1.Efficacy and Pharmacoeconomics Evaluation of Piperacillin Sodium/Sulbactam Sodium in the Treatment of Bacterial Infections

Objective:To observe the effectiveness and security of piperacillin sodium/sulbactam sodium in the treatment of bacte-rial infections and perform the pharmacoeconomics evaluation. Methods: Totally 126 patients with hand trauma bacterial infections were divided into the control group and the treatment group. The control group was given piperacillin sodium/tazobactam sodium 2. 5g, ivd, bid, while the treatment group was treated with piperacillin sodium/sulbactam sodium 1. 25g, ivd, bid. After 14-day treatment, the clinical therapeutic effect and adverse reactions of the two groups were observed, and the cost-minimization analysis was carried out. Results:The effective rate of the treatment group was 93. 65%while that of the control group was 92. 06%(P>0. 05). The bac-terial clearance rate of the treatment group was 96. 83%while that of the control group was 95. 24% (P>0. 05). The incidence of ad-verse reactions of the treatment group was 4. 76% (3/63) while that of the control group was 9. 52%(P>0. 05). The cost-effective-ness of piperacillin/sulbactam was significantly lower than that of the control group (P<0. 05). Conclusion: The effectiveness and safety of the two groups show no obvious differences in the treatment of hand trauma bacterial infections, and both have good antibacte-rial effect with low incidence of adverse reactions. For the better economic benefits, piperacillin /sulbactam is worthy of promoted ap-plication.
2.CHANGES OF PLASMA IMMUNOREACTIVE ?-ENDORPHIN LEVEL IN NIDDM AND ITS RELATIONSHIP WITH CHRONIC COMPLICATIONS

Plasma concentrations of immunoreactivc beta-endorphin in patients with NIDDM were measured by radioimmuno-assay. It was found that the plasma beta-endorphin levels decreased in 68 cases of NIDDM especially in those with long duration of illness and chronic complication. These results suggested that beta-endorphin may be related to diabetes and its chronic complication.
3.Imaging appearances of primary hepatic and gallbladder neuroendocrine carcinomas

Objective To review the imaging appearances and clinical symptoms of primary hepatic and gallbladder neuroendocrine carcinomas (PHGNC). Methods Five patients (4 females and 1 male) were examined. Long-term diarrhea not controlled by drug was found in four patients, and two of them accompanied by abdominal pain. The liver rupture occurred in one case due to the huge tumor bleeding. Five patients were examined by using ultrasonography (US), four patients by CT, and three patients by hepatic angiography. The tumor originated from liver in three cases, from liver and gallbladder in one case, and from gallbladder in one case. The tumors were diagnosed by surgery in 3 cases, by biopsy and by autopsy in 1 case, respectively. Results Multiple hyperechoic inhomogeneous masses in two cases and hypoechoic inhomogeneous masses in one case were revealed, all with some small fluid areas and abundant vessels. A huge cyst-solid tumor was clearly demonstrated by using US. A small mural nodule along the gallbladder wall was displayed by using US in a gallbladder tumor, and it showed no specific sign in the diagnosis. CT findings included hypodense inhomogeneous masses on plain CT scan, and slightly enhanced masses with some small fluid areas after injection of contrast agent. A huge cyst-solid tumor was clearly displayed by CT due to the hemorrhage, necrosis, and cystic changes in the tumor. Hepatoangiography displayed abundant vessels and stain in the solid part of the tumor, and absent vessel in the cystic region of the cyst-solid tumor accompanied by dislocation of surrounding vessels. Conclusion Imaging appearances of the liver tumor were inhomogeneous mass with abundant vessels and tumor stains. Imaging appearances of the gallbladder tumor were raised mural nodule along the gallbladder wall without specific sign in the diagnosis of the tumor. The tumor may develop hemorrhage, necrosis, and cystic changes if it is large enough. PHGNC was prone to hepatic metastases. The patients often presented with diarrhea and abdominal pain.
5.Effects of chronic compound stress on serum adiponectin level and adipoR2 expression in rats

Objective To explore the effect of chronic compound stress on serum adiponectin level and AdipoR2 expression in rats.Methods Forty male Sprague-Dawley rats were randomly divided into two groups:the stress group underwent chronic compound stress and the control group was fed normally.After 8 weeks of stress or feeding,the serum adiponectin,blood glucose ,insulin resistance and HbA1 C,HDL-C,LDL-C,triglyceride and total cholesterol were measured.Results ①After 8 weeks of stress,the score of open-field behaviors and 24-h urocortisol were significantly different between the stress and control groups (32.0 ± 8.6 vs.52.0 ± 12.7,5.8 ± 0.4 vs.(5.3 ±0.1 ) ng/L,repectively; P <0.05 or 0.01 ) ;②After 8 weeks of stress,the adiponectin,total cholesterol ,triglyceride,HDL-C,InIR and HbA1 C were significantly different between the two groups (P < 0.05 or 0.01 ) ;③After 8 weeks of stress,there were significant difference on the expression of AdipoR2mRNA and protein between the two groups (P <0.01 ) ,with the expression of AdipoR2mRNA of 0.67 ±0.04 and 1.00 ±0.11 and the expression of protein of 0.73±0.02 and 1.00 ± 0.04 in the stress and control group,respectively.Adiponectin was negatively related with HbA1 C,LDL-C and IR ( r = - 0.26,- 0.88 and - 0.37,P < 0.05 or 0.01 ),and positively related with HDL-C ( r =0.78 ,P <0.01 ).Conclusions In rats,chronic compound stress decreases the level of serum adiponectin and the expression of AdipoR2 in liver.
6.The diagnosis value of TI-RADS and sonographic characteristics for papillary thyroid carcinoma and follicular thyroid carcinoma

Objective To investigate the value of TI-RADS for sonographic diagnosis of papillary thyroid carcinoma (PTC) and follicular thyroid carcinoma (FTC) and to evaluate the sonographic findings of FTC compared to PTC.Methods The clinical and sonographic data of 363 cases of pathology confirmed thyroid nodules were retrospectively analyzed.The thyroid nodules were classified by TI-RADS grading criteria,the accuracy of TI-RADS and sonographic findings between FTC and PTC was analyzed.Results In 363 thyroid nodules,114 cases were confirmed as benign lesions;249 cases were confirmed as carcinomas,including 50 FTC cases and 199 PTC cases.The sensitivity,specificity,PPV,NPV,and accuracy of PTC were 97.0% (193/199),85.1% (97/114),91.9% (193/210),85.8% (97/113),95.7% (290/303) respectively.The sensitivity,specificity,PPV,NPV,and accuracy of FTC were 97.0%(193/199),85.1% (97/114),91.9% (193/210),85.8% (97/113),95.7% (290/303) respectively.There were significant differences of nodule size,shape,margin,echogenecity,calcification,halo and flow between FTC group and PTC group (P < 0.05).Conclusion TI-RADS has higher value in the sonographic diagnosis of PTC than that of FTC.FTC are greatly different from PTC in many sonographic findings making it less valuable for the diagnosis of FTC.
7.Biological characteristics of mesenchymal stem cells from psoriatic skinversus normal skin

BACKGROUND:Skin-derived mesenchymal stem cels may reflect the onset of psoriasis.
OBJECTIVE:To analyze the biological characteristics of skin-derived mesenchymal stem cels in psoriasis patients.
METHODS:Skin-derived mesenchymal stem cels from 30 patients with psoriasis and 20 healthy controls were isolated and cultured by trypsin. Flow cytometry was used to detect the celular immune phenotypes CD34, CD44, CD29, CD45, CD90, CD105, CD73 and HLA-DR. The mesenchymal stem cels were induced by the corresponding cartilage, osteogenic and osteogenic inducing agents, to identify the multi-directional differentiation ability. The cel proliferation curve was plotted at passage 3, and the levels of transforming growth factor-β1 and epidermal growth factor in culture supernatant were detected by ELISA assay.
RESULTS AND CONCLUSION: Under an inverted phase contrast microscope, primary skin-derived mesenchymal stem cels isolated from patients with psoriasis and normal controls both exhibited heterogeneity. In the two groups, CD29, CD90, CD44, CD73 and CD105 were highly expressed, and CD45, CD34, and HLA-DR were lowly expressed. Under certain conditions, skin-derived mesenchymal stem cels were induced to differentiate into adipocytes, osteoblasts or chondrocytes. Proliferation of skin-derived mesenchymal stem cels in the psoriasis group was significantly faster than that in control group, but the final number of cels in the two groups tended to be consistent. The levels of transforming growth factor-β1 and epidermal growth factor in the psoriatic skinhad no correlation with the severity of the disease (P > 0.05). Compared with the control group, the epidermal growth factor level in the cel supernatant was significantly higher in the psoriasis group (P < 0.01), while the level of transforming growth factor-β1 was significantly lower (P < 0.01). These results showed that there is heterogeneity in the morphology of skin-derived mesenchymal stem cels from psoriasis patients, and the biological activity of mesenchymal stem cels is abnormal.
8.Antioxidant activity of an exopolysaccharide from cultivated Cordyceps sisnensis fungus

Aim: To preliminarily investigate the antioxidant activity of the acidic exopolysaccharide(EPS-A) from cultivated Cordyceps sisnensis in vitro and in vivo. Methods: PC12 cells were injured by H_2O_2 after the cells were treated with different doses of EPS-A for 6 h, and then the levels of malondialdelyde( MDA) and superoxide dis-mutase (SOD) in PC12 cells were determined. H22 cells were inoculated into the right oxter of each mouse after ICR mice were treated with EPS-A for 7 days, and tumor-bearing mice were treated with EPS-A for 7 days. MDA level and SOD activity in liver, brain and serum were assayed, and tumor weight of each mouse was measured as well. Results: EPS-A significantly decreased the production of MDA and increased SOD activity in H_2O_2-injured PC12 cells. Moreover, EPS-A inhibited H22 tumor growth in mice, in which the level of MDA in liver and brain was reduced, and SOD activity in liver, brain and serum was enhanced. Conclusion: EPS-A exhibited antioxidant activity in cell cultures and mice.
9.The Assembly of a Novel Enzyme Biosensor for Aflatoxin B_1 Detection

A novel biosensor for aflatoxin B1 detecting has been reported. The biosensor electrode for AFB1 detecting was assembled by immobilized aflatoxin-oxidoreductase using open-ended multi-walled carbon nanotubes as matrix. Its linear range was between 0.16?M and 3.2?M. And if the specific anti-aflatoxin B1 antibody and aflatoxin oxidoreductase were both immobilized on the electrode with Multi-Walled carbon nanotubes, the detection limit of the modified electrode could be 16 nM with a 10 times improved sensitivity. The aflatoxin enzyme biosensor assembled this way strode one step forward its practical application.
10.Determination of β-sitosterol and total sterols content and antioxidant activity of oil in acai (Euterpe oleracea).

In order to establish a method for the determination of the sterols of the oil in the freeze-dried acai (Euterpe oleracea Mart.) and to evaluate its antioxidant activities, a saponification/extraction procedure and high performance liquid chromatography (HPLC) analysis method were developed and validated for the analysis of phytosterols in PEE (Petroleum ether extract). Separation was achieved on a Purosper STAR LP C18 column with a binary, gradient solvent system of acetonitrile and isopropanol. Evaporative light scattering detection (ELSD) was used to quantify β-sitosterol and the total sterols. Peak identification was verified by retention times and spikes with external standards. Standard curves were constructed (r = 0.999 2) to allow for sample quantification. Recovery of the saponification and extraction was demonstrated via analysis of spiked samples. The highest content of total sterols is β-sitosterol. The antioxidant activities of the extracts were evaluated using the total oxyradical scavenging capacity assay (TOSC assay). The result showed that the PEE exhibited significant antioxidant properties, sample concentration and the antioxidant capacity had a certain relevance.
