Objective:To investigate the cytokine profile expressed by CIK cells. Methods:CIK cells were induced from peripheral blood mononuclear cells in the presence of IFN-?,IL-1?,IL-2 and mAb against CD3. The phenotype and characterization of CIK cells were identified by flow cytometric analysis.Cytokine expression profiles were determined at mRNA level by semi-quantitative RT-PCR.Results:The percentage of CD3~+CD56~+ positive cells reached up to or higher than 50%.CIK cells couldn't express IFN-?、IFN-? and IFN-? under this kind of condition. But they could express IFNs principally on day 10 to 20. The level of ILs went down after steadily expressed on day 6 to 10(or 20).TNF-? and TRAIL constitutively expressed during the culture period in vitro. TNF-? maintained high expression on day 20.TGF-?_1 mainly expressed on day 30.Conclusion:CIK cells can widely express various kinds of cytokines in vitro.

Objective To determine if nitric oxide(NO)is involved in the protective effect of propefolpretreatment(PP)against ischemia-reperfusion injury(I/R).Methods Eighteen male SD rats weighing 250-350 gwere randomly divided into 3 groups(n=6 each):A normal control;B I/R and C PP+I/R.The animals werekilled by a knock on the head.Hearts were immediately removed and passively perfused in a Langendorff apparatusat 37℃ with oxygenated(95% O_2,5% CO_2)Krebs-Hensleit(KH)solution at 90 cm H_20.In group B and C thehearts were subjected to 35 min global ischemia by suspension of perfusion followed by 120 min reperfusion.Ingroup C the hearts were perfused with KH solution containing 50 ?mol?L~(-1) propofol for 10 min followed by 10 minpropefol wash-out before I/R.At the end of reperfusion myocardial specimen was obtained from left ventricle andhomogenized for determination of the activities of total NOS(iNOS+cNOS)aand SOD and expression of iNOS andheme-oxygenase-1(HO-1)and content of NO,The relationship between the NO content and activity and/orexpression of these protein and enzyme were analyzed by correlation analysis.Myocardium was examined with lightand electron microscope. Results The myocardial NO content,tNOS activity and tSOD activity were significantlylower in group B than in group A and C and there was no significant difference in NO content, and activity of tNOSand tSOD between group A and C.The NO content was positively correlated with tNOS and negatively correlatedwith Mn-SOD aetivety and HO-1 expression.Microscopic examination showed severe cell injury or necrosis in groupB but little injury in group C.Conclusion Ischemia and reperfusion decrease activity of tNOS and SOD butincrease HO-1 expression resulting in decrease in NO content.Propofol pretreatment protects the heart from I/Rinjury through increase in NOS and antioxidases(Mn-SOD,HO-1).

Objective To study the expression of the fused proteasome activator REGγ(11S regulator complex gamma subunit) using gene recombination technology and to further study the interaction between REGγ and casein kinase-2 interacting protein-1(CKIP-1)in vitro. Methods Firstly, the full length cDNA fragment of REGγ was amplified through PCR using the plasmid pCMV-Myc-REGγ as template and subcloned into the prokaryotic expression vector pGEX-4T-2 before being transformed into E.coli BL21 cells. The protein expression was induced by isopropyl-β-D-thiogalactoside(IPTG) .Secondly, the protein expression was monitored by SDS-PAGE and Western blotting after ultrasonication. Finally, the GST Pull-down assay was performed to investigate the interaction between REGγ and CKIP-1 in vitro. Results The prokaryotic expression construct pGEX-4T-2-REGγ was generated successfully and confirmed by DNA sequencing. Expression analysis showed that the GST-REGγ protein was easily expressed and isolated mainly in the lysate supernatant after sonication and centrifugation. The GST Pull-down assay revealed the strong mutual interaction between REGγ and CKIP-1 in vitro. Conclusion The proteasome activator REGγ could interact with the negative regulator of osteoblastogenesis CKIP-1 in vitro and the current study has shed light on further investigations of their physiological relevance.

Objective To improve the method to establish a rat model of vascular dementia and to better serve the experimental studies on vascular dementia. Methods We used the method of“repeatedly clipping the carotid artery com-bined with injection of sodium nitroprusside and with permanent unilateral carotid artery ligation” to prepare a rat model of vascular dementia. The drug piracetam was used to validate the established rat model in respect of the behavior and histopa-thology.Results Different from the reports of previous research, firstly, the results of this study suggested that injection dose of sodium nitroprusside should be 2.0 mg/kg, room temperature should be controlled at 28℃ during surgical opera-tion, and kept at 25℃postoperatively for 24 hours.Under these experimental conditions, the rats were stable and the death rate was reduced.Secondly,“repeatedly clipping carotid artery combined with permanent unilateral carotid artery ligation”could cut off about a third of cerebral blood supply, and causing chronic cerebral ischemia, which is seemingly more con-sistent with the pathogenesis of vascular dementia.Experimental results showed that compared with the control group, the navigation incubation period was extended and space search ability became worse in the model group, cell number was de-creased, with blurred cell contour and deeply stained cytoplasm, and cell nuclei were not clear in the hippocampal tissue. Conclusions Our findings indicate that the improved methodrepeatedly clipping the carotid artery combined with injec-tion of sodium nitroprusside and with permanent unilateral carotid artery ligationcan be used to efficiently establish a rat model of vascular dementia. The similar results of experiment using piracetam validate that this rat model can be used in vascular dementia-related experimental studies.
