1.Effect of beclin1 on vincristine-induced dopaminergic neurons injury in zebrafish.

To investigate vincristine-induced dopaminergic neurons toxicity and mechanism, and explore the molecular target to reduce the toxicity, zebrafish was chosen as a model animal, based on RT-PCR, Western blotting, whole mount in situ immunofluorescence and other technical means. The results showed that the transcription levels of tyrosine hydroxylase gene and dopamine transporter protein gene were inhibited. Furthermore, the number of dopaminergic neurons was decreased by vincristine. Autophagy was suppressed and beclin1 gene expression was inhibited in a dose-dependent manner by vincristine in larval zebrafish. Up-regulated beclin1 partly reduced vincristine-induced neurotoxicity, and down-regulated beclin1 increased toxicity. Beclin1 plays an important role in vincristine-induced dopaminergic neurons toxicity.

2.Improvement and optimization of performance verification on enzyme linked immunosorbent assay for determination of hepatitis B markers

Objective To evaluate the performance of enzyme linked immunosorbent assay (ELISA) kit in detection of Hepatitis B virus(HBV) markers by using improved and optimized method ,so as to provide a practical and feasible method and reagents for clinical laboratory .Methods ELISA test was used for the detection of HBV markers .The gradient dilution method was used to e‐valuate the lower limit .The verifiation of cut off value was carried out based on clinical and laboratory standards institute (CLSI) EP12‐A2 document .Samples with cut off values were collected to evaluate the precision ,including repeatability and intermediate precision .The coincidence rates were counted through comparing the results of ELISA with those of external quality assessment and those detected by using Abbott i4000SR chemiluminescence instrument .Results The lower detection limit of HBsAg ,HBsAb , HBeAg ,HBeAb and HBcAb were 0 .2 IU/mL ,20 mIU/mL ,1 NCU/mL ,0 .75 NCU/mL and 0 .05 NCU/mL respectively .The cut‐off value(C50 )± 20% concentration included the concentration range between C5 and C95 .The within‐run coefficient of variation (CV)≤15% ,in sandwich method the between‐run CV≤25% ,in competition method the between‐run CV≤35% .The positive and negative coincidence rates in accuracy and comparing with i 4000SR all were more than 95% and all κ>0 .75 .Conclusion ELISA tests for HBV markers in our laboratory could meet the requirements of the detection performance and clinical needs .

5.Effect of high-pressure steam sterilization on two kinds of orthodontic distal end cutters

BACKGROUND:Steam sterilization is a commonly used oral clinical method of disinfection, but whether it would produce more significant impact on equipments waiting for sterilization has been controversial.
OBJECTIVE:To investigate the effect of autoclave on two kinds of orthodontic distal end cutters by comparing shearing force and corrosion after sterilization.
METHODS:Tiantian (n=15) and 3M (n=15) distal end cutters commonly used in orthodontic treatment were chosen for testing and divided into three groups, respectively. Three groups for each kind of cutter were treated with high-pressure steam sterilization, steam sterilization+extraoral shear, and steam sterilization+intraoral shear, respectively. The shearing force and metal ography of the distal end cutters were recorded after 0, 20, 40, and 60 cycles of sterilization. An energy dispersive spectrometer was used to scan the surfaces of the working edges of the distal end cutters after 60 cycles.
RESULTS AND CONCLUSION:It was found that with an increase in the number of sterilization cycles, the shear force and extent of corrosion increased in al six groups of distal end cutters, but the situation was worse for Tiantian cutters than for 3M cutters. It was found that the difference of shear force between the Tiantian autoclave group and 3M autoclave group was statistical y significant (P<0.05) after 20, 40, and 60 sterilization cycles. Meanwhile, the metal ography showed that different degrees of discoloration and rust spots appeared on the work edges of the two brands of distal end cutters with an increase in the number of sterilization cycles, but the corrosion of the Tiantian distal-end cutter was more widespread, and even work-edge defects appeared. The carbon content of the Tiantian distal-end cutter was higher than that of the 3M cutter (P<0.05), but the chromium content was higher in the 3M cutter (P<0.05). In addition, aluminum and titanium elements only existed in the 3M cutter. The use of the autoclave affected the distal end cutters in terms of shearing force and corrosion, but the effect on 3M distal end cutters was obviously less than that on Tiantian cutters.
6.Profile and mechanism of imipenem resistance in clinical isolates of Serratia marcescens

Objective To investigate the profile and mechanism of imipenem resistance in the clinical isolates of Serratia marcescens. Methods A total of 152 strains of S. marcescens were isolated from January 2013 to December 2014. Disk diffusion method?and?automated?systems?were?used?to?determine?the?susceptibility?of?the?isolates.?The?modified?Hodge?test?and?EDTA?synergy?test?were?employed?to?test?carbapenemase?phenotype.?Agar?dilution?method?in?combination?with?efflux?pump?inhibitor?MC207110?was used to observe the change of imipenem MIC values. The genes encoding carbapenemase, AmpC beta-lactamase and outer membrane protein were detected by polymerase chain reaction. Results? Imipenem?resistance?was?identified?in?87?of?the?152?strains?of S. marcescens.?Modified?Hodge?test?was?positive?for?12?of?the?87?imipenem-resistant?S. marcescens strains. EDTA synergy test was?positive?for?9?of?the?strains.?Imipenem?MIC?was?reduced?to?1/4?to?1/64?in?46?strains?by?agar?dilution?method?in?presence?of?efflux?pump inhibitor MC207110. Five strains were positive for KPC genes, 8 strains positive for IMP gene, and 6 strains positive for DHA gene. Loss of outer membrane protein was found in 16 strains by PCR. Conclusions Imipenem-resistant strains of S. marcescens are?prevalent?in?our?hospital.?KPC,?IMP,?DHA?genes,?and?loss?of?outer?membrane?proteins?and?efflux?pumps?may?all?contribute?to?imipenem resistance in S. marcescens isolates.
7.Clinical characteristics and treatment outcome of 145 cases with multiple myositis and dermatomyositis

Objective To investigate the clinical characteristics of patients with polymyositis(PM) and dermatomyositis (DM), and compare the differences of PM/DM to help the understanding of clinical diagnosis and treatment. Methods One hundred and forty-five hospitalized PM/DM patients from Department of Rheumatology of the First Affiliated Hospital of Xiˊan Jiaotong University were collected from May 2008 to December 2014, and the clinical manifestations, muscle enzymes, electromyogram, muscle biopsy, treatment outcome were retrospectively analyzed. Mann-Whitney U test and χ2 test were used for statistical analysis. Results The most common initial symptom of PM was muscle weakness, accounted for 51.2%, while rash was the initial presentation in most DM patients(43.1%). The incidence of interstitial lung disease (ILD) (62.7% vs 39.5%, χ2=11.009, P=0.001), and the elevation of CRP (48.9% vs 26.8%, χ2=10.272, P=0.001) were all higher in DM than PM, while the elevation of level of CK (85.4% vs 61.8%, U=-2.668, P=0.008) and CKMB (82.9%vs 41.2%, U=-3.303, P=0.001) were more common in PM compared with DM. The pathological study showed degeneration of muscle fiber, connective tissue hyperplasia in most PM patients, and perimysium atrophy, vacuoles degeneration, muscle bundles, perivascular inflammatory cell infiltration were observed in most DM patients. During the follow-up, the clinical remission rate was 57.5%, the relapse rate and the mortality rate was 7.5%and 31.1%respectively. The mortality rate was higher in DM than PM (34.6% vs 21.4%, χ2=4.861, P=0.027). Infection and tumors were the major causes of death, and the lung was the most common site of infection. Conclusion Differences in the clinical features, muscle enzymes, CRP level, pathology and the mortality rate between PM and DM are evident, while ILD, infection and the higher mortality rate are more common in DM than in PM.
8.Effects of placental transfusion of umbilical cord milking on very low birth weight infants

ObjectiveTo study the effects of placental transfusion of umbilical cord milking on very low birth weight (VLBW) infants. Methods Fifty-seven VLBW infants born from September 2011 to May 2014 who had umbilical cord milking at birth were selected as experimental group. Sixty-one VLBW infants born from January 2008 to August 2011 who had normal cord clamping at birth were selected as control group. The complications of VLBW infants, blood transfusion, frequency of using pulmonary surfactant (PS), the duration of mechanical ventilation, the duration of oxygen and mortality were compared between two groups.Results The incidence of severe asphyxia, IVH and anemia was signiifcantly lower in experimental group than in control group (P< 0.05). The blood transfusion and transfusion volume, duration of mechanical ventilation, and duration of oxy-gen were signiifcantly lower in experimental group than in control group (P< 0.05).Conclusions Umbilical cord milking can reduce the incidence of severe asphyxia, IVH and anemia. It also can reduce the blood transfusion, the duration of mechanical ventilation, and the duration of oxygen in VLBW infants.

10.Study of cardiac torsion in fetuses with gestational diabetes mellitus by velocity vector imaging

Objective To investigate the clinical value of velocity vector imaging (VVI) in the assessment of cardiac torsion in fetuses with gestational diabetes mellitus(GDM).Methods Digital dynamic four-chamber views of 98 fetuses with gestational diabetes mellitus and 135 normal fetuses were collected and analyzed using VVI.Left ventricular(LV) peak apical rotation of endocardium and epicardium at basal and apical level were measured,LV peak torsion was calculated.Results 140 of 233 cases were analyzed successfully.Direction of LV peak apical and basal rotation of endocardium and epicardium in fetus could be clockwise or counter clockwise,however apical and basal level was opposite.LV peak apical and basal rotation of endocardium and epicardium in fetus were stable throughout gestation ( P > 0.05).LV peak rotation of endocardium was higher than that of epicardium in both apical and basal level.The peak rotation in apical level of endocardium and epicardium was higher than that in basal level( P <0.05).LV peak torsion of endocardium was significantly higher than that of epicardium( P <0.05).Compared with normal group,LV peak rotation of endocardium and LV peak torsion in fetus with GDM were higher( P <0.05). Conclusions VVI can measure the LV rotation and torsion parameters,and evaluate the cardiac function of GDM fetus.
