1.Analysis of efficacy and prognostic factors of postoperative intensity-modulated radiotherapy with or without chemotherapy in rectal cancer

Objective To explore the efficacy and prognostic factors of postoperative intensity?modulated radiotherapy ( IMRT) with or without chemotherapy in rectal cancer. Methods A retrospective analysis was performed on the clinical data of 218 patients with rectal cancer, who underwent postoperative IMRT in our hospital from January 2009 to December 2013. The Kaplan?Meier method was used to calculate survival rate;the log?rank test was used for survival difference analysis and univariate prognostic analysis;the Cox regression model was used for multivariate prognostic analysis. Results The follow?up rate was 97. 7%. The 1?and 3?year overall survival rates were 90. 8% and 75. 2%, respectively, the 1?and 3?year disease?free survival rates were 85. 3% and 70. 5%, respectively, and the 1?and 3?year locoregional recurrence?free survival rates were 96. 7% and 88. 1%, respectively. The incidence of grade 3?4 acute adverse reactions was 28. 4%, mainly manifested as leukopenia ( 13. 8%) and diarrhea ( 11. 0%) . Univariate prognostic analysis showed that preoperative carcinoembryonic antigen ( CEA) and CA199 levels, maximum tumor diameter, tumor location, degree of differentiation, depth of tumor invasion, number of lymph node metastases, TNM stage, perineural invasion, surgical procedure, total mesorectal excision, preoperative bowel obstruction, and preoperative anemia were the predictors of survival ( P=0. 006, 0. 000, 0. 000, 0. 017, 0. 000, 0. 016, 0. 000,0. 011,0. 001,0. 006,0. 037 and 0. 010) . Multivariate prognostic analysis showed that preoperative CEA level, tumor location, TNM stage, preoperative bowel obstruction, and preoperative anemia were the predictors of survival ( P=0. 000,0. 000,0. 000,0. 001 and 0. 001) . Conclusions Postoperative IMRT with or without chemotherapy is an effective method for rectal cancer with mild adverse reactions and high compliance. Preoperative CEA level, tumor position, TNM stage, preoperative bowel obstruction, and preoperative anemia are independent prognostic factors for the overall survival.
2.An Electrochemical Biosensor for Detection of Breast Cancer Cells Based on Exonuclease I-catalyzed DNA Degradation Reaction

In terms of the characteristic of exonuclease I that can selectively catalyze the degradation of single-stranded DNA, we report a new sensitive electrochemical biosensor for the detection of breast cancer cells by using the aptamers of MUC1 that is overexpressed on the surface of MCF-7 cells. Because the binding of aptamer and MUC1 that is over-expressed on the surface of breast cancer cell MCF-7 can prevent its hybridization with the complementary probe DNA, the unhybridized single-stranded probe DNA that is immobilized on the electrode surface will be selectively digested by Exonuclease I and result in the removal of the signal molecule methylene blue. Therefore, by tracing the changes in electrochemical responses, the biosensor can be used to sensitively detect MCF-7 cells in a linear range from 103 to 106 cell/mL with a detection limit of 330 cell/mL, and easily distinguish the control islet β cells with high specificity.
3.Clinical efficacy of treating pains suffered from metastatic bone cancer with composite kushen injection and pamidronate disodium injection

Objective To explore the clinical efficacy of treating pains suffered from metastatic bone cancer with composite kushen injection and pamidronate disodium injection.Methods The clinical information of 60 cases of metastatic bone cancer patients suffered with pains was collected retrospectively.Thirty patients were assigned to the treatment group and 30 to the control group according to the treatment they underwent.The control group were treated with pamidronate disodium injection for 3 cycle,the treatment group were additionaly treated with composite kushen injection.The differences of two groups cases were compared in respect of the relief of pains and the changes of performance status (PS) and the incidence of adverse effects after treatment for 3 cycles.Results The objective remission rate of bone pain was 60.0% (18/30) in the treatment group,which was significantly higher than that of 30.0% (9/30) in the control group was higher( x2 =5.455,P=0.020 ).The incidence of adverse effect was 40.0% (12/30) in the treatment group and 46.6% (14/30) in the control group,with no significant difference between the two groups( x2 =0.271,P =0.602).In the treatment group the performance status of patients was( 2.30 ± 0.70 ) after treatment,which was better than that of( 1.80 ± 0.80 )before treatment(t =15.000,P =0.042),wheras there was no significant difference on performance status in the control group.Conclusion Kushen injection has synergistic effect with pamidronate disodium injection in treating pains with matstatic bone cancer.It could improve the short term efficacy,and significantly relief the pain and improve the quality of life.
4.Serum uric acid level and related clinical features in high risk syndrome of neuromyelitis optica

Objective To investigate serum uric acid (UA) levels and related clinical features in patients with high risk syndrome of neuromyelitis optica. Methods UA levels were measured in 51 patients with high risk syndrome of neuromyelitis optica including 34 with longitudinally extensive transverse myelitis (LETM) and 17 with optic neuritis (ON), 48 with neuromyelitis optica (NMO), 45 with other neurological diseases (OND) and 65 with healthy controls (HC). The disability severity was assessed by the expanded disability status scale (EDSS). Spinal lesions were viewed by MRI. Serum aquaporin-4(AQP4) antibody was tested in cell based immunofluorescence assay. Results Serum UA levels in LETM ( ( 189. 84 ±85. 65) μmol/L) and ON patients ( (222. 12 ±61.68) μmol/L) were significantly lower than that in OND ((315.90±71.36) μ mol/L) and HC ((291.05 ±76.64) μ mol/L) subjects (P<0.01). No difference was found between LETM, ON and NMO groups. UA levels were significantly lower in females ( ( 158.24 ±55.92), (187.00±47.52), (198.21 ±62.62), (274.51 ±70.66)and (243.26±60.65) μmol/L)than in males ( ( 262. 09 ± 101.63 ), ( 262. 45 ± 62. 13 ), ( 298.90 ± 74. 14 ), ( 355.37 ± 50. 30 ) and (340. 34 ±58. 23) μmol/L) in all groups (t=3. 183, 2.578, 4.356, 4.365 and 6.579, all P<0.05).UA levels in patients with high risk syndrome of NMO were not correlated with mono or relapse course,duration or status of serum AQP4 antibody. UA were negatively correlated with EDSS in patients with LETM (r= -0.714, P<0.01). Conclusion Lower serum UA levels were found in patients with high risk syndrome of NMO and related to more severe symptoms in LETM group.
5.An evaluation of the effectiveness of prevention and control measures on coal-burning-borne endemic fluorosis in Guizhou Province

Objective To evaluate the effects of health education and improved stoves on control of coalburning-borne endemic fluorosis in Guizhou Province.Methods In 2014,according to the detectable rate of dental fluorosis of children aged 8-12 before implementing control measures,11 counties were divided into three layers:low detection rate (detection rate was less than 60%),medium detection rate (detection rate from 60% to 90%) and high detection rate (detection rate was greater than 90%).One county was selected from each layer and two towns were selected in each county.According to the Diagnosis of Dental Fluorosis (WS/T 208-2011),dental fluorosis of all children aged 8-12 was examined in all towns;At the same time,in the 2 villages in each town,20households and students of one class in grade 4 to 6 were selected to survey knowledge about prevention and control of coal-burning-borne endemic fluorosis;and to survey the situation of improved stoves,drying and elutriation of corn and chillies,and related life behavior.Results The detectable rate of dental fluorosis of children aged 8-12was 5.49% (198/3 607),8.33% (72/864),12.46% (179/1 437),and it was significantly lower than before implementing control measures [41.88% (931/2 223),61.91% (4 863/7 855),91.25%(2 439/2 673),x2 =1 166.43,909.56,2 508.96,all P < 0.01].Correct answer rates of students about knowledge on endemic fluorosis were 78.03% (792/1 015),84.93% (1 465/1 725) and 80.18% (910/1 135),the correct answer rates of knowledge were significantly higher than those before implementing control measures [23.19% (640/2 760),47.41% (2 368/4 995) and 40.25% (3 405/8 460),x2 =947.96,736.61,644.69,all P < 0.01].Correct answer rates of family householders about knowledge on endemic fluorosis were 86.75% 047/400),95.25% (381/400) and 86.25% (345/400),the correct answer rates of knowledge were higher than those before implementing control measures [17.78% (80/450),37.20% (279/750) and 25.27% (139/550);x2 =402.97,359.49,344.55,all P < 0.01].The percentages of households whose stoves were qualified and can be used correctly to the total households were 96.25% (77/80),96.25% (77/80) and 93.75% (75/80),and before implementing control measures [4.92% (348/7 078),21.51% (7 461/34 693) and 6.73% (475/7 062)],and the differences were statistically significant (x2 =1 181.55,262.63,842.82,all P < 0.01).The percentages of households using clean energy in heating season to the total households increased from 0 (0/7 078),0 (0/34 693),0(0/7 062) to 98.75% (79/80),98.75% (79/80),96.25% (77/80).The rates of the three counties whose inhabitants could bake the chillies correctly were 96.25% (77/80),95.00% (76/80) and 97.50% (78/80),which were significantly higher than before implementing control measures [5.55%(5/90),26.00% (39/150),52.73% (58/110),x2=139.52,99.36,45.64,all P < 0.01].Who cooked chillies after washing were 93.75% (75/80),95.00% (76/80),93.75% (75/80),which were significantly higher than before implementing control measures [74.44% (67/90),87.33% (131/150),34.55% (38/110),x2 =11.47,3.91,67.36,all P < 0.05].Conclusion The effects of health education and improving stoves on coal-burning-borne endemic fluorosis in the three counties of Guizhou Province are remarkable.
6.Curative effect of Ganciclovir combined with interferon-α1b inhalation in treating children with infectious mononucleosis

Objective To investigate the clinical efficacy and safety of Ganciclovir combined with interferon-α1 b inhalation for children with infectious mononucleosis(IM).Methods A total of 177 childhood cases of IM were selected,and they were divided into 3 groups,59 cases in each group according to the random number table.Three therapeutic methods were applied in different groups for 5-7 days in different groups:Ganciclovir (group A),Ganciclovir + interferon-α1 b inhalation (group B) and Ganciclovir + interferon-α1b intramuscularly (group C).The time of post-drug recovery from isthmitis,less than 0.05 of heterotypic lymphocytes,shrink of cervical lymph nodes shrink,liver retraction,spleen retraction among groups were compared.The Epstein-Barr virus (EBV)-DNA copy number and T lymphocyte subsets were compared before and after treatment.Adverse reactions were observed in each group.Results Compared with group A,the time to defervescence [(3.20 ± 1.81) d,(3.17 ± 1.76) d vs.(4.01 ± 2.34) d],duration of isthmitis was [(3.15 ± 1.33) d,(3.09 ± 1.37) d vs.(3.98 ± 1.31) d],and the time of heterotypic lymphocytes less than 0.05 [(3.12 ± 1.55) d,(3.10 ± 1.33) d vs.(3.95 ± 1.26) d] in group B and group C,were obvious shorter,and there were significant differences(F =4.150,4.580,4.060,all P < 0.05).EBV-DNA negative conversion rate of group B and group C were higher than that of group A [53 cases(89.8%),52 cases (88.1%) vs.41 cases (69.5%),x2 =10.403,P < 0.05],and the cellular immune function was improved significantly than that of group A after treatment for 7 days [CD3 +:(63.00 ±4.39)%,(62.75 ±4.84)% vs.(68.70 ± 7.70)%;CD4+:34.08(30.21,41.70)%,33.94(29.17,45.17)% vs.32.34(28.16,43.53)%;CD8+:30.59 (27.14,40.22)%,30.09(27.54,40.48)% vs.32.57(28.68,41.17)%;CD4+/CD8+:1.12(1.03,1.31),1.11 (0.99,1.64) vs.0.94 (0.87,1.59),F/x2 =11.020,1.217,1.121,6.728,all P < 0.05].The differences in indexes between B group and C group were not significant,and there was no statistical significance (all P > 0.05).There were 2 cases with fever in the group C,and 2 cases of granulocytopenia in all group.Conclusions Ganciclovir combined with interferon-α1 b inhalation or intramuscular injection is effective and safe in treating children with IM.It can improve clinical symptoms,cellular immune function and EBV-DNA negative conversion rate.Since inhalation is of less side effects and no pain,it can be accepted by children and their parents easily.Therefore,it is recommended that Ganciclovir be used together with interferon-α1 b inhalation in the treatment of children with IM.

Objective To investigate the effects of total flavonoids in Gingko Biloba on glucose and lipid metabolism and liver function in rats with insulin resistance. Methods Forty SD rats were randomly divided into normal control group,model group,total flavonoids in Gingko Biloba group and rosiglitazone group(positive drug control group)(n=10).Models of insulin resistance were established by high glucose and high fat diet in model group,total flavonoids in Gingko Biloba group and rosiglitazone group.After treatment for 12 weeks,serum glucose,serum lipids,and parameters of insulin resistance,liver function and anti-oxidation capability were detected in each group,and histologic observations of liver tissues were conducted with adipose staining. 8.Denervation effects on bone regeneration and expression of Runx2 during distraction osteogenesis

BACKGROUND:During the healing of fractures, removal of sciatic nerve can result in insufficient mechanical rigidity of newborn woven bone. However, there are less reports concerning the denervation effects during distraction osteogenesis. OBJECTIVE:To observe the effect of removal of the sciatic nerve on bone regeneration and the expression of Runt-related transcription factor 2 (Runx2) protein during distraction osteogenesis in a rabbit model. METHODS:Twenty-four adult male New Zealand rabbits were selected and underwent left tibial osteodistraction to construct animal models of distraction osteogenesis. Before distraction, the animals were randomly divided into group R (resecting the left sciatic nerve) and group I (intact left sciatic nerve). Six weeks after completion of distraction, the animals were kil ed and the lengthened tibias were harvested for radiography, three-dimensional CT reconstruction, histological evaluation, connectivity density (Conn.D) evaluation. RESULTS AND CONCLUSION:New regenerated bone was present and Runx2 protein was expressed in the distraction gaps of al animals at the end of the study, as revealed by radiography, three-dimensional CT reconstruction, and histological observation. However, less new bone formation and a lower degree of mineralization and expression of Runx2 protein were observed in group R compared with group I. The results suggest that the denervation appears to have an inhibitory effect on bone formation and the expression of Runx2 protein during distraction osteogenesis.
9.Gene Mutation of RANTES and CCR5 in Systemic Lupus Erythematosus

Objective To evaluate the gene mutation of RANTES and CCR5 in SLE and its significance. Methods One hundred and forty-six definitive SLE patients and 159 controls were collected. SNPs of RANTES promoter and polymorphism of CCR5 were performed by PCR or PCR/RFLP assay, and further confirmed by direct DNA sequencing. Results The frequence of RANTES-403G/G compounded with 28C/C and CCR5/CCR5 was significantly different between SLE and control groups (72.6% vs 58.5%, P 0.05). Conclusions These results indicate that the two SNPs are linkage disequilibrium. Interaction of two SNPs in RANTES and CCR5 is related with SLE. RANTES-403G/G compounded with 28C/C and CCR5/CCR5 may be one of risk factors of SLE. RANTES-403A is probably related with renal damage of SLE.
10.Serum uric acid level and related clinical features in neuromyelitis optica

Objective To investigate serum uric acid (UA) levels and related clinical characteristics of neuromyelitis optica (NMO). Methods The serum uric acid levels were measured in 65 patients with NMO, compared to control groups which were 76 cases with multiple sclerosis ( MS), 126 cases with cerebral vascular diseases (CVD) and 130 healthy controls(HC). The disability severity in NMO was assessed by the Expanded Disability Status Scale (EDSS). Magnetic resonance imaging ( MRI ) was performed to strengthen assessment the involved lesions. Serum AQP4 antibody was tested in a cell based immunofluorescence assay. Results In male groups, serum UA levels in NMO patients [ (298.90±74.14) μmol/L] were significantly lower than that in CVD [ (355.37 ±50. 30) μmol/L] and HC subjects [ (340.33 ± 58.23 ) μmol/L, P < 0.05 ]. No difference was found between NMO and MS [ ( 292.36 ±92.95) μmol/L] groups. In female groups, serum UA levels in NMO patients [(198.21 ± 62.62)μmol/L] were significantly lower than that in CVD [(274.51 ± 70.66) μmol/L] and HC subjects [(243.26 ±60.65) μmol/L,P <0.05]. No difference was found between NMO and MS [(232.29 ±71.95 ) μmol/L ] groups. UA levels were significantly lower in females [ ( 198.21 ± 62. 62) μ mol/L] than in males [ (298.90 ±74.14) μmol/L]. UA levels were significantly lower in patients with EDSS≥5 [ ( 195.48 ± 83.70 )μmol/L] than EDSS < 5 [ (241.00 ± 63.20)μmol/L] NMO patients. In our study UA levels were not correlated with longitude of spinal lesions, activity revealed by MRI and AQP4 antibody tires.Conclusion Lower serum UA levels were found in patients with NMO and related to more severe symptoms.
