of pregnant metaphase serum CRP and premature rupture of membranes and preterm birth.Methods Totally 300 cases maternals in pregnant metaphase were sampled to collected their fasting venous blood in the middle of elbow to detected the levels of serum CRP by immunoturbidimetry,and allof the maternals were received follow-up until end of the delivery.Of the 300 cases,there were 16 cases with premature rupture of membranes,28 cases with preterm birth,and 256 cases with full -term birth.The levels of serum CRP in pregnant metaphase of the three groups were compared,and the value of threshold to predict premature rupture of membranes and preterm birth was analyzed.Results The levels of serum CRP in pregnant metaphase of the groups of premature rupture of membranes and preterm birth were hoth higher than the group of full-term birth(P < 0.05),but there were not significant difference between the two groups(P > 0.05),and the threshold of the serum CRP were 13.9,10.8mg/L respectively.The sensitivity of serum CRP in pregnant metaphase to predict premature rupture of membranes was 68.75%,and the specificity was 98.94%.The sensitivity of serum CRP in pregnant metaphaseto predict preterm birth was 75.00%,and the specificity was 98.90%.Conclusion The level of pregnant metaphase serum CRP beyond a certain range may increase the possibility of premature rupture of membranes and preterm birth,which is valuable to predict premature rupture of membranes and preterm birth.
2.Analysis of Prognosis of Children with Non-Hodgkin′s Lymphoma

Objective To analyze the factors affecting prognosis of children with non-Hodgkin′s lymphomas(NHL).Methods Thirty patients of childhood NHL histologically proven in Shanghai children′s medical center from Jan.1996 to Dec.2000 were analyzed.The patients were divided into groups according to some clinical factors that may have predictive significance including age,gender,immunologic phenotype,general system symptom,the size of tumor bulk,bone marrow involvement,mediastinal involvement,hepatomegaly or liver involvement,splenomegaly or spleen involvement and early response to chemotherapy,then the difference of survival rate were compared between groups.Survival analysis was performed by the Kaplan-Meier method,and difference between groups were campared using the Log-rank test.Results The 3-year survival rate was better in patients without giant tumor bulk than that with giant tumor bulk(P0.05).Conclusions Bone marrow involvement and the size of tumor bulk have very significant influence on prognosis of childhood NHL,and splenomegaly or spleen involvement have significant influence on disease-free survival.It can′t be proven in childhood NHL in this study.
3.Transformation of icariin by immobilized β-glucosidase and snailase.

This study was performed to prepare immobilized β-glucosidase and snailase, then optimize and compare the process conditions for conversion of icariin. Immobilized β-glucosidase and snailase were prepared using crosslink-embedding method. The best conditions of the preparation process were optimized by single factor analysis and the properties of immobilized β-glucosidase and snailase were investigated. The reaction conditions including temperature, pH, substrate ratio, substrate concentration, reaction time and reusing times of the conversion of icariin using immobilized β-glucosidase or snailase were optimized. Immobilized β-glucosidase and snailase exhibited better heat stabilities and could remain about 60% activity after storage at 4 degrees C for 4 weeks. The optimized conditions for the conversion of icariin were as follows, the temperature of 50 degrees C, pH of 5.0, enzyme and substrate ratio of 1 : 1, substrate concentration of 0.1 mg x mL(-1), reaction time of 6 h for β-glucosidase and 2 h for snailase, respectively. In 5 experiments, the average conversion ratio of immobilized β-glucosidase and snailase was 70.76% and 74.97%. The results suggest an effect of promoted stabilities, prolonged lifetimes in both β-glucosidase and snailase after immobilization. The immobilized β-glucosidase and snailase exhibited a higher conversion rate and reusability compared to the free β-glucosidase and snailase. Moreover, the conversion rate of immobilized snailase was higher than that of immobilized β-glucosidase. The process of icariin conversion using immobilized β-glucosidase and snailase was moderate and feasible, which suggests that immobilized enzymes may hold a promise for industrial usage.

4.Specific allergic food Intolerance and correlated factors in Qingdao areas

ObjectiveTo study specific allergic IgG antibody test and its impact factors.MethodsA total of 1770 Qingdao residents were recruited in this cross-sectional study from January to October 2011.ELISA was used to test specific allergic IgG antibodies.Logistic regression was used for multifactor analysis.Results Of 1770 participants,9 individuals had insufficient data,resulting in a response rate of 99.5%.Identification rate of specific allergic IgG antibody was 60.0% (1056/1761 ),and the most commonly seen allergic foods were crab (49.4% ),egg (44.80% ),ling (38.0% ),shrimp ( 30.4% ) and soy ( 22.2% ).Intolerance to crab,egg,shrimp,creamery,com,and tomato showed gender difference ( x2 =18.978,P<0.05 ).Serum level of allergic IgG antibody was increased with age [20 - 40 years:( 118.61 ±45.67) U/ml; 41-60 years:(166.57 ±55.82) U/ml; >60 years:(183.67 ±49.34) U/ml; x2=13.597,P<0.01].In logistic regression,difference between individuals with normal body weight and obesity population ( OR =1.897,95% CI 1.215 - 2.578 ) or between those with or without allergic history ( OR =0.780,95% CI 0.648 - 0.912) was found.Conclusions Impact factor of food intolerance includes gender,age,constitution and body mass index.Six kinds of food intolerance are more common in women.41 -60 year group shows peak of food intolerance.No exposure to allergic food and antianaphylaxis may reduce the risk of food intolerance.
5.Comparative study on the symptoms of PTSD and depression of the children exposed to the Wenchuan earthquake 2 and 3 years after it

ObjectiveTo examine the mental health status and its changes among children exposed to the Wenchuan earthquake 2 and 3 years after it has passed,and evaluate the possible influencing factors of mental health.MethodsThe General questionnaire,the self-reported Children's revised impact of event scale(CRIES)and the Depression self-rating scale for children ( DSRSC ) were administered to the same children from the earthquake affected areas 2,3 years after the earthquake.ResultsBoth of the CRIES scores and DSRSC scores had decreased over the 2,3 years periods after the earthquake ( z =- 8.772,P =0.00; z=- 2.870,P =0.00 ).The risk factors associated with PTSD 3 years after the earthquake were:gender (OR =1.069,P =0.00 ),the severity of their family's injury( OR =1.141,P =0.02 ),the length of living in transferred safe places( OR =1.416,P =0.03 ),and whether or not witnessed collapse ( OR =1.014,P =0.01 ).The risk factors associated with depression 3 years after the earthquake were:gender( OR =1.367,P =0.00),age( OR =1.035,P =0.04),and despair( OR =2.490,P =0.01 ).ConclusionBoth of the symptoms of PTSD and depression among the children from earthquake affected areas have decreased over the 2,3 years periods after the earthquake.The mental health status of the children from the earthquake affected areas are influenced by their gender,age,the length of living in transferred safe places,the severity of their family's injury,whether or not witnessed collapse and despair.These factors should be taken into considerations when the long-term post-traumatic psychological interventions are planned.
6.A Novel Monoclonal Antibody With a Mono-specificity for a 46 ku-Cytokeratin

A mAb T2-2 was generated using hybridoma techniques, and its target was identified as a 46 ku-cytokeratin (CK), based on biochemical study and a completely overlapped binding pattern of mAb T2-2 with anti-pan-CKs antibodies. An epithelia-specificity of the mAb T2-2 was determined by screening 68 human normal and 65 tumor tissues using immunohistochemistry. Unlike most of anti-CKs antibodies, the mAb T2-2 recognized a mono-specific epitope only expressed on the 46 ku CK, suggesting that mAb T2-2 is superior to most anti-CKs antibodies that cross-reacted with many different kinds of CKs. In addition, it was found that the mAb T2-2 was multipurpose with a broad applicability to ELISA, immunohistochemistry, immunofluorescence, Western blotting, and was also compatible with various fixation reagents. These results strongly indicate that the mAb T2-2 has potential applications for studying CKs function and for diagnosis of tumor and other disorders.
7.Three-dimensional double echo steady state with water excitation MR imaging of facial nerve at 1.5T: A preliminary study

Objective To assess the value of three-dimensional double echo steady state with water excitation (3D-DESSwe) MR sequence in depicting the normal anatomy of facial nerve. Methods 3D-DESSwe MR sequence was performed in 15 healthy volunteers on a 1.5T MR. The 3D datasets obtained were constructed in relation to the course of the facial nerve using multi-planar reconstruction (MPR) and thin slice maximum intensity projection (MIP) programs, and the images acquired were compared with anatomical textbook diagrams. Two doctors analyzed the images in collaboration and recorded on an arbitrary scale of 0-2 grades (grade 2, homogeneity and facial nerve identified with certainty;grade 1, mild inhomogeneity and facial nerve probably identified;grade 0, severe inhomogeneity and facial nerve not identified). Results In all subjects, 3D-DESSwe images provided reliable definition of the normal facial nerve anatomy bilaterally and high contrast between the facial nerve and adjacent structures were obtained. The cisternal segment of facial nerve showed high signal intensity while the moving CSF flow surrounding it showed low signal intensity. The internal auditory canal (IAC) segment of facial nerve showed relatively low signal intensity while the stationary CSF surrounding it showed high signal intensity. The labyrinthine, tympanic, mastoid and extracranial segments of facial nerve showed high signal intensity while the bone and soft tissue surrounding it showed low signal intensity, and parts of facial nerve branches could be demonstrated. Conclusion 3D-DESSwe sequence can display the complete course of the normal facial nerve, especially for the intratemporal and extracranial parts.
8.Protective effect of micro needle array drug delivery technology on the blood vessel of arteriovenous fistula

Objective To investigate the protective effect of mucopolysaccharide polysulfate cream(MPC) and micro needle array drug delivery technologarray(MNADDT) on the blood vessel of arteriovenous fistula.Methods Eighty cases maintenance hemodialysis patients with autogenous arteriovenous fistula were randomly divided into control group(with the treatment of MPC) and study group(with the treatment of MPC plus MNADDT),40 patients in each group.Both groups were studied for the duration of 12 months.The vascular complications of puncture pain sense,puncture failure times,phlebitis,hardening of the arteries,internal fistula stenosis or embolism,pseudoaneurysm,swollen hands comprehensive syndrome of two groups were observed,and the changes of arteriovenous fistula blood flow,blood vessel diameter by Doppler ultrasound were observed in the two groups for 12 months.Results (1)The incidence of puncture pain(2.71±0.11 vs.2.76±0.14,t=2.21,P<0.05),puncture failure times(38 times vs.73 times,χ2=11.425,P<0.05),phlebitis(2 cases vs.8 cases,χ2=4.341,P<0.05) and blood vessel sclerosis(2 cases vs.9 cases,χ2=5.446,P<0.05) of the treatment group were significantly lower than that of the control group during the study(P<0.05).(2)Blood flow of arteriovenous fistulain of control group before and after treatment were (859.7±148.3) ml/min and (946.5±169.2) ml/min respectively,and the difference was significant(t=2.356,P<0.05).While blood flow of arteriovenous fistulain,cephalic vein diameter,head of venous flow velocity of study group before and after treatment were (824.6±171.5) ml/min and (1 218.1±241.9) ml/min,(5.59±0.74) mm and (6.02±0.57) mm,(94.23±27.35) cm/s and (115.85±36.63) cm/s,respectively,and the differences were significant(t=8.212,2.382,2.877,P<0.05),there were no significantly changes in the above indexes in the study group after treatment(t=5.3612,2.152,2.281,P<0.05).Conclusion MPC plus MNADDT can reduce the vascular complications of arteriovenous fistula,improve blood vessel diameter and increase blood flow.
9.Visualization and analysis of flow patterns of human carotid bifurcation by computational fluid dynamics

Objective To investigate flow patterns at carotid bifurcation in vivo by combining computational fluid dynamics(CFD)and MR angiography imaging.Methods Seven subjects underwent contrast-enhanced MR angiography of carotid artery in Siemens 3.0 T MR.Flow patterns of the carotid artery bifurcation were calculated and visualized by combining MR vascular imaging post-processing and CFD. Results The flow patterns of the carotid bifurcations in 7 subjects were varied with different phases of a cardiac cycle.The turbulent flow and back flow occurred at bifurcation and proximal of internal carotid artery (ICA)and external carotid artery(ECA),their occurrence and conformation were varied with different phase of a cardiac cycle.The turbulent flow and back flow faded out quickly when the blood flow to the distal of ICA and ECA.Conclusion CFD combined with MR angiography can be utilized to visualize the cyclical change of flow patterns of carotid bifurcation with different phases of a cardiac cycle.
10.Effect of terpene penetration enhancer and its mechanisms on membrane fluidity and potential of HaCaT keratinocytes.

The aim of this paper was to investigate the effect of terpene penetration enhancers on membrane fluidity and membrane potential using HaCaT keratinocytes, and study the potential mechanisms of these terpene compounds using as natural transdermal penetration enhancer. Six terpene compounds, namely menthol, limonene, 1,8-cineole, menthone, terpinen-4-ol and pulegone, were chosen in this study on account of their good penetration-enhancement activities. The cytotoxicity of these terpene compounds was measured using an MTT assay. The fluorescence recovery after photobleaching (FRAP) technique was employed to measure the change of membrane fluidity of HaCaT cells. The flow cytometer was used to study the alteration of membrane fluidity of HaCaT cells, and investigate the effect of terpene compounds on intracellular Ca2+. It was found that 6 terpene compounds possessed low cytotoxicity in comparison to the well-established and standard penetration enhancer azone. Those terpene compounds could significantly enhance HaCaT cells membrane fluidity and decrease HaCaT cells membrane potentials. Meanwhile, after treated with various terpene compounds, the Ca2(+)-ATPase activity and intracellular Ca2+ of HaCaT cells was decreased significantly. Terpene penetration enhancers perhaps changed the membrane fluidity and potentials of HaCaT cells by altering the Ca2+ balance of the cell inside and outside, resulting in the low skin permeability to increase the drug transdermal absorption.
