1.Effects of different antidiabetic therapies on the macular thickness in diabetes patients without fundus complication

Abstract?AIM: To observe the effects of different antidiabetic therapies on macular thickness in diabetes patients without fundus complication.?METHODS: The macular thickness was measured by optical coherence tomography ( OCT) .The retina volume on the center of macula was scanned, and it generated automatically the average thickness data of three rings and nine areas of 1mm, 3mm and 6mm.The data were statistically analyzed.?RESULTS: Healthy control group, oral hypoglycemic drug group and insulin treatment group were enrolled. Each group included 22 cases 22 eyes.The macular retinal thickness of healthy control group is 268.09±17.97μm (the 1st ring), 340.41 ±22.25μm ( the 2nd ring) and 298.14 ± 12.90μm ( the 3 rd ring ) , respectively. The macular thickness of oral hypoglycemic drug group is 260.00 ± 18.17μm (the 1st ring), 335.44±21.12μm (the 2nd ring) and 295.63 ±15.92μm ( the 3rd ring), respectively.The macular thickness of insulin therapy group is 271.01 ± 26.09μm (the 1st ring), 340.86±17.10μm (the 2nd ring) and 298.57 ±12.14μm ( the 3rd ring ), respectively. Comparison of the macular thickness of the 1st ring among 3 groups was insignificant (F=1.21, P=0.31), neither the comparison of the 2nd ring (F=0.35, P=0.71), neither the comparison of the 3rd ring (F=0.22, P=0.81).? CONCLUSION: Compared with healthy individuals, both oral antidiabetic medicines and insulin therapy don't alter the macular thickness of patients with diabetes while without fundus complication.
2.Impact of stress hyperglycemia on prognosis of patients in medical intensive care unit

Objective To evaluate the association between stress hyperglycemia and clinical out-comes among patients with and without diabetes mellitns admitted to medical intensive care unit (MICU).Methods We reviewed the medical ICU records of 1016 consecutive patients admitted to Wu Han Univer-sity Ren Min Hospital. According to admission glucose level, 126 patients with debates and 890 patients without known debates were divided into group 1 ( <6 mmol/L), group 2(6 ~ 8mmol/L), group 3(8 ~10mmol/L), and group 4( > 10mmol/L). The primary end point of the study was in-hospital mortality. Secondary end points included length of hospital stay, length of ICU stay, the ratio of mechanical ventila-tion, blood transfusions, antibiotics and blood glucose treatment. Results The hospital mortality among patients without a history of diabetes mellitus was 4. 7%, 12. 1%, 20. 5%, 30. 3% in group 1, 2, 3, 4,respectively. The ratio of mechanical ventilation, blood transfusions, and antibiotics increased progressively as glucose values increased. Conclusions Hyperglycemia was a common disease, and it may be an impor-tant marker of poor clinical outcome and mortality in patients without history of diabetes.
3.Effects of JAK-STAT signaling pathway, IL-1βand IL-6 on injury of PC12 cells with X-ray irradiation

AIM:To investigate the role of JAK-STAT pathway , IL-1βand IL-6 in the PC12 cells with X-ray irradiation.METHODS:The PC12 cells were irradiated with X-ray at doses of 2, 4 and 8 Gy.After 24 h, the levels of IL-1βand IL-6 were detected by ELISA .The protein levels of p-JAK1, p-JAK2, p-STAT1, p-STAT3 and p-STAT5 were measured by Western blot .RESULTS:Compared with control group , the levels of IL-1βand IL-6 increased .The protein levels of p-JAK1, p-JAK2, p-STAT1, p-STAT3 and p-STAT5 increased with the doses of X-ray exposed.CONCLUSION:JAK-STAT signaling pathway , IL-1βand IL-6 play a role in the injury of PC12 cells with X-ray irradiation.

Objective To investigate the effect of matrix metalloproteinase(MMP-2) in the attack of exprimental autoimmune myositis(EAM) rats, and the effects of methylprednisolone on expressions of gene mRNA and protein of MMP-2. Methods The expression levels of MMP-2 in the peripheral blood, spleen lymphocytes and muscles were detected by using RT-PCR and immunohistochemistry techniques and was compared among EAM group,EAM with methylprednisolone treatment group (EAMM) and control group. Results ⑴ The degree of episode in EAMM group was lower than that of EAM group , and the infiltration of the inflammatory cells and the necrosis of the muscles were also mild in EAMM group. ⑵There was significant upregulation of the expression of the MMP-2 gene mRNA in the lymphocytes of peripheral bood and spleen of EAM rats as compared with that of the control,Elevation of the expression of MMP-2 protein in rats muscles tissues of EAMM group was observed obviously. However, the expression of the mRNA and the protein of MMP-2 was suppressed significantly in EAM group compared with control group. Conclusions The upregulation of the expression of MMP-2 may be correlated with the onset of EAM. Methylprednisolone may relieve the degree of the pathology of EAM by the downregulation of MMP-2 .
6.Variation of asthma patient's quality oflife after anti-inflammation treatment

Aim To evaluate the influence of asthma and the effect of anti-inflammation therapy on the quality of life (QOL) in allergic asthma patients. Methods The QOL of 45 patients with asthma were investigated by collecting questionnaire before and after three months anti-inflammation treatment , at the same time the serum eosinophil cationic protein(ECP) level was detected in 20 patients. The QOL questionnaire included 4 domains, they were respiratory symptoms, physical activity limitation, emotional function and environmental stimuli. Results The factors which influenced the QOL in asthma patients ranging from high to low were environmental stimuli, respiratory symptom and emotion function and physical activity limitation. 7. Direction of new drug R&D in Chinese materia medica over therapeutic field of new drugs approved by CFDA during 2010-2014

To provide the reference for selecting the research and development trend and direction of new drug in Chinese materia medica (CMM). We sorted out the new drugs of CMM approved by the China Food and Drug Administration (CFDA) during 2010-2014 through database retrieval, analyzed their indications, declared period and approval rates, then discuss and suggest the predominance of CMM for research and development. In recent five years, among the new drugs of CMM approved by CFDA, their main indications are those for cardiovascular system, respiratory system, and urogenital system. The declared period from short to long is gynaecology system, respiratory system, and urogenital system. From the point of approval rates, urogential system and pediatric system are higher than others. Comprehensive the above three factors, we get that cardiovascular system is a classic indication for CMM; Urogenital system, pediatrics system and gynecologic system will be the potential fields for CMM research and development.

Alternative splicing is a process refering a pre-mRNA transforms to different mature mRNAs by different splicing sites combination, and then the mRNAs translate to various proteins.This process is regulated by a variety of cis acting elements and trans acting factors.Recent studies have shown that aberrant alternative splicing is prevalent in leukemia patients, and is closely associated with leukemic occurrence, development and chemotherapy resistance.This shows us that aberrant altemative splicing may be helpful to leukemia diagnosis and treatment.
