2.Effects of Intermediate Frequency Alternating Magnetic Field on Blood Analysis and Histopathological Analysis for Important Organs in Mice

0.05).Histopathological examination showed no pathological change in the brain,liver,kidneys,and spleen of the mice,but reduced spermatogenic cells spermatozoa.The sperm motility was(40.58?7.75)% in the control,(7.40?1.06)% in the experiment group 1(q=16.909,P0.05).No significant difference in the sperm densities was found among the groups.Conclusions Intermediated alternating magnetic field has adverse effects on the reproduction of mice.Testosterone is one of the target organs of electromagnetic radiation.It is sensitive to IF magnetic field(40 kHz,28 mT).Blood,brain,liver,kidney and spleen of mice are not effected by IF alternating magnetic field.
3.Effects of Iodine Deficiency and Hypothyroidism during Pregnancy on the Protein Expressions of cAMP Response Element-Binding Protein in the Entorhinal Cortex of Pups

Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of CREB in the entorhinal cortex of pups.Methods Twenty-eight female Wistar clean rats after pregnancy were randomly divided into control group,hypothyroid 1,2 group and iodine deficient group(7 in each group).From GD6 till PN28,iodine-deficient group was administered with iodine-deficient diet [iodine content:(14.11 ?1.96) ng/g] and tap water;hypothyroid 1 and 2 group were administered with 5 mg/L and 15 mg/L PTU in the drinking water and fed with normal diet [iodine content:(470.50?46.52) ng/g].Control group received tap water and normal diet during the experiment.Five pups from each group were sacrificed and intracardiac perfused at PN7,PN14,PN21,PN28 and PN42.Brains were removed,fixed and sectioned coronally.All sections were observed and were analyzed for the protein expression of CREB by immunohistochemistry in the entorhinal cortex.Results At PN7,there was no significant difference in the expression of Ng in all four groups.At PN14 and PN21,the expression of CREB in the entorhinal cortex in hypothyroid 1,2 group and iodine deficient group were significantly lower than those of controls(P
5.Analysis of crossmatch in irregular antibody-positive patients

Objective To investigate the clinical significance of irregular antibody screening positive patients in the blood transfusion.Methods From September 2003 to September 2012,5 260 samples of blood test were given pre-transfusion irregular antibody detection,and the patients with positive antibody screening were statistically analyzed.Results Of 5 260 samples,38 cases (6.8%) had irregular antibodies,the difference between males and females was statistically significant(x2 =2.646,P < 0.05).The Rh blood group had the highest proportion in irregular antibodies;pregnancy history and history of blood transfusion group,pregnancy history group,history of blood transfusion group were significantly higher than those without history of pregnancy and transfusion history groups,the difference was statistically significant (x2 =28.827,P < 0.05).Conclusion Screening of irregular antibodies plays an important role in the safety of clinical blood transfusion.
7.Hope for the patients with retinitis pigmentosa

Retinitis pigmentosa (RP) is a hereditary disease characterized by the progressive loss of photoreceptor cells. Disease pathology primarily affects rod photoreceptor cells first but light-insensitive cone photoreceptor bodies can survivor longer. Delivered by adeno-associated viral vector or lentiviral vector, expression of microbial-type rhodopsin, channelrhodopsin-2 or archaebacterial halorhodopsin in the survived retinal cells can substitute for the native phototransduction cascade in mouse models of retinitis pigmentosa. It can also restore light sensitivity and activate all retinal cone pathways and drive sophisticated retinal circuit functions. Thus, expression of channelrhodopsins or halorhodopsin might be a potential strategy for the restoration vision of patients with RP.
8.The Role of TGF?1 and IL-13 in Cellular Signal Transduction of Hepatic Fibrosis of Schistosomiasis

Liver fibrosis is characterized by an abnormal hepatic accumulation of extracellular matrix (ECM) that results from both increased deposition and reduced degradation of collagen fibres. Some studies show that transforming growth factor ?1(TGF-?1), alternatively activated macrophage (aaM) and interleukin 13(IL-13) play a key role in the evolution of fibrosis, of which TGF-?1 and IL-13 become research hotspots. TGF-?1 mainly activates hepatic stellate cells (HSC) through TGF-?1/Smad signal pathway, while IL-13 seems to play a rather crucial role through JAK-STAT6 signal pathway. aaM is an important source of TGF-?1 and activated with Il-13. This paper reviews the role of those signaling molecules in cellular signal transduction of hepatic fibrosis of schistosomiasis japonica, and provides some targets for future drug development.
9.Simplification of Protein Sequence and Alignment?-free Sequence Analysis

Alignment-free comparison is a recently developed method for sequence alignment, which has high computational efficiency and suitable to the low identical sequences. Alignment-free comparison was successfully applied in the DNA analysis. However, the accuracy of analysis is not high when it was applied in protein analysis because the complexity of protein is larger than DNA by consisting of 20 types of residues. Thus, residues are clustered into a few groups based on their similarity of physicochemical features. Using such simplified alphabets, the complexity of protein sequences is reduced and at the same time the key information encoded in the sequences remains. Therefore, the accuracy of alignment-free comparison is improved.
10.Taurine improves neuronal stem cell proliferation in fetal rats with growth restriction

Objective To detect the proliferation of neural stem cells (NSC) and analyze the effects of taurine on NSC proliferation in rats with fetal growth restriction (FGR).Methods For in vivo experiment,pregnant rats were randomly divided into three groups:control group (n=5),FGR group (n=7) and taurine group (n=6) [treated with taurine,300 mg/(kg · d)].Rats in the latter two groups were fed with 40% of food intake in control group throughout pregnancy to establish the FGR model.Fatty acid binding protein-7 (FABP-7) expression in brains was detected by immunohistochemistry.For in vitro experiment,pregnant rats were divided into two groups including control group and FGR group.FGR modeling was established in the same way as above.On the 14.5 day of pregnancy,NSC were isolated and cultured for five days to form neurospheres with different concentrations of taurine (control,0,5,10,15,20,30,40 mmol/L).Immunofluorescence staining was used to detect the expression of NSC-specific marker FABP-7.CCK8 test was used to detect the cell proliferating index.Neurospheres were collected at the end of culture and disaggregated to get the total number of viable cells.One-way ANOVA,Kruskal-Wallis Test,SNK-test and Tamhane's test were used for statistical analysis.Results Compared with the control group (18 012±3 016),the number of FABP-7-positive cells in FGR model group (10 173±1 728) was decreased (P<0.05),but increased in taurine group (19 518±3 201) (F=200.8,P<0.05).Free-floating neurospheres could be formed in both control and FGR groups and the cells within neurospheres were mostly FABP-7-positive and were then considered as NSC.The proliferation indexes in control,FGR,FGR+5 mmol/L taurine,FGR+10 mmol/L taurine,FGR+15 mmol/L taurine,FGR+20 mmol/L taurine,FGR+30 mmol/L taurine,FGR+40 mmol/L taurine groups were 1,0.80±0.07,0.92±0.11,1.33±0.24,1.20±0.15,1.11 ±0.14,0.80±0.12 and 0.82±0.08,respectively.The proliferation indexes in FGR groups treated with 10,15 or 20 mmol/L of taurine were significantly higher than that of FGR group without taurine interference (P<0.05).The total cell numbers in the eight groups were (24.5 ± 0.6)× 105,(20.23 ± 1.09)× 105,(22.80 ±0.86) × 105,(27.53± 1.22)× 10s,(25.30±1.05)× 105,(22.70±0.72)× 105,(21.80± 1.80)× 105 and (18.06 ± 0.90)× 105,respectively.Compared with the FGR group without taurine interference,those treated with 5,10,15 or 20 mmol/L of taurine showed higher total cell numbers (all P<0.05 or <0.01),while that treated with 40 mmol/L oftaurine was lower.Conclusions The number and proliferation of NSC in FGR fetal rat brains is significantly decreased,however,these could be reversed by antenatal supplementation of taurine.The best effect is shown when l0 mmol/L of taurine is given.
