1.Differentiation of adult adipose tissue-derived mesenchymal stem cells into cardiocyte-like cells in vitro

BACKGROUND: At present, many problems deserve to be solved before clinical utilization of adipose tissue-derived mesenchymal stem cells (ADMSCs) such as complete differentiation from ADMSCs into cardiomyocytes and ADMSCs with or without specific function of cardiomyocytes. It is of significance to solve the problems including how to elevate the differentiation rate of ADMSCs into cardiomyocytes and how to elevate the homing and survival rate after transplantation for clinical utilization of stem cells.OBJECTIVE: To observe the differentiation of ADMSCs into cardiomyocytes after in vitro culture and induction.DESIGN: Randomized controlled observation.SETTING: Laboratory of Cardiology, General Hospital of Chinese PLA.MATERIALS: Adipose tissue was collected from abdominal operative patients at Department of General Surgery of General Hospital of Chinese PLA with the agreement of patients. Iscove's Modified Dulbecco's Medium (IMDM), type Ⅰ collagenase, 5-azacytidine (5-aza) and polyclonal antibody of specific anti-myocardial Troponin T (TnT) were purchased from Hyclon company, Gibco company, Sigma and Fujian Maixin Biotechnology Company, respectively.Monoclone antibodies of CD44, CD45, CD34, HLA2DR and factor-Ⅷ, Desmin, anti-α-striated muscle actin and anti-myosin heavy chain (MHC) were bought from Beijing Zhongshan Golden Bridge Biotechnology Limited Corporation. DAB stain, reverse transcription-polymerase chain reaction (RT-PCR) kit and atrial natriuretic peptide (ANP) radioimmunity kit were purchased from Beijing Zhongshan Golden Bridge Biotechnology Limited Corporation,Invitrogen and Radioimmunity Research Institute of Science and Technology Development Center of General Hospital of Chinese PLA, respectively.METHODS: The experiment was performed at the Laboratory of Cardiology, General Hospital of Chinese PLA from March 2005 to April 2006. ADMSCs were isolated and cultured by digestion and attachment culture method. The third generation of cells were determined by immunocytochemical method for surface molecule CD44, CD13, CD105, CD45,CD34, HLA-DR, factor Ⅷ and induced by addition of 5-aza into culture medium. On the 7th, 14th, 21st and 28th days, gene of GATA4 and Nkx2.5 were tested by reverse transcription-polymerase chain reaction (RT-PCR), and the concentration of atrial natriuretic polypeptide (ANP) were also examined by radioimmunoassay.MAIN OUTCOME MEASURES: Determination of cell surface marker, gene of GATA4 and Nkx2.5, and ANP concentration.and factor Ⅷ negative for the cultured cells. After induction for 7 days with 5-aza, no cells expressed Desmin,α-Sarcomeric Actin, Myocin heavy chain (MHC) and TnT, most cells were positive stained for CD44. After induction for 14 days, small amounts of cells displayed positive for Desmin,α-Sarcomeric Actin and MHC but still negative for TnT,while partial cells expressed positive CD44. After induction for 21 days, most cells were positive for Desmin,α-Sarcomeric after induction and no significant difference was found compared with that at the 7th day [(0.022±0.01) ng/L (P>0.05].ANP concentration was respectively (7.92±0.21) and (8.12±0.50) ng/L at the 21st day and the 28th day (P>0.05), but there was significant difference compared with that at the 7th day (P<0.05).
2.Application of stem cell transplantation in treating cardiovascular diseases

BACKGROUND: As a group of primary cells,stem cells have the potentials of self-renewal and multi-directional differentiation, and can replace the damaged cardiomyocytes to construct new vessels, and enable the asthenic heart to obtain the blood pumping function again.This study summarized the progress of stem cell transplantation in treating cardiovascular diseasesDATA SOURCES:A computer-based online search of Pubmed database was undertaken to identify English articles published from January 2000 to December 2005, the keywords were "stem cell transplantation, cardiovascular diseases".STUDY SELECTION: The articles were primarily checked, and the references of each literature were looked up. Inclusive criteria:articles correlated with the classification and the transplanted pathways of stem cells,the present status,problems and prospect of its basic and clinical application in treating cardiovascular diseases. Exclusive criteria: repetitive study or Meta analysis.DATA EXTRACTION:Totally 50 relative literatures were collected, and 31 of them accorded with the inclusive criteria,and the other 19 were excluded due to the old or repetitive contents. Of the 31 involved literatures, 19 were about the classification of stem cells used in cardiovascular system, 1 about the transplanted pathways of stem cells, 8 about the application of stem cells in treating cardiovascular diseases, and 3 about the existing problems and prospect.DATA SYNTHESIS:With the continuous improvement of living level and aging, cardiovascular diseases have become the main causes for death and disability in modern society.Stem cells have the potentials of multi-directional differentiation, and have become the main seed cells for cell replacement therapy, and provided important investigative and applicating field for the treatment of cardiovascular diseases. At present, there are many stem cells to be used in cardiovascular system, including embryonic stem cells, umbilical cord blood stem cells, peripheral blood stem cells, bone marrow stem cells, skeletal muscle satellite cells, myocardial stem cells, adipose tissue-derived stem cells, etc. The common pathways for stem cell transplantation are direct injection into myocardium,intracoronary artery injection and intravenous injection.Stem cell transplantation can treat cardiovascular diseases by promoting angiogenesis and myocardial regeneration, decreasing myocardial apoptosis and reducing myocardial collagen, then ameliorate the cardiac function, and breakthrough and progress have been made in both animal models and clinical studies. However, problems sill exist to be solved, and there is agreat distance to the routine clinical application.CONCLUSION: Stem cells are the important fields of the basic research and clinical application for preventing and treating cardiovascular diseases,thus it will provide powerful theoretical and practical supports for the more effective prevention and treatment of cardiovascular diseases to solving the key points and difficulties in cardiovascular diseases.
3.Compare efficacy of oxytocin, dinoprostone suppositories and cervical dilation balloon in odinopoeia of late pregnancy

Objective To compare the effect of the oxytocin, dinoprostone suppositoriesository and cervical dilation balloon in odinopoeia of late pregnancy.Methods A total of 90 patients with late pregnancy singletons primipara were randomly divided into 3 groups with 30 cases in each group. Patients in the group A were treated by dinoprostone suppositoriesository, patients in the group B were treated by oxytocin, patients in the group C were treated by the cervical dilation balloon, the cervical, childbirth, complications and newborns in the 3 groups were observed and compared. Results Compared with group B and group C, late pregnancy induced labor curative effect in group A was better:cervical Bishop score was higher after intervention 6h and 12 h (P<0.05), and promote cervical mature rate and success rate of induced labor were higher(P<0.05), labor time was shorter (P<0.05).The incidence of delivery complications was lower(P<0.05).Conclusion Dinoprostone suppositories can reduce labor time significantly in the treatment of late pregnancy, and improve the success rate of induced labor, the effect is superior to oxytocin and cervical dilation balloon.
4.The influence of extract of schisandra chinensis and paeonia veitchii on the level of cytokines in serum of asthmatic rats

Objective To investigate the influence of extract of schisandra chinensis and paeonia veitchii on level of cytokines in asthmatic rats .Methods SD rats were randomly divided into three groups :Chinese medicine-treated ,untreated asthma ,physiologi-cal saline control groups .The tracheal pathology and symptom of different groups were observed ,then cytokine expressions in ser-um were detected with enzyme-linked immunosorbent assay (ELISA ) .Results Chinese herb extract obviously improved tracheal pathology and airway symptoms .The levels of IL-4 ,IL-6、IL-13 in serum of Chinese medicine-treated and untreated asthma groups were significantly increased ,while the IFN-γ expression was reduced .But after treatment of Chinese herb extract ,compared with untreated asthma group the IFN-γexpression was significantly increased in Chinese medicine-treated group .Conclusion The effect of extract of schisandra chinensis and paeonia veitchii could reduce airway inflammation of asthma rats may by significantly in-creased IFN-γexpression .
5.Chemical constituents from Vaccinium bracteatum.

The chemical constituents of Vaccinium bracteatum were studied by means of macroporous resin, ODS column chromatography and preparative HPLC. Eleven compounds were isolated from this plant. By using ESI-MS and NMR, the structures of the eleven compounds were determined as 10-O-trans-p-coumaroyl-6alpha-hydroxyl-dihydromonotropein (1), 10-O-cis-p-coumaroyl -6alpha-hydroxyl-dihydromonotropein (2), vaccinoside (3), 10-O-cis-p-coumaroyl monotropein (4), isolariciresinol-9-O-beta-D-xyloside (5), tectoridin (6), vicenin-3 (7), quercetin-3-O-alpha-L-rhamnoside (8), quercetin-3-O-alpha-L-arabinopyranoside (9), quercetin-3-O-beta-D-galactopyranoside (10), and quercetin-3-O-beta-D-glucuronide (11), respectively. Compounds 1 and 2 are new, and compounds 4, 6 and 7 are isolated from the genus Vaccinium for the first time.

6.Detection and significance of IL-25 ,eosinophil levels in induced sputum of patients with bronchial asthma

Objective To investigate the roles of interleukin(IL)-25,eosinophils in the occurrence of bronchial asthma.Methods Selected 30 cases of acute episode of asthma(asthma group),and ease a group of 23 cases of asthma treatment(remission group),20 cases of the normal control group.Using the method of gradient ultrasonic atomizing inhalation of Hyperosmotic saline collection of induced sputum specimens,by ELISA method for the determination IL-25 level in induced sputum,count of eosinophils,analysis of correlation between them.Results Group IL-25 in induced sputum of asthma and eosinophil levels[(313.12±75.64)ng/L vs(236.43±57.90)ng/L]were sig-nificandy higher than the control group[(0.386±0.267) × 109/L vs(0.005±0.002) × 109/L],and significantly higher than acute stage in remission stage[(268.63 + 40.19) ng/L,(0.120 + 0.016) × 109/L](P < 0.05).Conclusion IL-25 has an important role in asthma development.It provides a new way of thinking for monitoring and treatment.
7.Application and technical analysis of enhanced T*2 star weighted angiography sequence in the detection of hemorrhagic shearing lesions associated with diffuse axonal injury

Objective To compare the efficiency of enhanced T2* weighted angiography (ESWAN) sequence with that of a conventional T2*-weighted gradient-recalled-echo (GRE T2*WI) sequence for the detection of hemorrhagic shearing lesions in patients with diffuse axonal injury (DAI). And combined with MRI parameters, to further discuss the principles and virtues of ESWAN sequence. Methods Seventeen patients with DAI were enrolled in this study. The raw data acquired from ESWAN scan were postprocessed by using the mean square root of multi-echoes. Then, the postprocessed images were compared with the conventional GRE T2* weighted images. The global and regional (superficial, deep and posterior fossa) lesion numbers determined by both sequences were compared by using Wilcoxon signed ranks test (two-tailed). Differences were considered to be significant at P≤0.05. Results Hemorrhagic lesions were more obvious on ESWAN images than those on conventional GRE T2* weighted images. The median and range value of the detected lesion numbers on ESWAN images were 27 and (1-239) in whole brain, 13 and (1-89) in cerebral superficial region, 5 and (0-111) in cerebral deep region and 1 and (0-39) in posterior fossa region, respectively; whereas, on GRE T2* weighted images, they were 7 and (1-34) in whole brain, 5 and (1-27) in cerebral superficial region, 2 and (0-25) in cerebral deep region and 0 and (0-4) in posterior fossa region, respectively. There were significant statistical differences between the two sequences in revealing the lesions in all the four regions (Z=-3.519,-3.182,-3.185,-2.677,P<0.05). Conclusion In ESWAN sequence, multi-echo acquisition ensured sufficient magnetic susceptibility for detecting small hemorrhagic lesions. So, ESWAN is more sensitive to small hemorrhage, which revealed more hemorrhagic lesions than conventional GRE T2*WI and presented more valuable information for the diagnosis of DAI.
