1.Association of Apo-E gene polymorphism with biochemical and lipid metabolism parameters in patients with diabetic nephropathy of Hui and Han populations in Gansu Province

Objective To study the association of apolipoprotein ( Apo) E gene polymorphism with difference in biochemical metabolism of diabetic nephropathy of Hui and Han populations. Methods ApoE genotype was determined by PCR-RFLP in diabetic patients with or without diabetic nephropathy (DN) and normal peoples in Hui and Han peoples, the related biochemical parameters were simultaneously detected. Results (1) Huis had 3 genotypes, i. e. E2/E3, E3/E3 and E3/E4, and their frequencies were 0. 043, 0. 894 and 0. 064, respectively. Hans had 5 genetypes, i. e. E2/E2, E2/E3 , E2/E4, E3/E3 and E3/E4, and their frequencies were 0.075,0.060, 0.164, 0.642 and 0.060, respectively. The frequencies of 3 alleles, E2, E3 and E4, in Huis and Hans were 0.021, 0.947, 0.032 and 0.187, 0. 702, 0.112, respectively. 3.Effect of hydroxycamptothecin with thermotherapy on angiogenesis in vivo and in vitro

Purpose:To study the effects of hydroxycamptothecin with thermotherapy on anti-angiogenesis in vitro and in vivo. Methods:We chose human microvascular endothelial cell (HMVEC) culture and chick embryo choriallantoic membrane (CAM) model and used MTT and number-calculating methods to observe hydroxycamptothecine on HMVEC’s proliferation , sprouts and CAM blood vessels’ formation.Results:The survival rate of endothelial cells was in the range of (68.2%)-44.7% within the dose of 20-40 ng/ml and 5-80 ng/ml and was negatively correlated with the concentration (correlation coefficient was -0.906,-0.469,P=0.00003,0.0051). Hydroxycamptothecine could significantly suppress the endothelial cells’ proliferation and the sprouts. Hydroxycamptothecine could significantly suppress CAM vessels. The survival rate of HepG II cells is in the range 100%-90% within the dose of 5-80 ng/ml. There was no cytotoxicity.There was a synergestic anti-angiogenetic effect when hydroxycamptothecin (20 ng/ml) was combined with thermotherapy in vitro while there was additive effect when hydroxycamptothecin (40 ng/ml) was combined with thermotherapy in vitro.Conclusions:This experiment shows that small doses of hydroxycamptothecine (20-40 ng/ml) with thermotherapy has anti-angiogenetic synergestic or additive effect on proliferation and migration both in vivo and in vitro.
4.Effect and Mechanism of Music Exposure on Fearful Behavior of Prenatal Stressed Rats

Objective To investigate the effect of postnatal music exposure on fearful behavior of prenatal stressed rats in defensive withdrawal test and the expression of gastrin-releasing peptide receptor (GRPR) in the amygdala. Methods Wistar pregnant rats were divided in stress group (n=8) and control group (n=8). The stress group were immobilized and bright lighted 45 min, 3 times a day, when gestation 11-20 days, while the control group without any stress. The new born rats from the stress group were exposed in music (SM group) or white noise (SW group) for 45 min twice a day since 1 day after born, and the rats from the control (SC) group left undisturbed. They were tested with defensive withdrawal test 60 days after birth, and the GRPR levels in the amygdala were measured with immunofluorescence and real-time PCR. Results The latency was shorter and the frequence out of the chamber was the most in the SM group of the defensive withdrawal test (P<0.001), while GRPR-positive cells and GRPR mRNA increased in amygdala (P<0.001). Conclusion Postnatal music exposure can counteract the abnormal behavior response of prenatal stressed offspring and accompany with increased expression of GRPR in the amygdala.
5.Clinical predictors in early pregnancy for adverse pregnancy outcomes in women complicated with chronic ;nephropathy

Objective To investigate clinical predictors in early pregnancy for adverse pregnancy outcomes in women complicated with chronic nephropathy. Methods One hundred and eighty-four pregnancies complicated with chronic nephropathy who delivered between January, 2005 and January, 2014 in Peking University First Hospital were retrospectively analyzed. Two hundred pregnant women without chronic nephropathy were selected as the control group. Pregnancy outcomes were compared between the two groups. Relationships between clinical predictors in early pregnancy [age, history of kidney disease, drugs used before pregnancy (in three months), results of renal biopsy, blood pressure, serum creatinine, urea nitrogen, 24 h urinary protein, estimated glomerular filtration rate, stage of chronic nephropathy] and adverse pregnancy outcomes [maternal mortality;pregnancy complicated by severe preeclampsia;renal function decline, early preterm birth, very low birth weight infants, fetal loss after 20 weeks of pregnancy] were assessed by logistic regression analysis. T-test, Chi-square test and multivariate regressions were used for statistical analysis. Results There were 174 and 197 livebirths in the study and the control groups, respectively. The most popular type of chronic nephropathy in pregnant women was IgA nephropathy (38.6%, 71/184). Compared with the control group, the risk of premature labor [9.8%(17/174) vs 3.0%(6/197), χ2=7.184], low birth weight infants[18.4%(32/174) vs 7.1%(14/197),χ2=9.813], very low birth weight infants [5.7%(10/174) vs 1.5%(3/197),χ2=4.536], fetal loss after 20 weeks of pregnancy [5.7%(10/184) vs 1.5%(3/200), χ2=4.536] and severe preeclampsia [17.9%(33/184) vs 1.5%(3/200), χ2=33.544] increased significantly in the study group (all P < 0.05). Twenty-four-hour urinary protein content (OR=1.84, 95%CI: 1.36-2.50, P=0.001) and mean arterial pressure (OR=1.04, 95%CI: 1.00-1.07, P=0.027) in early pregnancy were risk factors for adverse pregnancy outcomes. The risk of renal function decline increased significantly in patients with higher stages of chronic nephropathy in early pregnancy (OR=6.50, 95%CI: 3.34-8.21, P<0.01). Mother complicated by preeclampsia during pregnancy was an independent risk factor (OR=11.10, 95%CI: 4.48-27.20, P<0.01). Compared with women whose 24 h urinary protein content less than 1g in early pregnancy (122 livebirths within 126 cases), the risk of premature labor [17.3%(9/52) vs 6.6%(8/122), χ2=4.780], increased significantly in women whose 24 h urinary protein content were more than or equal to 1 g in early pregnancy (52 livebirths within 58 cases) (P<0.05). Conclusions Elevated urinary protein level and mean arterial pressure in early pregnancy were risk factors of adverse pregnancy outcomes. The risk of renal function decline increased in patients with higher stages of chronic nephropathy in early pregnancy. Mother complicated by preeclampsia during pregnancy was an independent risk factor for poor prognosis of the fetus.

8.Expression of MMP-2 in aqueous humor from patients with high myopia

Objective To investigate the correlation between high myopia and MMP-2 in aqueous humor (AH), by comparing the protein level of MMP-2 in AH from patients with cataract combined with high myopia and with emmetropia. Design Experimental study. Participants 30 AH samples of patients with age-related cataract. Methods AH samples were collected from the patients with age-related cataract during phacoemulsification, including 15 AH samples from patients combined with high myopia (experimental group) and 15 AH samples from patients with emmetropia (control group). The expression of pro-MMP-2 and MMP-2 in AH were ana-lyzed by Western blot technique. Main Outcome Measures The gray value of MMP-2 protein band detected by Western blot. Results The level of pro MMP-2 was statistically higher than that of MMP-2 in experimental group (430.4±57.3 versus 294.5±35.2, t=10.400, P= 0.000) and control group (402.8±57.7 versus 280.3±49.7, t=8.400, P=0.000). There was no statistically difference in the level of pro-MMP-2(t=1.320, P=0.200)and MMP-2 (t=0.900,P=0.375) between the two groups. Conclusions No abnormal expression of MMP-2 was detected in AH from patients with high myopia, according to this study based on limited samples. Pro-MMP-2 was the main form of MMP-2 in AH of patients with cataract combined with high myopia or emmetropia, which possessed potential ability of transferring into form of active MMP-2. 9.Corneal aberrations and visual performance after flap creation of LASIK with a femtosecond laser and a mechanical microkeratome

Background Laser in situ keratomileusis (LASIK) for myopia and/or myopic astigmatism still is one of primary surgeries.Femtosecond laser (FS)-LASIK offers several advantages over microkeratomes for flap creation including better safety,reproducibility,and predictability.However,the visual performance after FS-LASIK is of high clinical concern.Objective This study was to compare the visual performance between Intralase FS60 femtosecond laser flap and mechanical microkeratome Moria M2 flap in LASIK.Methods A prospective nonrandomized controlled study was performed.Two hundred and four myopic eyes of 102 patients were enrolled in this study and divided into two groups according to patients’ opinion,with matched demography between the two groups.LASIK with FS60 femtosecond laser flap was performed on 100 eyes of 50 myopia and/or myopic astigmatism in the FS group,and Moria M2 flap LASIK was performed on 104 eyes of 52 patients in the microkeratomes group.Zernike coefficients and the root-mean-square (RMS) of higher order aberrations were measured by Wavescan wavefront aberrometer before and 3,6 and 12 months after surgery,and the contrast sensitivity (CS) in different spatial frequencies under the scotopia and scotopia + glare background was detected using Optec 6500 visual function instrument before and 3,6,12 months after surgery.This study protocol was approved by Ethic Committec of Beijing Tongren Hospital,and written informed consent was obtained from each patient prior to surgery.Results The uncorrected visual acuity (UCVA) levels after surgery reached or was superior to the preoperative best corrected visual acuity (BCVA) in 95.1%,94.2% and 93.9% patients in the FS group and in 94.2%,93.8% and 93.2% patients in the microkeratomes group 3,6,12 months after surgery,respectively.The mean spherical equivalent (SE) was (-6.37 ±2.06)D,(-0.26±0.45)D and (-0.45± 0.51)) in the FS group,(-6.25± 2.43) D,(-0.44± 0.64) D and (-0.35±0.59)D in the microkeratomes group before surgery and 3,12 months after surgery,respectively.There were no significant differences in the SE in various time points between the two groups (u =1.194,1.429;both at P> 0.05).The root means square high order (RMSh) and RMS3-6 in the FS group were significantly lower than those in the microkeratomes group 3,6 and 12 months after surgery (all at P<0.05).Under the background of scotopia or scotopia+glare,the LogCS values under different spatial frequencies were significantly higher in the FS group than those in the microkeratomes group 3 and 6 months after surgery (all at P<0.05).In 12 months after surgery,no significant difference was found in the LogCS under the 1.5 c/d in the background of scotopia between FS group and microkeratomes group (P>0.05),and the LogCS values under the 12.0 c/d and 18.0 c/d in the background of scotopia or scotopia+glare in the FS group were better than those in the microkeratomes group (all at P<0.05).Conclusions Femtosecond laser assistant LASIK has better visual performance and lower higher-order aberrations than microkeratomes assistant LASIK.
10.Inflammatory Factors and von Willebrand Factor in Hypertensive Patients Complicated with Metabolic Syndrome

Objective To test the hypothese that metabolic syndrome is closely associated with inflammation, we compare the difference of inflammatory factors and von Willebrand factor between essential hypertensives and hypertensive patients with metabolic syndrome. Methods According to the new definition of metabolic syndrome by IDF 2005, one hundred thirty eight consecutive hospitalized essential hypertensives were catelogorized into EH+MS group, n=99 and EH alone group, n=39.
