1.Effect of beclin1 on vincristine-induced dopaminergic neurons injury in zebrafish.
Acta Pharmaceutica Sinica 2014;49(6):843-848
To investigate vincristine-induced dopaminergic neurons toxicity and mechanism, and explore the molecular target to reduce the toxicity, zebrafish was chosen as a model animal, based on RT-PCR, Western blotting, whole mount in situ immunofluorescence and other technical means. The results showed that the transcription levels of tyrosine hydroxylase gene and dopamine transporter protein gene were inhibited. Furthermore, the number of dopaminergic neurons was decreased by vincristine. Autophagy was suppressed and beclin1 gene expression was inhibited in a dose-dependent manner by vincristine in larval zebrafish. Up-regulated beclin1 partly reduced vincristine-induced neurotoxicity, and down-regulated beclin1 increased toxicity. Beclin1 plays an important role in vincristine-induced dopaminergic neurons toxicity.

International Journal of Traditional Chinese Medicine 2016;38(3):212-215
Objective To discuss the relationship between the expression of programmed death-1 (PD-1) in liver with traditional Chinese medicine (TCM) syndromes and pathological diagnosis in chronic hepatitis B.Methods 156 CHB patients treated in our hospital of infectious diseases department were recruited as an observation group. Based on the principle of informed voluntary and approved by the ethics committee, liver biopsy was adopted to make clear liver tissue pathology. According to TCM classification criteria, CHB patients were divided into five groups: a blood stasis group, a damp heat resistance group, a liver and spleen deficiency group, a liver and kidney yin deficiency group, and a spleen and kidney yang deficiency group. In the same period, 12 healthy persons were recruited as a control group. The PD-1 expression was detected with immunohistochemical SP method, and the correlation between expression of PD-1 in liver tissue and TCM syndrome type and liver pathology was analyzed. Results Different degrees of positive cell expression were found in the liver tissue. With the liver inflammation and fibrosis severity, the number of PD-1 positive cells also increased. The PD-1 expression levels varied with mild, moderate, and severe CHB patients (0.24 ± 0.03, 0.36 ± 0.05 vs. 0.43 ± 0.05) , which were statistically significant (P<0.05) . PD-1 expression levels also varied among different TCM type CHB patients, of which, PD-1 expression of blood stasis type was the highest (0.35 ± 0.04), while the liver and spleen deficiency type was the lowest (0.23 ± 0.03).Conclusion The expression levels of PD-1 has a certain correlation with the patients illness, chronic mechanism, and TCM syndromes. CHB patients can be treated by controlling the expression of PD-1.

Objective:To review the application of antihypertensive drugs in Wuhan Xiehe Hospital in the period of 1996 to 1998.Methods:Statistically analyze the application of antihypertensive drugs during the period in terms of their types, sales income,DDDs*d-1 as well as the changes of daily expense on drugs. Results:During the period analyzed, the newly applied drugs are mainly ACEI drugs. The DDDs*d-1 of metoprotol, benazepril, nifedipine and nimodipine had been among the highest. The increase of DDDs*d-1 was rapid. The newly prescribed drugs were mainly oral and retard preparations. Conclusion:The Prescription of antihypertensive drugs in Wuhan Xiehe Hospital was rational and in accordance to the principles of convenience and cost-effectiveness.

AIM:To establish a qualitative fingerprint analysis with capillary GC. METHODS:HP-INNOWAX column(30 m?0.53 mm,1.0 ?m),column temperature:90 ℃(15 min)4 ℃/min180 ℃(5 min)2 ℃/min200 ℃(3 min)10 ℃/min220 ℃(5 min),FID detector temperature:230 ℃. RESULTS:Compared with reference peak of Sec-butyl-cis/trans-1-propentyl disulfide,17 common peaks on fingerprints of the oil from ferula sinkiangensis K.M.Shen were indicated by the method with satisfied stability, precision and repeatability. CONCLUSION:There are good similarities among 10 batches of essential oil from Ferula sinkiangensis K.M.Shen,and this method is reliable, simple and provides a reference standard for the quality control of Ferula sinkiangensis K.M.Shen.

Chinese Journal of Hypertension 2006;0(09):-
Background The ubiquitin-proteasome pathway(UPP)involves 80%-90% degradation of all in- tracellular proteins. Both ubiquitin and rat component 3 of proteasome(RC3)are hence considered to be central me- diators for cell biology. Objective To evaluate the effect of simvastatin on neointimal hyperplasia and the expres- sion of ubiquitin and rat component 3 of proteasome(RC3)after balloon injury in carotid artery. Methods Thirty- two male SD rats were randomly to receive, low dose(0.4 mg/ng,n=8),or moderate(4 mg/ng,n=8),or high- dose(40 mg/ng,n=8)of simvastatin treatment for 28 days.Common carotid aortic artery was injuried rat ballon. The ratio of intima-media(I/M)thickness was determined. The expression level of ubiquitin and RC3 mRNA were assessed by RT-PCR.The expression level of ubiquitin protein was examined with immunohistochemistry. Results Simvastatin inhibited the expression of ubiquitin and RC3 mRNA and ubiquitin protein in dose dependent manner. The intima-media ratio(-50.2 %)and the expression of ubiquitin(-60.3 %)and RC3 mRNA and ubiq- uitin protein(-60.5 %)was reduced by the high dose simvastatin [40 mg/(kg?d)](P

Objective To investigate the changes of serum trace elements and nutritional proteins in children with acute lymphoblastic leukemia and acute myeloid leukemia at the stage of remission. Methods Erythrocyte count, hemoglobin, serum levels of total protein, albumin, iron, ferritin, transferrin, lactate dehydrogenase, ceruloplasmin, cuprum, zinc and their ratio were measured in 43 patients with acute lymphoblastic leukemia,19patients with acute myeloid leukemia at stages of remission(remission groups),and 30 healthy controls(control group)enrolled from Shanghai Children's Medical Center using atomic absorption spectrometry, nophelometry assay, dry chemical method, and chemiluminescence method. The differences of these indicators between remission groups and control group were analyzed. Results Serum levels of total protein(P=0.454),iron(P=0.769),transferrin(P=0.903),and zinc(P=0.343)were not significantly different between the remission groups and the control group.Serum levels of ferritin(P=0.000),lactate dehydrogenase(P=0.000),ceruloplasmin(P=0.000),cuprum(P=0.002),and Cu/Zn ratio(P=0.003)in the remission groups were significantly higher than those in control group. On the contrary, erythrocyte count(P=0.000),hemoglobin(P=0.000)and albumin(P=0.046)were significantly lowerin remission groups than those of control group.Serum levels of all detected indicators were not significantly different between the acute lymphoblastic leukemia remission group and acute myeloid leukemia remission group(P>0.05)except for lactate dehydrogenase(P=0.025).Conclusion At the remission stage of acute lymphoblastic leukemia and acute myeloid leukemia, serum levels of some trace elements and nutritional proteins gradually returned to normal, and the original balance is established again.
