1.Development of regulation strategy questionnaire of sea-going seafarers' negative emotion

Objective To study the negative emotion regulation of sea-going seafarers,establish a regulation strategy questionnaire of sea-going seafarers' negative emotion and examine its reliability and validity.Methods Based on open-ended questionnaires and previous researches,the initial questionnaire was obtained.Randomly selected 188 sea-going seafarers,by adopting program analysis and factor analysis,the final questionnaire was determined.Tested 121 sea-going seafarers by using the final questionnaire,while 55 of them by using coping style questionnaire at the same time.After two weeks,47 of them were tested by final questionnaire again.The reliability and validity of the questionnaire were investigated during the research,and the criterion validity between the coping style questionnaire and regulation strategy questionnaire of sea-going seafarers' negative emotion was also studied.Results The factor analysis results showed that the questionnaire was composed of seeking for support,abreaction,inobservance,transfer and oppression.Cronbach α of each factors was 0.857,0.841,0.825,0.806,0.754 respectively;and Cronbach o of the entire questionnaire was 0.873.The split-half correlation coefficients were 0.697,-0.839 (P < 0.01) ; test-retest reliability coefficients were 0.706,-0.822 (P < 0.01).The correlation between subscales were-0.031,-0.459 ; and the correlation between subscales and total scale were 0.590,-0.806 (P < 0.01).Criterion validity showed that despite for the abreaction,each factor was significantly correlated with the factors of coping style questionnaire.Conclusion The sea-going seafarers' negative emotion regulation strategy questionnaire has acceptable psychometrics properties on reliability and validity.
2.Experimental Studies on the Effect of“Liver-Softening Anti-Fibrotic Decoction”on Active and Inactive Fibrotic Rats

Liver-Softening Anti-Fibrotic Decoction", a preparation consisted mainly of medicinal qerbs with touifying "Qi", flourishing"Yin", activating blood and eliminating stasis, was given by gavage to Dimethyl nitrosamine (DMN) induced fibrotic rat to assess its effect on ative and inactive liver fibrosis. It was found that this preparation, besides its promoting growth aud protein metabolism activeties, can also lower serum alkaline phosphatase, decrease serum hyaluronate and liver hydroxy proline, with a better curative effect on experimental liver fibrosis in rats than its prophylactic effect
3.Study on the method of decoloration for immunohistochemical HE slides

Objective To explore the different methods of decoloration for immunohistochemical HE slides,and their influence on tissue antigen expression.Methods Antigen retrieval solution (EDTA or citric acid solution) was used in immunohistochemistry to decolor HE slides,and was compared with the method of hydrochloric acid solution decoloration in normal temperature.Results Antigen heat retrieval decolors completely.Compared with other methods,it has shorter decoloration time (1 ~ 2min),more than 90% of completely decoloration rate,the difference was significant(x2 =27.6,23.9,all P < 0.01).Meanwhile,the method dose not damage slides,and the immunohistoehemical results are exact,without background coloring and unusual coloring,antigen expression rate and intensity are high.Conclusion Antigen heat retrieval decoloration not only simplifies the coloration process,but also protects tissue antigen activity better.It is the preferable method of decoloration and protecting antigen resource,which possesses some practical value in the pathology diagnosis.
4.Biological role of β-arrestin1 in human gastric cancer BGC-823 cells

Objective To investigate the effects of β-arrestin1 on proliferation,migration,invasion and apoptosis of human gastric cancer BGC-823 cell line.Methods The expression of β-arrestin1 in human gastric epithelial cell line GES,human gastric cancer cell line BGC-823,MKN-28 and SGC-7901 was detected by realtime-polymerase chain reaction (PCR) and Western blot.The stable β- arrestin1 and negative control interfered BGC-823 cell line were established by RNA interference technology.The cell proliferation,migration,invasion and cell apoptosis of β-arrestin1 stable interfered BGC-823 cell line was examined by cell counting,scratch test,Transwell chamber test and flow cytometry assays.The data were analyzed by t test.Results The expression of β-arrestin1 in cell line GES,MKN-28,SGC-7901 and BGC-823 was 0.001 ± 0.001,0.002 ± 0.000,0.003± 0.002 and 0.005 ± 0.000 respectively.The inhibition ratio of proliferation in β-arrestin1 interfered BGC-823 cells and negative control cells were -30.2 % and 100.0 %.The invasion ability was also inhibited,the number of migratory cells was 126.25±3.24 and 213.50±6.27 (t=0.000,P<0.01),and the apoptosis rate was (41.350±1.053)% and (11.497±0.589) % (t=0.015,P<0.05).Conclusions β-arrestin1 is highly expressed in gastric carcinoma,and the expression increased along with the malignancy degree.The cell proliferation,migration and invasion is inhibited by interference of β-arrestin1 in BGC-823 cells,while the cell apoptosis is promoted.
5.Effect evaluation on employee assistance program (EAP) for medical workers

Objective To evaluate the mental health of medical workers, and develop an EAP suitable for heahhcare sector. Methods A 1-year EAP was implemented in a hospital in Beijing, in which SF-36 and JSS scales were used to study the health-related quality of life and job satisfaction of these workers. Results Work pressure is found to be the main source of pressure for medical workers, accounting for 63.7%. Scores of the group receiving EAP service are found to be significantly higher (P<0.01) in all dimensions (excepting work nature and supervisor dimension) of both SF-36 and JSS. Conclusions Medical workers are exposed to high pressure of work, having strong need for EAP service as result. EAP is an effective means to maintain mental health and improve quality of life and job performance, if applied widely.
6.Study on the cognition status of implementing case-based payment policy among healthcare providers:A case in Chengdu, Sichuan Province

Objective:To understand the implementation status of case-based payment among healthcare provid-ers and their cognition on the matter. Methods:Semi-structured interview was conducted on 30 purposely selected staff from 9 hospitals in Chengdu. Results:After one-year implementation of case-based payment, hospitals at different level carried out the policy vigorously:executed corresponding expense control measures and management. Neverthe-less, the proportion of cases that were paid with case-based payment was low, moreover the inclusion criteria for case was of disunity and the formulation of the expense standard was ambiguous to some extent. Conclusion:Certain achievements were accompanied with problems, so it is essential to refine reimbursement standards, improve case-based payment, make clear the inclusion criteria for case and extend the covering range of case-based payment in Chengdu. In addition, the medical insurance agency should strengthen the supervision of healthcare providers,and guide them to set up effective incentive mechanism.
7.Enhancers on the transmembrane transport of chlorogenic acid.

To investigate the influence of the difference enhancers on the transport mechanism of chlorogenic acid (CGA) across Caco-2 cells model, a RP-HPLC method was adopted to detect the concentrations of CGA. At the concentrations of 20 to 80 microg x mL(-1), the difference of absorption rate constants (K(a)) was not statistically significant. At the concentrations of 40 and 20 microg x mL(-1), the ratios of apparent permeability coefficients (P(app)) of the apical to basolateral and the basolateral to apical were 1.14 and 1.18, respectively. With the effect of enhancers K(a) and P(app) increased, the absorption half-life (T1/2) decreased. CGA passed through the Caco-2 cell membrane mainly by passive transport. It showed that monocarboxylic acid transporter (MCT) could be involved in the across membrane transport process of CGA. Borneol had no effect on the cell membrane transport processes. The order of increasing absorption of CGA caused by the enhancers was sodium lauryl sulphate > sodium taurocholate > carbomer.
8.Aspirin resistance after off-pump coronary artery bypass graft surgery

ObjectiveThe present study was designed evaluate the aspirin effectiveness in the inhibition of platelet aggregation in patients after OPCAB.Methods290 patients were recruited.145 patients underwent first time OPCAB (surgery group).Arachidonic acid induced platelet aggregation and urine 11-dehydro thromboxane B2 (11-dehydroTxB2) were measured before operation and on aspirin re-administered days 1,4, 10, and 6 months after surgery.The same tests were also detected in 145 patients from the cardiology department (non-surgery group) received medicine therapy as controls.Results Ninety-nine patients were defined as aspirin sensitive after OPCAB (AS Group).Postoperative aspirin resistance was identified in 46 (32%) patients at the first day after aspirin treatment started (AR Group).19 (13%) and 5 (3%) patients remained as AR at day 4 and 10 after aspirin re-administration, respectively.Patients in the AR group had higher 11-dehydroTxB2 levels than those in the AS group (P = 0.049).Six months follow-up showed ARA-induced platelet aggregation was (11.5 ± 3.4) %.Urine level of 11-dehydroTxB2 was (50.3 ± 15.4) ng/L.No resistance was found.All cardiologic patients were identified as aspirin sensitive, the change of platelet aggregation and 11-dehydroTxB2 were similar as those in the AS group.Weight >75 kg and postoperative drainage >500 ml were risk factors of aspirin resistance after OPCAB.ConclusionAnti-platelet effect of aspirin was reduced during the early postoperative period in certain patients undergoing OPCAB.In case of resistance,antiplatelet treatment strategy should be intensified or modified.
9.Antenatal taurine supplementation reduces cerebral cell apoptosis in fetal rats with growth restriction

Objective To explore the effect of antenatal taurine supplementation on cerebral apoptosis and the expression of glial cell line-derived neurotrophic factor (GDNF) and caspase-3 in fetal rats with fetal growth restriction (FGR).Methods Fifteen pregnant Sprague-Dawley rats were randomly divided into three groups:control group,FGR model group (model group) and FGR with antenatal taurine supplementation group (taurine group).Taurine was added into the diet of taurine group at a dose of 300 mg/(kg · d) from the 12th day of gestation until natural delivery.Two appropriate for gestational age (AGA) newborn rats were randomly selected from each mother in control group and two FGR fetal rats were randomly selected from each mother both in model and taurine groups.Apoptosis of neural cells in the brain was detected by terminal deoxynucleotidyl transferase mediated nick end labeling (TUNEL).Changes in protein expression of GDNF and caspase-3 were detected by immunohistochemistry.Levene method,one-way ANOVA and SNK-test,or Kruskal Wallis rank sum test and Tamhane’ s test were applied for statistical analysis.Results (1)The total amounts of fetal rats in control group,model group and taurine group were 65,60 and 59.The mean body weight of fetal rats were (6.36±0.44) g,(4.55 ± 0.45) g and (5.11±0.67) g,respectively.All fetal rats developed FGR in model group,while 76.3%(45/59) of fetal rats were FGR in taurine group.Therefore,FGR model was successfully established.(2) In control group,there were few expression of TUNEL positive cells in cerebral cortex.A large amount of TUNEL positive cells were found in the cortex,hippocampal and white matter area in model group,but less positive cells were identified in taurine group than in model group.The amount of apoptotic brain cells in the three groups were (0.46 ± 0.11),(14.76 ± 3.42) and (6.78 ± 1.93),respectively(H=429.80,P=0.000).(3)There were only small amount of GDNF positive cells in cerebral cortex in control group and more in model group.The amount of GDNF positive cells was further increased in taurine group.The amount of GDNF positive cells in cerebral cortex in the three groups were (93.56± 6.73),(120.36± 6.23)and(139.56± 5.28),respectively (H=715.17,P=0.000).(4) Few caspase-3 positive cells were found in cerebral cortex in control group.A large amount of positive cells were found in model group and less positive cells were found in taurine group,which was still more than those in control group.The amounts of caspase-3 positive cells in the three groups were (7.50±2.31),(151.32±24.43)and(37.28±11.21),respectively (F=132.54,P=0.000).Conclusions The number of apoptotic neural cells in brain tissue of baby rats with FGR were significantly increased,which can be significantly alleviated by maternal antenatal taurine supplementation through upregulation of GDNF and downregulation of caspase-3 expression.
10.EUS elastographic patterns of normal pancreas and focal pancreatic lesions

Objective To investigate the EUS elastographic patterns of normal pancreas and focal pancreatic lesions, and its value for characterizing and differentiating between benign and malignant pancreatic tissues. Methods Between January and June 2009, 6 patients with normal pancreas and 9 patients with focal pancreatic lesions were enrolled. Real-time elastography was carried out during the conventional EUS examination. The elastographic images were scored with 1 to 5 based on the elastographic pattern. Results Nine focal pancreatic lesions were finally diagnosed as pancreatic cancer (n = 4), cyst-adenocarcinoma (n =1), cyst-adenoma (n = 2) and focal pancreatitis (n = 2), respectively. Pancreatic cancers were scored as 3(n = 1) and 4 (n = 3). Cyst-adenocarcinoma was scored as 5 and cyst-adenomas were scored both as 2,while focal pancreatitis were scored as 2 and 3, respectively. Elastographic pattern was scored as 1 (n = 5)and 2 (n = 1) in 6 normal pancreas. When scores 1 and 2 were assigned to benign lesion and 3 to 5 to malignancy, the overall diagnostic accuracy of EUS elastography for focal pancreatic lesions was 88. 9%(8/9). Conclusion There are apparent differences in elastographic patterns between benign and malignant pancreatic tissues. EUS elastography is a promising method that allows characterization and differentiation of benignancy and malignancy.
