1.Chemical constituents from aerial parts of Fallopia convolvulus.

OBJECTIVETo investigate the chemical constituents in the herbs of Fallopia convolvulus.
METHODIts 80% ethanol extract was separated by means of silica gel and Sephadex LH-20 column chromatography to give twelve compounds, whose structures were identified by physicochemical properties and spectroscopic evidence.
RESULTTwelve compounds were isolated and identified as: quercetin (1), luteolin (2), chrysoeriol (3), apigenin (4), kaempferol-3-O-beta-D-glucoside (5), quercetin-3-O-rhamnoside (6), N-trans-coumaroyl tyramine (7), loliolide (8), ethylparaben (9), beta-sitosterol (10), daucosterol (11), n-hexadecanoic acid (12).
CONCLUSIONAll these compounds were isolated firstly from this plant, compounds 6-9 were obtained from this genus for the first time.

2.Appilcation of intraoperative transesophageal echocardiography in aortic valve replacement with bovine pericardium

Objective To evaluate the appilcation value of intraoperative transesophageal echocardiography (IOTEE) in aortic valve replacement with bovine pericardium.Methods Totally 106 patients were ready to underwent the surgery of aortic valve replacement.The parameters of the aortic valves (diameter of aortic annular,aortic sinus and aortic sinus tube,effective height [eH]) were measured before cardiopulmonary by using IOTEE and during intraoperation.Results Eight cases were removed from the study due to supplemental preoperative diagnosis and modification of operative method,the rest of the 98 cases were enrolled.The coincidence rate of initial diagnosis hy using IOTEE and intraoperative diagnosis was 100 %;5 cases (5/98,5.10 %) of secondary diagnosis were inconsistent with intraoperative diagnosis.The coincidence rate of the number of aortic valves with preoperative IOTEE diagnosis and intraoperative diagnosis was 100 %,the aortic valves perforation fistulas was 80.00%,the aortic valves vegetation was 85.71%,the senile calcified valvular disease was 100%,and the rheumatic disease was 100%.Diameter of aortic annular,aortic sinus and aortic sinus tube,eH measured by preoperative IOTEE had good correlation with intraoperative measurement (all r>0.8,all P<0.05).Conclusion IOTEE has feasibility and guiding value in aortic valve replacement with bovine pericardium,and is also of great significance to evaluate the prognosis of surgery.
3.Association between strength quality with comorbid depressive and anxiety and its subtypes among junior high school students in Jiangxi Province

Objective:
To analyze the association of strength quality of junior high school students with comorbid depressive and anxiety (CDA) and its subtypes, so as to provide reference for mental health improvement among junior high students.
Methods:
A convenient sampling method was used to select 10 007 junior high students aged 13-16 from four cities (Nanchang, Ganzhou, Jiujiang, Shangrao) in Jiangxi Province during September to November 2023. A questionnaire survey and physical fitness tests were employed to collect basic information and assess students depressive and anxiety status, as well as strength quality. The χ 2 test was used to compare the distribution differences of CDA and its subtypes among different demographic characteristics, sleep, physical activity, and overweight/obesity conditions. Logistic regression models were applied to infer the association strength between strength quality and CDA and its subtypes.
Results:
The overall detection rate of CDA among junior high students was 11.04%. Among the CDA subtypes, the detection rates for mild depression-mild anxiety type, anxiety dominant type, depressiondominant type, and severe depression-severe anxiety type were 3.67%, 3.35%, 0.39% and 3.64%, respectively. Compared to the grip strength and standing long jump scores in the > P 75 group, the risk of CDA occurrence increased in other groups (grip strength: P 25 - P 75 , OR =1.20; < P 25 , OR =1.28; standing long jump: P 25 - P 75 , OR =1.31; < P 25 , OR =1.51; P <0.05). In stratified analysis of different CDA subtypes, compared to the grip strength > P 75 group, the risks for mild depression-mild anxiety type increased in the P 25 - P 75 and < P 25 groups ( OR=1.32, 1.44, P <0.05). Compared to the standing long jump > P 75 group, the risks for anxiety dominant type and severe depression-severe anxiety type increased in other groups (anxiety dominant type: P 25 - P 75 , OR =1.32; severe depression-severe anxiety type: < P 25 , OR =1.87; P < 0.05 ).
Conclusions
Lower levels of grip strength and standing long jump performance among junior high students are positively correlated with CDA and its subtypes. Lower levels of lower limb strength may be an important predictor of CDA in severe depression and anxiety.
4.Determination of phenols and triterpenoid saponins in stems of Sargentodoxa cuneata.

The methods to determine the total phenols, total saponins, and marker constituents salidroside, chlorogenic acid and 3, 4-dihydroxy-phenylethyl-β-D-glucopyranoside in the samples of Sargentodoxae Caulis were established to provide the evidence for the improvement and revision of the quality standard of the crude material recorded in the Chinese Pharmacopoeia (2015 Edition). The content of total phenols was determined by ultraviolet spectrophotometry, using gallic acid as a reference substance. The content of total saponins was determined by ultraviolet spectrophotometry, using 3-O-[β-D-xylopyranosyl-(1-2)-O-β-D-glucuronopyranosyl]-28-O-[β-D-glucopyranosyl] asiatic acid as a reference substance. The contents of salidroside, chlorogenic acid and 3,4-dihydroxy-phenylethyl-β-D-glucopyranoside were detected by HPLC. The linear ranges were 1.01-7.04 mg x L(-1) for total phenols, 37.7-201 μg for total saponins, 0.025 8-1.55 μg for salidroside, 0.076 2-5.44 μg for chlorogenic acid, and 0.064 9-3.47 μg for 3,4-dihydroxy-phenylethyl-βP-D-glucopyranoside, respectively. Their average recoveries were 99.12%, 99.11% 105.5%, 99.08%, and 101.6%, respectively. The contents of total phenols and total saponins were 3. 04% -11. 9% and 0. 87% -3. 63%. The contents of salidroside, chlorogenic acid and 3,4-dihydroxy-phenylethyl-β-D-glucopyranoside fluctuated from 0.018% to 0. 572%, from 0.041% to 1.75% and from 0.035% to 1.32%. The established methods were reproducible, and they could be used for the quality control of Sargentodoxae Caulis. The present investigation suggested that total phenols, salidroside, and chlorogenic acid should be recorded in the quality standard of Sargentodoxae Caulis and their contents should not be less than 6.8% for total phenols, 0.040% for salidroside, and 0.21% for chlorogenic acid.

5.Study on effects of sulfur fumigation on chemical constituents of Chrysanthemum morifolium cv. Boju.

A comprehensively comparison of the chemical profiles between sun-drying BJ (NBJ) and sulfur-fumigated BJ (SBJ) was conducted by HPLC analysis and the discrepant peaks were identified or tentatively assigned by HPLC-ESI-MSn. A total of 32 chemical components were used for qualitative comparison. Meanwhile, a quantitative comparison of BJwere conducted by HPLC analysis and determining seven compounds from 3 NBJ and 3 SBJ samples dramatic chemical changes were found. After sulfur fumigation, the contents of flavonoids glycosides and phenolic acids were remarkably reduced, but the contents of flavonoids aglycones were significantly increased. Multivariate statistics, including principle component analysis (PCA) and partial least squares discriminate analysis (PLS-DA) were used to investigate the potential damaging effect of sulfur-fumigating process. The PCA score plots showed six samples were clearly classified into the sun-drying and sulfur-fumigating groups. And according to VIP >1, the most important chemical markers were apigenin, luteolin and 3,5-dicaffeoylquninic acid which could be used to distinguish NBJ and SBJ samples. Combining the results of qualitative and quantitative analysis, it showed that the sulfur fumigation has a significant effect on BJ.

6.Preparation of 99Tcm labeled survivin mRNA antisense PNA and gene imaging in nude mice bearing lung carcinoma A549 xenografts

Objective To prepare the 99Tcm-survivin mRNA antisense peptide nucleic acid (PNA)and investigate its value as a gene imaging agent in tumor bearing mice and early diagnosis in tumor.Methods Survivin mRNA antisense PNA and mismatch PNA were synthesized.Four amino acids (Gly- (D)Ala-Gly-Gly) and Aba (4-aminobutyric acid) were linked to the 5' end of PNA.Gly- (D)Ala-Gly-Gly served as a chelating moiety for strong chelation of 99Tcm and Aba acted as a spacer to minimize the steric hindrance.PNAs were labeled with 99Tcm by the ligand-exchange method.The labeling efficiency and radiochemical purity were measured by HPLC and ITLC methods.There were five BALB/c nude mice bearing human lung carcinoma ( A549 ) in each of antisense PNA and mismatch PNA groups.Gene imaging of 99Tcm-survivin mRNA antisense and mismatch PNAs were performed at 1,2 and 4 h post the injection,respectively,and the T/NT ratio was measured by the method of ROI.The statistical comparisons of average values were performed with the two-group t-test for independent sample by SPSS 13.0.Results The product kept stable in vitro.The labeling efficiency of 99Tcm-survivin mRNA antisense PNA was (95.48 ±1.92)% and more than 85% after the incubation for24 h in serum.The radiochemical purity was > 95%.The labeling efficiency of mismatch PNA was similar to the antisense PNA.99Tcm-survivin mRNA antisense PNA was especially uptaken by tumor lesion,and its accumulation reached the top at 4 h post the injection.T/NT ratios at 1,2,and 4 h were 2.70 ± 0.28,3.44 ± 0.35,4.21 ± 0.63,respectively.In the comparison,the T/NT ratio of 99Tcm-survivin mRNA mismatch PNA at 4 h (3.12 ±0.50) was significantly lower (t =2.918,P =0.019).Conclusions 99Tcm-survivin mRNA antisense PNA has high labeling efficiency,good stability and no need of purification.Its characteristic of especial uptake by tumor lesion provides the potential value in early diagnosis of tumor.
7.Inhibition of HCCR-2 gene expression by siRNA suppresses liver metastasis of ovarian cancer

Objective To investigate the impact of human cervical cancer oncogene-2 (HCCR-2) inhibited by small interference RNA (siRNA) on the liver metastatic potential of ovarian cancer. Methods High metastatic ability human ovarian cancer cell line HO-8910PM was infected using HCCR-2-siRNA lentiviral expression vector and empty negative control vector to establish small interference (SI) group and negative control (NC) group. The expression of HCCR-2 protein in two groups was detected by Western blot assay. Transwell assay was used to deifne the cell invasion ability. Liver metastases model of ovarian cancer was established to detect the cell metastatic ability. The data between two groups were compared using t test, and the rates were compared using Chi-square test. Results The average relative expression of HCCR-2 protein in SI group (0.16±0.02) signiifcantly decreased compared with that in NC group (0.86±0.05) (t=-8.06, P<0.05). The membrane permeating cell count in SI group (58±3) was signiifcantly less than that in NC group (262±19) (t=-37.77, P<0.05). The incidence of liver metastases in SI group (2/6) was signiifcantly lower than that in NC group (6/6) (P<0.05). The number of metastases in SI group (1.3 ± 0.2) was signiifcantly less than that in NC group (9.4 ± 0.8) (t=-10.25, P<0.05). Conclusion Inhibition of HCCR-2 gene expression by siRNA can reduce the invasion and liver metastatic potential of ovarian cancer cells.
8.Removal of cadmium from extracts of Gentianae Radix et Rhizoma by gamma-mercaptopropyl-modified silica gel.

OBJECTIVETo study the optimum process of removing cadmium irons from extracts of Gentianae Radix et Rhizoma with gamma-mercaptopropyl-modified silica gel (MPS) and assess its cadmium ion-removing property.
METHODStatic and dynamic adsorptions were adopted to detect the cadmium-removing rate. MPS' cadmium ion-removing property was assessed with such indicators as the cadmium-removing rate, the solid content and the HPLC fingerprint.
RESULTThe process parameters of the static adsorption were as follows: 0.20 g x mL(-1) of concentration of extracts, 120 minutes of adsorption time and 15:1 between raw materials and MPS. The process parameters of the dynamic adsorption were as follows: 1:3.5 times between diameter and height, 0.20 g x mL(-1) of concentration of extracts, 0.9 mL x min(-1) of flow rate of the extracts and 50:1 between raw materials and MPS. Before and after the cadmium ion-removing process, the extracts showed no notable difference in solid content and HPLC fingerprint.
CONCLUSIONgamma-mercaptopropyl-modified silica gel (MPS) can effectively remove cadmium ion from the extracts of Gentianae Radix et Rhizoma with an excellent cadmium ion-removing property.

9.Feasibility study of QAMS for quantitative analysis of multiple structural types of ingredients in Zhizi Jinhua pill.

OBJECTIVETo study the technical feasibility of QAMS to determine different structural types of ingredients in Zhizi Jinhua pill, a Chinese patent medicine.
METHODEmodin was chosen as the internal reference substance. The relative correction factors (RCF) of berberine, baicalin, wogonoside, baicalein, aloe-emodin, wogonin, rhein, chrysophanol and physcion to emodin were calculated with the peak areas determined at 254 nm. The ruggedness of these RCFs was validated. The contents of emodin in the samples of Zhizi Jinhua pill were determined by using the external standard method, and the contents of the other nine ingredients were calculated by their RCFs. The contents of these ten ingredients in all samples were also determined by the external standard method.
RESULTNo significant differences were observed between the quantitative results of the two methods. The obtained RCFs were credible.
CONCLUSIONThe established QAMS method can be used for quantitative determination of different structural types of multi-compounds in Zhizi Jinhua pill. It can provide reference for the quality control of Chinese patent medicine.

10.Study on revising the criteria of classification of hazard conditions of productive dust.

OBJECTIVETo provide scientific basis data for revising the national hygiene criteria of "Classification of hazard conditions of productive dust" (GB5817-86).
METHODSThe data of the retrospective study and the field survey data were analyzed with correlation and regression analysis. The product of total dust concentration of respiratory exposure (mg/m(3)), total ventilation during exposure (m(3)/d per psrson), and level of free SiO(2) in dust (%) was the respiratory exposure dose of free SiO(2) (mg per day per person) which was used as dose criteria value of classification of hazard degree of dust.
RESULTSUsing free SiO(2) exposure dose and the dose-effect relationship, the hazard degrees of the dust were divided into 5 grades: 0, I, II, III, IV (0 - 8.0, 8.1 - 12.0, 12.1 - 16.0, > 24.1 mg per day per person).
CONCLUSIONThe exposure dose of free SiO(2) is closely related to the pathogenesis of silicosis. Using the exposure dose of free SiO(2) as the classification indicator of hazard degree of dust is reliable, simple and easy to execute.
