1.The detection of expression and significance of TGase3 mRNA in esophageal carcinoma by tissue microarray and in situ hybridization methods

Objective To study the expression of TGase3 mRNA in esophageal carcinoma and to investigate it's role and the significance of in the carcinogenesis and progression of esophageal carcinoma.Methods The expression of TGase3 mRNA in 70 ca8es of esophageal carcinoma tissue and 10 cases of normal esophageal tissue were detected by tissue miemarray(TMA)and in situ hybridization(ISH)methods.Results The loss expression rate of TGase3mRNA in esophageal carcinoma tissue was 68.57%(48/70),which was significantly higher than in normal control(P<0.05).The expression of TGase3 mRNA was no correlation with patients'sex,age,tumor site,lymph node metastasis.But it's significant correlation with clinical stage,histology grade,Pathology grade(P<0.05).Conclusion There was a high frequency of TGase3 mRNA expression loss in esophageal carcinoma,TGase3 mRNA gene may be involved in carcinogenesis and progression of esophageal carcinoma.
2.Clinical Study on Xianglian Mixture in Treatment of Premature Beat Accompanied Anxiety Disorder

Objective To study the clinical effect of Xianglian in Mixture (Mixture of Nutgrass Galingale Rhizome and Golden Thread) in treatment of premature beat accompanied anxiety disorder.Methods Totally 100 patients of premature beat accompanied anxiety disorder were randomly assigned into two groups.The 50 patients in the treatment group were treated with Xianglian Mixture and the 50 in the control group were treated with Rytmonorm and Alprazolam for 4 weeks.The TCM symptom scores,premature beats in Holter,scores of Hamilton anxiety scale (HAMA) and the therapeutic effect on all these indices of both groups were observed.Results After treatment,the TCM symptom scores,premature beats in Holter,scores of HAMA of both groups were decreased significantly (P0.05).Conclusion Xianglian Mixture can reduce premature beats and relieve anxiety as well.
3.Thalidomide inhibits the expression of hypoxia-inducible factor-1?in synovial tissues of rats with collagen-induced arthritis

Objective To observe the therapeutic effect of thalidomide(Thd)on rheumatoid arthritis and its relation with the expression of hypoxia-inducible factor-1?(HIF-1?).Methods Female SD rats were randomly divided into two groups:a normal group and a model group.Each group was further divided into three subgroups:the control group,the vehicle group and the Thd group.The model group was subjected to collagen-induced arthritis.The Thd group and the vehicle group were given Thd(600 mg/kg)once daily and its vehicle DMSO respectively.The profile of arthritis was assessed by observing the clinical symptoms and histopathology and a qualitative clinical score of arthritis.The expression of HIF-1?in synovial tissuses was further detected by Western blot and immunohistochemistry technics.Results Compared to the control group with arthritis,clinical signs and pathological changes of arthritis were improved one and two weeks after ad- ministration of Thd respectively,and the qualitative clinical score of arthritis was evidently lowered.The ex- pression of HIF-1?in the inflammed synovial tissuses was found to be enhanced(P
4.The neural mechanisms of voluntary control of saccadic eye movements

Voluntary control of saccadic eye movements is a kind of saccades controlled by conscious and always used to investigate the brain areas related to saccades and ability of response inhibition and work memory.Researching the neural mechanisms of voluntary control of saccadic eye movements could provide a basis to study the advanced cognitive function of brain and facilitate to explore basic neural mechanisms of abnormal saccadic in brain disorders.As neuroimaging technologies develops,intensive studies about the neural mechanisms of voluntary control of saccadic eye movements have been made great progress.It is showed that several brain areas such as prefrontal cortex,subcortical areas and parietal cortex mainly in participate.These multiple brain regions are involved in voluntary control of saccadic eye movements at different stages.Antisaccades and memory-guided saccades including different brain regions because of their different characteristics of saccadic.The future direction of the research is to combine study with neural imaging technology,and to study more about the brain regions and their functional connectivity involved in voluntary control of saccadic eye movements.

6.The neurophysiological features of shoulder-hand syndrome after stroke

Objective T_O analyze the electrophysiological features of shoulder-hand syndrome(SHS)fol- lowing stroke and investigate the relationships between peripheral nerve damage and the factors causing SHS.Meth- ods Fifty-eight stroke patients were divided into an SHS group(39 patients with shoulder-hand syndrome)and a model group(19 patients without shoulder-hand syndrome).Standard sensory and motor nerve conduction studies were performed with 58 of the patients,including sensory nerve conduction velocity(SCV),amplitude of the sensory nerve action potential(SNAP),distal motor latency(DMI)and the amplitude of the compound muscle action poten- tial(CMAP)of the median nerve.A needle electromyogram(EMG)test was performed on the abductor pollicis of the affected side with both groups.Results The needle EMG showed abnormal insertion potential,fibrillation po- tential and positive sharp waves in all 39 cases of the SHS group,which was significantly higher than in the model group.The amplitude of the sensory nerve action potential(7.77?4.34 mV)and the amplitude of the compound muscle action potential(10.13?3.15 mV)were significantly lower than those of the model group.Abnormal ampli- tude was more severe in sensory nerves than in motor nerves.Conclusion Peripheral nerve damage was found in the shoulder-hand syndrome patients,and this damage was mainly the dystrophy of axonal neuropathy.The damage severity was more in sensory nerves than in motor nerves.The study could offer an useful clue of prevention and treat- ment of shoulder-hand syndrome.
7.Detection and analysis of alkaline phosphatase and isozymes in urine and renal disease

Objective To investigate the correlation of alkal ine phosphatase(ALP) and isozymes with renal disease.Methods In 1997-03~2003-05 the activity of ALP was determined b y biochemical automatic analyzer in Heilongjiang Province Hospital.The activity of ALP isozymes were determined by polyacrylamide disc gel electrophoresis and s canning measurement.Results The samples from patients with renal disease were divid ed into glomerular injury group,renal tubular injury group and severe widespread damage group based on the pathological changes.The activity of ALPs in urine i n the glomerular injury group and renal tubular injury group were significantly different from those in the control group (P0.05).Conclusion Isozymes of ALP in urine can be a useful marker for different renal segments injury.
8.Cost-effectiveness Analysis of Two Chemotherapy Regimens in the Treatment of Initially Diagnosed Multiple Myeloma

OBJECTIVE:To evaluate the cost and effectiveness of two chemotherapy regimens in the treatment of initially di-agnosed multiple myeloma (MM). METHODS:63 patients who were newly diagnosed as MM without transplant chance in our hospital were analyzed retrospectively,and then divided into MPT group (alkeran+metacortandracin+thalidomide,41 cases) and VDT group (bortezomib +dexamethasone+thalidomide,22 cases) according to chemotherapy regimens. Clinical efficacy,survival time and ADR of 2 groups were observed,and cost-effectiveness analysis of them were conducted. RESULTS:The overall re-sponse rates of MPT group and VDT group were 63.4% and 77.3%,respectively(P=0.260);the complete response rate of them were 12.2% and 36.4%,with statistical difference(P=0.024). The progression free survival of MPT group and VDT group were (14.314±0.488)months and(18.557±0.848)months,with statistical significance(P<0.001);the overall survival of MPT group and VDT group were(33.536±1.143)months and(41.048±1.868)months,with statistical significance(P=0.007). Leukocytope-nia,nausea and vomit and peripheral neurotoxicity of VDT group were both higher than those of MPT group,with statistical signif-icance (P<0.05). The cost-effectiveness ratio of MPT group was 247.48,and that of VDT group was 2 922.77;the incremental cost-effectiveness ratio was 15 125.18. The results were consistent with the sensitivity test. CONCLUSIONS:Clinical efficacy and survival time of VDT regimen is better than that of MPT regimen,but MPT regimen is superior to VDT regimen in respect of cost-effectiveness analysis and the incidence of ADR. which will more suitable for the MM patients without transplant chance or goood economic conditions.
9.Application of peer education in guiding community diabetic patients to adopt diet therapy using food exchange method

Objective To use peer education to guide community diabetic patients to adopt diet therapy using food exchange method in order to explore effective method for diet therapy of diabetes mellitus. Meth-ods 91 diabetic patients were selected using simple sampling method and randomly divided into the peer ed-ucation(45 cases) and the common education group(46 cases), the two groups adopted food exchange method for diet therapy for 1 year. The blood glucose, blood lipid, glycosylated hemoglobin and body weight index were compared between the two groups using t test. Results The blood glucose, blood lipid, glycosylated hemoglobin and body weight index all decreased in the two groups after intervention, but the the decreased de-gree in the peer education group was more obvious. The fasting blood glucose, 2 hour postprandial blood glu-cose, glycosylated hemoglobin, triglyceride, low-donordensity lipid protein and body weight index in the peer education were lower thanthose of the common education group, but HDL-C was higher. Conclusions Peer education is more effective in health education of food exchange method.
10.Targeted damage of the cerebrospinal fluid-contacting nucleus contributes to the pain behavior and the expression of 5-HT and c-Fos in spinal dorsal horn of rats.

OBJECTIVEThe changes of pain threshold and expression of 5-hydroxytryptamine(5-HT) and c-Fos in spinal dorsal horn of rats were observed after targetedly damaged the cerebraspinal fluid-contacting nucleus (CSF-contacting nucleus) to provide experimental evidence for the mechanism of regulating pain CSF-contacting nucleus involved in.
METHODSMale adult SD rats were divided into control, sham, choleratoxin subunit B conjugated with horse-radish peroxidase (CB-HRP)and damage groups randomly. The pain threshold using mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were recorded and analyzed. Immunofluorescence method was used to observe the expression of 5-HT and c-Fos in spinal dorsal horn.
RESULTSCompared with the control, sham and CB-HRP groups, the MWT and TWL of the damage group were significantly increased (P < 0.05). The results of immunofluorescence showed that 5-HT was detected in neurons of CSF-contacting nucleus. In the damage group, the number of neurons of CSF-contacting nucleus reduced gradually, and no survived neurons were observed at the 10th day. Meanwhile, both the expression of 5-HT and c-Fos in spinal dorsal horn increased gradually, and negatively correlated with the change of pain threshold.
CONCLUSIONThe method of targeted damaging CSF-contacting nucleus by cholera toxin subnit B conjugated with saporin(CB-SAP) is scientific and reliable, and it results in the changes of pain threshold and expression of 5-HT and c-Fos in spinal dorsal horn of rats. This study suggests that CSF-contacting nucleus participate in the regulation of pain, moreover, 5-HT and c-Fos play important roles in this regulation.
