1.HISTOLOGICAL STUDY ON THE EFFECT OF A HIGH DOSAGE OF LH-RH-A IN ENDOME-TRIUM ON PSEUDOPREGNANT RATS

The decidua of pseudopregnant rats and of those effected by high dosage of luteinizing hormone-releasing hormone analogue (LH-RH-A) were studied histologically.After inducement of pseudopregnancy, 29 Wistar virgin rats were divided into experimental and control groups. The experimental group was injected with 200?g. of LH-RH-A subcutaneously daily for four days, while to the control group, physiological saline was injected instead. Both experimental and control animals were killed on the 9th, 11th and 13th day of pseudopregnancy.The uteri were fixed with appropriate fixatives and paraffin sections were processed with HE, phloxine-tartrazine, Foots and aldehyde fuchsin staihing methods.On the 9th day decidua of the control rats are well developed with many endometrial granulated cells, while in the decidua of the experimental group, the decidual cells become pyknotic, and the granulated cells are barely seen. The results suggest that a high dosage of LH-RH-A advances the necrosis of decidua of the pseudopregnant rats.
2.Study on the mechanism of intraventicular administration of insulin improve the cardiopulmonary resuscitation rats’ neurological function

Objective:To explore the effect of intraventricular administration of insulin on pro-apoptotic expression of Caspase-3 mRNA and neuronal apoptosis in hippocampus CA1, and neurological function after rats’ CPR. Methods:Thirty male SD rats were randomly divided into three groups:sham group;the resuscitation with saline-treated group and resuscitation with insulin-treated group. Six minute cardiac arrest was induced by ventricular fibrillation (VF) via pacing electrode placed in rats’ esophagus in saline and insulin group. Results:1. The NDS revealed a clear neurological deficit after reperfusion 24 h, 72 h in insulin and saline group as compare to the sham group, Comparing to the saline group, Insulin could improve the rats’ neurologic function after CPR 24 (insulin vs. saline group, 70(64~72)vs.56(50~58), P<0.001);2. Through TUNEL stain, insulin inhibited apoptosis in CA1 hippocampus as compare to the saline after CPR 7d. (F=5.853, P=0.02) 3.Caspase-3 expression in insulin-treated groups were significantly decreased compared to the saline-treated group after reperfusion 24h and 72h[(70(64~72), 56(50~58);P<0.001]. Conclusion:Intraventricular administration of insulin could inhibit the mRNA expression levels of the apoptotic activities of Caspase-3 after cardiac arrest, prevent neuronal apoptosis in the CA1 hippocampus and improve the rats’ neurological function (at least 24hours) after CPR.
3.Protective Effect of Resveratrol on Oxidative Injury in Astrocytes

Objective To investigate the protective effect of resveratrol ( RES) against hydrogen peroxide ( H2 O2 )-induced oxidative injury to astrocytes and the related mechanism. Methods Subcultured astrocytes were randomly divided into four groups:negative control group ( treated with normal culture medium) , model control group ( treated with 100 μmol·L-1 H2 O2 for 12 h), resveratrol low dose group (treated with 20 μmol·L-1 RES for 24 h H2O2 for 12 h) and resveratrol high dose group ( treated with 40 μmol·L-1 RES for 24 h before H2 O2 for 12 h) . Cell viability was detected by MTT assay, apoptosis rate was detected by flow cytometry, apoptotic cell morphology was detected by hochest33258 staining, and the expression of apoptosis-related factors such as caspses-3 and caspase-9 were measured by colorimetric detection. Results MTT assay showed that after treatment with 5, 10, 20, and 40 μmol·L-1 RES for 24 h, cell viability was (100. 46±3. 17)%, (101. 33± 3.14)%, (101. 33±1. 30)%, and (99. 67±2. 62)%, respectively, and the difference was not statistically significant as compared with the negative control group [(98. 33±2. 13)%, P>0. 05]. RES showed no effect on astrocyte activity, after treatment with 20 and 40 μmol·L-1 RES, astrocyte activity was significantly elevated to (54. 67±4. 11)% and (70.33± 2. 61)% as compared with model control group (t=3. 59, 7. 13, P<0. 05), RES inhibited hydrogen peroxide-induced decrease in cell viability. Flow cytometry results showed that after treatment with 20, 40 μmol·L-1 RES, the apoptosis rate of astrocytes significantly decreased to (35.51±3. 56)% and (14. 12%±3. 19)% (t=4. 26, 6. 33, P<0. 01) as compared with model control group (46. 31±4. 16)%. Hochest 33258 staining showed that RES inhibited hydrogen peroxide-induced cell apoptosis, besides, the RES treatment also could reduce H2 O2-induced expression of caspses-3 and caspase-9 in astrocytesin a time-dependent manner. Conclusion RES can inhibit hydrogen peroxide-induced astrocytes apoptosis through inhibiting the expression of caspses-3 and caspase-9, which can provide experimental evidence for its treatment of central nervous disorders.
4.HISTOCHEMICAL STUDIES OF THE EFFECT OF HIGH DOSAGE LUTEINIZING HORMONE RELEASING HORMONE ANALOGUE(LH-RH-A)ON THE ENDOMETRIUM IN PSEUDOPREGNANT RATS

29 virgin female Wistar rats were divided into experimental and control groupson the 5th day of pseudopregnancy.The experimental animals were injected with200 ?g.of LH-RH-A subcutaneously daily in 5th-8th day,while in the controlgroup physiological saline was used instead.Both experimental and control animalswere killed on 9th,11th,13th days of pseudopregnancy.Histochemical techniques were applied to cryostat sections to show the activityof the following enzymes:SDH,LDH,NE,~5△A-3?-HSD,G6Pase,TPPase,AlPand AcP;PAS,Oil red O,and gallocyanin-chromalum staining methods were usedto show carbohydrates,fats and RNA respectively.The above enzymatic reactions of the decidual cells of the experimental animalswere negative,while those of the stromal cells were positive on 9th day.Incomparison,the results of the decidua of the control group on 13th day are verysimilar to the decidua of the experimental group on the 9th day.The resultssuggest that a high dosage of LH-RH-A only advances degeneration of thedecidua.
5.Long-term effects on health-related quality of life after short-segment pedicle fixation of thoracolumbar fractures without neurologic injury.

OBJECTIVETo evaluate the effects on health-related quality of life after short-segment pedicle fixation of thoracolumbar fractures without neurologic injury.
METHODSA total of 55 patients suffered from thoracolumbar fractures without neurological deficit from January 2007 to October 2010 were included in this retrospective study. Of the 55 patients, 35 were male and 20 were female. The mean age was (44.0±10.5) years old (ranged, 7 to 75). Preoperatively, the main clinical symptoms in patients were thoracolumbar pain without neurologic injury and the imaging findings mainly showed that the injured vertebra became wedging or flat with the loss of height. Three questionnaires (ODI, EQ-5D and SF-36) were applied to follow-up in this study.
RESULTSAll operations were successful and incisions got primary healing. The function of thoracolumbar spine fully or basiclly restored and postoperative imagings showed fractures healed without loose screws, broken screws or broken rods. The completion rates of all the queasionnaires were 100% while the mean follow-up period was 4.2 years (ranged, 3 to 6). The ODI scores were 25.8±18.7, which were statistically higher than those of the general population in our country (P<0.05). The EQ-5D index scores were 0.70, which were statistically lower than those of the normal Chinese population (P<0.05). Compared with the normal scores in our country, the scores of 8 items in the SF-36 questionnaire were obviously lower with significant difference (P<0.05).
CONCLUSIONCompared with the normal Chinese population, it should be paid more attention to that the health-related quality of life in patients after short-segment pedicle fixation of thoracolumbar fractures without neurologic injury because of lower scores.

6.Experimental study of the pipe replacement cycle for the suspension of ventilator

Objective To compare the detection results by bacteria pathogenic rate of ventilatorassociated pneumonia (VAP) pipeline and ventilator patients with mechanical ventilation,to investigate whether need to replace the ventilator weaning patients underwent another test of pipeline during mechanical ventilation.Methods This study selected using the ventilation mechanical ventilation for more than 48 hours,try (48 ± 4) hours required to mechanical ventilation in 69 patients.They were divided into two groups according to admission time,group A was in need of replacement ventilation pipeline of new again during mechanical ventilation,group B used the original pipeline.In offline,try again during mechanical ventilation after mechanical ventilation,once daily for 4 days to collect ventilator pipe air outlet samples for bacterial culture counts were observed,while offline,use again after 2 hours,12 hours of ventilator,24 hours,2 days,3 days,4 days of body temperature,white blood cell count,neutrophil classification and other indicators.The incidence of VAP was compared between two groups.Results The temperature,white blood cell count,neutrophil classification were (38.5 ± 1.6)℃,(11.8 ± 3.5)×109/L,0.85±0.16 in group A,while (38.4 ± 2.0) ℃,(12.1 ± 3.7)×109/L,0.84 ± 0.20 in group B,and there were no significant differences between two groups,P>0.05.The incidence of VAP was 37.1%(13/35) and 35.3%(12/34) in group A and group B,and there was no significant difference,P> 0.05.Conclusion There is no necessary for trial offline patients without respiratory infection to change the ventilator pipe when to re-ventilate during the 7 days using period of the ventilator pile.
7.Clinical Study on Postpartum Depression and its Related Factors

Objective The prevalence rate, related factors and clinical characteristics of postpartum depression were studied in order to promote obstetrician′s understanding of the disease, and resort to preventive and therapeutic measure earlier. Methods Two hundred and ten puerpera were selected from September to November 1998 for mood study by self rating depression scale, self rating anxiety scale, Hamilton depression scale, and self made questionnaires. Results The prevalence rate of postpartum depression was 37 14% (78/210), of which, 91 03% (71/78) was mild, and 8 97% (7/78) moderate. In addition, 10 48% (22/210) of women got anxiety after delivery. The symptoms, including decreased activity, difficulty in thinking, problems in decision making, difficulty in doing things, feeling of uselessness and hopelessness were chief complaints of postpartum depression patients. Bad social behavior and mood controlling, inadequate psychological preparation for delivery, lack of knowledge of delivery were closely correlated to the onset of postpartum depression, which are the risk factors of postpartum depression. Conclusions Postpartum depression is a spiritual and psychological obstacle with relatively high prevalence rate. Bad social psychological factors can bring out and aggravate postpartum depression. It is important to carry out prenatal psychological care and to propagate the knowledge of delivery, to find out and cure of postpartum depression as soon as possible.
8.Observation of the Effect of PGE1 in Treatment of 34 Cases of Heart Failure Caused by Cor Pulmonale

Objective:To observe the effect of PGE1 in treatment of heart failure caused by cor pulmonale.Method:100-200 μg of PGE1 was added to 250 mL of 10% glucose and administered by intravenous drip to patients once per day for 7-10 days.Results:Of the 34 cases,21 cases were significantly improved,10 cases improved while only 3 cases remained the same as before. The total effective rate was 91%. The mean time of improvement was 7 days.Conclusion:PGE1 is effective in treating cor pulmonale heart failure,it can help decrease the danger of digitalis toxication.
9.The role of type 3 muscarinic acetylcholine receptor polypeptide in the pathogenesis of Sj(o)gren's syndrome

Objective To immunize BALB/c mice with type 3 musearinic acetylcholine receptor polypeptide (M3RP) and to evaluate the role of M3RP in SS. Methods Four-week-old BALB/e mice were immunized with M3R polypeptide 213-228 (M3RP) on days 0, 14, 35, 56, and were re-immuniged on days 65, 84, 105, and one mouse was killed every 2 to 3 weeks. The mice of the control group were immunized with submaxillary gland homogenate, GST, and PBS. The animals were analyzed for the presence of anti-SSA,anti-SSB, RF, ANA, anti-a-fodrin and anti-M3RP in sera by immunofluorescenee or ELISA. The cytokines of IFN-γ IL-2 and IL-10 were measured with ELISA. Salivary glands were examined by H&E staining and immunohistochemical analysis. Volume of water drinking by each groups were calculated. Results BALB/e mice immunized with M3RP and submaxillary gland homogenate developed an immune response directed against M3RP, α-fodrin and ANA, but no antibodies against SSA, SSB and RF were found. Furthermore, lym-phocytic infiltrates in the salivary glands of immunized animals were observed 50 days after first immunization of M3RP and submaxillary gland homogenate. The serum IFN-α in mice of M3RP, submaxillary gland ho-mogenate, GST and PBS was (62±6), (89±5), (30±5) and (19±6) pg/ml respectively, and IL-2 was (12.6±1.6), (19.8±0.4), (3.9±0.9), and (4.9±1.1) pg/ml respectively (P<0.05). No difference was found in the level of serum IL-10 among the four group. Expression of α-fodrin was found only in submaxillary gland in M3RP and submaxillary gland homogenate groups of mice, but not in PBS and GST controls when studied by immunohistochemical analysis. Conclusion These results suggest that BALB/c mice immunized with M3RP are reminiscent of human SS, and M3RP as an autoantigen participates the development of SS.
10.Antimutagenicity of 14 Compounds in Salmonella typhimurium TA100

The antimutagenicity of 14 compounds, cysteine (1), cinnamic acid (2), rutin (3), tannic acid (4), germanium dioxide (5),fluoro uracil (6), sodium copper chlorophylline (7),?-sitosterol (8), vitamin C (9), coumarin (10), vitamin E (11), L-glutathine (oxidized form) (12), L-glutathine (reduced form) (13) and organic germanium (14), were investigated using Salmonella typhimurium/microsome assay in TA100.Three modes of action, i.e. inhibitory, antimutagenic and desmutagenic effects, were observed-for their antimutagenicity. Results showed that all of test compounds inhibited mutagenicity induced by MNNG and B(a)P with the exception of germanium dioxide. Percent inhibition of compounds 1, 2, 3, 4, 6, 8, 10 and 12 were greater than 90%. 14 compounds exhibited antimutagenic effects on the mutagenic activity of MNNG, and 12 compounds exhibited antimutagenic effects on B(a)P. Germanium dioxide and organic germanium had no such effect. 14 compounds all exerted desmutagenic effects on B (a)P directly before B(a)P acted on cells. According to the potential and modes of action, cysteine, cinnamic acid, rutin, tannic acid and coumarin were better among 14 compounds. The doseresponse relationships of inhibitory and antimutagenic effects on mutation induced by MNNG and B(a)P were found. Each compound has its sufficient range of dosage. These studies suggest that it is necessary to select effective antimutagens to make mixtures, and their synergistic effects should be investigated.
