1.Influence of Asphyxia on Nitric Oxide and Nitric Oxide Synthase Levels in Renal of Neonatal Rats

Objective To study the effects of nitric oxide synthase(NOS) and nitric oxide(NO) in post-asphyxial renal injury in neonatal rats.Methods Forty-eight Wistar neonatal rats were randomly divided into 4 groups:controls,2 h,24 h and 48 h post-asphyxia groups (12 in each group).The rats were decapitated in different times(2 h,24 h and 48 h) after asphyxia for 30 minutes.The renals were dissected to determined the concentrations of NO and NOS.And the scores of renal tubules were measured under light microscope.Results Compared with control group,the levels of NO and NOS significantly increased at 2 h and 24 h after asphyxia.The scores of renal tubules were significant difference at 24 h and 48 h after asphyxia compared to controls.Conclusion These findings suggest NOS and NO may play an important role in the development of post-asphyxia renal injury.
3.Effect of zhenggu powder (Chinese characters: see text) on the inflammatory cell factor of soft tissue in experimental acute injury.

OBJECTIVETo probe the recovery mechanism of Zhenggu powder (ZGP) on acute soft tissue injury in cell levels.
METHODSForty rabbits which established animal model of acute soft tissue injury with hammer hitting,were divided randomly into normal group (A), model group (B), vaseline group (C)and ZGP group (D). Injured part was applied external drug daily after model was made. All animals were killed after using drug for 4 days. The local tissue of injured part was taken pathologic study, and was measured the content of IL-1beta, IL-6, TNFalpha by ELISA method and TXB2, 6-keto-PGF1alpha by RIA method.
RESULTSMuscular tissue of group A was normal. But that of group B and C was aberrant,such as swelling and broken of muscular fiber or infiltration of inflammatory cell. Such histological change of group D was lightly and hyperplasia of blood vessel was found. The contents of IL-1beta, TNFalpha, TXB2 and TXB2/6-keto-PGF1alpha in group D were lower than that of group B and group C. On the contrary, the contents of 6-keto-PGF1alpha in the group D were higher than that of group B and group C. The difference of content of IL-6 between groups was not obvious.
CONCLUSIONZGP could promote not only the dilution and the transportation of inflammatory cell factors,but also the repair and the regeneration of the injured tissue structures. The therapeutic effect of ZGP was not relative to IL-6.

We recently found that calcitonin gene-related peptide (CGRP)-like immunoreactivity was present in lymphocytes of rat thymus and lymph node. In order to investigate whether these cells were capable of synthesizing CGRP, CGRP mRNA of rat dorsal root ganglia, thymocytes and mesenteric lymph node lymphocytes were determined by reverse transcription and polymerase chain reaction (RT-PCR) utilizing synthetic oligonucleotides bracketing a portion of the calcitonin/CGRP gene. A discrete band of the expected size of 90 base pairs was found in the dorsal root ganglia (positive control), and in both thymocytes and mesenteric lymph node lymphocytes. These data strongly suggest that CGRP is not only an important neuropeptide, but it is also synthesized in lymphocytes of both thymus and lymph nodes, which is identical to that in sensory neurons. CGRP from lymphocytes may act as an immunomodulator and serve as a common ligand in immune and nervous systems.
7.Expression of CK20 mRNA in the peripheral blood around postoperative chemotherapy in patients with gastric cancer and its clinical significance.

OBJECTIVETo investigate whether CK20 mRNA expression level could be considered as an effective molecular indicator for evaluation of chemotherapy sensitivity.
METHODSAll samples of peripheral blood were taken from 31 gastric cancer patients undergone radical operation a week before postoperative chemotherapy, at the first day of chemotherapy point, and after the first cycle of chemotherapy respectively, and subjected to FQ RT-PCR assay for CK20 mRNA. The chemotherapy scheme was FOLFOX 4. The control group was 15 healthy volunteers.
RESULTSAomng the 31 gastric cancer patients, the value of CK20 mRNA before postoperative chemotherapy was increased (2.96+/-2.27 vs 2.22+/-2.12, t=2.10, P<0.05) in 25 positive cases, and then declined after chemotherapy(2.05+/-1.86 vs 2.96+/-2.27, t=2.50, P<0.05) in 24 positive cases. The expression level of CK20 mRNA in patients before chemotherapy was increased in 16 cases(51.6%), declined in 9 cases(29.0%) and stabilized as negative in 6 cases(19.4%). After chemotherapy the level of CK20 mRNA was increased in 7 cases(22.6%), declined in 17 cases (54.8%) and stabilized as negative in 7 cases(22.6%), there was significant difference between the two groups(chi(2)=6.06, P<0.05).
CONCLUSIONSThe expression level of CK20 mRNA in the peripheral blood detected by FQ RT-PCR in patients with gastric cancer declines after postoperative adjuvant chemotherapy. Different individuals have different sensitivity to chemotherapeutics. Dynamic monitoring CK20 mRNA should be considered as an effective index to evaluate the efficacy of postoperative adjuvant chemotherapy.

8.Clinical features of children with incomplete Kawasaki disease.

OBJECTIVETo investigate the clinical features of children with incomplete Kawasaki disease (IKD), and to provide reference for the early diagnosis of IKD.
METHODSThe clinical data of 22 hospitalized children with IKD were analyzed retrospectively and compared with the data of 63 children with Kawasaki disease (KD) who were hospitalized during the same period of time. Another 20 children with pyrexia were enrolled as the control group.
RESULTSPyrexia was observed in all children. Compared with the KD group, the IKD group had significantly lower proportions of children with changes in the limbs, conjunctival hyperaemia, and cervical lymphadenectasis (P<0.05), a significantly higher serum level of glutamic-pyruvic transaminase (P<0.05), and significantly lower levels of plasma albumin, serum sodium, and interleukin-6 (P<0.05). There was no significant difference in the rate of γ-globulin application between the IKD and KD groups; however, the IKD group had a significantly higher incidence rate of coronary artery lesion than the KD group (P<0.05).
CONCLUSIONSThe symptoms and signs in children with IKD are untypical. The liver function test and serum hyponatremia and IL-6 measurements may be useful for the diagnosis of IKD.

9.Investigation on occupational hazards of ultraviolet light, sunscreen awareness and behaviors in Wuhan city traffic police.

OBJECTIVETo understand the awareness of occupational hazards to ultraviolet (UV) and sunscreen awareness, protective measures in Wuhan City traffic police on duty outside.
METHODSThe investigation included questionnaire survey in Wuhan City 367 traffic police on duty outside, talk with them face to face, fill in the questionnaires, and medical examine skin of exposed parts of body of them and 134 Wuhan City administration staffs.
RESULTSThey understand UV harm to the human body and skin well (94.8% of them know that UV harm to skin), did not understand sun skin care and protective measures enough, and did not adopt enough sun skin care and protective measures (only 3.8% of them use sun skin care more than twice); but contrast to older persons, younger traffic police had better understanding of UV radiation damage on the human body and the skin, and sunscreen products and protective measures, paid more attention to sunscreen, and had less chance of sunburn (in the past 5 years, 18.3% of younger traffic police had sunburnt more than 3 times, but for older traffic police, the number is 30.3%). Traffic police had more skin problems than administration staffs in exposed parts of body (Traffic police face appears oily and large pores, facial pigmentation spots, face telangiectasia, deep wrinkles crude rates respectively were 73.7%, 40.4%, 36.5%, 10.4%, but for administration staffs, the numbers respectively were 26.1%, 15.7%, 15.7%, 1.5%).
CONCLUSIONUV can induce skin problems in exposed parts of body. The traffic police should be enhanced the publicity and education on UV-related knowledge and occupational hazards, especially for older traffic police.

10.Association between surfactant protein B gene polymorphisms and susceptibility to bronchopulmonary dysplasia in preterm infant

Objective To investigate the distribution of surfactant protein B (SP-B) gene single nucleotide polymorphisms and to study the association between the SP-B gene polymorphisms and bronchopulmonary dysplasia (BPD) in preterm infant.Methods Forty-two preterm infants with BPD and 68 preterm infants without BPD (control group) were selected.Polymerase chain reaction with restriction fragment length polymorphism was used to establish the genotype and allele frequencies of the SP-B-18A/C,SP-B 1580C/T,SP-B 8714G/C single nucleotide the polymorphisms,and then the association between the polymorphisms and BPD was analyzed.Results The frequencies of genotype CC,AC,AA of SP-B-18A/C in BPD group and control group were 35.7%,52.4%,11.9% and 32.3%,47.7%,20.0%,respectively.The frequencies of genotype CC,GC,GG of SP-B 8714G/C in BPD group and the control group were 26.2%,54.8%,19.0% and 27.7%,58.5%,13.8%,respectively.The frequencies of genotype CC,CT,TT of SP-B 1580C/T in BPD group and control group were 66.7 %,26.2%,7.1% and 40.0%,47.7 %,12.3 %,respectively.The frequencies of allele C and allele A of SP-B-18A/C in BPD group and control group were 61.9%,38.1% and 56.2%,43.8%,respectively.The frequencies of allele G and allele C of SP-B 8714G/C in BPD group and control group were 53.6%,46.8% and 56.9%,43.1%,respectively.The frequencies of allele C and allele T of SP-B 1580C/T in BPD group and control group were 79.8%,20.2% and 63.8%,36.2%,respectively.SP-B-18A/C,SP-B 1580C/T,SP-B 8714G/C were found to be polymorphic.The frequencies of genotype CC of SP-B 1580C/T in BPD group were significantly higher than those in the controls (x2 =7.26,P < 0.05).The frequencies of allele C of SP-B 1580C/T were also statistically different between BPD group and control group(x2 =6.17,P <0.05).The C allele increased the risk of BPD(OR =2.23,95% CI:1.18-4.24).There was no significant difference between the 2 groups in the frequencies of genotype and allele of SP-B-18A/C and SP-B 8714G/C.Conclusions SP-B 1580C/T polymorphism has association with the etiology of BPD and may serve as the susceptibility gene for BPD,the C allele increase the risk of BPD.SP-B-18A/C and SP-B 8714G/C do not have association with the etiology of BPD.
