1.Comparison of the effects of virus inactivation of plasma and albumin in plasma exchange liquid gallinger therapy on the treatment of severe systemic lupus erythematosus

Objective To compare the clinical curative effect of virus inactivating activity of plasma and albumin compound sodium chloride injection in liquid plasma exchange(PE)for treatment of severe systemic lupus erythematosus(SLE).Methods A total of 75 cases of severe SLE patients were randomized according to the number table method patients for virus inactivation of plasma group A(37 cases)and liquid albumin plasma group B (38 cases). The changes of immunoglobulin protein(IgG,IgA,IgM),ESR,complement C3,C -reactive protein(CRP),blood urea nitrogen(BUN)and serum creatinine(CR)index in the two groups were observed before and after PE.Clinical SLE-DAI(systemic lupus erythematosus disease activity index)score changes,adverse reactions occurred rate and clinical efficacy of two groups were compared.Results After treatment of group A was treated with PE,laboratory indicators except IgM(t =0.509,P =0.612),other indexes were significantly decreased,differences were statistically significant (t =29.447,186,56.447,41.867,12.841,92.675,92.980,all P <0.01).In group B,the IgM,IgA,CRP,C3 and preoperative showed no statistical significance(t =0.565,1.165,0.902,all P >0.05),while IgG and ESR,bun,crea and replacement of the differences have statistical significance(t value were 28.999,194.9,40.634,40.634104.918, P value <0.01 ).Ttotal efficiency of group A was 91.9%,the average clinical SLEDAI score was (9.4 ± 1.2)points,total efficiency of group B was 77.3%,clinical SLEDAI score was (13.8 ±1.1 )points and the difference between the two groups were statistically significant(χ2 =8.831,t =15.560,P <0.01).In group A,the adverse reaction rate was 15.3%,that of group B was 16.2%,there was no significant difference between the two groups(χ2 =0.509,P >0.05).Conclusion The clinical curative effect of virus inactivation of plasma replacement solution in treatment of severe SLE is better than that of albumin by Ringer's solution.
3.Effect of platelet activation on the levels of VEGR,TGF-?_1 and PDGF in apheresis platelets

Objective To investigate the changes of vascular endothelial growth factor (VEGF),transforming growth factor-?1 (TGF-?1) and platelet derived growth factor (PDGF) in plasma and apheresis platelets with or without activation by shaking. Methods A total of 10 units of double therapeutic dose platelets were collected and each unit was divided into 3 groups to take samples of plasma (3ml/person,as control group),fresh apheresis platelets (5ml/person,as experimental group1) and activated apheresis platelets (platelets stored by shaking at 22 ℃ for 5 days,5ml/person,as experimental group2). The levels of VEGF,TGF-?1 and PDGF in the samples of the 3 groups were measured by ELISA. The expression of CD62p in 2 apheresis experimental groups was measured by flow cytometry. The differences between the two groups were compared. Eukaryotic elongation factor 2 kinase (eEF2K) is well known as a Ca2+/calmodulin (CaM)-dependent kinase. eEF2K catalyzes the phosphorylation of eEF2 and subsequently inactivates eEF2 by impairing its ability to bind to the ribosome, thereby negatively modulates protein synthesis. The high expression of eEF2K has been found recently in several types of malignancies. As participating in the progress of tumor, eEF2K emerges a potential target for future cancer therapy. The relationship between eEF2K and tumor, and the latest progress of eEF2K inhibitors were summarized in this article.

7.Efficacy of modified PLT additive solution for platelets storage at low temperature(10℃) on rabbit thrombocytopenia model

Objective To evaluate the in vivo efficacy of modified platelet(PLT) additive solution(PAS-ⅢM) with trehalose as a substitute for plasma for the storage of platelet concentrates at low temperature(10℃).Methods Rabbit platelet concentrates were collected and stored in different media and temperature conditions,plasma(22℃),70%PAS-ⅢM/30% plasma(10℃) and plasma/trehalose(-85℃).Platelets in plasma(22℃) on storage day 3,in 70%PAS-ⅢM/30% plasma(10℃) on storage day 3,7,9,and in plasma/trehalose(-85℃) thawed on day 20 were transfused to rabbit thrombocytopenia model.The in vivo recoveries and survivals of the three preserved platelets and fresh platelets were measured and evaluated after transfusion into rabit model of thrombocytopenia.Results The survival of platelets stored in 70% PAS-ⅢM/30% plasma(10℃) on day 9 was lower than that of fresh platelets(P0.05).Recovery and survival of frozen platelets were significant lower than those of the other groups(P

Age-related macular degeneration (AMD) has become a leading cause of irreversible visual loss in senior population and a serious issue of public health as the aging of society worldwide,especially wet AMD.Wet AMD is characterized by choroidal neovascularization(CNV)with a rapidly developing process andsevere impairment of visual acuity.The effective therapy that can destroy choroidal neovascularization and improve visual acuity for very long time is lack.In the recent 20 years,more attentions focuse on the treatment of choroidal neovascularization.From bench to bedside,the research on targeted treatment of angiogenesis brings the bloom of wet AMD medical therapy.This paper will summarize the update information of wet AMD.
9.Therapeutic observation on superficial needling with different frequencies for intractable facial paralysis

Objective:To obverse the therapeutic effect of superficial needling with different frequencies for intractable facial paralysis. Methods: A total of 120 patients with intractable peripheral facial paralysis were allocated into a superficial needling with high frequency group (150 times/min), a moderate frequency group (100 times/min) and a low frequency group (50 times/min) according to the random number table method. The Toronto facial grading system (TFGS) was used to evaluate facial nerve functions before treatment and after 2 weeks and 4 weeks of treatment respectively. The electromyography (EMG) test of the mandibular branch of facial nerve was used to compare the motor nerve conduction velocity (MCV), sensory nerve conduction velocity (SCV) and monophasic action potential (MAP) among different groups, and was done before treatment and after 4 weeks of treatment. The clinical efficacy was also compared. Results: After 2 weeks and 4 weeks of treatment, the changes of TFGS scores in the three groups all showed statistical significance (all P<0.05), and the TFGS score in the low frequency group was substantially higher than that in the other two groups. After treatment, the changes of the MCV and SCV in the three groups all showed statistical significance (all P<0.05), and the results in the low frequency group were higher than those in the other two groups; the change of MAP in the three groups showed no statistical significance (P>0.05). The total effective rate was 65.0%, 80.0% and 95.0% in the high frequency group, moderate frequency group and low frequency group respectively, and the between-group differences showed statistical significance (P<0.05). Conclusion: Compared with the superficial needling with high and moderate frequencies, superficial needling with low frequency can produce more significant clinical efficacy for intractable facial paralysis.
