1.Diagnosis and Treatment of Small Intestinal Atresia in Neonates

Objective To explore the clinical feature,treatment and prognosis of neonatal small intestinal atresia.Methods Retrospective study was carried out of 80 neonatal small intestinal atresias over the last 14 years,including diagnosis,treatment and prognosis.Results Of them,62 cases were healed(77.5%),18 cases were dead(22.5%),24 cases(30%)associated with other anomalities.The living cases were followed up for 6 months to 3 years.These symptoms had not relapse.Conclusions Satisfactry effect can be obtained by early diagnosis and operative treatment.Sonography is an ideal way for early finding intestinal atresias in a fetus.End-to-end single layer anastomosis is an ideal way for monointestinal atresia,and poly-end-to-end single layer anastomosis plus long gastrointestinal silicone tube for supporting is an ideal way for multipal intestinal atresia.
2.Effects of valsartan on adrenomedullin receptor expression in diabetic rat's renal cortex

Objective Valsartan, the angiotensin Ⅱ type 1 receptor blocker, is recently proved to reduce urinary albumin at the microalbuminuria stage in human diabetic nephropathy without altering glucose metabolism. But the pathway is still uncertain. In present study, we examined the changes of adrenomedullin receptor (ADMR) mRNA and protein expressions in the renal cortex of diabetic rats to investigate the protective effects of valsartan on an experimental model of diabetic renal injury. Method The SD rats were randomly divided into following groups: normal rats, STZ-induced diabetic rats, and diabetic rats treated with valsartan. STZ-induced diabetic rats were treated with valsartan (10mg/kg body weight) or vehicle for 8 weeks. The expressions of ADMR mRNA in renal cortex were analyzed by RT-PCR, as well as ADMR protein expressions were detected through western blot. Results We found (1) Valsartan treatments reduced urinary albumin excretion in 24h, compared with the untreated. But no notable difference was seen in HbA1c and blood sugar of diabetic rats between the two groups. (2) Valsartan treatments increased the expressions of ADMR mRNA and protein in diabetic rats renal cortex. Conclusion These results indicate that valsartan treatment can upgrade the expressions of ADMR in the renal cortex of diabetic rats. It may be one of renal protective pathways of Ang Ⅱ type 1 receptor blocker.

4.Enhancing effect of DHEA ON DbcAMP-induced cell differentiation

Aim To observe whether DHEA has enhancing effect on DbcAMP -induced differentiation of NG108-15 cells, including neurite outgrowth, and study its possible mechanisms. Methods NG108-15 cells (a h ybrid cell line of mouse neuroblastoma and rat glioma) were used as a substitute for primary culture neuron in vitro. The morphology of NG108-15 cells was o bserved and neurite outgrowth was determined in an inversed microscope after treatme nt with various drugs. Gelatin-substrate gel electrophoresis was used to detect gelatinases (MMP-9 and MMP-2). Results ① DHEA and DbcAMP inhibited NG108-15 proliferation.②DHEA had enhancing effect on the promoting activity of neuronal differentiation and neurite outgrowth by DbcAMP. DbcAMP could increase neurite elongation of NG108-15 cells. Compared with this, the combined treatment with DHEA and DbcAMP significantly enhanced the neurite outgrowth of NG108-15 cells, including neurite length and numbers of cells with neurite, in a DHEA dose-dependent manner. ③ MMPs were involved in neuronal differentiation. DbcAMP induced the increase in MMP-9 and MMP-2 activities and such elevation was enhanced by DHEA in a dose-dependent manner. Conclusion DHEA enhances the effect of DbcAMP in promoting the neurite outgrowth of NG108-15 cells, which might be related to the increase in MMP-9 and MMP-2 activities.
5.Pathological observation of two huge spontaneous tumors in a Wistar and a GK rats

Objective To study the pathological features of two huge spontaneous tumors in Wistar and GK rats. Methods Forty Wistar rats and 40 GK rats were included in this study. Among those rats, two huge spontaneous tumors were observed in a Wistar rat at 14 months of age and in a GK rat at 22 months of age. The growth and survival status of the tumor?bearing rats were recorded. The tumors were surgically removed, and their pathological features were examined using HE and immunohistochemical staining (vimentin, CK19, α?SMA, CD31, CD34, S?100, NF及Ki?67). Results Both the two tumors were completely resected by surgery without much difficulties, and both host rats survived well after the operation. The weight of the two huge tumors was 502 g and 119 g, which corresponding to 64% and 24% of the body weight of their host rats, respectively. The tumors surface had a complete capsule, with a clear boundary separating from the normal surrounding tissues, and no vascular pedicle structure was found. According to the results of immunohistochemical staining, both the two tumors were diagnosed as benign fibroma. Conclusion This type of huge spontaneous tumors is benign fibroma. Besides the impact on the activities of the rats, the tumors have no significant impact on the living conditions in the hosts.
6.Cloning and Expression of ywtD Gene from B. subtilis NX-2 and the Enzymatic Degradation of ?-Polyglutamic Acid

The B.subtilis ywtD gene,encoding a ?-polyglutamic acid(?-PGA)depolymerase,was amplified from the genome of B.subtilis NX-2 by PCR.The comparability between the cloned ywtD gene sequence to the reported sequence is high to 99.0%.Only one of the substituted nucleotide base caused the change to the amino acid sequence.The recombinant plasmid pET-15b-ywtD was then transformed into E.coli Rosetta(DE3)and the ywtD gene product could be expressed with the induction of 0.5mmol/L IPTG.The YwtD protein exhibited a remarkable activity in ?-polyglutamic acid degradation.The molecular weight of ?-PGA could be reduced from 700kDa to 20kDa after 72h through the enzymatic hydrolysis and consequently trended to be constant.

8.Preparation, characterization and in vitro evaluation of poly(2-ethyl-2-oxazoline) modified curcumin liposomes

Objective To prepare Curcumin liposome (Cur-L) and poly(2-ethyl-oxazoline)-cholesteryl methyl carbonate (PEOz-CHMC) was used to modified Cur-L and to evaluate their associated properties in vitro.MethodsEncapsulation efficiency and particle size were taken as evaluation indicators to optimize the formulation and preparation conditions of Cur-L by orthogonal test.The EE, particle size and shape of the liposomes were determined by sephadex G-50 mini-column centrifugation method, ZLS dynamic light scattering instrument and transmission electron microscopy (TEM), respectively.The release of the liposome in vitro was detected by The dialysis method.MTT assay was used to determine the cell inhibition of two Cur-L.ResultsThe optimized preparing method of Cur-L is as following: 1.56(w/w) as drug-lipid ratio, 5.1(w/w) as the ratio of mass of phosphatide and cholesterol, 7.4 as the pH of PBS buffer.The EE of Cur-L was (75.05±0.64)%, while the modification of PEOZ hasno influences on EE.Through TEM, PEOZ-Cur-L has aobviouslipid bilayer structure.The average particle diameter of PEOZ-Cur-L was 84.89 nm.In vitro release experiments showed that in 24h, the accumulative release rate of Cur-L is more than 70% with pH 7.4, while that of PEOZ-Cur-L was less than 25%.The cytotoxicity experiment showed that PEOZ-Cur-L can inhibit HCT116 Human colon cancer cells more effectively.ConclusionThe optimized preparing method of Cur-L is reasonable.PEOZ can provide stability to liposomes well and does not hamper its inhibitive effects.
9.Heme oxygenase-1 regulates oxidant and antioxidant imbalance in guinea pigs with asthma

Objective To explor the antioxidant effects of heme oxygenase-1(HO-1) for asthma and its mechanism.Methods Thirty two months old healthy male guinea pigs were randomly assigned to 3 groups,all of them were feed under the same management condition.Asthmatic group,refering to Huston′s research results,were sensitized by exposure to aerosolized ovalbumin(OVA) in order to build the guinea pig modle of asthma.Dexamethasone group(DG),were also sensitized by exposur to aerosolized ovalbumin as well as the asthmatic group had done and then treated with dexamethasone.Normal control group(NCG):were treated with 0.9% sterile sodium chloride.All the tested animals were measured the activity of lung HO-1,levels of malondialdehyde (MDA) in the lung tissue or serum and bilirubin in the serum by spectrophotometry.And the pathology and expression of HO-1protein in the lung tissue were observed.Results In the OVA induced asthmatic group,the HO-1 activity and level of MDA in lung tissue or serum were markedly increased compared with the groups of NCG and DG (P<0.01) and the expression of lung HO-1 protein was as 3~4 grade.While level of bilirubin was markedly reduced compared with NCG and DG.For example,in asthmatic group,the activity of lung HO-1 was,the level of bilirubin formed,(901.12±321.53)pmol/(mg MDA was(69.38±7.49)nmol/mg protein,and the level of bilirubin in serum was (0.54±0.23)pmol/L.In DG,the data of HO-1 and MDA as described above were markedly reduced compared with the OVA induced asthmatic group(P<0.01),and lung protein expression was 1~2 grade,while the level of bilirubin in the serum was increased(P<0.01).But,all datas of DG had no significant difference compared with NCG(P>0.05).Conclusion The guinea pigs′ in vivo lipid peroxide level increases and the level of bilirubin,an antioxidant,markedly decrease during asthma attacks,which induce the imbalance of oxidant and antioxidant.Content level and activity of HO-1 protein can increase in asthma body in order to compensate the reduce of bilirubin level,regulate the imbalance of oxidant and antioxidant,protect lung tissues from oxidizing reaction damage and relieve allergy or asthma symptoms.
10.Analysis on Salmonella carrying situation among employees in Chengdu railway area during 2012-2015

Objective To understand the Salmonella carrying situation among the employees of public places and dietetic in dustries in Chengdu railway area to enhance the health management among employees in catering and service industries and provide the basis for preventing the occurrence and outbreak of food poisoning.Methods The anal swab culture was adopted to perform the health carriage examination among the employees of public places and dietetic industries during 2011-2015.The isolated Salmonellas were identified by stereotyping and the relationship between Salmonella carrier state with seasons and years was analyzed.Results Among 86 971 detected samples,43 strains of Salmonella were detected with the detection rate of 0.49‰.Nineteen serotypes were identified,which were dominated by Salmonella derby,Salmonella Senftenberg and Salmonella typhimurium.The highest detection rate was in summer and autumn.Conclusion The stains types of Salmonella carried by employees in catering and service in dustries in Chengdu railway area were complex,and the detection rate was closely related to seasonal variation.It is needed to strengthen the health examination and prevent food poisoning occurrence.
