2.Non-linear developable model of skeletal muscle

Objective To discussing the methods used in creating non linear developable biomechanical models of skeletal muscle.Methods One of the basic tools of the analytical approach in analytical mechanics, the principle of virtual work for deformable systems, was used to create the model. By means of method of Lagrange multipliers, the problems of constraints, such as the incompressibility of skeletal muscle, were effectively tackled. Results and Conclusion The biomechanical models of skeletal muscle has been developed. The program of this model can be used to obtain nodal displacement coordinates of given nodal coordinates and parameters.

Objective To study the effects of valsartan and triptergium glycosides on the abnormal expression of connective tissue growth factor(CTGF) in rats with diabetic nephropathy. Objective To investigate in vitro anti-hydatid efficacy on Echinococcus granulosus protoscolex(EgPSC) by using albendazole sulfoxide(ASOX) and its two enantiomeric antipodes, L-ASOX and D-ASOX.Methods Eg protoscoleces were divided into eight groups and cultured in the DMEM culture media under two concentrations(50 ?g/ml and 100 ?g/ml) of ASOX, L-ASOX and D-ASOX respectively.The appropriate controls included(i) a culture containing an equal amount of DMSO and(ii) a culture medium alone.The mortality of EgPSC in each group was daily counted until 100% EgPSC death in some groups.Results Significant difference of EgPSC mortality was found among the three drugs with various concentrations compared to control group(P0.05), but between ASOX group and L-ASOX group(P

Objective To investigate the effects of rosiglitazone,an agonist of proliferators activated receptor?(PPAR?),on expression of nuclear factor-kappa B(NF-?B)in peripheral blood mononuclear cells(PBMC)of multiple organ dysfunction syndrome(MODS) rats.Methods Forty male SD rats were randomly divided into four groups(n = 6 per group): vehicle control group,endotoxin(LPS) group,rosiglitazone(ROSI) pretreatment group,and PPAR-?antagonist GW9662 pretreatment group.Blood was taken 4 hours after operation,and mononuclear cells were separated.The expression of NF-?B p65in PBMC was detected by immunocytochemical method and image analysis was carried out.Results The expression of NF-?B p65 was low in vehicle control group.In LPS group,the expression of NF-?B p65 was significantly higher than that in vehicle control group(P0.05).Conclusion Rosiglitazone protected the MODS rats by inhibiting NF-?B activation.
7.Effects of sevoflurane postconditioning on mitophagy during ischemia-reperfusion in rats

Objective To evaluate the effects of sevoflurane postconditioning on mitophagy during myocardial ischemia-reperfusion (I/R) in rats.Methods Forty-two pathogen-free adult male SpragueDawley rats, weighing 250-300 g, were randomly divided into 3 groups (n=14 each) using a random number table: sham operation group (group S) , I/R group and sevoflurane postconditioning group (group SP).Myocardial I/R was induced by 30 min ligation of the left anterior descending branch of the coronary artery followed by 2 h of reperfusion.In group SP, 2.4% sevoflurane was inhaled for 15 min starting from the onset of reperfusion, while 33% oxygen was inhaled in group I/R.The rats were sacrificed at the end of reperfusion, and the hearts were removed for measurement of myocardial infarct size (by 1% 2, 3, 5 triphenyltetrazolium chloride) , expression of LC3 Ⅱ/LC3 Ⅰ , Beclin-1, p62 and Parkin (by Western blot) ,and mitochondrial menbrane potential (by using JC-1 probe) , and for examination of the uhrastructure of cardiomyocytes (with transmission electron microscope).Results Compared with group S, the myocardial infarct size was significantly increased, mitochondrial membrane potential was decreased, the expression of LC3 Ⅱ/LC3 Ⅰ , Beclin-1 and Parkin was up-regulated, and the expression of p62 was down-regulated in group I/R.Compared with group I/R, the myocardial infarct size was significantly decreased, the mitochondrial membrane potential was increased, and the expression of LC3 Ⅱ/LC3 Ⅰ , Beclin-1, p62 and Parkin was down-regulated in group SP.Conclusion Sevoflurane postconditioning can mitigate I/R injury in rats, and inhibition of excessive activation of mitophagy may be involved in the nechanism.
8.Expression of transforming growth factor-alpha and its receptor during the process of human liver cirrhosis

BACKGROUND: The expression and significance of transforming growth factor-alpha (TGF-α) and epidermal growth factor receptor (EGF-R) in different diseases are different.OBJ ECTTVE: To investigate the expression of TGF-α and EGF-R in human liver cirrhosis tissues.DESIGN : Non-randomized controlled trial.SETTING: Department of General Surgery, Tangdu Hospital, Fourth Military Medical University of Chinese PLA.MATERIALS: This trial was carried out in the Department of Pathology, Xijing Hospital, Fourth Military Medical University of Chinese PLA during March 2003 to May 2004. Sixty-three samples of human liver cirrhosis tissue were obtained from Department of General Surgery, Xijing Hospital, Fourth Military Medical University of Chinese PLA (Informed consents were obtained). Five human normal liver tissues were obtained form autopsy in Department of Pathology, Fourth Military Medical University of Chinese PLA (patients with liver diseases were excluded).METHODS: Sixty-three samples of human liver cirrhosis specimens and five human normal liver tissues were studied with immunohistochemical method and in situ hybridization technique. The positive rates of TGF-α and EGF-R were calculated and managed statistically.MAIN OUTCOME MEASURE: Detection of the expressions of TGF-α and EGF-R in liver cirrhosis tissues were detected with immunohistochemical method and in situ hybridization technique.RESULTS: The expression rate of TGF-α and EGF-R in 63 samples of human liver cirrhosis tissues was 84% (53/63)and 52% (33/63) respectively. These positive granules were brown and most of them were in cytoplasm of hepatocytes.There was significantly positive correlation between TGF-α and EGF-R (r =0.32,P < 0.05). In five normal liver tissues,TGF-α and EGF-R were not detectable in hepatocytes and bile ducts. In situ hybridization showed the levels of TGF-αmRNA and EGF-R mRNA were higher (86%, 54%) than those of immunohistochemistry. There was no significant difference between two detective methods (P>0.05).CONCLUSrON: Liver cirrhosis might be under the autocrine regulation of TGF-α/EGF-R. Increasing expression of TGF-αand EGF-R might be one of important factors in liver cirrhosis pathogenesis.
9.THE APPLICATION OF NUCLEAR MEDICAL TECHNOLOGY IN PRIMARY HYPERPARATHYROIDISM

Objective To expound emphatically the application of nuclear medical examination in primary hyperparathyroidism.Methods 22 patients (19 adenomas,3 hyperplasia) with primary hyperparathyroidism confirmed by clinical examination and surgery underwent the detection of parathormone (PTH) by immunoradiometric assay (IRMA) and double phase 99 Tcm-MIBI parathyroid imaging,of whom 15 performed 99 TcmO-4 thyroid imaging and 8 99 Tcm-MDP bone imaging.Results The level of serum PTH in all patients (428.21±256.48) was significantly higher as compared to normal value and in cases with adenomas (781.83±194.56) was markedly elevated.The sensitivity of 99 Tcm-MIBI early and delayed phase imaging in detecting lesions of parathyroid adenomas was 84.2% and 100%,respectively.The sensitivity in detecting lesions of hyperplasia was only 33.3%.With 99 Tcm-O-4 thryoid imaging,the cold nodules were present in 2 and normality in 13.As to bone imaging,the scans were abnormal in 7.Their severities on bone scintigraph were correlated with the duration of the disease.Conclusions The level of serum PTH detected by IRMA and double phase 99 Tcm-MIBI parathyroid imaging are both sensitive methods in qualitative diagnosis and localization of hyperthyroidism.99Tcm-MIBI imaging has limited value for detecting the hyperplasia.The thyroid and bone imaging may well improve the diagnostic sensitivity and accuracy,and reflect the stage and progress of hyperparathyroidism.
10.Quantifying the effect of antiplatelet therapy by whole blood platelet aggregometry

Objective To study the impedance detected by the whole blood electrical impedance aggregometry in evaluating the effect of antiplatelet therapy.Methods The imprecision of the two different assays(the whole blood electrical impedance aggregometry and the plasma light transmission aggregometry) was compared.Thirty healthy volunteers and 60 patients with coronary artery disease under antiplatelet treatment were enrolled in the present study,whose anti-aggregated blood were collected for the measurement of electrical impedance in the presence of ADP,arachidonic acid and collagen,respectively.Finally,we analysed the agreement between the results of the two different assays which were used to measured the antiplatelet effect of aspirin and clopidgrel.Results The imprecision analysis showed the CV,(the whole blood electrical impedance aggregometry was 4.8%,superior to the plasma light transmission aggregometry (CV = 7.7%).In healthy control group,the mean impedances were(9.6?3.4)?、(10.4?2.8)? and (13.5?2.2)? in the presence of ADP,arachidonic acid and collagen as in the patients taking aspirin 100 mg/day,the 95% patients' results were 0 ?.Furthermore,the mean impedance of patients taking aspirin 100 mg/day and/or clopidogrel 75 mg/day was significantly lower than that of healthy volunteers(ADP as agonist,t = 2.391,P
