AIM: To observe the effects of Dispel Stasis Decoction(Radix et Rhizowa Salviae multiorrhizae,Rhizoma Sparganii,Rhizoma Curcumae, etc) on TNF-? and E_2 in rats with endometriosis(EMS),and to observe its anti-inflammatory and analgesic effect. METHODS: Endometriosis rat models were made by autograft of endometrium in abdomen.Four weeks after operation,EMS model rats were randomly divided into 5 groups: a model group,3 Dispel Stasis Decoction groups and a danazol group. All the rats were treated for three weeks respectively, and then anaesthetised and killed. The ectopic endometrium's focuses were observed by microscope, the levels of TNF-? and E_2 in the serum of all groups were measured by radioimmunoassay. The method of body twist and ear edema of mouse were used to the analysis the anti-inflammatory and analgesic effect of Dispel Stasis Decoction. RESULTS: The survived graft shrinked and the number of the glanduls decreased in rats of Dispel Stasis Decoction group. The levels of TNF-? and E_2 in the serum were significantly lower than that of the model group. Also,Dispel Stasis Decoction could inhibite the ear edema of the mouse induced by dimethylbenzene and the body twist of the mouse caused by acetic acid notablely. CONCLUSION: Dispel Stasis Decoction inhibited growth of the graft and showed a significant therapeutic effects on EMS model rat.It also has notable functions of anti-inflammatory and analgesic. This may be related to its inhibition of TNF-? and E_2 levels.

Basic & Clinical Medicine 2006;0(11):-
Objective To reconfirm the mesangial cell differentiation potential of BMSCs and to study the mechanism of cell induction. Methods Rat BMSCs were isolated and incubated with platelet-derived growth factor(PDGF)-BB to induce the mesangial cell phenotype. At day 7,the cell morphology, mesangial cell-specific markers and cell contraction function were examined.MAPK/ERK kinase inhibitor(PD98059) was added to examine its impact on cell growth and mesangial cells marker expression. Results After cultivation under the differentiation condition for 7 days, the induced cells expressed ?-actin and Desmin, which highly resembled cultured mesangial cells in rat.The induced cells with increasing level of PDGF receptors mRNA expression changed into a wide range of shapes from spindle to stellate and were contracted in response to angiotensin Ⅱ.PD98059 perturbed the induced cells mesangial cell-specific markers mRNA expression. Conclusion BMSCs can differentiate into mesangial-like cells with PDGF-BB induction in vitro. The mechanism involved in the differentiation was mediated through MAPK/ERK1/2 signal transduction pathway.

Objective To investigate the effects of different doses of dexamethasone (Dex) on bone mineral density (BMD) and body composition in Sprague-Dawley (SD) rats. Methods Forty 3-month-old female SD rats were divided into four groups:control group (Cont, saline), low dose Dex group (LDG, 1 mg/kg), medium dose Dex group (MDG, 2.5 mg/kg) and high dose Dex group (HDG, 5 mg/kg). Ten rats for each group. Dex was injected intramuscularly twice a week. The values of BMD and body composition were measured by DEXA densitometer at the beginning and 4-week of treatment. Results The body weights of different doses of Dex intervention groups were decreased after 1 to 4-week intervention compared with those of Cont group (P<0.01). After 4-week intervention, the total BMD, femur BMD, total bone mineral content (BMC), to-tal fat mass, trunk fat mass and leg fat mass were significantly increased in Cont group (P<0.01), while the total lean mass, trunk lean mass were significantly decreased (P<0.01). The total BMC, femur BMD, leg lean mass, leg fat mass were signifi-cantly lower in LDG group and MDG group than those of Cont group (P<0.05). The femur BMD, leg fat mass were signifi-cantly lower in LDG group and MDG group than those of HDG group (P<0.05). Conclusion The doses of 1 mg/kg and 2.5 mg/kg Dex have greater impact on the femur BMD and the leg composition in SD rats than that of Dex (dose of 5 mg/kg ).

To investigate the morphogenetic process of human adenovirus type 41 (HAdV-41), 293TE cells were infected with purified wild-type HAdV-41, and ultrathin sections of infected cells were prepared and observed under a transmission electron microscope. Results showed that HAdV-41 entered host cells mainly through three ways: non-clathrin-coated pit, clathrin-coated pit, and direct penetration of plasma membrane. In addition, cell microvilli might help HAdV-41 enter cells. After entering into cells, HAdV-41 virus particles could be found in vacuoles or lysosomes or be in a free state in cytoplasm. Only free virus particles could be found near nuclear pores (NP), suggesting that the virus needed to escape from lysosomes for effective infection and viral nucleoprotein entered the nucleus through NP. Progeny viruses were as-sembled in the nucleus. Three types of inclusion bodies, which were termed as fibrillous inclusion body, condense inclusion body, and stripped condense inclusion body, were involved in HAdV-41 morphogenesis. In the late phase of viral replication, the membrane integrity of the infected cells was lost and viral particles were released extracellularly. This study reveals the partial process of HAdV-41 morphogenesis and provides more biological information on HAdV-41.
