Objective To explore the effect of transforming growth factor ?_1 (TGF-?_1) on expression of paxillin(Pax) in human kidney proximal tubular epithelial cell line (HKC cells) in different phases. Methods HKC cells cultured in vitro were divided into four groups at random. Control group(C group): HKC cells were cultured with serum free medium (FSM), and TGF-?_1-treated groups (T1, T2, T3 group): HKC cells were cultured with FSM containing 10ng/ml TGF-?_1. In the latter groups, duration of treatment varied as follows: T1 group for 24h, T2 group for 48h, T3 group for 72h. The proliferation of HKC cells induced by TGF-?_1 was assessed by MTT and the expression of Pax was determined. The gene expression of Pax was determined by reverse transcripton polymerase chain reaction (RT-PCR) and the protein expression of Pax was assessed by immunohistochemical staining and Western blot. Results TGF-?_1 could induce the proliferation of HKC cells in a time-dependent manner. In C group there was expression of Pax mRNA and protein in HKC cells. The expression of Pax mRNA and protein in HKC cells was positive in all the T groups, especially in T2 group and T3 group they were significantly increased compared with T1 group (P

Objective:To search for the efficient immune routes for rotavirus DNA vaccine by evaluating efficacy in inducing immune responses to rotavirus.Methods:In this study BALB/c mice were immunized by intramuscular and intranasal routes with rotavirus DNA vaccine,pcDNA 1/VP7,then rotavirus specific systemic immunity was measured.Results:Serum rotavirus specific IgG but not IgA titers of mice immunized by both routes were significantly higher than that of the unimmunized controls.Conclusion:Both intramuscular and intranasal routes are candidates for rotavirus DNA vaccine.
5.Surgical correction of congenital vertical talus in infants

Objective To investigate the surgical treatment of congenital vertical talus (CVT). Methods Ten feet in six patients (4 girls and 2 boys) with CVT were reviewed. Four of the feet were associated with neuromuscular disorders(Ⅱ), five with malformation syndromes. one foot was idiopathic. The age at surgery ranged from 13 to 39 months with an average of 16.5 months. All were operated with a single-stage soft tissue procedures by using double of posterior and dorsal incisions. Elongation of tendo Achillis and capsulotomy of the ankle and subtalar joints were performed through a straight posterior incision. The dorsal approach was made over the anterior inferior ankle crease from the tip of fibula to talonavicular joint laterally. The extensor digitorum longus and peroneal tendons were lengthened, whereas the peroneus tertius was always tenotomized. The calcaneocuboid capsule, subtalar capsule, and talonavicular capsule also were released. The tibilis anterior tendon was placed through a drill hole in the neck of the talus. One K-wire was passed through the navicular and then retrograde across the talonavicular joint to maintain the reduced position. The other K-wire drove from the heel pad across the subtalar joint. A cast was applied with the ankle in slight plantarflexion for 3 months. Then special shoe was applied for 12 months. Results Six cases (10 feet) were available for follow-up from 6 to 33 months (mean, 23.1 months). The postoperative clinical outcome was evaluated by using a version of Adalaar score. All patients got good cosmetic appearance of the feet and the medial longitudinal arch of feet, had no prominent talar head, and had no the ankle and subtalar motion loss. The presence of mild hindfoot valgus on weight-bearing was noted in only two feet. Seven of ten feet were wearing special shoes. Radiographically, two parameters-talar and calcaneal axis-first metatarsal base angles (TAMBA and CAMBA) were measured from the lateral view of the feet. Preoperative and final follow-up radiographs were obtained. None had avascular necrosis of the talus. TAMBA changed from 66?(range, 50?-98?) to 1.6?(range, -11?-16?), CAMBA changed from 26.1?(range, 14?-60?) to -5.45?(range, -13?-3?). Conclusion Operative reduction has been advocated as the only effective treatment for CVT. It is the best choice to use a single-stage soft tissue surgery for infants with CVT. It is concluded that the results of this procedure are satisfactory.
6.Morphology and mechanism research of gastric cancer vasculogenic mimicry

Objective:Vasculogenic mimicry(VM) is a new microcirculation form of tumor cells which found resent years. This research observed whether gastric cancer SGC-7901 cells can form vasculogenic mimicry in 3-dimentional culture and had a primer examination on its mechanism. Methods:Put SGC-7901 cells in 3-dimentinal culture and observed if it could form vasculogenic mimicry. Immunohischemistry tested the MMP-2 and MT1-MMP level of 3-dimentional cultured SGC-7901 cells. Observe the effect of PI3K way inhibitor on MMP-2、MT1-MMP level and VM formation of gastric cancer cells. Results:Gastric cancer SGC-7901 cells can form VM in 3-demintional culture. MMP-2 and MT1-MMP highly expressed in gastric cancer cells that took part in the tube structure. PI3K way inhibitor decreased MT1-MMPmRNA and MMP-2 protein level and inhibited VM formation of SGC-7901cells. Conclusion:Gastric cancer SGC-7901 cells had the ability to form vasculogenic mimicry in vitro 3-dimentional culture.One of its mechanism was probably through decreasing MT1-MMP and MMP-2 level.
7.Clinical Effect of Allevyn Thin and Nursing on Patients with Phlebitis Caused by PICC

Objective To investigate the clinical effect of Allevyn Thin on phlebitis caused by PICC. Methods Total 60 patients with phlebitis caused by PICC were randomly divided into 2 groups:experiment group and control group. The patients in the experiment group were treated with Allevyn Thin, while the patients in the control group were treated with hydropathic compress of 50% magnesium sulfate solution. The curative effect and healing time of the two methods were observed and compared. Results The average healing time of the experiment group was significantly shorter than that of the control group, and the curative effect of the experiment group was significantly better than that of the control group (P<0.05). Conclusion Allevyn Thin can shorten the healing time and improve the curative rate in treating phlebitis caused by PICC, suggesting that Allevyn Thin has a good potential of extensive clinical application.
8.The effects of resveratrol on the express of inflammatory factors of human umbilical vein endothelial cells induced by Aβ1 -42

Objective To explore the effects of resveratrol on the express of inflammatory factors (IL‐1 ,IL‐6 ,TNF‐α)of hu‐man umbilical vein endothelial cells(HUVEC) induced by Aβ1 -42 .Methods HUVEC were stimulated with Aβ1 -42 5 × 101 μmol/L , last 24 h and administrated with resveratrol 160 ,80 ,40 ,20 μg/L ,HUVEC viability were detected by CCK‐8 ;the concentration of IL‐1 ,IL‐6 and TNF‐α were detected by ELISA .Results Compared with model group ,160 ,80 ,40 ,20 μg/L resveratrol could in‐crease HUVEC survival rate ,reduced HUVEC damage by Aβ1 -42 ,inhibited the concentration of IL‐1 ,IL‐6 and TNF‐α(P<0 .05) . Conclusion Resveratrol over 20 μg/L could reduce the release of IL‐1 ,IL‐6 and TNF‐αof HUVEC inducing by Aβ1 -42 .

Objective To explore the effect of applying PDCA Circulation Theory in practical teaching of Basic Nursing at Higher Vocational College.Methods By using cluster random sampling method 2 classes of each 50 people from the nursing class of Grade 2013 were selected as the control group and the experimental group respectively.The traditional method of practice teaching was used in control group,that is,from the presentation of teachers,students practice,the guidance of teachers,periodic assessment to the final examination,while the PDCA Circulation Theory was used in experimental group.With periodic skill examination result,the final skill examination result and students' learning experience as evaluation index,two groups were compared with t test,using SPSS 17.0 for data analysis,expressed as percentage and-x ± s.Results The results of both single skill examination and the final skill examination as well as study experience of experimental group were higher than those of control group,and the differences are statistically significant.Conclusion Using PDCA Circulation Theory in practical teaching of Basic Nursing at Higher Vocational College can stimulate the students' consciousness and initiative,cultivate students' critical thinking,promote the student to study in a planned way with purpose and system and improve the learning effect.
