1.Concentration Determination of Catecholamine in Human Urine by HPLC-Electrochemistry

OBJECTIVE:To establish a method for the concentration determination of catecholamines in human urine. METH-ODS:After extracted by aluminum oxide,using 3,4-dihydroxybenzylamine as internal standard,HPLC-electrochemistry was per-formed on the column of Shim-Pack C18 with mixed solution [every 500 ml mixed solution containing ion-pairing reagents(octane sulfonate) 0.5 mmol,methanol 10 ml,0.06 mmol/L Di-n-butylamine 50 μl,sodium acetate anhydrous 2.0 g,citric acid 5.5 g, EDTA-Na2 60 mg and sodium chloride 584.4 mg,pH=3.5] at a flow rate of 0.8 ml/min,column temperature was 30 ℃,detection voltage was 750 mV,and injection volume was 40 μl. RESULTS:The linear ranges were 3.125-100 ng/ml for both norepinephrine (r=0.998 5)and epinephrine(r=0.999 2),lower limits of quantitation were 3.125 ng/ml;RSDs of within days and daytime were lower than 10%,method recoveries were 99.2%-101.6% and 98.9%-100.5%,extraction recoveries were 63.8%-66.8% and 60.5%-62.6%. CONCLUSIONS:The method shows strong specificity,high sensitivity and low detection cost,and can be used for the concentration determination of catecholamines in human urine.
2.Effects of OGP derivate H86A on Bone Metrology in Ovariectomized Rats

Objective To investigate the effects of OGP (10-14) and its derivate H86A on bone metrology in rats with ovariecto-my-reduced osteoporosis. Methods Thirty-two female Sprague-Dawley rats, three months old, were randomly divided into 4 groups. Rats in groups 1 to 3 were bilaterally ovariectomized(OVX) and the others in group 4 were sham-operated. After the operation, rats in group 1 and group 2 were received derivate of OGP(10-14) H86A daily at different concentration. Group 3 were given placebo. Rats were killed after 12 weeks. Left tibias were disconnected and made into non-decalcified bone slice, and then they were detected and analyzed on indexes which were relative with bone metrology. Objective To investigate the association of ILK expression in human renal interstitium and the pathological injury of renal interstitium in the patients with chronic nephropathy.Methods The expressions of ILK,TGF?1,E-cadherin and fibronectin(FN) in renal tissue were investigated with immunohistochemistry and computer image analysis system in 49 patients with chronic kidney disease.According to the score of the renal interstitial pathologic injury,49 patients were divided into four groups: 4 in no injury group(the normal control group),17 in slight injury group,16 in mild injury group,12 in severe injury group.Results ILK was hardly detected in normal control group,and ILK was observed in tubulointerstitial area in 3 injury groups.The expression of ILK was prominent in tubular epithelial cells,next in the damaged tubulointerstitial area.A significant positive correlation was found between the expression of ILK in renal interstitial tissue and the degree of renal tubulointerstitial pathological injury(r=0.736),as well as that of TGF?1(P
4.Effect of betulinic acid on proliferation,apoptosis,and cell cycle of human lymphoma cell line Raji

Objective To investigate the anticancer effects and molecular mechanism of betulinic acid (BA)on Raji cells in vitro.Methods The effects of BA on the growth of Raji cells were studied by MTT assay.Apoptosis was assessed by Annexin-V/PI double-labeled cytometry.The influence on cell cycle was studied by flow cytometer.The cyclin D3 mRNA expression was checked by Western blotting and RT-PCR techniques.Results BA showed obvious inhibition on proliferation,as well as induction potency of apoptosis on Raji cells in vitro in a time-and dose-dependent manner by Annexin-V/PI double-labeled method.With the IC_(50)value for 24 h being(39.44?0.65)?g/mL,Raji cells treated with BA showed ac- cumulation in G_0/G_1 phase and reduction in the percentage of cells in S phase.The cyclin D3 mRNA ex- pression and protein were sharply decreased in Raji cells treated with BA.Conclusion BA could inhibit the proliferation of Raji cells by regulating the cell cycle that arrests cells at G_0/G_1 phase and induces apop- tosis of Raji cells.The antitumor effects of BA may be related to down-regulation of the expression of cy- clin D3.
5.Investigating the correlation between PTPN22 C1858T polymorphism and rheumatoid arthritis in Han nationality in Xiamen area

Objective To assess the correlation between FFPN22 C1858T polymorphism and rheumatoid arthritis(RA)in Han people in Xiamen area by TaqMan-MGB real-time PCR.Methods A casecontrol study was carried out in Xiamen Han population.Their blood samples(100 RAs and 100 controls respectively)were collected and the PTPN22 C1858T polymnrphism was tested by TaqMan-MGB real-time PCR.Results①A technique of TaqMan-MGB teal-time PCR was established to investigate PTPN22 C1858T polymorphism;② Onlv 1858 C allele was presented in all the RAs and controls,and no T allele was detected.Conclusion There is no PTPN22 1858T allele in Han people in Xiamen area.It is suggested that there's no association between PTPN22 C1858T polymorphism and RA.
6.Clinical significance and changes of inflammatory cytokines in patients suffered from progressive cerebral ;infarction

Objective To observe and analyze the changes of inflammatory cytokines levels in patients with progressive cerebral infarction (PCI ).Methods The patients with progressive cerebral infarction (progressive cerebral infarction group),the patients with non-progressive cerebral infarction(non progressive cerebral infarction group)and the healthy individuals(control group)were selected as the research subjects.Sixty individuals were included in each group.The levels of IL-8,hs -CRP,SAA of the research subjects were detected and observed. Results The levels of IL -8[(201.89 ±46.41)mg/L],hs -CRP[(16.55 ±4.49)mg/L],SAA[(20.65 ± 4.17 )mg/L]of the progressive cerebral infarction group were significantly higher than those of non -progressive cerebral infarction group[(132.02 ±40.26 )mg/L,(8.09 ±4.06)mg/L and (15.35 ±4.07 )mg/L).]The differ-ences were statistically significant (t=4.289,P=0.000;t=2.276,P=0.006 and t=2.269,P=0.031).Compared with the control group[(50.70 ±31.14 )mg/L,(0.70 ±0.11 )mg/L and (5.15 ±2.21)mg/L],the levels of IL-8, hs-CRP,SAA in the progressive cerebral infarction group and non -progressive cerebral infarction group were significantly higher on the first day (PCI group vs control:t=3.614,P=0.000;t=3.406,P=0.007;t=2.559, P=0.009;NPCI group vs control:t=3.602,P=0.000;t=3.181,P=0.009;t=1.520,P=0.026).Conclusion The changes of the inflammatory factors levels such as IL-8,hs-CRP,SAA have certain predicative value on the development of the PCI.

9.Evaluation and analysis of transvaginal three-dimensional ultrasound in late pregnancy women of pelvic hiatus morphological structure

Objective To evaluate and analyse the morphological structure in late pregnancy women of pelvic hiatus by appli-cation of transvaginal three-dimensional ultrasound .Methods The 60 cases pregnant women who were checked pregnant weeks for 31 to 40 weeks from January 2012 to December 2014 came to our hospital were chosen as the observation group ,and other 60 cases women who were nulliparous women because of irregular menstruation at the same period treatment as control group .All patients underwent transvaginal three-dimensional ultrasound examination by observed the morphological structure of pelvic hiatus in each period ,and the research object of pelvic hiatus left and right diameter ,anteroposterior diameter and area were compared and meas-ured between the two groups .Results (1)The normal pelvic hiatus morphology was rhombic column crack-like structure ,but the observation group who had 43 .33% abnormal pelvic hiatus morphology and there was kind oval ;(2)The observation group were pelvic diaphragm hiatus of left and right diameter ,anteroposterior diameter and area were significantly larger than the control group in three conditions of the resting period ,anal contraction period and tension period ,and the difference was statistically significant(P< 0 .05) ;all the research object of the pelvic diaphragm hiatus left and right diameter ,anteroposterior diameter and area were de-crease in the anal contraction and increases in tension period for compared with the resting period ,so the difference was statistically significant (P< 0 .05) .Conclusion The three-dimensional ultrasound can be clearly observed the morphology of the female pelvic diaphragm hiatus ,and can be effectively evaluated .The late pregnancy women who will be significantly increases of pelvic dia-phragm ,occurs the relaxation phenomenon and some abnormal morphology for compared with nulliparous women .
10.Gender Differences in Clinical Features of Acute Type 1 Diabetes Mellitus

Objective To analyze whether there are any gender differences in clinical features of acute type 1 diabetes mellitus.Methods Clinical data of 193 patients with acute type 1 diabetes mellitus were collected in our hospital from 2002 to 2007,and subjected to statistical analysis by SPSS.Results The ratio of male to female in all the subjects was 1.6:1.There was no significant gender difference in such clinical aspects as age,fasting blood glucose level,HbAlc concentration,positive percentage of serum autoantibodies against pancreatic islet or morbidity of diabetic neuropathy.In contrast to the males,the onset age was significantly younger,the diabetic duration longer,fasting serum C peptide level lower and the incidence of ketotic acidosis higher in female patients.The patients with onset age less than 14 years (puberty) did not show any significant gender differences in the above-mentioned clinical aspects.Among those with onset age more than 14 years,the females had significantly longer duration and lower fasting serum C peptide level than males.The male patients with onset age more than 14 years had significantly higher fasting serum C peptide level than those less than 14,whereas it was the same for total positive percentages of pancreatic islet autoantibodies and morbidity of diabetic neuropathy in females.Conclusion Gender differences may exist in some clinical features of type 1 diabetic patients with acute onset after puberty, which may partially result from the effects of sex hormones.
