1.The vasodilating effects induced by etomidate in aortas of insulin resistant rats
Chinese Journal of Biochemical Pharmaceutics 2014;(2):51-54
Objection To investigate the effects of etomidate on aorta isolated from IR rats, and explore its underlying endothelium-related mechanism(s). Methods The IR animal model was made by feeding rats with high fructose diet for 8 weeks. Aortic rings were isolated and suspended in a tissue bath, and tensions were recorded isometrically. The effects of etomidate on provoked contractions of the rings were assessed in absence or presence of potassium channel blockers or NO-synthase inhibitors. Results Etomidate-induced relaxation in IR rings was greater than NC rings. NG-nitro-L-arginine methyl ester (L-NAME) or glibenclamide (Gli) inhibit significantly etomidate-induced relaxation in IR rings, and the inhibition of Gli was disappeared in endothelial-denuded aortic rings. Conclusion Etomidate cause vasodilation in IR rat aortas by an endothelial-dependent and independent manner. Impaired NO- and KATP channel-mediated relaxation and etomidate-induced increased availability of may involve in endothelial-dependent relaxation of etomidate in IR rat aortas.

OBJECTIVETo observe the therapeutic effect of a drug pair of Radix Astragali and Rehmanniae Radix combined with glucocorticoid (GC) in treating lupus nephritis (LN) patients and its influence on some experimental indices.
METHODSTotally 52 LN patients were randomly assigned to the treatment group (treated by routine Western medicine and a drug pair of Radix Astragali and Rehmanniae Radix, 25 cases) and the control group (treated by Western medicine, mainly by GC and cyclophosphamide, 27 cases). All patients received 6-month therapy. The GC dosage, the withdrawal and reduction dosage of GC, clinical efficacy, systemic lupus erythematosus disease activity index (SLEDAI) score, adverse reactions, and laboratory indicators were recorded.
RESULTS(1) All patients got relieved to some degree with the dosage of GC reduced. The total withdrawal and reduction dosage of GC was slightly higher in the treatment group than in the control group [(50.23 +/- 12.43) mg vs (48.76 +/- 13.61) mg, P > 0.05]. Besides, the prednisone dosage in the treatment group was lower than that in the control group, but without statistical difference (P > 0.05). The ratio of patients in need of adding prednisone for aggravating disease was 24.0%, significantly lower than that in the control group (44.44%, P < 0.05). (2) There was no statistical difference in the SLEDAI score, inflammatory indicators, liver and renal functions, blood electrolytes, blood glucose, blood and urine routines between the two groups (P > 0.05). The 24-h urinary protein count was (1.06 +/- 0.22) g/L in the treatment group, obviously lower than that in the control group (1.43 +/- 0.55 g/L, P < 0.05). (3) There was no statistical difference in the incidence rate of infection, gastrointestinal hemorrhage, psychoneuroses, Cushing's syndrome, cardiovascular anomalies, and femoral head necrosis between the two groups (P > 0.05). But the incidence of adverse reactions such as insomnia, tidal fever, spontaneous sweat, and obesity was less in the treatment group than in the control group (P < 0.05).
CONCLUSIONSUsing a drug pair of Radix Astragali and Rehmanniae Radix combined with GC in treating LN could withdraw the dosage of GC and relieve symptoms it induced. It was advantageous in reducing the dosage of GC and stabilizing patients' conditions.

Chinese Journal of Tissue Engineering Research 2006;10(31):190-192
BACKGROUND: Calcium pyruvate has great effect on reducing bodymass, and chitosan can regulate immunity system, promote bone growth, decrease blood sugar and adjust blood lipid. Calcium pyruvate and chi tosan capsule (CCC) was made by calcium pyruvate and chitosan combined with Chinese medicine, which can be used both as food and medicine. However, its effect still needs further observation. OBJECTIVE: To investigate the weight-reducing effect of CCC on obesity rats, and explore its mechanism.DESIGN: A randomized controlled trial.SETTING: Teaching and Research Section of Preventive Medicine, Department of Public Health, Xi'an Medical College, and Department of Public Health, Medical School of Xi' an Jiaotong University.MATERIALS: The experiment was conducted in the Department of Public Health, Medical School of Xi'an Jiaotong University from April to July 2001. Totally 60 healthy male weaning SD rats with body mass of 50-80 g,were provided by the Animal Center of Xi'an Jiaotong University. CCC,mainly composed of calcium pyruvate and chitosan, mixed with Chinese medicine extract after prepared with distilled water, was made by the author.METHODS: ①Grouping and modeling: The 60 rats were randomly divided into 5 groups: blank control group, model control group, high, middle and low dose CCC supplement groups, respectively with 12 rats in each group. The rats in the blank control group were fed w ith basal diet; the other groups were fed with high fat and nutrition diet to establish rat models of nutritional obesity. ②Administration: The rats in the blank and model control groups were given suspension mixed with starch matrix in capsule and distilled water at same dose. The rats in the high, middle and low dose CCC groups were intragastrically infused with 3, 1.5 and 0.75 g/kg CCC, once aily for 30 days. ③Detection: Body mass, body length and wet weight of fat tissue were measured before and after administration to calculate the obese index and ratio of fat weight/body mass. The adipocyte number and adipocyte size were observed by 400-fold microscope and ocular micrometer, respectively. The detection of serum triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and serum leptin levels were performed according to the leptin radioimmunity analyzer kit.MAIN OUTCOME MEASURES: ①Body mass, body length and obesity index before and after administration. ②Wet weight of fat, fat/body mass, adipocyte number and size. ③TG, TC, HDL-C and serum leptin.RESULTS: All the 60 rats were involved in the result analysis. Before administration, the body mass of the blank control group was obviously lower than other 4 groups (P < 0.01), and there was no significant difference among the 4 groups (P > 0.05). After administration, the mean body mass, obesity index, wet weight of fat tissue, ratio of fat/body mass and leptin level of the high, middle and low CCC groups were lower than the model group (P < 0.05 or 0.01), and the adipocyte size were significantly smaller than those in the model group (P < 0.05 or 0.01), but the adipocyte number was more than the model group (P < 0.05 or 0.01). Meanwhile, level of TG and TC of the high and middle dose CCC groups were lower than the model group (P < 0.05 or 0.01), there was no significant difference in the body length and HDL-C of each group.CONCLUSION: ①CCC shows evident weight-reducing effect on rats with nutritional obesity, which is not related to the ability of CCC to enhance the serum leptin level. ②CCC can also lower the blood lipid level.

To observe the incidence of high intraocular pressure ( lOP ) and its risk factors in the early stage (within 7d) after 23 - gauge sutureless microincisional vitrectomy.
●METHODS: Retrospective case series study. Totally 98 patients ( 98 eyes ) who had undergone 23 - gauge sutureless microincisional vitrectomy were enrolled. lOP was measured by non-contact tonometry. High lOP was defined as an lOP > 25mmHg at any time within 7d after surgery. The influence of age, sex, side of operation, course of disease, primary disease pre - operation, reoperation, surgical options, type of tamponade, status of lens, surgical time on postoperative high lOP were analyzed.
● RESULTS: High lOP was found in 33 eyes ( 33. 7%) within 7d after surgery. High lOP was found in 8 eyes on postoperative 1d, the incidence was 24. 2% (8 / 33), and 25 eyes was found on postoperative 3d, the incidence was 75. 8% (25 / 33). The incidence of high lOP of male and female were 32. 8% ( 20 / 61 ) and 35. 1% ( 13 / 37 ) respectively. The incidence of high lOP of right and left eye were 36. 8% (21 / 57) and 29. 3% (12 / 41) respectively. There were no significant differences comparing age (Z =-0. 22), sex( χ2 = 0. 057), side of operation( χ2 = 0. 612), course of disease (Z= -0.079) and surgical time (Z= -0. 553) between patients with high lOP and those without it (P>0. 05). Similarly, no statistical significance was found between eyes with gas tamponade and silicone oil tamponade (χ2 = 1. 04), traumatic eyes and non traumatic eyes(χ2 = 0. 044), and retinal detachment eyes and without retinal detachment eyes ( χ2 = 2. 282, P > 0. 05 ). The incidence of high lOP in eyes with several operations was higher than that in eyes with the first operation ( χ2 =5. 211,P< 0. 05), in eyes with combined operations eyes was significantly higher than that in eyes with pure vitrectomy (χ2 = 4. 57,P< 0. 05), and in eyes with aphakic eye was higher than that in eyes with phakic eyes ( χ2 =4. 224,P<0. 05).
●CONCLUSlON:High lOP occurs commonly in 3d after 23- gauge sutureless microincisional vitrectomy. The risk factors of high lOP are reoperation, combined operations and aphakic eye.
