Objective:To study the inhibitory effect of siRNA on Heparanase (HPA) expression in SKOV3 cells. Methods:Two pairs of 21 bp reverse repeated sequence targeting HPA RNA (spaced by 9 bp nueleotide) were synthe- sized and were cloned into plasmid PGenesil-1 to construct recombinant plasmid PGenesil-1(+)-HPA expressing 2 hair- pin siRNAs.The inhibition of HPA gene was detected by RT-PCR,real-time PCR and immunohistochemical staining after PGenesil-1(+)-HPA was stably transfected into SKOV3 cells. Results:The recombinant plasmid PGenesil-1(+)-HPA (expressing 2 hairpin siRNAs) was successfully constructed.RT-PCR,real-time PCR and immunohistochemical staining showed that there was a significant decrease in HPA mRNA and protein level in experimental group compared with those in control group. Conclusion:siRNA targeting HPA mRNA can specically suppress the expression of HPA gene in SKOV3 cells;RNA interference method provides a new way for studying the role of HPA and gene therapy of cancer.

Objective To characterize non-headache symptoms during the migraine attack phase in migraine patients. Methods We enrolled in the present investigation 71 patients with migraine with and without aura who had sought medical treatment in our department from January 2010 to January 2011. We used a self-made migraine questionnaire to investigate associations of the non-headache symptoms with the sociodemographic factors, subtypes, severities and attacks of the headache in the patients. Results There were no significant differences in the number of non-headache symptoms among patients with different ages, genders, education levels, subtypes, severities, mean attack frequencies per month (P>0.05). The most frequently reported non-headache symptoms were nausea (87.3%),phonophobia (81.7%) and mood change (69.0%).The incidences ofphonophobia, sleep disorder and osmophobia were significantly higher in patients without aura than in patients with aura (P<0.05).Conclusion Migraine attacks are often accompanied by a variety of non-headache symptoms of which the most frequently reported ones are nausea, phonophobia and mood change.Patients without aura tend to have more phonophobia, sleep disorder and osmophobia than those with aura.

Objective To investigate the role of C-Jun N-terminal kinase (JNK) in epithelial mesenchymal transition (EMT) induced by transforming growth factor β1 (TGF-β1) in rat peritoneal mesothelial cells(RPMCs). Methods RPMCs were harvested from the peritoneum of male Sprague-Dawley rats, then cultured in DMEM/F12 medium with 15% (V/V) FBS. After stimulation with TGF-β1, the expression of a-smooth muscle actin (α-SMA), E-cadherin and collagen I were detected in RPMCs. In some groups, the ceils were pretreated with SP600125, a specific inhibitor of JNK, for 4 hours before incubation with TGF-β1. The protein expression of phosphorylated JNK was detected by Western blotting. The mRNA and protein expression ofα-SMA, E-cadherin and collagen I were examined with RT-PCR and Western blotting, respectively. The intracellular distribution and expression of α-SMA was determined by indirect immunofluorescence. Results TGF-β1 could significantly increase the expression of α-SMA and collagen I, and decrease the expression of E-cadherin in RPMCs. TGF-α1 could stimulate the expression of phosphorylated JNK at 5 minutes with the peak at 10 minutes (P<0.01). The addition of SP600125 effectively inhibited TGF-β1-induced high expression of α-SMA and collagen I (P<0.05), and prevented TGF-β1-induced down-regulation of E-cadherin expression in RPMCs (P<0.05). The indirect immunofluorescence showed that the expression of intracellular α-SMA in RPMCs stimulated by TGF-β1 for 48 h increased significantly, which could be inhibited by SP600125. Conclusions JNK regulates epithelial mesenchymal transition induced by TGF-β1 in rat peritoneal mesothelial cells. JNK inhibitor may be used as a novel therapeutic agent for peritoneal fibrosis.
10.Correlation between circadian rhythm and rapid palatal expansion.
Chinese Journal of Stomatology 2010;45(11):655-658
OBJECTIVETo investigate the correlation between circadian rhythm and rapid palatal expansion (RPE).
METHODSOne hundred and eleven consecutive patients (55 males and 56 females) using the same rapid palatal expansion devices were included in the study. The mean age of males was (11.2 ± 2.1) years and the mean age of the females was (11.4 ± 2.7) years, According to the activation time (0, 3, 6, 9, 12, 15, 18 and 21 o'clock), the patients were divided into eight groups. The data were analyzed using Halberg methods for cosinor-rhythmometry and t test.
RESULTSThe results of the tooth movement in eight different groups fitted to a cosine curve. The acrophase and bathyphase appeared at 21 and 9 o'clock, respectively.
CONCLUSIONSThe human circadian rhythm could affect the rapid palatal expansion. This study demonstrated that the best activation time for patients with RPE was around 21 o'clock.
