2.It’s Time for This“ROSE”to Flower:Rapid on Site Evaluation in Interventional Pulmonology

Rapid on site evaluation (ROSE) technology of interventional pulmonology includes“cytological ROSE”(C-ROSE) and“microbiological ROSE”(M-ROSE). Recently, this“ROSE”has gradually become one of core technologies in modern interventional pulmonology. In this commentary, perspectives on origin and development, classification and clini-cal value, operational approach, clinical application, and how to carry out effective work related to ROSE were summarized and remarked.
3.Study on feasible prophylactic proposal for the periventricular-intraventricular hemorrhage in preterm infants

Objective To study the feasible prophylactic proposal for the periventricular-intraventricular hemorrhage(PIVH)in preterm infants with the aim to reduce the mortality and improve the prognosis.Methods From January 2004 to June 2005,pregnant women at high risk for preterm delivery and with gestational age of less than 35 weeks and their infants were enrolled and divided into five groups randomly.Pregnant women received antenatal intramuscular or intravenously injection of vitamin K_1 10 mg per day for 2 to 7 days were Group A.Group B received antenatal intramuscular or intravenously injection of dexamethasone 10 mg per day for 1 day.Group C received antenatal intramuscular or intravenously injection of dexamethasone 10 mg per day for 2 days.Group D received dexamethasone 10 mg per day for 1 or 2 days and vitamin K_1 10 mg per day for 2 to 7 days.Infants in the phenobarbital group received intravenous injection of phenobarbital within 3 hours after birth for 3 to 5 days.Infants in the control group received neither phenobarbital after birth nor dexamethasone or vitamin K_1 injection antenatally.Results PIVH was diagnosed in 17 of 40(42.5%)in Group A,34 of 63(54.0%)in Group B,36 of 70(51.4%)in Group C,14 of 44(31.8%)in Group D,they all higher than control group(?2=15.10,P=0.004).More infants in the control group had severe PIVH(Grade Ⅲ and Ⅳ)than other groups(?2=9.527,P=0.049).The incidence of PIVH and severe PIVH in Group B and Group C were no significant difference.Furthermore,the incidence of PIVH was no difference in phenobarbital group(60.0%)and control group(65.2%)(?2=0.361,P=0.548).But the incidence of severe PIVH in phenobarbital group(5.0%)was less than control group(18.4%)(?2=4.04,P=0.044).Conclusions The combined antenatal administration of dexamethasone and vitamin K_1 can reduce the incidence of PIVH in preterm infants.Postnatal administration of phenobarbital can not decrease the incidence of PIVH,but decrease the frequency of severe PIVH.
4.Hypoglycemic Activity of Polysaccharide of Roots of Common Anemarrhena(Anemarrhena asphodeloides)

Root stem of Anemarrhena asPhodeloides was extracted with hot water,treated with ethanol and dilutealkali to temove protein. The polysaccharide thus obtained was given to mice by gastric gavage. Result showedthat the polysaccharide can marked1y lower the blood sugar and liver glycogen of mice without affecting bloodlipid level. When given intraperitoneally, it also showed hypoglycemic activity- For alloxan diabetic mice, thepolysaccharide can also markedly lower its b1ood sugar by gastric gavage.

In recent years,zebrafish has become ideal animal models of human disease with its unique characteristics,such as small body,fecundity,fast development and growth,embryo transparency,and so on.Furthermore,the structure and gene of zebrafish eye are highly conservative with human eye,which make ophthalmologists to pay close attentions to zebrafish.This review focused on the studies and applications on zebrafish embryonic development of eyes,and also the applications of currently used genetic regulation and editing techniques and tools.

7.Clinical application of wire guided localization of breast biopsy under X-ray guidance

Objective To study the diagnostic and treatment value of wire guided localization of breast biopsy for non-palpable breast lesions.Methods The clinical data of 314 cases of non-palpable breast lesion resection operation in our hospital were retrospectively analyzed.The accuracy rate of resection and postoperative pathological examination results were analyzed.Results All lesions were positioned accurately and completely resected,48 cases in 314 cases were breast cancer(15.3 %),in which 42 cases were early breast cancer which were confirmed by pathology (87.5%).Conclusion Wire guided localization of breast biopsy has high accuracy,low cost,which is useful for the diagnosis of early breast cancer,there is a certain value in clinical application.
8.Content Determination of Diosgenin in Paridis Rhizoma Medicinal Plants in Different Habitats of Three Gorges Reservoir

Objective To establish an HPLC method to determine the content of diosgenin in Paridis Rhizoma; To examine contents of diosgenin in different varieties of Paridis Rhizoma medicinal plants and from different habitats. Methods Shimadzu Inertsil ODS-C18 guard column (150 mm×4.6 mm, 5μm) was used in HPLC system; elution conditions were the mobile phase using 92% acetonitrile and 8% water (isocraticelution) at a flow rate of 1.0 mL/min with 203 nm detection wavelength. The temperature of the column oven was maintained at 30℃.Results Different varieties of Paridis Rhizoma medicinal plants and Paridis Rhizoma medicinal plants from different habitats contained diosgenin. TheParis polyphylla var.pseudothibetica in Yubei district had the highest content (6.813 7 mg/g);Paris polyphylla var.stenophylla in Chengkou County had the middle content (5.758 4 mg/g);Paris yunnanensisFranch in Shizhu county showed the lowest content (1.952 2 mg/g). The results showed that the obvious difference of chemical quality between various cultivated varieties of Paridis Rhizoma medicinal plants, which might be related with origin, variety and the cultivation technology, and so on.Conclusion The diosgenin contents ofParis polyphylla var. pseudothibetica andParis polyphylla var.stenophylla were higher than that of varieties recorded in Chinese Pharmacopeia.Paris polyphylla var.pseudothibetica andParis polyphylla var.stenophylla had important medical value.
9.Influence of RNAi on the silencing expression of E-cadherin and the proliferation ability of Hep-2 trained in vitro

OBJECTIVE To explore the effect of E-cadherin on the proliferation ability of Hep-2 by method of RNA interference technology to silence the expression of E-cadherin. METHODS The specific siRNA sequences and non-silencing siRNA were designed and synthesized. Hep-2 cells were transfected and then the down expression of E-cadherin gene in vitro cultured Hep-2 cells were got. The silencing effect of E-cadherin gene was explored by Fluorescence Quantitative Polymerase Chain Reaction and the proliferation of the transfected Hep-2 cells were detected in vitro by MTT assay. RESULTS 1.When transfected with the ratio of recombinant plasmid and the quality of liposome volume at 1:1, the transfection efficiency at the siRNA-3 group was the highest and can be up to 65%. 2.The results of Fluorescence Quantitative Polymerase Chain Reaction: recombinant plasmid pRNAT-U6.1/Neo-siRNA1, pRNAT-U6.1/Neo-siRNA2 and pRNAT-U6.1/Neo-siRNA3 can down regulate the expression of E-cadherin mRAN. Set blank control group as a baseline (set to 1), the changes of expression of E-cadherin relative to β-actin in siRNA-1group was 0.00092, siRNA-2 group was 0.00143, siRNA-3 group was 0.00045 and the negative control group was 3.44898. The difference was statistically significant (P<0.05). 3. MTT: The growth rate of Hep-2 cells treated by specific siRNA was faster than that of the control group, and the difference was statistically significant (P<0.05). CONCLUSION Effectively inhibition the expression of E-cadherin's mRAN can enhance the proliferation of Hep-2 cells.
10.Immune enhancing effects of interleukin 2 genetic adjuvant on DNA vaccine against Chlamydia trachomatis serovar E

ObjectiveTo study the effect of a recombinant plasmid encoding mouse interleukin 2 (mlL-2) on the immunogenicity of DNA vaccine against Chlamydia trachomatis(Ct) serovar E.Methods BALB/c mice were divided into 4 groups to be intramuscularly inoculated with blank plasmid(negative control group),DNA vaccine against Ct serovar E(DNA vaccine group),DNA vaccine against Ct serovar E and a recombinant plasmid containing mIL-2(combination group),and inactivated Ct serovar E elementary bodies (positive control group),respectively.The immunological effects were evaluated by posterior foot pad thickness,proliferation level of spleen lymphocytes,serum level of IL-4 and interferon (IFN)-γ in mice,and the capability to clear Ct genital tract infection.ResultsThe proliferation index of spleen lymphocytes in the combination group and positive control group was similar(3.64 ± 0.41 vs.3.77 ± 0.34),but was significantly different from that in the blank control group and DNA vaccine group (1.37 ± 0.21 and 2.52 ± 0.30).The serum level of IL-4 was(38.49 ± 12.24) pg/ml in the positive control group,significantly higher than in the negative control group,DNA vaccine group and combination group ((25.37 ± 18.93),(24.75 ± 8.49),(21.74 ± 6.43) pg/ml,respectively).With respect to the serum level of IFN-γ,the combination group and positive control group were similar ((1923.3 ± 518.1) pg/ml vs.(2712.5 ± 887.2) pg/ml),but were significantly different from the negative control group and vaccine group((310.8 ± 160.7) pg/ml and(601.3 ± 357.9) pg/ml).Six days after Ct challenge,the exfoliated cells from genital tract were positive for Ct culture in the negative control group,but negative in the other 3 groups.ConclusionIL-2 genetic adjuvant can enhance the immune response,especially Th1 type response,induced by the DNA vaccine against Ct serovar E.
