1.Development and Design of Diffusion Optical Tomography System

Objective To design and establish a set of diffusion optical tomography(DOT) system.Methods Near-infrared(NIR) laser was employed as the light source and only one photomultiplier tube(PMT) as the detector.An optical multiplexer was used to alter the detector channels.The 32 channels of the system,which are consisted of 16 launch channels and 16 detector channels,worked under the continuous-wave(CW) model and were used to acquire 256 boundary data.Results Experiments were performed based on the proposed imaging system.The intralipid was used as the tissue-like medium and the India ink as the absorber.Two sets of data on the boundary were sampled,respectively,before and after the absorber was embedded inside the tissue-like medium.The two sets of data were normalized and then used to reconstruct the absorption coefficient distribution.The recovered image reflected the real location and size of the absorber.Conclusion The proposed imaging platform can image the tissue optical parameters effectively.However the resolution of the reconstructed image was not high because the inverse problem was gravely underdetermined and the noise was not considered in the reconstruction algorithm.The recovered result in the next generation system could be improved by making more use of prior information and enhancing the performance of the system.
2.Left Ventricular Hypertrophy to Heart Failure in Gradually Pressure Afterload Heart of Wistar Rat

Objective:Our purpose was to establish an ideal chronic pressure-afterload heart failure rat model which has the transition from cardiac hypertrophy to heart failure. Methods: Chronic pressure-afterload heart failure rat model was induced by gradually constricting the ascending aorta of young rats. Young rats were randomly divided into 2 groups: the constricted and sham-operated groups. Clinical manifestation, tail-cuff blood pressure, organ weight, and hemodynamic data were observed at various time after operation. Results: The overall survival rate was 87%. Tail-cuff pressure began to increase in 4 weeks after operation. Left ventricular hypertrophy appeared in 12 weeks and heart failure in 5 months. Conclusion:It's a practical and reproducible model of cardiac hypertrophy that progresses to chronic heart failure.
3.Factors Affecting Measurements of Lung Tumor Standardized Uptake Value:A Monte-Carlo Simulation Study

Objective To evaluate the factors affecting the measurement of lung tumor activity concentration (AC) with FDG-PET. Methods High count Monte-Carlo simulations,based on VIP-Man (marked VHP atlas) added with spherical lung tumors of different diameter and activity,were performed to generate the essentially noiseless projection data for PET. The factors related with the measurement of tumor AC were evaluated. Results Using the maximum value in the tumor region of interest (ROI) generally resulted in the over-estimation of AC,and the degree of over-estimation would also increase as the noise became severer. For the small tumors with low AC,the proper partial volume correction was needed. Drawing ROI after post-reconstruction filtering could improve the reproducibility of AC measurement compared with no filtering first. Conclusion The accuracy and reliability of AC measurement depends strongly on factors including tumor size,post-reconstruction filter,noise level,ROI definition,PVE correction,etc.
4.Anti-lung cancer effect and anti-angiogenesis therapy study of perillyl alcohol

Objective:To investigate the inhibitory effect of perillyl alcohol (PA) on the proliferation and invasion of tung cancer cell A549,and the influence of PA on tumor angiogenesis was studied.Methods:Different concentrations of PA and erlotinib were added into lung cancer cell A549,the inhibiting effect of drug group on lung cancer cell A549 was found by MTT assay.The inhibiting effect of PA on lung cancer cell A549 invasion was measured by Transwell assay.ROS changes of PA on lung cancer cell A549 was detected by fluorescent.Influence of PA on Caspase-3 activity of lung cancer cell A549 was measured by spectrophotometry,VEGF,HIF-1 α,COX-2 expression in lung cancer cell A549 was measured by Western blot,and the NF-κB activity of lung cancer cell A549 was measured by EMSA.Results:Compared with blank control group,cell growth inhibition rate of PA and erlotinib on lung cancer cell A549 was increasing with the increased concentrations (10,50,100 μ,g/ml),the difference was statistically significant (P< 0.05),the invasion ability of lung cancer cell A549 was decreased continuously,the difference was statistically significant (P<0.05).The ROS level of lung cancer cell A549 had no obvious change with the increasing density of erlotinib,but obviously increased with the increasing concentrations of PA (10,50,100 μg/ml).With the increasing concentrations of PA,the expression of COX-2,VEGF and HIF-1α were continuously decreased.EMSA assay showed that NF-κB was continuously decreased with the increasing concentrations of PA.Conclusion:The antitumor mechanism of PA on lung cancer cell A549 might be related to increase the expression level of ROS and reduce the expression of activity of NF-κB,COX-2,VEGF and HIF-1α with angiogenesis signaling pathway.
5.Mechanism of miR-34a on invasion and migration ability of human lung carcinoma by Snail induced EMT

Objective:To investigate the exression of miR-34a on lung cancer and normal lung tissues,and the effect and mechanism of miR-34a in lung cancer cell invasion and migration.Methods: qPCR was used to detect the expression of miR-34a on lung cancer.miR-34a-mimic and miR-34a-inhibitor were used to overexpress and knockdown miR-34a.qPCR was used to detect the effectiveness.Western blot was used to detect the expression of Snail after induced with miR-34a-mimic and miR-34a-inhibitor.Luciferase reporter gene was used to detect interaction between miR-34a and Snail.Transwell invasion assay was used to detect invasion ability after induced with miR-34a-mimic and miR-34a-inhibitor.Scratch assay was used to detect migration ability after induced with miR-34a-mimic and miR-34a-inhibitor.The expression of E-cadherin,Vimentin and Twist were detected by Western blot.Results: miR-34a expression was significantly reduced in lung cancer.With the stage of lung cancer progression,the expression of miR-34a reduced.With the differentiation of lung cancer progression,the expression of miR-34a decreased.Decreasing of miR-34a was associated with lung cancer lymph node metastasis.miR-34a-mimic and miR-34a-inhibitor could overexpress and knockdown miR-34a.miR-34a could regulate expression of Snail.Snail was the direct target of miR-34a;miR-34a could regulate the invasion ability of human lung carcinoma H1650 cells;miR-34a could regulate the migration of human lung carcinoma H1650 cells;miR-34a could regulate the expression of E-cadherin,Vimentin and Twist.Conclusion: miR-34a plays the role of tumor suppressor factor in lung cancer.miR-34a can regulate the invasion and migration ability of lung carcinoma H1650 cells by Snail induced EMT.

8.Comparisons between NIR and HPLC fingerprint on quality stability of Shudihuang decoction pieces.

OBJECTIVETo establish a new method for evaluating quality stability of Shudihuang decoction pieces with near-infrared spectrum.
METHODClustering analysis was used to distinguish different Shudihuang samples. And comparisons between NIR and HPLC fringerprint.
RESULTResults of clustering reflected the similar degree among these samples. Compared with HPLC fingerprint, NIR analytical process fast and convenient with exact results.
CONCLUSIONNIR is a feasible and effective approach to examine the quality stability of Shudihuang decoction pieces with unique virtues.

9.Plasmid and stable cell linereconstruction of RET point mutation in medullary thyroid carcinoma pedigree

OBJECTIVEThe plasmid construction and validation of RET point mutation in vitro.METHODS The RET gene exon in the mainland familial medullary thyroid carcinoma family was analized with muon capture two generation sequencing of. In vitro, the RET relative point mutationswere reconstructed in NIH3T3 cells and the expression levels were studied.RESULTSThe corresponding lentiviral plasmids of RET point mutations were successfully obtained after point mutating the wild RET plasmids, it was verified that the target genes were expressed in NIH3T3 cells stably by Western Blot. CONCLUSIONSStable cell lines carrying RET point mutations were reconstructedsuccessfully, which provide a good platform for studying various point mutations.
10.Risk factors for failed internal fixation in surgery of senile femoral intertrochanteric fractures

Objective To analyze the risk factors for failures of intramedullary and extramedullary internal fixation in surgery of femoral intertrochanteric fractures in elderly patients.Methods A retrospective study was conducted of the 205 elderly patients with osteoporotic femoral intertrochanteric fracture who had accepted closed reduction together with intramedullary and extramedullary internal fixation between September 2005 and August 2014.They were 89 men and 116 women,from 65 to 98 year of age (average,78.8 years).By AO classification,137 cases were of types AI.1-A2.1 (stable fractures),and 68 of type A2.2-A3.3(unstable fractures).The incidence of internal fixation failure and Harris scores at the last follow-up were recorded.The factors possibly contributing to the failure were analyzed using the univariate analysis and multivariate logistic regression analysis.Results Of the patients,192 obtained a mean follow-up of 39 months (from 14 to 60 months),but 13 were lost after a 10-month follow-up.Of the 205 patients,internal fixation failure occurred in 12 (incidence of 5.9%).Five failed cases received DHS fixation and 7 accepted PENA fixation.At the last follow-up when the 13 cases were lost after 10-month follow-up,the mean Harris hip score was 81.6 (from 57 to 92),and the excellent to good rate was 84.9% (29 excellent cases,145 good ones,10 fair ones and 21 poor ones).The multivariate regression analysis revealed that tip-apex distance (TAD) > 25 mm(OR = 333.33),severe osteoporosis (OR =267.44),AO types A2.2-A3.3 (OR = 22.24),functional reduction of fracture (OR =20.79),and concomitant medical diseases (OR =4.59) were independent risk factors for failures of internal fixation.Conclusions DHS and PFNA fixations are effective treatments for elderly patients with femoral intertrochanteric fractures.TAD> 25 mm,severe osteoporosis,unstable factures,functional reduction of fracture,and concomitant medical diseases may lead to internal fixation failure in surgery of femoral intertrochanteric fractures in elderly patients.
