1.Effects of sevoflurane on systemic inflammatory response and cardiopulmonary function in septic shock rats

Objective To investigate the effects of sevoflurane on the systemic inflammatory response and cardiopulmonary function in septic shock rats. Methods Thirty-two SD rats, 8-10 months old, weighing 250-300 g, were randomly divided into 4 groups (n = 8 each): sham operation group (group S), cecal ligation and puncture (CLP) induced septic shock group (group CLP) , sevoflurane I group (group SEV, ) and sevoflurane II group (group SEV,). The abdomen was opened but CLP was not performed in group S. The septic shock was induced by CLP as described by Baker et al. Group SEV, and SEV, inhaled 2.4% sevoflurane for 30 min at 1 h and 3 h after the successful establishment of the model respectively. At 1, 3 and 5 h after septic shock, MAP and HR were recorded and arterial blood samples were taken for blood gas analysis and determination of plasma concentrations of TNF-α, IL-1, MDA and NO. The left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD), left ventricular fractional shortening (LVFS) and cardiac output (CO) were also detected 5 h after septic shock. The animals were killed after the detection of cardiac function. The lungs were removed for determination of W/D lung weight ratio and Evans blue (EB) content. The tissues from the heart, lung, liver and kidney were taken for detection of NF-kB activity by electrophoretic mobility shift assay (EMSA) ResultsMAP was significantly lower, HR higher, LVEDD, LVESD, LVFS, CO, pH value, PaO2 and PaCO2 lower, and W/D lung weight ratio, EB content, plasma concentrations of TNF-α, IL-1, MDA and NO, and NF-kB activity in the heart, lung, liver and kidney tissues higher in group CLP, SEV, and SEV2 than in group S (P < 0.05). NF-kB activity in the heart, lung, liver and kidney tissues and plasma concentrations of TNF-α, IL-1, MDA and NO were significantly lower in group SEV, than in group CLP and SEV2 ( P < 0.05 ), but no significant differences were found in the other indices between group SEV, and CLP and between group SEV1 and SEV2 ( P > 0.05). Conclusion Inhalation of 2.4% sevoflurane for 30 min 1 h after septic shock can inhibit the systemic inflammatory response slightly, but can not improve the cardiopulmonary function in rats with CLP-induced septic shock.
2.Inhibitory Effect of Hydroxycamptothecin on C6 Gliomas Cells

OBJECTIVE: To investigate the inhibitory effect of hydroxycamptothecin (HCPT) on C6 gliomas cells in rats. METHODS: The cytotoxicity of HCPT at different concentration(1~0.031 25 mg?mL-1) on C6 in vitro was measured by MTT with conventional therapy for C6 gliomas cells as control. Meanwhile,the antitumor effect of O-HCPT(open ring form) and that of C-HCPT(closed ring form) were compared. RESULTS: At a high concentration,HCPT showed an inhibition ratio of as high as(69.43?4.5)% on C6 gliomas cells in concentration-dependent manner,showing no significant differences as compared with control. The antitumor effect of O-HCPT was lower than that of C-HCPT,and their IC50 were 3.626 mg?mL-1 and 0.135 mg?mL-1,respectively. CONCLUSION: HCPT had remarkable inhibitory effect on C6,and the antitumor effect of C-HCPT is potent than that of O-HCPT.
3.Preparation Technology and Optimization of Formulation of Hydroxycamptothecin Sustained-released Tablets

OBJECTIVE:To prepare hydroxycamptothecin sustained-release tablets and to optimize its preparation technology and formulation.METHODS:The preparation technology and formulation were optimized by orthogonal experiment taking the type of excipient polylactic acid (A),tabletting pressure (B),ratio of HCPT to PLA (C) as factors with the deviation degree of the accumulative release percentage as evaluation index.Then the optimized results were verified.RESULTS:The optimal technology and formulation were as follows:A was PLA (20 000),B was 250 N,C was 1∶5.The deviation degree of the formulation was verified to be 2.963.CONCLUSION:The hydroxycamptothecin sustained-release tablets prepared in optimized preparation technology and formulation are up to the standard on preparation.

5.Notch signaling pathway,articular cartilage development and osteoarthritis

BACKGROUND:Osteoarthritis,often accompanied by cartilage abnormal signal transduction,suggesting that intracellular signal transduction may play an important role in osteoarthritis genesis and development.OBJECTIVE:To review recent advances in the research of Notch signaling pathway,and explore the mechanisms of chondrogenic differentiation of bone marrow mesenchymal stem cells and its role in cartilage development and chondrogenesis.To analysis the role of dysregulated Notch signaling in the development of osteoarthritis.METHODS:Pubmed was undertaken to identify the relevant articles published from March 1919 to February 2009 with the key words of"Notch signaling pathway,osteoarthritis,chondrocytes,chondrogenesis,bone marrow stem cells"in English.Simultaneously,CNKI Database was searched for relevant articles published from February 2004 to September 2008 with the key words of"Notch signaling pathway,osteoarthritis,chondrocytes,chondrogenesis,bone marrow stem cells"in Chinese Articles with the Notch signaling pathway-related research,Notch signaling pathway of bone marrow-derived mesenchymal stem cells,differentiation of cartilage relative to the mechanism studies,and the Notch signaling pathway disorders and osteoarthritis research were included,Reproducibility articles were excluded Development regulation of articular cartilage and cartilage cell proliferation were considered as evaluation indexes.RESULTS AND CONCLUSION:Of 632 articles retrieved,30 were included in our study according to the inclusive and exclusive criteria.The Notch pathway is a highly consewed signaling mechanism involved in many processes determining cell fate during development and maintenance of homeostasis of mature tissues in the adult organism.Activation of Notch occurs following Notch receptor/ligand engagement upon cell-cell contact,which initiates translocation of the Notch intracellular domain to the nucleus and activation of target genes.It has been demonstrated that the Notch signaling plays an important role in regulating articular cartilage development and chondrocyte proliferation and differentiation In recent studies abnormality of notch signaling was also found in disease,which suggests that notch signaling is involved in osteoarthritis initiation and development.Further study of the specific mechanism of notch signaling in the initiation and development of osteoarthritis will enable us to develop targeted approaches in the treatment of osteoarthritis,thus bring more strategies in the future treatment of osteoarthritis.
6.Effect of hydroxycamptothecin with thermotherapy on angiogenesis in vivo and in vitro

Purpose:To study the effects of hydroxycamptothecin with thermotherapy on anti-angiogenesis in vitro and in vivo. Methods:We chose human microvascular endothelial cell (HMVEC) culture and chick embryo choriallantoic membrane (CAM) model and used MTT and number-calculating methods to observe hydroxycamptothecine on HMVEC’s proliferation , sprouts and CAM blood vessels’ formation.Results:The survival rate of endothelial cells was in the range of (68.2%)-44.7% within the dose of 20-40 ng/ml and 5-80 ng/ml and was negatively correlated with the concentration (correlation coefficient was -0.906,-0.469,P=0.00003,0.0051). Hydroxycamptothecine could significantly suppress the endothelial cells’ proliferation and the sprouts. Hydroxycamptothecine could significantly suppress CAM vessels. The survival rate of HepG II cells is in the range 100%-90% within the dose of 5-80 ng/ml. There was no cytotoxicity.There was a synergestic anti-angiogenetic effect when hydroxycamptothecin (20 ng/ml) was combined with thermotherapy in vitro while there was additive effect when hydroxycamptothecin (40 ng/ml) was combined with thermotherapy in vitro.Conclusions:This experiment shows that small doses of hydroxycamptothecine (20-40 ng/ml) with thermotherapy has anti-angiogenetic synergestic or additive effect on proliferation and migration both in vivo and in vitro.

Objective:To compare the difference of microalbuminuria (MAU) and von Willebrand factor (vWF) between essential hypertensives and hypertensive patients with metabolic syndrome (MS), and to explore the association of these factors with MS. Methods: According to the new definition of MS by International Diabetes Federation (IDF) for 2005, 133 consecutive essential hypertensives were divi-ded into two groups: hypertensive patients with metabolic syndrome (EH-MS group, n=97), and essential hypertensives (EH group, n=36). Biochemistry assay, plasma vWF, 24 hours urine albumin excretion (UAE) and morning urine albumin/creatinine ratio (ACR) were measured. The association of MAU and vWF with MS was analysed. Results: Waist circumference, body mass index, triglyceride (TG), low density lipoprotein cholesterol (LDL-C), plasma vWF and UAE were increased(P0.05, respectively). OBJECTIVETo reveal the quality variation of polysaccharides, triterpenoids and proteins in spores and fruiting bodies of Ganoderma lucidum from producing areas, different varieties, harvesting parts and periods, and wall-breaking treatments.
METHODSpores and fruiting bodies from varieties of Longzhi No. 1 and Hunong No. 1 were collected as test samples, together with wall-broken spores sold in domestic main producing areas. The anthrone-sulfuric acid colorimetric method was used to determine the content of total polysaccharides. The vanillin-glacial acetic acid-perchloric acid colorimetric method was used to determine the content of total triterpenoids. The Lowry method was used to determine the content of total proteins.
RESULTThe content ranges of total polysaccharides, total triterpenoids, and total proteins from 6 domestic main producing areas were 0.40% - 2.25%, 1.36%-3.15% and 0.74% -1.91% respectively. The content ranges of total polysaccharides, triterpenoids, and proteins in the fruiting bodies from 2 varieties cultured in Zhejiang were 0.25% -1.42%, 0.44% -1.42% and 1.82% -3.67% respectively. In addition, the ranges of samples from wall-unbroken spores were 0.41% - 0.91%, 0.09% - 0.12%, 0.78% - 0.90% respectively and wall-broken spores are 1.03% - 2.25%, 1.89% - 3.15%, 0.96% - 1.04% respectively.
CONCLUSIONThere are significant differences in the contents of main chemical ingredients of wall-broken G. lucidum spores saled in the markets. The samples from Zhejiang contain high content of total polysaccharides and triterpenoids, and samples from Fujian contains more proteins. Between the 2 major varieties cultured in Zhejiang, Longzhi No. 1 contains higher content of triterpenoids, but Hunong No. 1 has more polysaccharides. Contents of triterpenoids and polysaccharides from wall-broken spores are much higher than those of fruiting bodies. The stipes from fruiting bodies contains more polysaccharides than those of the pileus, while the triterpenoids contents are higher in the pileus than stipes. The pileus and stipes collected in the second year contain higher content of polysaccharides than the first year's samples, but the contents of triterpenoids are lower. Wall-breaking treatment would significantly improve the extraction and dissolution rate of total triterpenoids and polysaccharides.
