2.Preparation and performance detection of carbon fiber-polylactic acid-polyethylene glycol composite scaffold

BACKGROUND:The mechanical performance and cytocompatibility of polylactic acid-polyethylene glycol (PLA-PEG) scaffold is poor via long-term experiments; therefore, other materials are added to the scaffold in order to improve its biological activity and mechanical properties. OBJECTIVE:To investigate the preparation process of carbon fiber (CF)-PLA-PEG scaffold, and to test its performance. METHODS:Modified CF-PLA-PEG composite scaffolds were prepared using solution injection-particle leaching method. Ultra-structure, porosity, water-absorbing quality, degradation rate and mechanical properties of CF-PLA-PEG composite scaffolds and PLA-PEG scaffolds were compared. Rat osteoblasts were co-cultured with these two kinds of scaffoldsin vitro, and the cel adhesion rate was detected by precipitation method after 12 hours. Cel counting was detected using MTT method to determine the cel proliferation at culture days 1, 3, 5, 7, 9. RESULTS AND CONCLUSION:The surface structure of PLA-PEG distributed uniformly, and the aperture was (404.0±10.5) μm. The carbon fiber surface of modified CF-PLA-PEG composite scaffolds had a lot of longitudinal grooves, the surface structure also distributed uniformly, and the aperture was (433.0±3.0) μm. Difference in scaffold aperture was of significance between the two groups (P < 0.05). The porosity, water-absorbing quality, elasticity modulus, compressive strength, degradation rate, cel adhesion rate and cel growth rate of CF-PLA-PEG were better than those of PLA-PEG (P < 0.05). These results show that the CF-PLA-PEG scaffolds have good mechanical property and cel compatibility.
3.Analysis on the adverse events of cupping therapy in the application.

The deep analysis has been done on the cases of adverse events and common injury of cupping therapy encountered in recent years in terms of manipulation and patient's constitution. The adverse events of cupping therapy are commonly caused by improper manipulation of medical practitioners, ignoring contraindication and patient's constitution. Clinical practitioners should use cupping therapy cautiously, follow strictly the rules of standard manipulation and medical core system, pay attention to the contraindication and take strict precautions against the occurrence of adverse events.

5.Playing the guiding roles of national criteria and precisely eliminating schis-tosomiasis in P. R. China

Schistosomiasis Control and Elimination(GB 15976-2015)and Diagnostic Criteria for Schistosomiasis(WS 261-2006)are the only two national health criteria related to schistosomiasis control program implemented in P. R. China. The roles of criteria to guide and accelerate the transition from schistosomiasis control to elimination are concluded ,based on this system?atic review how the criteria led the implementation of the medium?and long?term national plan and provided the guidance when drafting the thirteen?five years national plan for schistosomiasis,and the suggestion to draft more criteria related to schistosomia?sis elimination program and strengthening the implementation of current criteria,so as to precisely guide the schistosomiasis elimination program in P. R. China.
6.Clinical study of occupational therapy in conjuct with Taoist cognitive psychotherapy on emotional disorder in rehabilitation of hemiplegic stroke patients

Objective To evaluate the effects of occupational therapy and Taoist cognitive psychotherapy on emotional disorder of stroke patients.Methods A total of 136 hemiplegic patients with emotional disorder were randomly divided into a general treatment group,a cognitive psychotherapy group,an occupational therapy group and a combined therapy group(occupational therapy combined Taoist cognitive psychotherapy group).All patients were treated accordingly for 8 weeks and followed up for 6 months.All the patients were evaluated with Fugl-Meyer Assess- ment,COPM and SCL-90.Results It was found that the Fugl-Meyer Assessment and COPM scores were improved gradually in all groups.The combined therapy group scored higher than occupational therapy group(P
7.A Survey on Nephrolithiasis and Its Related Influencing Factors in Putuo District,Zhoushan City

Objective To study the epidemiological characters and risk factors of nephrolithiasis in an island area,Putuo district, Zhoushan city.Methods With stratified cluster random sampling method,5 915 residents aged above 15 yrs and resided 5 yrs and above in Putuo district,Zhoushan city were selected,and surveyed with Ultrasound examination and questionnaire.X~2 tendency test and non-condition Logistic regression were conducted by means of SPSS software.Results After single factor analysis and Multi- factor analysis,the factors entered the model finally were sex(OR=0.543,95%CI:0.454~0.649),education status(OR=1.253,95%CI: 1.151~1.364),dietary habit(OR=0.882,95%CI:0.823~0.944),BMI(OR=1.274,95%CI:1.142~0.1.421),urinary tract infection(OR= 2.126,95%CI:1.663~2.718).Conclusion Risk factors for nephrolithiasis include dietary habit,education status,and urinary tract infection.The vegetables are the protection factor.
8. The killing effect of two different formulations paclitaxel in KB cell in vitro

OBJECTIVE: To investigate the killing effect of albumin-bound paclitaxel and paclitaxel solution on human oral epithelial carcinoma KB cells in vitro. METHODS: The KB cell line was cultured with different concentrations of albumin-bound paclitaxel or paclitaxel solution. MTT assay, inverted microscopic observation and flow cytometry assay were then used to monitor the growth and proliferation of the cell line. RESULTS: When treated with different concentrations of albumin-bound paclitaxel or paclitaxel solution for 36 and 72 h, KB cells survival rate decreased significantly, however, the difference between their IC50 was no significant; degeneration and necrosis of KB cells were found under microscope; the rate of cell apoptosis was increased gradually; the rate of S phase was reduced, and the percentage of G2/M cells treated with 0.1, 1 and 10 μmol · L-1 albumin-bound paclitaxel or paclitaxel was increased, which significantly higher than that of G2/M cells treated with 100 μmol · L-1 albumin-bound paclitaxel or paclitaxel. However, the two groups with the same concentration showed no significant differences in apoptosis rate and cell cycle (P > 0.05). CONCLUSION: Compared with paclitaxel, albumin-bound paclitaxel have the similar ability to inhibit proliferate and promote apoptosis in KB cells. 9.Effect of lovastatin on proliferation and apoptosis of glioma stem cells

BACKGROUND:Increasing evidence has shown that lovastatin with less toxicity to normal cels has crucial effects on proliferation, apoptosis and differentiation of various cancer cels. However, its roles in glioma stem cels remain unclear.
OBJECTIVE:To explore the effect of lovastatin on proliferation and apoptosis of glioma stem cels.
METHODS: Flow cytometric sorting was used to separate glioma stem cels from human glioblastoma cel line U87. Effects of lovastatin on the proliferation and apoptosis of glioma stem cels were determined by MTT and flow cytometry, respectively. Furthermore, expression levels of Ki67, Bax and Bcl-2 in glioma stem cels treated with lovastatin were detected using western blot analysis.
RESULTS AND CONCLUSION: The CD133-positive glioma stem cels were sorted from human glioblastoma cel line U87 with a positive percentage of 85%. MTT assay showed that lovastatin inhibited the proliferation of glioma stem cels in dose (5, 10, 20 μmol/L)- and time (24, 48, 72, 96 hours)-dependent manners. Flow cytometry analysis showed that 10 μmol/L lovastatin (48 hours) induced apoptosis in glioma stem cels. In addition, the expression level of Ki67 was decreased by lovastatin treatment in a dose-dependent manner, and the Bcl-2 and Bax expression levels were reduced and increased by 10 μmol/L lovastatin treatment, respectively. In conclusion, lovastatin can inhibit cel proliferation and induce apoptosis of glioma stem cels, and lovastatin may be a potential drug for treatment of brain tumors.
10.Anti-SARS-CoV-2 activity of small molecule inhibitors of cathepsin L

The coronavirus disease 2019 (COVID-19) is an acute infectious disease caused by the new severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, which has led to serious worldwide economic burden. Due to the continuous emergence of variants, vaccines and monoclonal antibodies are only partial effective against infections caused by distinct strains of SARS-CoV-2. Therefore, it is still of great importance to call for the development of broad-spectrum and effective small molecule drugs to combat both current and future outbreaks triggered by SARS-CoV-2. Cathepsin L (CatL) cleaves the spike glycoprotein (S) of SARS-CoV-2, playing an indispensable role in enhancing virus entry into host cells. Therefore CatL is one of the ideal targets for the development of pan-coronavirus inhibitor-based drugs. In this study, a CatL enzyme inhibitor screening model was established based on fluorescein labeled substrate. Two CatL inhibitors IMB 6290 and IMB 8014 with low cytotoxicity were obtained through high-throughput screening, the half inhibition concentrations (IC50) of which were 11.53 ± 0.68 and 1.56 ± 1.10 μmol·L-1, respectively. SDS-PAGE and cell-cell fusion experiments confirmed that the compounds inhibited the hydrolysis of S protein by CatL in a concentration-dependent manner. Surface plasmon resonance (SPR) detection showed that both compounds exhibited moderate binding affinity with CatL. Molecular docking revealed the binding mode between the compound and the CatL active pocket. The pseudovirus experiment further confirmed the inhibitory effects of IMB 8014 on the S protein mediated entry process.
