2.Effect of Exelon on cognitive function of chronic cerebral hypoperfusion rats

Objective To investigate the effect of Exelon on cognitive function of chronic cerebral hypoperfusion rats and its cholinergic mechanism.Methods 112 rats were randomly separated into four groups (normal, hypoperfusion, normal saline and Exelon). Bilateral common carotid arteries were ligated for models of cerebral hypoperfusion in the rats except normal group. 4 weeks later, Exelon and normal saline were used to treat the rats in Exelon group and normal saline group respectively for 6 weeks. Then the values of AchE lever and Ach fiber density together with the results of experiments in water maze and the values of event-related potential (ERP) were recorded and compared with each group.Results Comparing with the normal group, the water maze completion time of the hypoperfusion group was significantly delayed, error frequency significantly increased, the latency of ERP much extended, the AchE level and Ach fiber density evidently decreased(all P

Objective To investigate the effect of Aricept on the level of cholinesterase and the density of cholinergic fibres in cortex of frontal lobe and hippocampus zone in rats undergoing chronic cerebral hypoperfusion rats. Methods One hundred and twelve male SD rats were divided into a normal group, a model group, a saline treatment group (saline, 3mg/kg/day for six weeks) and an Aricept treatment group (Aricept, 3mg/kg/day for six weeks). Objective To determine if propofol-remifentanil and isoflurane have any different effects on intrapulmonary shunt during one-lung ventilation (OLV). Methods Twenty-four ASA Ⅰ or Ⅱ patients (18 male, 6 female) aged 42-69 yr undergoing radical esophagus cancer resection via left thoracotomy were randomly divided into 2 equal groups ( n = 8 each) : propofol group (Pro) and isoflurane group (Iso) . The preoperative lung function was normal in both groups. The patients were premedicated with intramuscular diazepam 10 mg and atropine 0.5 mg. Radial artery was cannulated and S-G catheter was placed via right internal jugular vein in pulmonary artery. Anesthesia was induced with propofol 1.5-2.0 mg?kg-1, fentanyl 4 ?g?kg-1 and succinylcholine 1.5 mg?kg-1 and maintained with TCI of propofol and remifentanil (target plasma concentration was set at 3.2 ?g?ml-1 and 4.5 ng?ml-1 ) or isoflurane inhalation ( end- tidal isoflurane concentration = 1.5% -2.5 % ) and intermittent i. v. boluses of fentanyl. The patients were mechanically ventilated after endobronchial intubation with double-lumen tube (VT = 8-10 ml, RR= 10-12 bpm, I:E = 1:2) . During OLV VT was reduced to 6-8 ml and RR increased to 14-16 bpm.PaCO2 was maintained at 35-45 mm Hg. ECG, HR, MAP, SpO2, auditory-evoked potential index (AAI), cardiac index (CI), airway pressure and end-tidal isoflurane concentration were continuously monitored during operation. Blood samples were taken from radial artery and pulmonary artery at 10 min after S-G catheter placement (T0, baseline) at 10 min bilateral ventilation (in right lateral position) (T1) at 15, 30, 60, 90 min of OLV (T2-5) for measurement of blood gases and calculation of Qs/Qt.Results The two groups were comparable with respect to age, M/F ratio, body weight and preoperative lung function. AAI was below 30 during operation and PaCO2 and pH were within normal range in both groups. Qs/Qt was significantly increased while PaO2 was significantly decreased during operation (from T1 to T5 ) as compared to baseline values (T0) but Qs/Qt was gradually decreasing while PaO2 was gradually increasing from T2-5 in both groups. Qs/Qt was significantly lower in Pro group than in Iso group at each interval (T2-5) but there was no significant difference in PaO2 at T2-5 between the two groups. Conclusion There is less intrapulmonary shunt during OLV under general anesthesia with propofol-remifentanil than with isoflurane but the difference is not significant enough to affect PaO2.

Objective To investigate the effect of partial liquid ventilation(PLV) with perfluorocarbon(PFC) on pulmonary surfactant during beatment of acute lung injury(ALI) induced by oleic acid. 6.Comparative Study on HPLC Characteristic Spectrum of Erycibe obtusifolia from Different Habitats by HPLC

OBJECTIVE:To establish and compare the HPLC characteristic Spectrum Erycibe obtusifolia from different habi-tats. METHODS:HPLC was performed on the column of Hypersil ODS C18 with the mobile phase of acetonitrile-0.1% phosphoric acid at flow rate of 1.0 ml/min,detection wavelength was 347 nm,the column temperature was 30℃and the sample size was 20μl. 12 different batches of samples were determined and the characteristic spectrums of those were established. TCM fingerprint evalu-ation software(2004 edition)was used to evaluate the similarity of 12 batches of samples. RESULTS:10 characteristic peaks were identified in the characteristic spectrum of E. obtusifolia. Peak 3 was scopolamine glycosides,peak 4 was chlorogenic acid and peak 6 was scopoletin. The similarities of 10 batches of samples were proved to be higher than 0.90 and 2 batches were proved to be less than 0.90. CONCLUSIONS:The method is simple,accurate and reproducible,and can provide scientific evidence for effectively controlling the internal quality standards.
7.MDA accelerates the glomerular mesangial cell apoptosis through inhibition of Nrf 2/ARE

Objective To explore the function of MDA on diabetic nephropathy .Methods Glomerular mesangial cells ( GMC) were pretreated with MDA at a final concentrations of 0μmol/L, 1μmol/L, 5μmol/L, 10μmol/L and 50 μmol/L.MTT assay was used to examine the viability of GMC ) .AnnexinV-FITC was used to evaluate effect of MDA on cell apoptosis .RT-PCR and western blot were used to analyze the expression of Nrf 2, HO-1 andγGCL.Results MDA treatment inhibited GMC viability in a dose-dependent manner .MDA at the concentration of more than 5 μmol/L induced mass production of ROS in GMC ( P<0.05 ) .In addition , antioxygen of tBHQ may relieve MDA-induced reduction of cell viability .MDA inhibited the expression of HO-1 , γGCLand Nrf2 ( P <0.05 ) .Conclusions MDA inhibites GMC viability and promotes the cell apoptosis by ROS production through in-hibiting Nrf2/HO-1-γGCL.
8.Myringotomy with Grommet Insertion for Treatment of Tympanic Membrane

Objective To evaluate the efficacy of myringotomy with grommet insertion in the treatment of tympanic membrane atelectasis .Methods From July 2011 to July 2013 ,62 patient with tympanic membrane atelecta-sis were treated with myringotomy with grommet insertion in our hospital ,we evaluated 54 patients(56 ears)whose follow -up period was 1 year or more .Of the 54 patients included in this study ,28 (30 ears))were female and 26(26 ears)were male .The age of the patients ranged from 8 to 54 ,with a mean age of 25 .8 ± 6 .6 years old ,and 18 pa-tients (32% ) were under the age of 16 .The air -bone gap(ABG)was compared at pre -operation and 1 year after post-operation .Results The average ABG of pre-operation and post -operation ,including all patients ,were 27 . 07 ± 5 .03 dB and 15 .52 ± 5 .73 dB ,respectively (P<0 .05) ,and the difference was statistically significant .All the 3 groups ,the ABG of pre- and post-operation ,the difference of each group was statistically significant .Conclusion Myringotomy with grommet insertion was effective in treating tympanic membrane atelectasis .
10.Construction of recombinant eukaryotic expression plasmid containing 1.3-fold-overlength genome of HBV and its expression in Huh7 cells

Objective To construct 1.3-fold-overlength infectious clone of hepatitis B virus isolated from Chinese patients , observe the expression of plasmid in Huh7 of liver cancer cells and establish genome of HBV in vitro. Methods HBV DNA in serum was extracted from HBV patient. SOE-PCR was performed to produce a 1.3-fold-overlength genome of HBV. The plasmid was named pHBV1.3 (C). After that,pHBV1.3 (C) was transfected into Huh7 cells, HBV related viral antigens and DNA were detected by ELISA,Western Blot and Fluorescence quantitative PCR. Furthermore, adefovir dipivoxil, a clinic anti-viral drug, was utilized to test the sensitivity of the new infectious clone. Results An infectious clone of pHBV1.3 (C) was successfully constructed. HBV gene carried in pHBV1.3 (C) could be efficiently replicated and expressed in Huh7 cells. Adefovir could inhibit HBV replication in this HBV cell model. Conclusions A recombinant plasmid containing 1.3-fold-overlength of HBV genotype C was successfully constructed. This construct is competent to support viral transcription and replication in vitro , suggesting that infectious cells are expected to be a new model of HBV infection in vitro.
