1.Correlation between allergy and HLA genetic polymorphism
Chinese Pharmacological Bulletin 1986;0(04):-
Allergy increasingly threatens human health. At present, the molecular mechanisms of allergy have been one of the hot topics. It is believed that genetic factor and environmental factor play important roles in allergy. It is not surprising that particular HLA (human leukocyte antigen) class II alleles have been implicated in susceptility to allergic diseases and in restriction of the IgE responses to a variety of allergens. It will provide an original measure if the correlation between allergy and HLA genetic polymorphism was illuminated.

Chinese Pharmacological Bulletin 1986;0(06):-
Aim To explore the correlation of positive specific IgE antibodies with penicillin allergy and HLA-DRB genetic polymorphism. Methods Sequence specific primer-polymerase chain reaction(PCR-SSP) was used to type human leucocyte antigen-DBR(HLA-DRB) alleles in 80 patients with positive IgE antibodies to penicillin and 101 healthy subjects without any allergic reactions. Results When different penicillin antigens were analyzed separately, the gene frequencies of DR16 in patients with positive BPO-IgE antibody and DR14.1 in patients with positive PVO-IgE antibody were found greatly lower than those of the control subjects(0 vs 8.62%,P

Objective To investigate the potential mechanisms of vascular dementia (VD) by comparing the neuroethological and pathological changes of two VD models, established with spontaneously hypertensive rat (SHR) and the rat, of which the bilateral carotid arteries were ligated. Methods Ten male SHRs were employed and defined as hypertension control group. Thirty male Wistar rats were randomly assigned to operation group (bilateral carotid arteries were ligated), sham group and control group. All rats enrolled in the operation group were undergone a permanent ligation of bilateral carotid artery (2-VO). The memory function of rats was estimated by the mean escape latency of Morris water maze. The morphological changes of neuron cells in the frontal lobe, temporal lobes, hippocampus and thalami were observed with HE staining, while the changes of white matter around cerebral ventricles were checked with LFB staining, and the demyelination of nerve was calculated. Results The memory function of rats in SHR control group and Wistar operation group was decreased compared with that of the rats in sham group and control group (P0.05). Conclusions SHRs without intervention and Wistar rats undergone 2-VO can be used to reproduce the ideal VD model. Both chronic permanent hypoperfusion and hypertension may lead to the loss of neurons and myelin which may result in memory dysfunction. Hypoperfusion is even more harmful than hypertension. The present study suggests that loss of neurons and myelin may play a role in the VD.

Aim To evaluate the value of expressions of CD63 on basophils in diagnosing allergy to penicillins. Methods The expressions of CD63 on basophils activated with 9 kinds of antigens(PG:penicillin G,PV:phenoxomethylpenicillin,AMP:ampicillin,AX:amoxicilloyl,6-APA:6-aminopenicillanic acid,PHA:phenylacetic acid,PHOA:phenoxacetic acid,PHPG:N-(?-hydroxyphenyl)glycine,NPG:N-phenylglycine) were detected by FAST(Flow cytometric allergen stimulation test) in 43 patients with penicillins allergy.Eight types of specific IgE and IgG antibodies to penicillins were determinated by RAST and ELISA,respectively. Results The sensitivity and specificity of CD63 used as a marker in patients with penicillins allergy were 65.12% and 93.33%,respectively.Moreover,the sensitivities of CD63 and specific IgE were higher than those of specific IgG in patients with positive skin test(P

Objective To study the effect of solution focused theory on improving the sleep quality of ACS patients with clinical pathway. Methods 100 ACS patients with clinical pathway in Tangshan Gongren hospital from September 2010 to January 2011 were selected as study objects, and they were randomly divided into the control group(51 cases)and the intervention group(49 cases). Then the sleep quality was assessed during the first 24 hours being admitted and on the second and forth week after the intervention with the Athens Insomnia Scale(ASI).Results At the second week after intervention,4 items of the ASI Score of the observation group were different from the control group significantly. The 4 items were nighttime sleep disruption, waking up early, evaluation of total sleep quality, effect on the day of emotion. On the forth week after intervention, only the item of effects on daytime function was not different from the control group significantly, the scores of other 7 items were significantly different from those of the observation group. Conclusions The effect of solution focused theory in improving the sleep quality of ACS patients with clinical pathway is significant, and it plays an active role in improving the state of disease.

Objective To prospectively investigate the characteristics and correlative influential factors for the changes in pulmonary artery hypertension (PAH)in chronic kidney disease (CKD) patients without end-stage renal disease. Methods Complete clinical and laboratory data of 133 outpatients without ESRD (CKD stage 2,3 and 4)in Beijing Hospital between January 2012 and January 2013 were retrospectively analyzed.Pulmonary artery systolic pressure(PASP)was assessed by echocardiography according to the guideline from the American Society of Echocardiography in 2010,and PASP more than 35 mmHg(1 mmHg=0.133 kPa)was diagnosed as PAH.Results PAH was found in twelve(9.0 %) patients, including 7 females and 5 males, with mean age(82.9 ± 5.4 years). The prevalence of PAH was 2.3%,10.9% and 17.6% in the patients of CKD stage 2,3 and 4(P< 0.05) respectively.PASP could be calculated in 67 patients in whom tricuspid regurgitation was present. The mean PASP was (27.1 ± 5.4) mmHg,(31.0 ± 8.3) mmHg and (37.3 ± 13.8) mmHg in patients with CKD stage 2,3 and 4,respectively(P < 0.05).Body mass index(BMI),eGFR and BUN showed significant differences between PAH group and non-PAH group with statistical significance (P<0.05).Multivariate logistic regression analysis revealed that the lower BMI and the higher stage of CKD were independent risk factors for PAH (P < 0.05).Conclusions PAH is not a rare complication in the CKD patients without end-stage renal disease. The incidence of PAH is significantly associated with lower BMI and the higher stage of CKD.Doppler echocardiography should be performed periodically to detect PAH in CKD patients in order to improve the prognosis of these patients.

AIM:To investigate the effect of cyclopamine , a Hedgehog ( Hh) signaling pathway inhibitor , on the biological behavior of intrahepatic cholangiocarcinoma cell line RBE .METHODS:The proliferation of RBE cells was detected by cell counting with Typan blue staining and MTT assay , and the apoptosis was analyzed by the flow cytometry . The Transwell invasive cabin assay was used to detect the invasion ability , and Western blot was used to determine the pro-tein expression of Gli 1 and MMP-9 in the RBE cells before and after cyclopamine treatment .RESULTS:Cyclopamine in-hibited the growth of RBE cells in a time-and dose-dependent manner .After cyclopamine treatment for 24 h, 48 h and 72 h, the apoptotic rates were significantly higher than those in control group .In control group , the number of cells invading through the Matrigel of invasion chamber was 154.52 ±13.61, while in experimental group it was 62.00 ±12.17, indica-ting that the invasion ability of the cells declined significantly .Furthermore , Western blot showed that the protein levels of Glil and MMP-9 in the RBE cells were decreased after treatment with cyclopamine for 24 h and 48 h.CONCLUSION:Blockage of the Hh signaling pathway with cyclopamine suppresses the proliferation , promotes the apoptosis and inhibits the invasion ability of RBE cells .
