1.Analgesic effect of electroacupuncture on gastric ulcer rats with liver-depression syndrome.

OBJECTIVETo explore the analgesic effect and action mechanism of electroacupuncture (EA) on gastric ulcer rats with liver-depression syndrome.
METHODSThrough open-field experimental method, 45 qualified SPF-grade male SD rats were selected and divided into a blank group, a model group and an EA group according to random number table method, 15 rats in each group. The model of gastric ulcer rats with liver-depression syndrome was established in the model group and the EA group by using chronic unpredictable stimulation combined with acetic acid burning method. Rats in the blank group did not receive intervention. Rats in the model group were treated with fixation and immobilization for 13 days. Rats in the EA group were treated with EA at "Liangqiu" (ST 34) and "Ganshu" (BL 18); EA voltage was 2 V; disperse-dense wave was selected with 4 Hz of disperse wave and 15 Hz of dense wave, and the intensity of EA was according to the slight vibration of local skin and; muscles; the needles were retained for 20 min, once a day for consecutive 6 days; there was an interval of 1 day' and the treatment was given for 2 weeks. The general condition, open-field experimental result and gastric ulcer index were observed; the western blotting method was applied to measure the expression of vanilloid receptor subtype 1 (VR1) in hypothalamus and gastric antral mucosal, and ELISA method was applied to test the expression of 5-hydroxytryptamine (5-HT) and norepinephrine (NE) in hippocampus.
RESULTSAfter model establishment, the general behavior condition in the model group was inferior to that in the blank group, which was obviously improved after EA. The range of motion in the model group was less than that in the blank group (P<0.01) while that in the EA group was higher than that in the model group (P<0.01). The ulcer inhibition rate was. 54.95%, and the ulcer index in the EA group was lower than that in the model group (P<0.01). Compared with; the blank group, the expression of VR1 in hypothalamus and gastric antral mucosal in the model group was increased (P<0.05); compared with the model group, the expression of VR1 in the EA group was reduced (P<0.05). Compared with the blank group, the expression of 5-HT an NE in hippocampus in the model group was significantly reduced (both P<0.01); compared with the model group, the expression of 5-HT and NE in the EA group was increased (both P<0.01).
CONCLUSIONEA at "Liangqiu" (ST 34) and "Ganshu" (BL 18) has certain analgesic effect in gastric ulcer rats with liver-depression syndrome, which is likely to be related with lowering the contents of VR1 in hypothalamus and gastric antral mucosal and increasing the content of 5-HT and NE in hippocampus.

2.The relationship between aggression and thyroid hormone and cortisol in schizophrenic patients

Objective To explore the relationship between aggressive behavior with thyroid hormone or cortisol and in schizophrenic patients. Methods According to the past violence history and modified overt aggression scale( MOAS) weighted total scores,108 schizophrenic patients were divided into aggressive group ( n=69) and non-aggressive group( n=39) . Positive and Negative Syndrome Scale ( PANSS) total scores, serum thyroid hormone levels and cortisol concentration were compared between the two groups before and af-ter 2,4 weeks treatment. And correlations of PANSS and sub-scales scores,thyroid hormone levels and corti-sol concentration with MOAS weighted total scores and subscales scores were analyzed before these patients receiving treatment. Results Before treatment,total scores of PANSS,positive symptoms and general psy-chopathology scores in aggressive group((94.19±12.71),(23.77±4.94),(52.61±6.45))were higher than that in non-aggressive group ((83.26±11.21),(21.36±7.10),(45.49±6.84)) and these differences were significant(P<0.05). But the difference of PANSS total scores after 2,4 weeks treatment between the two groups was not significant (P>0. 05 ) . Free thyroxine ( FT4 ) level of aggressive group ( ( 14. 41 ± 3. 58 ) pmol/L) was higher than that of non-aggressive group ((12.95±2.66)pmol/L) before treatment and the difference was significant(P<0.05) . Meanwhile,there was no significant differences between the two groups in thyrotropic-stimulating hormone(TSH),free triiodothyronine(FT3),total triiodothyronine(TT3) and total thyroxine( TT4) levels( all P>0.05) . After 2 or 4 weeks treatment,the differences between the two groups in TSH,FT3,FT4,TT3,TT4 levels were not significant(P>0.05). The differences between the two groups of cortisol concentration were not significant before and after 2,4 weeks treatment(P>0.05) . MOAS weighted to-tal scores were positively correlated with PANSS total scores,negative symptoms scores,general psychopathol-ogy scores and level of FT4,and their r values were 0.471,0.204,0.531,0.239(all P<0.05). Verbal aggres-sion was positively correlated with PANSS total scores,positive symptoms scores and general psychopathology scores,and their r values were 0.213,0.215,0.292(P<0.05). Auto-aggression was positively correlated with PANSS total scores and general psychopathology scores,and their r values were 0.278,0.382(P<0.05) . Psy-chical aggression was positively correlated with PANSS total scores,negative symptoms scores,general psy-chopathology scores and level of FT4,and their r values were 0.361,0.193,0.338,0.276(P<0.05). Conclusion The total scores of PANSS,positive symptoms,general psychopathology scores,level of FT4 and their variances can reflect severity of aggression and predict aggressive behavior in schizophrenic patients. Concen-tration of cortisol is not associated with aggressive behavior and can not be used as a predictor of aggressive behavior in schizophrenic patients.
3.Study on determination methods of chymosin potency

Objective To investigate the influencing factors and improve potency methods of chymosin, to verify the stability and applicability of the national standard of chymosin.Methods The effects of different formula milk powder substrate and enzyme concentration on the determination of the activity of chymosin were studied.3 ×3 dose-response parallel line method was established.The results were compared with the different methods of absolute and relative methods.Results The different formula milk powder had a significant effect on the determination of the absolute potency of the activity of chymosin.The concentration of the enzyme was a power function relationship with the milk clotting time.Compared with the absolute potency, reproducibility of the relative potency of the results was better in different laboratories.The suitable doses in 3 ×3 dose-response parallel line method were 0.35,0.44,0.55U/mL.The confidence limit rate was less than 5%.The potency of the national standard of chymosin (140712-201302) was not significantly different between 2013 and 2015.In a certain dose range, the dose-response of the national standard of chimosin and gastropylor complex or lamb'tripe extract was linear, and the two lines were parallel.Conclusion A lot of factors can affect on the potency of chymosin.Relative potency is determinate by reference standard which can eliminate the influence of different substrates, different operators and endpoint judgment on the determination in order to make results have comparability between laboratories.The test design of 3 ×3 dose-response parallel line can control the test deviation better than the single point determination.The stability of the national standard of chymosin(140712-201302) is good, and is suitable for the potency of chymosin of the products of gastropylor complex and the extract of the lamb.
4.Establishment and Evaluation of Gastric Ulcer Liver Depression Syndrome Rat Model

Objective To discuss the establishment of gastric ulcer liver depression syndrome rat model and the therapeutical effects of electroacupuncture. Methods As the result of Open-field experimental method, 60 SPF male SD rats were selected as experimental animals. After 7 days of adaptive feeding, 60 rats were randomly divided into blank group, model group, model control group, and electoracupuncture group, 15 rats in each group. Except for the blank group, the other three groups were stimulated by chronic unpredictable stimulation to establish rat depression model. Then model control group was once again stimulated by chronic unpredictable stimulation for 13 days;electoracupuncture group was treated at Gan Shu (BL18) and Liang Qiu (ST34) acupoints for 13 days. General state of rats, Open-field experimental results and gastric ulcer index were observed. BDNF was measured by RT-PCR. GAS content was measured by ELISA. Results After modeling for 21 days, except for the blank group, the other three groups appeared listlessness, limb weakness, and fur dry and lose luster, appetites reduced, etc. After modeling for 34 days, compared with model group, model control group had more severe general state than model group. After acupuncture treatment, the general state of acupuncture group was improved. The horizontal and vertical activity sore in model group decreased significantly compared with the blank group (P<0.01). Compared with the model group, the horizontal and vertical activity sore in model control group decreased significantly (P<0.05), and increased significantly in the electroacupuncture group (P<0.01). Ulcer index in the model group increased significantly compared with the blank group (P<0.01). Ulcer index in the model control group increased significantly compared with the model group (P<0.01). Ulcer index in electroacupuncture group decreased significantly (P<0.01). BDNF mRNA expression in hippocampus of model group decreased significantly compared with the blank group (P<0.01). Compared with model group, the BDNF mRNA expression in hippocampus of model control group decreased significantly (P<0.05). The BDNF mRNA expression in the electroacupuncture group increased significantly (P<0.01). The GAS content in the model group increased significantly compared with the blank group (P<0.01). Compared with the model group, the GAS content in the model control group increased significantly (P<0.05). The GAS content in the electroacupuncture group decreased significantly (P<0.05). Conclusion The establishment of depressive rat gastric ulcer model which combining disease and syndrome together proves the feasibility of this animal experimental model. Electroacupuncture for Gan Shu (BL18) and Liang Qiu (ST34) can improve the efficacy of this model.
5.Enhancers on the transmembrane transport of chlorogenic acid.

To investigate the influence of the difference enhancers on the transport mechanism of chlorogenic acid (CGA) across Caco-2 cells model, a RP-HPLC method was adopted to detect the concentrations of CGA. At the concentrations of 20 to 80 microg x mL(-1), the difference of absorption rate constants (K(a)) was not statistically significant. At the concentrations of 40 and 20 microg x mL(-1), the ratios of apparent permeability coefficients (P(app)) of the apical to basolateral and the basolateral to apical were 1.14 and 1.18, respectively. With the effect of enhancers K(a) and P(app) increased, the absorption half-life (T1/2) decreased. CGA passed through the Caco-2 cell membrane mainly by passive transport. It showed that monocarboxylic acid transporter (MCT) could be involved in the across membrane transport process of CGA. Borneol had no effect on the cell membrane transport processes. The order of increasing absorption of CGA caused by the enhancers was sodium lauryl sulphate > sodium taurocholate > carbomer.

6.Effect of different volume of fresh gas on postoperative analepsia period of general anesthesia with sevoflurane in patients undergoing abdominal surgery

Objective To investigate the effect of different volume of fresh gas on postoperative analepsia period of general anesthesia with sevoflurane in patients undergoing abdominal surgery. Methods 181 ASAⅠ-Ⅱpatients aged 20 - 70 undergoing elective abdominal surgery were divided into group A (112 patients) and group B (69 patients). Both groups received sevoflurane general anesthesia during surgery. At the end of surgery , group A received fresh gas for no more than 2L/min , and group B received the gas for more than 2L/min. The recovery time of spontaneous breathing, eye-opening time (from the end of surger to eye opening), and time of endotracheal extubation (from the end of surgery to endotracheal extubation ) were recorded. Results There were no significant differences in the recovery time of spontaneous breathing ,eye-opening time and time of endotracheal extubation between group A and group B (P > 0.05); Gender and time of intraoperative continuous uses of sevoflurane had effects on the recovery time of spontaneous breathing , which showed statistical significances (P < 0.05). Conclusions Uses of fresh gas for no more than 2L/min or more than 2L/min after surgery have no marked effects on the recovery time. However , gender and time of intraoperative continuous uses of sevoflurane may have effects on the recovery time of spontaneous breathing.
7.PCR-HRM Analysis for Gene Mutation Screening in a Child with Osteogenesis Imperfecta

Objective To investigate COL1A1 gene mutation by PCR-high resolution melting (PCR-HRM) and an-alyze the correlation between genotype and clinical phenotype in a child (proband) with osteogenesis imperfecta (OI). Methods The family history of OI pedigree along with the clinical data was collected. Blood samples from the proband and his family members, as well as 50 normal controls, were collected. The mutation of COL1A1 gene was screened using PCR-HRM and validated by the gene sequence. Results The detection of PCR-HRM showed the abnormal result of COL1A1 17 exon in proband with a lower melting temperature (Tm) value than that of normal controls by 0.4℃. There were signifi-cant differences in the standardization melting curve and the different melting curve between the proband and the normal controls. The sequencing result was c.1138G>A, which meant that cDNA of 1138 base G mutation into A. The mutations transformed the amino acid glycine into a serine at amino acid 380(P. Gly 380 Ser), which resulted in missense mutations. The proband’s father and grandmother had the same mutation of COL1A1 gene. The mutation was not found in the proband’s mother and normal controls. There was no report for such mutation in Chinese population. Pedigree analysis showed the fami-ly genetic characteristics of autosomal dominant inheritance. The proband was clinically diagnosed as OI type Ⅳwith more severe clinical phenotype. Conclusion PCR-HRM analysis is a new effective method for genetic screening of OI. COL1A1 mutation of c.1138G>A is a newly discovered mutation in Chinese population. Gly replaced inαhelical domain may lead to a more severe clinical phenotype.
8.Correlation between anxiety, coping style and perceived social support in vocational nursing college graduates

Objective To examine status of anxiety, coping style, perceived social support and their relationships among vocational nursing college graduates.Methods A total of 556 vocational nursing college graduates were asked to fill in State Anxiety Inventory, Simplified Coping Style Questionnaire and Perceived Social Support Scale.Results The prevalence rate of anxiety was 20.5% among the graduates. The mean scores of anxiety were (45.73±9.28),which was higher than normal state anxiety scores(39.71±8.89),there were significant differences(t=6.05,P<0.01); There was a significant difference in the prevalence of anxiety between graduates from countryside and city[(49.81±8.38) vs (44.58±7.52) scores,t=-2.35,P<0.05]. Graduates' anxiety was significantly correlated with positive coping style, negative coping style, family support, friend support, and other support. Family support, friend support and positive coping style play a positive role in the prediction of anxiety.Conclusions Anxiety is closely related to coping style and perceived social support. Positive coping ability and social support could help prevent and reduce anxiety among vocational nursing college graduates.

10.Effects of targeted care combined with low molecular weight heparin on the incidence of deep venous thrombosis of lower extremities after cesarean section

Objective To explore effects of the nursing care combined with low molecular weight heparin on the lower extremity deep venous thrombosis after cesarean section incidence. Methods 192 patients who underwent cesarean section from October 2015 to October 2016 in Yongkang maternal and child health hospital, excluding the relevant taboos, according to the different nursing methods, were divided into the control group (n=96) and the study group (n=96). Two groups of patients after 24 h of cesarean section were treated with low molecular weight heparin calcium 5000 IU subcutaneous injection, once a day, continuous treatment for 5 days; the control group was given routine cesarean section nursing after the operation; the study group was given targeted nursing (psychological guidance, preoperative guidance, postoperative care, dietary guidance, exercise guidance, and environmental care). Results The incidence of deep venous thrombosis of patients (2.08%) in the study group was significantly lower than (9.38%) in the control group with statistical significance (P<0.05); the plasma D dimer in the study group was significantly lower than the control group with statistical significance (P<0.05); nursing satisfaction rate of patients in the study group was (95.83%) higher than (81.25%) of the control group with statistical significance (P<0.05). Conclusion The combination of targeted care plus low molecular weight heparin could improve the satisfaction of nursing and the quality of life, reduce the incidence of postoperative deep vein thrombosis. It is worthy of clinical application.
