The separation of digestive system cancer stem cells (CSCs) is the first step of the study of stem cell.At present,there are three methods of separation of the digestive system CSCs:cell phenotype sorting,side population cell sorting and serum-free culture.The separation of digestive system CSCs is based on these three methods or be improved to a higher proportion of CSCs.
2.Effect of nitrotyrosine on the expression of ?1D-adrenoceptor mRNA in vascular smooth muscle of rat

Objective To investigate the effect of nitrotyrosine (3-NT) on the expression of ?1D adrenoceptor (?1D-AR) mRNA in cultured vascular smooth muscle cells (SMCS) .Methods SMCS were obtained from the tunica media of thoracic aorta of 1 month old SD rats and cultured in DMEM medium. The experiment consisted of two parts. In part Ⅰ SMCS were incubated with 0,1, 10, 100 or 200 ?mol?L-1 3-NT for 24 h and in part Ⅱ SMCS were incubated with 100 ?mol?L-1 3-NT forO,12, 24, 48 or 72 h. The total RNA was isolated by using Trizol reagent. The expression of ?1D-AR mRNA was determined by RT-PCR and agarose gel electrophoresis. Results In part I incubation with 1 and 10 ?mol?L- 3-NT for 24 h had no significant effect on the expression of ?1D-AR mRNA while incubation with 100 or 200 ?mol?L-1 3-NT for 24 h decreased the expression of ?1D-AR mRNA compared with 0?mol?L-1 3-NT (P
4.Risk Factors and Pathogens Distribution of Urinary Tract Infection in Patients with Spinal Cord Injury

Objective To analyze the risk factors and distribution of pathogenic bacteria of urinary tract infection (UTI) patients with spi-nal cord injury (SCI). Methods A total of 452 patients with SCI collected from December, 2015 to June, 2016 were retrospectively analyzed. Results 104 cases (23.1%) were diagnosed as UTI. 119 strains were identified, including 96 strains of Gram-negative bacteria (80.7%) and 22 strains of Gram-positive bacteria (19.3%). Female, invasive operation, depressed lower limb muscle strength, neurogenic bladder and pre-vention of antibacterial agents use were considered to be the main risk infection factors. The invasive operation included indwelling urethral catheterization, intermittent catheterization and other transurethral operation. The incidence rate of UTI in patients with SCI who also accept-ed indwelling catheter was 100.0%. Conclusion Gram-negative bacteria is the dominated pathogenic bacteria in SCI patients with UTI. It should be noticed the characteristics of pathogen, which both may have the high drug resistance rate and high separation rate, to select exact-ly antimicrobial agents to control the spread and drug resistance of pathogenic bacteria. In addition, patients with underlying diseases, inva-sive operation and lower strength should be monitored targeted.
5.Determination of geniposide in Diaojinyao powder by HPLC

Objective To establish a high performance liquid chromatography (HPLC) method for the determination of geniposide in Diaojinyao powder. Methods The HPLC system consisted of the Fortis-C18 (4.6 mm × 250 mm, 5μm) column, the mobile phase consisted of HCN∶H2O (12∶88) , the flow rate was 1.0 ml/min, the column temperature was 30℃, The UV detector was set at 238 nm. Results The linear response range was 0.049~0.980mg/ml ( r=0.999 9). The average recovery of geniposide was 99.1%, and RSD was 1.66%. Conclusion The method is simple, rapid, accurate and repeatable. It can be applied in determination of Geniposide in Diaojinyao Powder.
6.Meta-analysis of titanium plate and elastic intramedullary nail in the treatment of midshaft clavicular fractures

BACKGROUND:Elastic stable intramedulary nailing and titanium plate fixation are two methods for the treatment of midshaft clavicular fractures. Current research about comparison of these two methods is not too much, and most of cases were retrospectively analyzed. OBJECTIVE:To compare the efficacy of elastic stable intramedulary nailing and titanium plate fixation for midshaft clavicular fractures. METHODS:The PubMed database, CBM, EMbase database, Cochrane Library database, CNKI database and Wangfang database were searched to collect the trials on midshaft clavicular fractures. The searching time ranged from the date of building to September 2015. The quality of trails was evaluated. RESULTS AND CONCLUSION:(1) Seven trials involving 512 patients were included. (2) Meta-analysis results showed the postoperative functional recovery was better in the elastic stable intramedulary nailing group than that of the plate fixation group (P< 0.01). (3) The average bone union time, operation time, incision length,intraoperative blood loss and the hospital stay of the elastic stable intramedulary nailing group were less than those of the plate fixation group (P< 0.01). (4) There were no significant differences in incidence rate of complications between two groups.(5) It is concluded that elastic stable intramedulary nailing for treating midshaft clavicular fractures is superior to the plate fixation in the efficacy. As the first choice for treatment of midshaft clavicular fractures, above conclusions are needed to be verified by large-scale multi-center randomized controled trials.
7.Proteomic techniques based protein biomarkerssearching and validation in pleural effusion

Objective Through analyzing benign and malignant pleural effusion samples by proteomic techniques, finding protein biomarkers to provide help and new clues for effusion differential diagnosis.Methods Two-dimensional electrophoresis(2-DE) and matrix-assisted laser desorption ionization-time of flight-mass spectrometry(MALDI-TOF-MS) were used to search and identify protein biomarkers, enzyme linked immunosorbent assay(ELISA) was to validate the biomarkers.Results By comparing malignant group with benign group, 43 significantly different protein spots(Up or down regulated≥2 times) were found.Including 9 up regulated spots and 34 down regulated spots.And 7 spots were identified(Up or down regulated≥3 times) by MALDI-TOF-MS and validated 2 spots immunoglobulin λ(Igλ) and haptoglobin(Hp) by ELISA.The results showed that Igλ showed no statistical significance between two groups, while Hp showed the statistical significance(P<0.05).The diagnostic sensitivity and specificity of Hp in malignant pleural effusion were 75.00% and 52.38% at diagnostic cut-off point of 389.02 μg/L.Conclusion The application of proteomics technology has a great help with protein biomarkers searching in pleural effusion.HP has a certain value for the differential diagnosis of benign and malignant pleural effusionand and worthy of further study.
8.Effect and mechanism of PEDF on proliferationof human umbilical veinendothelial cellsand lung cancer SK-MES-1 cells

Objective To investigate the effect and potential mechanism of pigment epithelium derived factor(PEDF) acting upon SK-MES-1 cell and human umbilical vein endothelial cells(HUVECs).Methods CCK-8 was used to detect the effect of varying concentrations of PEDF upon HUVECs and SK-MES-1 cell, measuring the degree of cell proliferation and inhibition effect across varying times.The flow cytometry tests were carried out to invest gate the apoptosis of these two kinds of cells when exposed to varying concentration of PEDF.qRT-PCR were carried out to assess the vascular endothelial growth factor(VEGF) gene expression level in these two kinds of cells after treatment of PEDF.Results CCK-8 results revealed that PEDF had a concentration-dependent and time-dependent cell proliferation inhibition effect on SK-MES-1 cell and HUVECs(P<0.05);Flow cytometry showed that the apoptosis of the cells in the treatment group were higher than that of control group(P<0.05), and the apoptosis rate of high concentration group was higher than that of the low concentration group(P<0.05);qRT-PCR results showed that PEDF was able to inhibit expression of mRNA of VEGF in both HUVECs and SK-MES-1 cell compared with control samples(P<0.05).Conclusion The antitumor properties of PEDF is mainly related to the inhibition of tumor angiogenesis and direct effects on tumor cells, the effect of PEDF on HUVECs and SK-MES-1 cell maybe related to the effects of PEDF on downregulating expression of VEGF.
