1.Experiment of turning scleral flap of self-body over drainage in trabeculectomy of glaucoma

Objective To investigate the effect of turning scleral flap of self-body over drainage in trabeculectomy.Methods The surgery was performed on one eye of a rabbit while the other eye of the same rabbit was conducted with the routine trabeculectomy as the control group. The filtering bleb, intraocular pressure and pathology were observed. Results The intraocular pressure in the experimental eye was much lower than that in the control group 3~12 weeks after the operations (P<0.05).As it was within 2 weeks after the surgeries, but there was no significant filtering difference (P>0.05).The rate of eyes with functional bleb in the experimental group was significantly higher than that in the control group at the postoperative 2~7 weeks .Conclusion This study suggests the turning of self-body over drainage surgery can help to maintain filtering function for a long time and to prevent complication after filtering surgery.
2.Inhibition of the expression of inflammation cytokines by rhubarb in experimental acute necrotizing pancreatitis rats

Objective To observe the changes of inflammation cytokines in pancreas tissues of experimental acute necrotizing pancreatitis (ANP) rats treated with rhubarb.MethodsThirty-three rats were randomly divided into sham operation group,ANP group and rhubarb treatment group with 11 rats in each group.ANP model was induced by retrograde injection of 3% sodium taurocholate into the biliopancreatic duct,and jejunostomy was performed.Rats in rhubarb treatment group were fed with 1 g/ml rhubarb at the dose of 1 ml/kg body weight via jejunum route.Thirty-six hours later the rats were sacrificed.The serum level of amylase was measured; part of the pancreatic tissue was harvested for routine pathologic examination; total RNA was extracted from pancreatic tissue.The expression of IL-6,IL-8,and TNF-α mRNA was measured by real-time PCR.ResultsAfter ANP induction,the serum level of amylase was significantly increased,and pancreatic tissue necrosis,bleeding and inflammatory cell infiltration were observed,which was consistent with changes of ANP.The expressions of IL-6,IL-8,and TNF-α mRNA were 0.29 ±0.13,0.35 ±0.15,1.09 ±0.32 in sham operation group,while they were 2.23 ±0.49,2.26 ±0.51,5.24 ±0.59 in ANP group,which were significantly higher than those in sham operation group ( P < 0.05 ).The corresponding values were 0.97 ±0.30,1.02 ±0.34,2.59 ±0.36 in rhubarb treatment group,which were significantly lower than those in ANP group,but they were still significantly higher than those in sham operation group ( P < 0.05 ).ConclusionsRhubarb lavage via jejunum route can decrease the expressions of IL-6,IL-8,and TNF-α,therefore attenuate the pancreatic pathologic injuries.

4.Computer aided design and rapid manufacturing of removable partial denture frameworks.

OBJECTIVETo introduce a method of digital modeling and fabricating removable partial denture (RPD) frameworks using self-developed software for RPD design and rapid manufacturing system.
METHODSThe three-dimensional data of two partially dentate dental casts were obtained using a three-dimensional crossing section scanner. Self-developed software package for RPD design was used to decide the path of insertion and to design different components of RPD frameworks. The components included occlusal rest, clasp, lingual bar, polymeric retention framework and maxillary major connector. The design procedure for the components was as following: first, determine the outline of the component. Second, build the tissue surface of the component using the scanned data within the outline. Third, preset cross section was used to produce the polished surface. Finally, different RPD components were modeled respectively and connected by minor connectors to form an integrated RPD framework. The finished data were imported into a self-developed selective laser melting (SLM) machine and metal frameworks were fabricated directly.
RESULTSRPD frameworks for the two scanned dental casts were modeled with this self-developed program and metal RPD frameworks were successfully fabricated using SLM method. The finished metal frameworks fit well on the plaster models.
CONCLUSIONSThe self-developed computer aided design and computer aided manufacture (CAD-CAM) system for RPD design and fabrication has completely independent intellectual property rights. It provides a new method of manufacturing metal RPD frameworks.

5.Electronic surveying of digital partially dentate casts.

OBJECTIVETo develop a method of surveying the digitalized partially dentate cast in order to accomplish computer aided design (CAD) of removable partial denture (RPD) frameworks.
METHODSThe stone cast of a partially dentate patient was scanned using a three-dimensional laser scanner. Points on the surface of the digitalized cast, termed as a "point cloud", were obtained. The point cloud was then imported into self-developed Tanglong software to sample and uniform. New module of identifying the surveying lines was specially written in Tanglong using C++. A straight line was created in the center of the cloud point. Then the cloud point surrounding the straight line was divided into many parts. Local coordinates were established to indicate the information of angle and distance of every point to the straight line. Surveying lines were produced step by step electronically by identifying the closest and farthest points relative to the straight line and then connected together. Different surveying lines were obtained by adjusting the angle of the straight line. After the surveying lines were decided, the undercut areas could be marked and the depth of undercut was calculated automatically. The blockout of the undercuts could also be achieved by moving the location of the point cloud in undercut area.
RESULTSSurvey lines of digitalized partially dentate casts were generated in computer. The undercut area and its depth were identified and the undercut could be blocked out. The shape of survey lines on the digitalized casts was similar to that on the physical casts drawn using traditional method.
CONCLUSIONSThe new module in Tanglong software was developed specifically for surveying partially dentate casts. It had a user-friendly interface with the easy-to-understand menus. The success of surveying dental casts digitally would make it possible to CAD of RPD frameworks.

6.Femtosecond pulsed laser ablation of dental hard tissues with numerical control: a roughness and morphology study.

OBJECTIVETo establish the femtosecond laser experimental platform in vitro for numerical controlled cavity preparation, and to evaluate the roughness quantitatively and observe the microscopic morphology of the cutting surface.
METHODSEnamel and dentin planes were prepared on human third molars. A universal motion controller was used to control the samples to do rectangle wave motion perpendicular to the incident direction of the laser at focus. The surface roughness was observed with confocal laser scanning microscope.
RESULTSPrecise ablation of the dental hard tissues can be achieved with the established femtosecond laser numerical control platform. For enamel, the surface roughness of the cavity inside laser scanning line was 7.173 µm at the bottom and 2.675 µm on the wall of the cavity. The surface roughness of the cavity between laser scanning lines was 13.667 µm at the bottom and 33.927 µm on the wall. For dentin, the surface roughness of the cavity bottom was 51.182 µm and 25.629 µm for the wall. Scanning electron microscope images showed no micro-cracks or carbonization on enamel, while carbonization, cracks and a small amount of crystalline particles were observed on dentin.
CONCLUSIONSPrecise tooth preparation can be achieved with femtosecond laser numerical control flatform. The surface roughness of cavity wall was less than that of the bottom and can meet the clinical needs. Suitable femtosecond laser output power should be set for different cutting objects, otherwise it may result in tissue damages.

7.XBP-1 interacts with estrogen receptor alpha (ERalpha).

Estrogen receptor alpha (ERalpha) has been a primary target of treatment as well as a prognostic indicator for breast cancer. The level of human X-box binding protein 1 (XBP-1) mRNA was related with that of ERalpha in breast tumors and was over-expressed in some breast tumors. These previous studies suggested that XBP-1 may interact with ERalpha. XBP-1 has two isoforms, XBP-1S and XBP-1U, as the result of unique splicing. GST pull-down assay showed that both XBP-1S and XBP-1U bound to ERalpha in vitro. The binding of XBP-1S to ERalpha was stronger than that of XBP-1U to ERalpha. Co-immunoprecipitation revealed that the binding was in a ligand-independent manner. XBP-1S and XBP-1U interacted with the region of ERalpha that contains a DNA-binding domain. The ERalpha-interacting regions on XBP-1S and XBP-1U have been mapped to two regions, the N-terminal basic region leucine zipper domain (bzip) and the C-terminal activation domain. These findings suggest that XBP-1S and XBP-1U may participate in ERalpha signaling pathway through the mediation of ERalpha.

8.A computer aided design approach of all-ceramics abutment for maxilla central incisor.

OBJECTIVETo establish the computer aided design (CAD) software platform of individualized abutment for the maxilla central incisor.
METHODSThree-dimentional data of the incisor was collected by scanning and geometric transformation. Data mainly included the occlusal part of the healing abutment, the location carinae of the bedpiece, the occlusal 1/3 part of the artificial gingiva's inner surface, and so on. The all-ceramic crown designed in advanced was "virtual cutback" to get the original data of the abutment's supragingival part. The abutment's in-gum part was designed to simulate the individual natural tooth root. The functions such as "data offset", "bi-rail sweep surface" and "loft surface" were used in the process of CAD.
RESULTSThe CAD route of the individualized all-ceramic abutment was set up. The functions and application methods were decided and the complete CAD process was realized.
CONCLUSIONSThe software platform was basically set up according to the requests of the dental clinic.

9.Transmural heterogeneity of calcium handling proteins in the mechanism of porcine model of ventricular fibrillation.

OBJECTIVETo investigate the role of heterogeneous expression of calcium handling proteins and spatial heterogeneity of APD restitution in the maintenance mechanism of ventricular fibrillation.
METHODSDuring programmed electrical simulation, APD restitution curves in the endocardium and epicardium of LV were constructed by plotting APD100 and diastolic interval. APD alternans, delayed after depolarization events were recorded simultaneously. Endocardial and epicardial myocytes were isolated from LV base and apex. Real time-PCR and Western blotting were performed to determine the relative messenger RNA and protein expression levels of calcium handling proteins.
RESULTSThe normal hearts have spatial heterogeneity of action potential restitution property and transmural heterogeneity of calcium handling proteins. The slopes of the APD restitution curve in the endocardium were significantly steeper than those in epicardium, and the slopes of APD curve at the LV apex were significantly steeper than those in LV base. However, delayed after depolarization events with larger amplitude and earlier onset consistently occurred in the endocardium of LV base. After programmed electrical simulation, the expression of messenger RNA of RyR2, SERCA2a except for Calstabin2 significantly decreased (by 57% and 41%, respectively, P < 0.05) in the endocardium of the base, while the expression of RyR2, SERCA2a, Calstabin2 significantly increased (by 90%, 78%and 64%, respectively, P < 0.05) in the epicardium of LV base. Although transmural heterogeneity of calcium handling proteins at the LV apex were also observed after rapid pacing, there is no significant differences in the transmural heterogeneity at the LV apex compared to the LV base. The base of LV has unique calcium handling properties.
CONCLUSIONSIt has been shown that Calcium cycling could modulate APD restitution property in the intact heart. The interaction between action potential and calcium dynamics instabilities is one of the most important reasons why simple criterion such as the APD restitution slope > 1 may fail to accurately predict the onset of APD alternans.

10.Separation and amplification of CD4(+)CD25(+) regulatory T cells from sensitized mice.

The aim of this study was to separate and amplify CD4(+)CD25(+)Treg cells from splenocytes of sensitized mice. The percentage of CD4(+)CD25(+)Treg cells was detected by flow cytometry in sensitized and normal control mice. CD4(+)T, CD4(+)CD25(+)Treg and CD4(+)CD25(-) T cells were isolated from mouse splenocytes by MACS. CD4(+)CD25(+)Treg cells were expanded in vitro cultures in addition of CD3/CD28 MACSiBead and IL-2. The activity of cells was detected with 0.4 trypan blue staining. The purity of cells after sorting, the main surface marker and the level of Foxp3 were detected by flow cytometry. The results showed that CD4(+)CD25(+)Treg cell proportion was higher in sensitized mice than normal control mice (P < 0.05). The average purity of CD4(+)CD25(+)Treg cells was 87. The activity of these cells was more than 97, and the expression of Foxp3 in these cells was high. The amplification multiples achieved 42 times after 2 weeks in vitro. The percentage of CD4(+)CD25(+) regulatory T cells was 85.32, and the expression of Foxp3 decreased from (76.92 ± 1.72) to (75.33 ± 2.11) (P > 0.05). It is concluded that the sorting of CD4(+)CD25(+)Treg cells is isolated successfully by MACS without affecting the vitality of target cells. The amplification of CD4(+)CD25(+)Treg cells is successful in vitro. Expression of surface markers and Foxp3 gene does not obviously change after amplification, so that to establish a practical method to recover and enlarge the amount of CD4(+)CD25(+)Treg cells in good condition.
