3.Application of admission test in labor room

Objective To investigate the application of admission test as a medical management in the labor room.Methods Eight hundred and fifty singleton pregnant women underwent admission test in labor room at Women's Hospital,School of Medicine,Zhejiang University from Dec.2009 to Dec.2011 were enrolled into this study.They were divided into two groups:normal group (admission test was normal,n =658) and abnormal group (admission test was abnormal or undetermined,n =192).Deliveries and perinatal outcomes of them were observed.The difference between the two groups were compared with two independent sample t test and Chi-square test.Results There were no significant differences in the mean age at delivery of the women and mean birth weight of neonates between the two groups (all P>0.05).The cesarean section rate of normal group (34.2%,225/658) was lower than that (59.4%,114/192) of abnormal group (x2=3.93,P<0.05).The rates of prematurc rupture of membranes (72.4%,139/ 192),fetaldistress (42.2%,81/192),neonatal asphyxia (16.5%,27/192) and neonatal complications (31.8%,61/192) in abnormal group were significantly higher than those [31.0% (204/658),24.2% (159/ 658),0.2%(1/658) and 2.6% (17/658)] in normal group (x2 =105.78,40.84,52.54 and 151.92,P<0.01respectively).Conclusions Admission test is a good method to forecast placental function during labor and perinatal outcomes,which might play an important role in medical management in labor room.

Objective:To investigate the immunoregulation effect of ovalbumin immune tolerance factors (OVA ITFs).Methods:Components that were smaller than 3 kD were isolated from the splenic lymphocytes lysates of OVA tolerance mice or naive mice,respectively,named as OVA ITFs or OVA ITFs conttrol.Naive BALB/c mice were divided into 5 groups,group A,B,C,D were injected i.v.by OVA ITFs,OVA ITFs control,splenic lymphocytes from OVA tolerant mice or PBS,respectively,group E as the blank control.The percentages of CD4+CD25+ T cell subpopulation from spleens before and after adoptive transfer were measured with flow cytometer;OVA specific lymphocyte responses were assessed by MTT assay.The levels of IL-10 and TGF-?1 in the culture supernatants were tested by ELISA kits.Results:For OVA ITFs or splenic lymphocytes from OVA tolerant mice,the percentages of CD4+CD25+ T cell subpopulation from spleens after adoptive transfer were raised significantly compared with that before adoptive transfer (15.32%?1.03% and 15.35%?0.62% vs 9.97%?1.38%,P
5.Exploration of research innovation capacity building for young talents in research-oriented hospitals

Objective To analyze youth talents' scientific research projects in research oriented hospital for recent five years,provide scientific basis for hospitals to strengthen the youth talents' capacity building of research innovation.Methods Identify those youth talents who have been funded in the First Affiliated Hospital of Xinjiang Medical University,and statistically analyze the demographic information,granted scientific research projects and hospital funding situations.Results There are 140 young talents who have got scientific research projects from 2011 to 2015 in the hospital.The national project number and the trend of approval rate has no obvious change;the provincial level project number and approval rate showed a trend of a significant increase in the last two years;Setting up the hospital fund promoted the youth talents' scientific research ability,but there are still spaces for further improvement in the funding field and founded group.Conclusions There has been a group of youth talents to stand out from the crowd during the construction and the development of the researchoriented hospital,but youth talents remains scarce for the national key projects and talent projects,and that will put forward new requirements for the hospital in the future to strengthen the youth talents' capacity building of research innovation,which needs to gradually establish a suitable cultivation mode and mechanism for youth talents' growth.
6.Molecular identification of aucklandiae radix, vladimiriae radix, inulae radix, aristolochiae radix and kadsurae radix using ITS2 barcode.

In order to identify Aucklandiae Radix, Vladimiriae Radix, Inulae Radix, Aristolochiae Radix and Kadsurae Radix using ITS2 barcodes, genomic DNA from sixty samples was extracted and the ITS2 (internal transcribed spacer) regions were amplified and sequenced. The genetic distances were computed using MEGA 5.0 in accordance with the kimura 2-parameter (K2P) model and the neighbor-joining (NJ) phylogenetic tree was constructed. The results indicated that for Aucklandiae Radix (Aucklandia lappa), Vladimiriae Radix (Vladimiria souliei and V. souliei var. cinerea), Inulae Radix (Inula helenium), Aristolochiae Radix (Aristolochia debilis) and Kadsurae Radix (Kadsura longipedunculata), the intra-specific variation was smaller than inter-specific one. There are 162 variable sites among 272 bp after alignment of all ITS2 sequence haplotypes. For each species, the intra-specific genetic distances were also smaller than inter-specific one. Furthermore, the NJ tree strongly supported that Aucklandiae Radix, Vladimiriae Radix, Inulae Radix, Aristolochiae Radix and Kadsurae Radix can be differentiated. At the same time, V. souliei (Dolomiaea souliei) and V. souliei var. cinerea( D. souliei var. cinerea) belonging to Vladimiriae Radix were clearly identified. In conclusion, ITS2 barcode could be used to identify Aucklandiae Radix, Vladimiriae Radix, Inulae Radix, Aristolochiae Radix and Kadsurae Radix. Our study may provide a scientific foundation for clinical safe use of the traditional Chinese medicines.

In our study, we adopted a schedule plan management of the Project Management Body of Knowledge(PMBOK)in the management practice of the expefimental research on "The Research in Teaching and Inheritance of Experience of Famous and Veteran Doctors of TCM "which was supposed by " tenth five years "plan of national science and technology as a key program. We tried to establish a project planning based on the special features of TCM and the characters of the project. This planning played a significant support role in the top project design and ensuring the smooth implementation of the research.

9.Effects of different doses of dexmedetomidine combined with propofol and remifentanil target controlled infusion on postanesthesia recovery of patients in functional endoscopic sinuses surgery

Objective To investigate the effects of different doses of dexmedetomidine combined with propofol and remifentanil target controlled infusion (TCI) on postanesthesia recovery of patients in functional endoscopic sinus surgery. Methods Eighty ASA Ⅰor Ⅱ grade patients, scheduled for the endoscopic sinus surgery, were divided into control group (20 cases) and dexmedetomidine group (60 cases) according to the random digits table method. The patients in dexmedetomidine group were given loading dose dexmedetomidine 0.6 μg/kg, then were given dexmedetomidine of different maintenance doses:0.3 μg/(kg·h) in D1 group, 0.6 μg/(kg·h) in D2 group and 0.9 μg/(kg·h) in D3 group. The patients in dexmedetomidine group were given TCI propofol and remifentanil during the maintenance of general anesthesia, rocuronium was administrated intermittently during operation, and bispectral index (BIS) was controlled at 40 - 50. The changes of hemodynamics 5 - 10 min after entering operation room (T0), before induction (T1), 1 min after intubation (T2), 5 min after intubation (T3), before extubation (T4) and 5 min after extubation (T5) were observed. The spontaneous breathing recovery time, call of eye-opening time, extubation time and adverse reaction after surgery were recorded. Moreover, the visual analogue score (VAS) and Ramsay sedation score were used to evaluate the comfort level of patients. Results The mean arterial pressure (MAP) at T5 in control group and D1 group were significantly higher that in D2 group and D3 group, and there were statistical differences (P<0.05). The heart rate at T1-T5 in D1-D3 group were significantly lower than that in control group, and there were statistical differences (P<0.05). The spontaneous breathing recovery time, call of eye-opening time, extubation time in D1 - D3 group were longer than those in control group, but there were no significant differences (P>0.05). The VAS 15 min after extubation in D1 - D3 group were significantly lower than that in control group: (3.7 ± 0.3), (3.1 ± 0.4) and (3.0 ± 0.5) scores vs. (6.2 ± 0.6) scores, and there were statistical differences (P<0.05). The Ramsay sedation scores in D1 - D3 group were significantly higher than that in control group:(2.5 ± 0.2), (2.7 ±0.2) and (5.3 ± 0.3) scores vs. (1.4 ± 0.3) scores. Moreover, Ramsay sedation score in D3 group was significantly higher than that in D1 group and D2 group, and Ramsay sedation score in D2 group was significantly higher than that in D1 group. There were statistical differences (P<0.05). Four and 2 patients occurred nausea vomiting respectively in control group and D1 group within 24 h after operation. Conclusions Dexmedetomidine combined with propofol and remifentanil TCI can maintain hemodynamic stability and improve anesthesia recovery quality.
10.The biological response of loading ADSCs to surface topography of biomimetic matrix combined PRP with RGD modification

Objective To evaluate the biological potential of surface topography of biomimetic matrix combined PRP gels with RGD modification. Methods Surface topography of nβ-TCP/Cs/PCL matrix was made by Nd: YAG laser, tissue-engineered bone was constructed in the following ways: ADSCs were loaded to nβ-TCP/Cs/PCL matrix with PRP gels plus RGD modification (group A), ADSCs were cultured to nβ-TCP/Cs/PCL matrix with RGD modification (group B), ADSCs implantation with topography-surfaced treatment of matrix (group C), ADSCs cultivation with smooth-surfaced treatment of matrix (group D). SEM and CLM were used to observe cellular pattern, survival rate, cell activity, ALP and collagen type Ⅰ level were detected at 1, 4, 8, 12, 16, 20, 24,28 days. Runx2 and OPG expressions were assayed at different interval. Results Under observation of SEM and CLM, new tissue showed more remarkable cell proliferation with abundant ECM in group A. Compared with other groups, the survival rate in group A was significantly higher (88.16 ± 1.29, P < 0.05),and the level of cell activity, ALP, collagen type Ⅰ were significantly higher (0.92± 0.13, 87.27 ± 3.08, 93.27 ± 3.91, P< 0.05), and remarkable expression of Runx2 and OPG was also seen. Conclusion Topography-surfaced treatment of matrix combined PRP gels with RGD modification enhances the cell proliferation and acts as a feasible osteopromotive method.
