1.Effect of Living in Village on Atopic Sensitization of School-age Children

Objective To explore whether living in rural areas may reduce the risk of atopic sensitization. Methods The standardized questionnaires were answered by the parents of 2 986 school children during 2004 to 2005. Radioallergosorbent technique-fluorescence enzyme immunoassay (RAST-FEIA) was used to determine the level of specific IgE in the serum of the children. The risk factors were analyzed by Logistic regression. Results Compared with the subjects living in the towns from birth, the risk of atopic sensitization was lowest in the subjects living in the villages from birth (OR=0.32, 95%CI: 0.11-0.62) and it was moderate in the subjects who had relocated from village to town (OR=0.51, 95%CI: 0.28-0.92), there was a significant trend among them (P for trend

Objective:To investigate the effects of 17-? estradiol (E2) on the proliferation and metastasis of adenoid cystic carcinoma SAcc83 cells.Methods:The effects of E2 on the proliferation of SAcc83 cells were studied by MTT assay and flow cytometry.The invasion potential of the cells was investigated with chicken embryo heart invasion assay.The in vivo growth and metastasis of SAcc83 cell induced tumor were studied in ovary-resected female nude mice. Results:E2 at 10 -7 ,10 -8 and 10 -9 mol/L promoted the proliferation of SAcc83 cells. E2 at 10 -7 mol/L showed the strongest growth stimulation effects and increased the cell proliferation by 34.4%.With exposure time of 1,2 and 3 d it increased the proliferation index by 33.3%,40.9% and 17.8% respectively;with the exposure time of 12,18 and 24 h,it increased S phase cells in cell cycle by 12.3%,3.3% and 9.7% respectively. Neoplasm developed in all nude mice after subcutaneous transplantation of 106 cells.In the mice treated with 0.2 ml of E2 at 9.55 ?10 -5 mol/L every day for 15 days neoplasm volume doubling time was 61.5 h,that in control mice 76.5 h.In lung metastasis trial, 42 days after tail injection of the cells the metastasis foci in E2 treated mice inreased by 41.2%.Conclusion:E2 may increase the proliferation and metastasis of adenoid cystic carcinoma cells.
3.The Influence of Bacteria Lipopolysaccharide on the Cytokine Production of Flour Mite Allergic Rhinitis Patients

Objective To explore the influence of bacteria lipopolysaccharide on the cytokine production of flour mite allergic rhinitis patients. Methods 8 cases with perennial allergic rhinitis were selected. Then the heparin anticoagulation venous blood was taken from the same patient and was added to fetal calf serum,with ultimate volume 1 ml. The two experimental groups were added 10μg LPS and 1 μg LPS respectively, forming the high LPS dose group and low LPS dose group,while the control group was added nothing. Results The contents of IL-4、IL-5、IL-10、IL-12、IFN-γwere tested by enzyme-linked immunosorbent assay. The findings were that there was significant difference in the productive contents of IL-4、IL-5、IL-10、IL-12 and no significant difference in that of IFN-γ between the high LPS dose group and the low LPS dose group. When compared in two groups,significant difference was found in the productive contents of IL-4 ,IL-10 and IL-12 between the two experimental groups and between the high LPS dose group and the control group, while no obvious difference existed between the low LPS group and control group. However, significant difference was altogether found in the productive content of IL-5 when compared one with another among the three groups,and the result was that low dose of LPS obviously promoted the production of cytokine while high dose of LPS restrained the production of cytokine; no significant difference was found in the production of IFN-γ in all comparisons between the three groups. Conclusion This research, through the experiments in vitro, proved that LPS could effectively adjust the cytokine production of perennial flour mite allergic rhinitis patients and the balance of Th1/Th2 ,whose effect depends on the dose of LPS. This research suggested that the practical future in which special cytokine target treatment will be adopted on perennial flour mite allergic rhinitis patients.
4.Constitution Factors for Metabolic Syndrome: A Case-control Study

Objective To investigate the constitution factors for metabolic syndrome(MS). Methods 271 MS cases and 271 non-MS controls were selected. They were assessed according to the diagnosis standard of constitution of TCM. Results Multiple logistic regression analysis revealed that phlegm-dampness constitution and dampness-heat constitution were dangerous factors for MS, normal constitution was protective factor for MS (P<0.05). Conclusion Research on constitution play a role in investigating pathogenesis of MS.
5.Establishment of Social Stress Induced Depression-like Animal Model in Mice of C57BL/6 Strain and Behavioral Assessments.

OBJECTIVETo establish social stress induced depression-like model in mice of C57BL/6 strain, and to assess its reliability using differenf behavioral methods.
METHODSTotally 20 male mice of C57BL/6 strain were divided into the normal group and the stress model group by random digit table,10 in each group. Another 10 CD1 mice were subjected to social stress. Mice in the normal control group received no stress, while those in the model group received social stress for 10 successive days. Behavioral assessment was performed using social interaction test (SIT), the elevated plus-maze (EPM) test, tail suspension test (TST), respectively. Serum cortisol level was detected by ELISA to assess the reliability of the model.
RESULTSIn the social interaction test when the social target (CDI mice) was inexistent, mice in the normal control group spent longer time in the social interaction zone and less time in the corner zone (P < 0.05); mice in the model group spent less time in the social interaction zone and more time in the corner zone (P < 0.05). Compared with the normal group when CDI mice existed, mice in the model group spent less time in the social interaction zone and more time in the corner zone (P < 0.05). Compared with the normal control group, the total times for entry into open arms, close arms, and the maze were obviously reduced (P < 0.05), and the proportion of entering open arms was significantly reduced (P < 0.05) in the model group. In TST, the motionless time within the last 4 mm was prolonged in the model group (P < 0.05). The serum cortisol level in the model group was obviously elevated (P < 0.01).
CONCLUSIONSocial stress induced depression-like animal model in mice of C57BL/6 straineasquite reliable and possibly suitable to be used in integrative medicine research of combination of disease and syndrome model.

6.Questionnaire Survey for Researchers of Medicine Clinical Trial in Guang’anmen Hospital of China Academy of Chinese Medical Sciences

ObjectiveTo understand the status and requirement of the clinical researchers in Guang’anmen Hospital of China Academy of Chinese Medical Sciences (hereinafter referred to as Guang’anmen Hospital).MethodsSelf-filling questionnaire was used. All medical care personnel in the clinical specialty with the GCP qualification were set as respondents. The three aspects of recognition degree of clinical trials quality, initiative of participation and knowledge demand for GCP were investigated. Data were analyzed through SPSS11.5.Results 95.6% of clinical researchers considered that the course of clinical trial was legal;89.9% of clinical researchers believed that the quality of clinical trial was good. Most clinical researchers participated in the clinical trials actively (86.9%) and they considered that the trials were helpful for them to enhance research ability and to issue paper.Conclusion Clinical researchers are familiar with GCP knowledge and can participate in the clinical trials actively. They still expect to learn more.
7.The myocardial protection of polarizing adenosine-lidocaine cardioplegia in experimental animals

Objective To determine whether polarized arrest using adenosine and lidocaine cold crystalloid cardioplegia would give satisfactory myocardial protection in the cardiac surgery of experimental animals.Methods Twelve pigs were randomized divided into 2 groups (n =6) to receive adenosine-lidocaine cardioplegia or St.Thomas cardioplegia.Left ventricular systolic and diastolic function,cardiac output,cardiac troponin 1 were monitored at before operation (T1),1 hour after crossclamp was removed (T2) and 2 hours after cross-clamp was removed (T3).After T3 the left ventricular ultrastructure was observed,mitochondrial ultrastructure was analyzed by CMIAS image system.Results There were no statistical differences between the two groups in T1.The left ventricular systolic pressure,± dp/dt and cardiac output in adenosine-lidocaine group wasmuch higher than St.Thomas group in T2 and T3,meanwhile the left ventricular diastolic pressure,capillary wedge pressureand cardiac troponin Ⅰ value was lower than St.Thomas group.The hemodynamic values was more stable in adenosine-lidocaine group.St.Thomas group got more myocardial injury alterations in intracellular structure than adenosine-lidocaine group.The image analysis of mitochondrial ultrastructure shows St.Thomas group got lower shape factor,higher area density and perimeter density compared with baseline and adenosine-lidocaine group values.Conclusion Adenosine-lidocaine cold crystalloid car-dioplegia gave satisfactory cardiac arrest,got better myocardial protection than St.Thomas cardioplegia.
8.Probucol promotes reverse cholesterol transport from macrophage to feces in mice in vivo

Aim To investigate the effect of probucol on reverse cholesterol transport from macrophage to feces in vivo,serum lipid profiles of mice were tested before and after probucol administration for 4 weeks,and the 3H-contents in serum,liver and feces in mice were quantitated after 24 hours intraperitoneally injected macrophages which were labeled with 3H cholesterol.Methods 32 C57BL/6 mice were treated with different probucol(0,0.1%,0.5%,1% W/W)in chow diet for 4 weeks,then these mice were injected intraperitoneally with RAW264.7 macrophages(0.5 ml?mice,cells 5.0?106 with 6.2?106 cpm)which were loaded with cholesterol by incubation with acetylated LDL,labeled with 3H-cholesterol.Serum profiles were quantitated by enzymatic method;serum and liver tissues were harvested at 24th hour,feces were collected(0~24 hs),and all were analyzed for tracer counts(all were expressed as the percentage of the total injected counts per minute).Results After 4 weeks 0.1% probucol administration,the levels of total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)decreased markedly by 34.3%,52.8%,(P
9.Characteristics in initial sandtray of children with attention deficit hyperactivity disorder

ObjectiveTo explore the particular inner world of the children with attention deficit hyperactivity disorder (ADHD)with the sandplay therapy and the initial sandtray's clinical diagnosis role and value on the ADHD.MethodsThe differences of the initial sandtrays between 28 children with ADHD and 24 normal children were compared.Results 1.On the use of toys,the experimental group was significantly higher in transports ((5.04 ±6.60),(1.38 ±2.12),t=2.73) and lower in stones((0.00 ±0.00),(8.00 ± 12.33),t=-3.18).2.On the scene of sandtrays,the experimental group was significantly higher in the fight( x2 =12.96,P < 0.01 ),dynamic scenes ( x2 =13.37,P < 0.01 ).3.On the use of self-images,the experimental group was lower than the normal children,and the differences were significant ( x2 =4.75,P < 0.05 ).4.There were no significant differences in the use of sand between two groups( x2 =3.46,P > 0.05).ConclusionThe results show that the children with ADHD have significantly characteristics in sandtrays.Initial sandtray is valuable to be an aid to clinical diagnosis and intervention means.
10.Family environment and behavioral study of different subtypes of attention deficit hyperactivity disorder in children

Objective To investigate the risk factors which indicate behavior problems and degree of conflict function defects in different subtypes of attention deficit hyperactivity disorder( ADHD),and to compare behavioral problems,family environment and conflict function in children with and without ADHD.Methods 87children with ADHD and 38 matched controls performed Simon-Stroop task while questionnaires (Parent Symptom Questionnaire and Family environment Scale)were acquired from their parents.Results Questionnaire indicated that children with ADHD-combined(ADHD-C) had the highest score of conduct problem( 1.05 ± 0.44),learning problem ( 1.97 ± 0.52 ),hyperactivity-impulsion ( 1.56 ± 0.60) and hyperactivity index ( 1.45 ± 0.40 ) than ADHDⅠ(0.65±0.32,1.58 ±0.58,1.06 ±0.46,1.01 ±0.31) and matched controls ( 0.42 ±0.24,0.77 ±0.49,0.58±0.40,0.55 ±0.34) in PSQ.ADHD-Ⅰ and ADHD-C had higher score of contradiction(3.28 ± 1.70,4.61 ±2.56 ) and lower score of family cohesion (6.75 ± 2.27,6.61 ± 2.03 ),emotional expression (5.03 ± 1.54,5.06 ±1.91 ),knowledge (3.83 ± 2.30,3.61 ± 1.81 ) and entertainment (3.70 ± 2.12,3.47 ± 2.32 ) than matched controls (2.36 ± 1.44,8.06 ± 1.71,6.03 ± 1.54,4.86 ± 1.91,4.83 ± 2.29) in FES-CV.Behavior data indicated that the correct rate of children with ADHD-Ⅰ and ADHD-C in Simon-incongruent trials( Siin:0.76 ± 0.13,0.69 ±0.13 ) and Stroop-inconsistent trials( Stin:0.82 ± 0.10,0.78 ± 0.08 )were lower and reaction time of children with ADHD-Ⅰ and ADHD-C in the Siin ( (876.4 ± 97.34) ms,( 893.8 ± 130.1 ) ms) and Stin ( ( 864.4 ± 91.82 ) ms,(860.2 ± 125.0) ms) were longer than matched controls ( (Siin (0.81 ± 0.11 ) ms,(810.4 ± 136.1 ) ms; Stin (0.87 ± 0.08 ) ms,(797.4 ± 136.1 ) ms).Pearson correlation analysis suggested that some factors in the FES-CV had correlation with behavioral problems.Conclusion This finding indicates that poor family environment is the risk factor that indicate behavioral problems in children with ADHD and behavioral problems,conflict function defeet of ADHD-C are more serious than ADHD-Ⅰ and matched controls.
