Shanghai Journal of Acupuncture and Moxibustion 2017;36(2):229-235
Objective To investigate the analgesic effect of warm needling and explore its relationship with the function of mast cells in the acupoint area and the specificity of acupoints.Method Sixty SD rats were randomized into blank (C), model (M), warm needling Zusanli (Z), contralateral warm needling (O), manual acupuncture (A), warm needling Futu (F), warm needling Yanglingquan(Y), warm needling Weizhong (W), disodium cromoglycate + warm needling (DSCG+Z) and saline + warm needling (Saline+Z) groups. A rat with adjuvant arthritis was used as a model of inflammatory pain (AA model). Hindpaw withdrawal latency was measured in therats during warm needling at Zusanli (ST36), Yanglingquan (SP9), Futu (ST32) and Weizhong (BL40). Pre-treatment and post-treatment rates of mast cell degranulation in the acupoint area were compared and the effect of acupoint area injection of disodium cromoglycate on the degranulation was observed by acupoint tissue section staining.Result Obvious degranulation of mast cells in the acupoint area appeared after warm needling (P>0.05). The analgesic effect of warm needling was better than that of manualacupuncture (P>0.05). After treatment, pain threshold was significantly higher in the warm needling Zusanli group than in the warm needling Futu, warm needling Yanglingquan and warm needling Weizhong groups (P>0.05). Acupuncture-induced mast cell degranulation could be prevented by disodium cromoglycate (P>0.05).Conclusion Warm needling at point Zusanli can produce an analgesic effect in adjuvant arthritis rats. Its analgesic effect is better than that of manual acupuncture. The rate of mast cell degranulation in the acupoint area is higher in the warm needling Zusanli group than in the manual acupuncture group. The analgesic effect of warm needling is reduced after pretreatment with disodium cromoglycate, a mast cell stabilizer. In an AA model, the analgesic effect of warm needling at point Zusanli is better than those of warm needling at points Yanglingquan, Futu and Weizhong.

5. Protective effect of resveratrol on oxidative stress injury of cardiomyocytes induced by high fat and its correlation with AMPK/mTORC1/p70S6K signaling pathway
Chinese Pharmacological Bulletin 2019;35(7):995-1000
Aim To investigate the protective effect of resveratrol on oxidative stress injury of cardiomyocytes induced by high fat and its correlation with AMPK/mTC)RCl/p70S6K signaling pathway. Methods The model of cardiomyocyte lipotoxicity injury was established by palmitic acid (PA). Resveratrol pretreatment and MTT assay were used to detect cell proliferation. The expression of reactive oxygen species ( ROS) was detected by fluorescence and flow cytometry, and the levels of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) were detected by kit. The protein expression of AMPK/mTORCl/P70S6K signaling pathway and apoptosis related proteins were determined by Western blot. Results MIT results showed that the cell viability decreased significantly at 0. 4 mmol L-1 and 24 h-48 h stimulation time, re-spectively. Fluorescence and flow cytometry showed that the levels of ROS and MDA increased significantly and SOD activity decreased significantly when PA (0. 4 mmol • L"1) stimulated cells for 24 h compared with control group. Western blot showed that the expression of p-AMPK and Bcl-2 decreased and the expression of p-mTORCl, p-p70S6K, Bax and cleaved caspase-3 increased when PA (0.4 mmol • L"1) stimulated cells for 24 h compared with control group. Resveratrol pre-treatment could significantly reverse the above chan-ges. Conclusions Resveratrol can significantly inhibit the apoptosis of cardiomyocytes induced by high fat. Its mechanism may be related to the regulation of AMPK/mTORCl/p70S6K signaling pathway.

Chinese Journal of Tissue Engineering Research 2009;13(16):3185-3188
BACKGROUND: Both cyclodextrin high polymer and polyvinyl alcohol (PVA) have been widely applied as biomedical materials owing to their characteristics including no toxicity and good biocompatibility and mechanical property and have become important functional materials of biological drug manufacture and medication research.OBJECTIVE: To observe the drug controlled release property of β-cyclodextrin (β-CD) loaded onto PVA by the covalent bond and to investigate the possible mechanisms of action. DESIGN, TIME AND SETTING: An observational experiment was performed at the Laboratories of Applied Chemistry, College of Science, Northwestern Polytechnical University, and College of Chemistry & Chemical Engineering, Xiangyang Normal University in 2008. MATERIALS: PVA (Tianda Experimental and Chemical Plant, Tianjin, China), β-CD (Sanpu Chemical Engineering Co.,Ltd., Shanghai, China) and camptothecin (Modern Times Chemical Institute, Xi'an, China) were used in this study. METHODS: Mono-6-O-tosyl-β-CD and mono-6-formyl-β-CD were synthesized separately. Silylatedβ-CD was loaded onto polymer chain of PVA by acetalization to produce PVA containingβ-CD (PVA-β-CD) linear high polymer. The clathration between PVA-β-CD and camptothecin was observed. Drug-release content of PVA-β-CD membranes under different pH values was determined through the use of ultraviolet-visible spectrophotometer. MAIN OUTCOME MEASURES: Synthesis condition and drug release rate of PVA-β-CD. RESULTS: The best condition for synthesis of PVA-β-CD linear high polymer comprised reaction time 2 hours, temperature 70 ℃, and the mass ratio of mono-6-formyl-β-cyclodextrin to PVA not higher than 4:1. Experimental results regarding drug release revealed that PVA-β-CD promoted water-insoluble drug release owing to its clathration and solubilization. Withβ-CD content increasing, camptothecin release amount and release velocity had no significant change in pH 11 medium but increased greatly in pH 2 medium. CONCLUSION: As for compact PVA membrane, addition ofβ-CD possibly leads to pore formation, thus promoting water molecule infiltration and drug diffusion, beneficial to drug release, and simultaneously, the solubilization of β-CD plays an important role in promoting the release of insoluble drug.

International Journal of Biomedical Engineering 2010;33(1):31-35
Objective To investigate the autonomic modulation and influence during myocardial ischemia based on analysis on heart rate dynamics with heart rate variability(HRV). Methods From 13 free download ECG records in long-term ST-T(LTST)database, 193 ischemia episodes were extracted for analysis, and 5 min intervals before and after each selected isehemia episode were selected as control ones. Using symbolic dynamic analysis method, the indexes during different stages were calculated. The repeated measurement analysis of variance was introduced for statistic test. And the distribution of autonomic reflex mode to isehemia duration was analyzed by linear fitting. Results Symbolic analysis detected an increase in the percentage of non-varlable patterns (0 V%) and a decrease in the percentage of very variable patterns(2 UV%), indicating a more sympathetic predominance compared with the baseline. But the trend of the shift in autonomic balance towards sympathetic predominance reduced with the extension of ischemic duration, suggesting a possible protection for heart. Conclusion Symbolic dynamic analysis is capable of identifying changes in autonomic modulation and seems appropriate for elucidating the neural pathophysiological mechanisms occurring in myocardial ischemic episodes.
