2.CD4+CD25+CD127low/- regulatory T cells and related cytokines in patients with psoriasis vulgaris

Objective To explore the role of CD4+CD25+CD127low/-regulatory T cells (Tregs) in the pathogenesis of psoriasis vulgaris(PV). Methods Flow cytometry analysis was used to detect the amount of Tregs in peripheral blood and ELISA to test the levels of IL-10 and TGF-β1 in blood serum; the suppressive function of Tregs on autologous CD4+CD25-T cells was determined by MTT method. Results No significant difference was found in the proportion of Tregs in PV patients and healthy controls(P>0.05). There was a diminished suppression of Tregs from patients on autologous CD4+CD25- responder T cell proliferation in PV patients when compared with that in controls (P < 0.01). The serum level of IL-10 in patients was lower than that in controls (P < 0.01) while that of TGF-β1 in PV patients was significantly higher than that in controls(P < 0.01). Conclusion Abnormal function of Tregs and low secretion of IL-10 in PV patients might be related to the pathogenesis of psoriasis.
3.Closure of large full thickness stomach wall defect with a newly designed hemostatic clip

Objective To design a hemostatic clip and evaluate its efficacy and success rate of closure of stomach wall defect after full thickness resection (FTR).Methods A full thickness circular or linear resection (3 to 5 cm) was made on each model's antrum with needle knife and insulated-tip knife.The specimens were divided into 2 groups, using either an interrupted or continuous suturing method.Then the closure condition, suturing time, number of clips required and success rate of closure were compared.Results All 12 defects were successfully closed.The average closing time of interrupted and continuous suturing group were 13.33 ± 1.09 and 10.17 ±2.11 minutes, and the mean number of clips used were 4.67 ± 0.82 and 2.67 ± 0.82.The success rate was 100%.Conclusion This newly designed clip is a fast, reliable and convenient tool for stomach wall defect closure after FTR.
4.Progress of autophagy screening systems.

Accordingly, there is a growing demand to identify, quantify and manipulate the process accurately. Meanwhile, there is great interest in identifying compounds that modulate autophagy because they may have applications in the treatment of a variety of autophagy-related diseases. In this review, we summarize the current status of autophagy screening systems to facilitate identification of autophagy modulators.
5.Chemical constituents in root and stem of Piper laetispicum (Ⅱ)

Objective To study the chemcial constituents in the root and stem of Piper laetispicum. Methods Compounds were separated and purified by silica gel column. Their structures were identified on the basis of spectral methods. Results Six compounds were isolated and identified from the root and stem of P. laetispicum: N-isobutyl-(3, 4-methylendioxyphenyl)-2E, 7E-nonadienamide (Ⅰ), pipercallosidine (Ⅱ), N-isobutyl-7-(3, 4-methylendioxyphenyl)-2E, 4E-heptadienamide (Ⅲ), 4, 5-dihydropiperlonguminine (Ⅳ), 1-cinnamoylpyrrolidine (Ⅴ), piperitol (Ⅵ). Conclusion Compound Ⅰ is a novel one, named as laetispiamide B and compound Ⅱ is isolated from P. laetispicum for the first time.

7.Effect of Radix Isatidis on the expression of moesin mRNA induced by LPS in the tissues of mice.

To investigate the effect of the anti-endotoxic part of Radix Isatidis on the expression of moesin mRNA in murine tissues induced by lipopolysaccharide (LPS), the sample solution of F(022) part from Radix Isatidis was intraperitoneally administered to experimental mice, and the lipopoly-saccharide (LPS) were injected into the tail vein, and then the tissues of liver, kidney and spleen were colleted and cut into slices. The mRNA was detected by moesin mRNA hybridization in situ. The staining results were observed under microscope. It was found that moesin mRNA expression was increased in the tissues of liver, kidndy and spleen in mice treated with LPS, while in the mice pre-treated with F(022) part from Radix Isatidis, the LPS-induced moesin mRNA expressions in these tissues were inhibited in a dose-dependant manner. Our study showed that F(022) part from Radix Isatidis can inhibit the LPS-induced expression of moesin mRNA in the tissues of liver, kidney and spleen in mice.
8.The Effect of Quercetin Targeted p53 Gene and Protein Expression in NB4 Cell Lines

Objective In order to study the value of drug treatment for leukemia, quercetin was delivered to the cultured NB4 cell line. MethodsUsing RT- PCR and western blot, we studied the expression of mutant p53 gene and protein after inducing 24, 48, 72, 96 and 120h by the quercetin with different concentrations (30, 60, 90μmol/L). Results We verified the results that quercetin with concentration of 30 ~90μmol/L could successfully inhibit the expression of mutant p53 protein, but the mutant p53 gene did not have. ConclusionThat data suggested that quercetin would be an effective method for the therapy of leukemia.
