2.Analysis of risk factors of pressure ulcer healing

Objective To investigate the risk factors of wound healing 30 days after treatment in patients with pressure ulcer. Methods Two hundred and forty-five hospitalized patients with pressure ulcer between January 2015 and October 2016 were selected. They were divided into the healed group and the unhealed group 30 days after the treatment. The influencing factors 13 indexes including age, gender, edema, the number of diseases, the number of pressure ulcer, the size of pressure ulcer, PUSH score (Pressure Ulcer Scale Healing), hemoglobin, serum albumin, low density lipoprotein cholesterol and blood homocysteine underwent single factor and Logistic regression analysis. Results Single factor analysis of 12 indicators of risk factors affecting healing were age, Hb, Alb, Hcy, area of ulcer and PUSH scores of the 6 indicators in the regression equation (Z=3.11-6.63, All P < 0.01), gender also entered the regression equation (χ2=0.19, P<0.01);further multivariate regression analysis showed that there were statistically significant differences in PUSH score index (χ2=10.47, P<0.01, OR=0.001, 95%confidence interval were 0.001-0.037). Conclusions Age, gender, Hb, Alb, Hcy and PU area were the risk factors of pressure ulcer healing.
3.Investigation on cognitive status of emergency nurses on intravenous infusion of high-alert drugs in maternal and child hospital

Objective:To describe the status and influencing factors of emergency nurses′ high-alert drugs knowledge level in maternal and child hospital.Methods:Totally 171 nurses working in emergency department from 6 hospitals in Chengdu including West China Second University Hospital, Sichuan University, Sichuan Maternal and Child Health Hospital, Chengdu Women and Children Central Hospital, Chengdu Longquanyi Maternal and Child Health Hospital, Chengdu Chenghua Maternal and Child Health Hospital, Chengdu Pengzhou Maternal and Child Health Hospital were selected through convenient sampling to fill out the scale.Results:Nurses′ average score of high-alert drugs knowledge was 29.52±2.74. The accuracy of each item ranged from 22.2% to 100.0%. Multivariate linear regression analysis showed that major influencing factors for their knowledge level included monthly income, length of service in the emergency department, professional title, the model explained 12.7% amount of variance ( R2 value was 0.127, P<0.01). Conclusions:Emergency department nurses in maternal and child hospitals have insufficient awareness of intravenous infusion of high-alert drugs, and they have a high demand for knowledge. In order to ensure the safety of clinical drug use, it is urgent to formulate targeted training programs to improve nurses' knowledge level of high-alert drugs.
5.Clinical research of different concentration of heparin sodium solution sealing up intravenous indwelling needle in patients with Ⅳ thrombocytopenia

Objective To explore the optimal concentration of heparin sodium solution, which to seal up intravenous indwelling needle, in patients with Ⅳ thrombocytopenia. Methods A total of 90 patients withⅣthrombocytopenia who required intravenous indwelling needle were randomly divided into three groups which namely A, B and C groups with 30 cases each. A group was sealed up the tube of intravenous indwelling needle with 1 ml heparin sodium solution of 6.25 U/ml, B group with 12.50 U/ml and C group with 25.00 U/ml. The change of the platelet count, plasma prothrombin time (PT), activated partial thromboplastin time (APTT), plasma fibrinogen (Fbg) after extubation were observed in three groups and the incidence of blocking up and retention time were compared among three groups. Results The change of the platelet count, PT, APTT, Fbg were showed no statistical significance among three groups (P>0.05). The incidence of blocking up was 66.7%(22/30) in A group , 43.3%(13/30) in B group and 16.7%(5/30) in C group. They were showed statistical significance (χ2=15.000, P < 0.01). Retention time of tube above or equal to 3 days were observed respectively 13 person (43.3%,13/30) in A group, 21 cases(70.0%, 21/30) in B group, 25 patients(83.3%, 25/30) in C group. All above were showed statistical significance (χ2=11.000, P<0.01). Conclusions Heparin sodium 25.00 U/ml sealing up the tube of intravenous indwelling needle is safe in patient with Ⅳthrombocytopenia, which will reduce the ration of blocking tube and extend the life of tube.
6.Clinical application of visual evoked potential in orbital cellulitis of infants

AlM: To explore the visual evoked potential in infantile orbital cellulitis' clinical applications by monitoring the visual evoked potential changes in infantile orbital cellulitis before, during and after treatment.
METHODS:Twenty-three cases of CT diagnosed single orbital cellulitis were examined by the visual evoked potentials. The affected eyes as observation group, and healthy eyes as control group. Comparative observation of visual evoked potential changes in amplitude and incubation period before, during and after the treatment. RESULTS: Compared with the control group, the observation group's visual evoked potential changes included reduced amplitude, extended incubation period. With the treatment progress, the observation group had gradual increase in amplitude, gradual reduction in incubation period.
CONCLUSlON: ln infantile orbital cellulitis, the use of visual evoked potentials is a simple, feasible and effective method to monitoring the visual function during the treatment.

8.Evaluation of immunogenicity of an immune complexed hepatitis B vaccine

Objective To evaluate the immunogenicity of an immune complexed hepatitis B vac-cine ( HBsAg-HBIG immune complexes , IC) in mouse and cynomolgus monkeys by using recombinant hepa-titis B vaccine ( Saccharomyces cerevisiae, HBsAg) as the control .Methods BALB/c mice were vaccinated with single dose of IC and single dose of HBsAg respectively and then serum samples were collected at differ -ent time points for the detection of dynamic anti-HBs by using ELISA .The serum anti-HBs titers in BALB/c mice vaccinated with different immunization strategies were also analyzed .ELISPOT assay was performed to detect the numbers of IFN-γSFC and IFN-γpositive rate in splenocytes of BALB/c mice intramuscularly im-munized with IC, HBsAg or standard hepatitis B vaccine at 5μg/mouse.ED50 was measured to evaluate the stability of IC.Twelve cynomolgus monkeys were equally divided into two groups and immunized with high dose (100 μg) and low dose (20 μg) of IC respectively and then , serum anti-HBs levels at different time points were detected .Results The serum anti-HBs titers in IC immunized group at different time points were higher than those immunized with HBsAg .Moreover, the anti-HBs titer induced by two doses of IC reached a level comparable to that elicited by three doses of HBsAg .ELISPOT assay showed that both the numbers of IFN-γSFC and IFN-γpositive rate were the highest in IC immunized group as compared with those immunized with HBsAg and standard hepatitis B vaccine .IC had a lower ED50 than HBsAg, indicating a good long term stability .Cynomolgus monkeys immunized with high or low dose of IC produced high levels of anti-HBs titer during a long time period .Conclusion IC has a higher immunogenicity inducing both hu-moral immunity and cellular immunity as compared with HBsAg or standard hepatitis B vaccine .
9.Expression analyses of BcUGT3 and BcUGT6, and their in vitro expression in Escherichia coli.

The tissue-specific and MeJA-induced transcriptional levels of BcUGT3 and BcUGT6 in Bupleurum chinense were analyzed in the present study. The transcriptional levels of BcUGT3 in root, leaf, flower and fruit were similar and they all were higher than those in stem. The transcriptional level of BcUGT6 was the highest in leaf and the lowest in flower among in all tested tissues. With non-treated adventitious roots as control, BcUGT6's transcriptional levels were elevated to nearly 2 folds for 2 h, 8 h, 24 h, 2 d and 4 d in MeJA-treated adventitious roots of B. chinense. It showed that the transcriptional level of BcUGT6 was slightly affected by MeJA. While, BcUGT3's transcriptional levels were gradually elevated, and till 4 d after MeJA treatment, the expression level was about 7 folds than that of non-treated control. Using pET-28a (+), the expressions of two genes was investigated. Induced by IPTG, the target proteins were expressed in E. coli and then purified. All the results obtained in the present study will be helpful for follow-up bio-function analysis of BcUGT3 and BcUGT6.

10.High-risk human papillomaviral infection characterized by pseudocondyloma of vulvae

Objective To investigate the clinical features and human papillomavims (HPV) types in 12 cases of HPV infection characterized by pseudocondyloma of vulvae. Methods Twelve women, who had a history of sexual behavior with non-spouses and presented with pseudocondyloma of vulvae, were included in this study. Tissue specimens were scraped or resected from the lesions of pseudocondyloma of vulvae. PCR-based reverse hybridization assay was performed to determine the genotypes of HPV. Results All the 12 specimens were HPV positive. The reverse hybridization assay generated 13 subgenotypes of HPV, including 3 low-risk genotypes (11, 42 and 43) and 10 high-risk genotypes (16, 18, 31, 33, 35, 39, 51, 52, 58 and 59). All the patients were found to be infected by at least one high-risk genotype. Of the 12 patients, 4 were infected by single type of HPV, 6 by 2 types of HPV, and 2 by 3 or 4 genotypes of HPV. Conclusion The patients characterized by pseudocondyloma of vulvae may suffer from high-risk HPV infection.
