Objectives To study the expression features of C-type natriuretic peptide (CNP)/natriuretic peptide receptor-B (NPR-B) axis and two parallel elimination pathways, natriuretic peptide receptor-C (NPR-C) and neutral endopeptidase (NEP) in unilateral ureteral obstruction (UUO) rats. Methods CNP, NPR-B, NPR-C, NEP, Col-IV and type IV collagen (Col-IV) mRNA and proteins were determined by in situ hybridization, real-time PCR, immunohistochemistry and western blot in UUO rats at 24h, 72h, 1w, 2w, 3w, 1m, 2m and 3m. Results CNP expression tended to be higher immediately after ligation and de-clined along with the progression of disease, occurring predominantly in tubular epithelial cells. A high-level CNP may attribute to the elevated expression of NPR-B in the early phase of UUO. Conclusions NEP and NPR participate in the regulation of CNP expression in tubulointerstitial fibrosis. The gradual increased expression of NPR-C and NEP may cause the subsequent de-cline of CNP.

Objective To compare the QTc and QTcd between type 2 diabetic and non-diabetic patients with post-myocardial infarction (post-MI) ,and to compare the QTcd in type 2 diabetic patients with post-MI treated with insulin, sulfonylurea, mefformin,or diet alone. Methods We measured the QTc and QTcd through simultaneous 12-lead Electrocardiogram in 138 post-MI patients, including 70 type 2 diabetic (of which,23 were assigned to re-ceive insulin,20 glipizide,16 mefformin,11 diet control) and 68 non-diabetic patients. Result Compared with post-MI patients without diabetes, those with type 2 diabetes had significantly higher QTc [(377.2±24.3) ms vs (342.9±27.5)ms,t=7.79,P<0.01] and QTcd [(48.8±19.7)ms vs (40.3±26.6)ms,t=2.14,P<0.05]. There were no significant difference between the mefformin group and the diet control group (P>0.05). The QTc and QTcd in the insulin group were significantly higher than those in the other three group s(P<0.05),and the QTc and QTcd in the glipizide group were higher than those in the mefformin group or diet control group(P<0.05,and P<0.01,respectively). Conclusion Type 2 diabetes is associated with an additional increase in the QTcd in post-MI patients, suggesting higher mortality risk in post-MI patients with type 2 diabetes. Insulin and glipizide may in-crease the QTc and QTcd in post-MI patients with diabetes. These effects were more significant in the insulin therapy group.

Objective To investigate the differences in dosimetry between conformal radiotherapy (CRT),field-in-field intensity-modulated radiotherapy (FIF-IMRT),and intensity-modulated radiotherapy (IMRT) after breast-conserving surgery for left-sided breast cancer. Methods A total of 31 patients who underwent breast-conserving surgery for left-sided breast cancer were randomly selected, and the plans for CRT,FIF-IMRT,and IMRT were developed. The dose-volume histogram (DVH) was used for self-control study, and the non-parametric test was used to compare the differences in target volume and doses to organs at risk (OARs).Results All the three methods met the requirements of the prescribed doses. The CRT group had a higher V105 of the target volume and higher heart V30 and Dmax (P=0.000,0.000,0.000).The IMRT group had higher V5 and Dmean (P=0.000,0.000),as well as a higher lung V5 and a lower lung V40 (P=0.000,0.000).The FIF-IMRT group had the lowest Dmean (P=0.000),and the IMRT group had significantly higher Dmean and Dmax of the right lung and the spinal cord than the other two groups (P=0.000,0.000,0.000,0.000).The FIF-IMRT group had a significantly lower single hop count than the other two groups (P=0.000).Conclusions CRT has a good dose distribution in the target volume, but greatly injures the surrounding tissues.FIF-IMRT can well protect OARs and cause less damage to the device.

With the advance of drug development and research techniques, the drug metabolic processes and mechanism can be more deeply achieved. As the drug metabolism and pharmacokinetics process are mediated by drug metabolizing enzymes and transporters, study of drug metabolizing enzymes and transporters has become an important part for drug development. The traditional immunoassays with low sensitivity and poor specificity can not reflect the accurate expression level of drug metabolizing enzymes and transporters. We now give a brief review on the quantitative study of drug metabolizing enzymes and transporters by mass spectrometry-based proteomic approach.

Acta Pharmaceutica Sinica 2004;39(9):730-733
AIMTo study the electrochemical behavior of daunorubicin at Co/GC ion implantation modified electrode.
METHODSWith Co/GC ion implantation modified electrode as working electrode, daunorubicin was determined by voltammetry in 0.05 mol x L(-1) Na2HPO4-KH2PO4 (pH 6.82) solution.
RESULTSA sensitive reductive peak of daunorubicin was obtained by linear sweep voltammetry. The peak potential was -0.60 V (vs SCE). The peak current was proportional to the concentration of daunorubicin over the range of 2.84 x 10(-8) - 1.42 x 10(-6) mol x L(-1) and 1.42 x 10(-6) - 1.28 x 10(-5) mol x L(-1) with the detection limit of 1.42 x 10(-8) mol x L(-1). The reduction wave was applied to the determination of daunorubicin. The electrochemical behavior and reaction mechanism were studied by linear sweep and cyclic voltammetry.
CONCLUSIONThe reduction process was quasi-reversible with adsorption characteristics.
